  • A computer code that is based on the Pseudo-spectral finite difference algorithm using staggered grid is developed for the wave propagation modeling in the time domain. The advantage of a finite difference approximation is that any geometrically complicated media can be modeled. Staggered grids are advantageous as it provides much more accuracy than using a regular grid. Pseudo-spectral methods are those that evaluate spatial derivatives by multiplying a wavenumber by the Fourier transform of a pressure wave-field and performing the inverse Fourier transform. This method is very stable and reduces memory and the number of computations. The synthetic results by this algorithm agree with the analytic solution in the infinite and half space. The time domain modeling was implemented in various models. such as half-space. Pekeris waveguide, and range dependent environment. The snapshots showing the total wave-field reveals the Propagation characteristic or the acoustic waves through the complex ocean environment.

Studies on the Cultural Characteristics of Cellulase Production by Roseofomes subflexibilis (Roseofomes subflexibilis로부터 Cellulase 생산을 위한 배양학적 성질)

  • For the purpose of utilizing cellulose resources by cellulolytic enzymes of Roseofomes subflexibilis, it's cultural conditions for the production of cellulolytic enzymes in synthetic media were investigated. The results obtained were summarized as follows : The optimum temperature and pH for the enzyme production were $30^{\circ}C$ and pH 4.0, respectively. Among the carbon sources, glucose was good for the production of cellulase. The optimum concentration of saccharose as 1.2%. As a organic nitrogen source, yeast extract was good for the mycelial growth. The optimum concentration of yeast extract as 1.5%. As a inorganic nitrogen source, $NH_{4}H_{2}PO_{4}$ was good for the mycelial growth. The optimum concentration of $NH_{4}H_{2}PO_{4}$ were 1.1%. The mineral salt of $Al_{2}(SO_{4})_{3}$ was effective and the optimum concentration was 0.1 M.

Effect of Flask Type on the Production of Value-added Hydroxy Fatty Acid by Pseudomonas aeruginosa PR3 (Pseudomonas aeruginosa PR3에 의한 기능성지방산 Hydroxy fatty acid 생산에 있어 배양용기 형태에 따른 영향)

  • Hydroxy fatty acids have gained important attentions because of their special properties such as higher viscosity and reactivity compared to non-hydroxy fatty acids. The new bacterial isolate Pseudomonas aeruginosa (PR3) had been well studied to produce mono-, di-, and tri-hydroxy fatty acids from different unsaturated fatty acids. Of those hydroxy fatty acids, 7,10-dihydroxy-8(E)-octadecenoic acid (DOD) was produced with high yield from oleic acid by P. aeruginosa PR3. Mostly, the substrates used for the production of hydroxy fatty acid by microorganisms were free fatty acids or synthetic lipids. However, it is highly plausible to use vegetable oils containing oleic acid such as olive oil for DOD production by P. aeruginosa PR3. As a first step to address the utilization of olive oil as a substrate for DOD production, we tried to determine the effect of vessel type on DOD production from olive oil by P. aeruginosa PR3. Of two different flask types of normal flask and baffled-flask, baffled-flask was highly effective for DOD production with all the media tested. Maximum increase of productivity by baffled-flask represented 221% with the medium supplemented with whey powder instead of lactose. Results from this study demonstrated that vessel type and medium composition could be both significant factors for DOD production from olive oil by P. aeruginosa PR3.

  • The application of constructed subsurface-flow wetlands for treatment of wastewater from abandoned mines is being increased. Crushed limestone, oak chips, and mushroom composites are often employed in a bulk form, as the substrates in the bed media. Efficiency of the subsurface-flow treatment system drops with time as the hydraulic conductivity of the wetland soil decreases significantly, presumably due to chemical reactions with the wastewater. The purpose of this study is to investigate the applicability of immobilized beads carrying sulfate reducing bacteria for acid mine drainage treatment system. The ingredients of immobilized beads are organic materials such as mushroom composite and oak chips, limestone powder for a pH buffer, mixed with a modified Coleville Synthetic Brine. It was found that immobilized beads are more efficient than the bulk form for pH recovery, sulfate and heavy metal removal.

Efficient Target-Site Assay of Chemicals for Melanin Biosynthesis Inhibition of Magnaporthe grisea

  • A rapid and efficient assay to determine melanin biosynthesis inhibition of Magnaporthe grisea, a causal agent of the rice blast, by chemicals was developed. Wells in 24-well plates were loaded with spore suspension of the fungus and three known melanin biosynthesis inhibitors of KC10017, tricyclazole, and carpropamid. Subsequent color changes of mycelia and culture media in the wells were observed 7 days after incubation. The wells treated with KC10017 (an inhibitor of polyketide synthesis step and/or pentaketide cyclization step) became colorless, whereas tricyclazole (an inhibitor of 1, 3, 8-trihydroxynaphthalene reductase) or carpropamid (an inhibitor of scytalone dehydratase)-treated wells exhibited red color. They did not show any inhibitory effect on fungal growth. The inhibition of reaction steps prior to 1, 3, 6, 8-tetrahydroxynaphthalene formation was easily determined by colorless medium and mycelia. However, it was impossible to distinguish between inhibition of reduction steps and inhibition of dehydration steps by colors of the cultures. It was accomplished through HPLC analysis of the melanin biosynthesis-involving pentaketide metabolites accumulated by the inhibitors. Through screening of a number of synthetic chemicals using the in vitro assay, we could find a novel chemical group of melanin biosynthesis inhibitor.

Dendropanax morbifera Extract Inhibits Intimal Hyperplasia in Balloon-Injured Rat Carotid Arteries by Modulating Phenotypic Changes in Vascular Smooth Muscle Cells

  • The plant Dendropanax morbifera Léveille is effective folk medicines for the treatment of several conditions, such as infectious diseases, skin diseases, and other illnesses. Although the inhibitory effects of D. morbifera on the proliferation and migration of vascular smooth muscle cells (VSMCs) have been shown in our previous study, its effects in vivo remain to be elucidated. In this study, we aimed to investigate the protective effects of the extracts from D. morbifera (EDM) on neointimal hyperplasia of rat carotid artery and explore the underlying mechanisms. We observed that the ratio of intima to media thickness (I/M) was significantly decreased in the EDM-treated groups by ~80% compared to that of the control. The expression of Ki-67 and proliferating cell nuclear antigen was decreased by ~70% in the EDM-treated groups compared to that of the control. In addition, matrix metalloproteinase (MMP)2 and MMP9 significantly reduced in the neointimal layer of the EDM-treated groups. Moreover, the decreased levels of contractile phenotypic markers of VSMCs, such as α-smooth muscle actin, myocardin, and smooth muscle-myosin heavy chain, were successfully restored by EDM treatment. Furthermore, the levels of synthetic phenotypic markers, cellular retinal binding protein 1 and connexin 43 were also restored to normal levels. These results suggest that EDM inhibits vascular neointimal hyperplasia induced by balloon injury in rats via phenotypic modulation of VSMCs. Therefore, EDM may be a potential drug candidate for the prevention of restenosis.

Evaluation of Edge Detector′s Smoothness using Fuzzy Ambiguity (퍼지 애매성을 이용한 에지검출기의 평활화 정도평가)

  • While the conventional edge detection can be considered as the problem of determining the existence of edges at certain locations, the fuzzy edge modeling can be considered as the problem of determining the membership values of edges. Thus, if the location of an edge is unclear, or if the intensity function is different from the ideal edge model, the degree of edgeness at the location is represented as a fuzzy membership value. Using the concept of fuzzy edgeness, an automatic smoothing parameter evaluation and selection method for a conventional edge detector is proposed. This evaluation method uses the fuzzy edge modeling, and can analyze the effect of smoothing parameter to determine an optimal parameter for a given image. By using the selected parameter we can detect least ambiguous edges of a detection method for an image. The effectiveness of the parameter evaluation method is analyzed and demonstrated using a set of synthetic and real images.

  • This study is to design and implement the synthetic courseware that covers repeating learning, individual teaching, and effective teaching-learning games by using multimedia authoring tool KAS for numerical data of computer science which is especially a little boring and difficult to understand through blackboard-based lesson for senior high school students. It also overcomes the drawbacks of the existing CAI focused on text and image by combining various kinds of media like image, animation and sound effects. Animation of numerical data in the fields of radix conversion makes it possible for learners to check the individual learning process and visual interest. So it comes to increase the effectiveness of learning. Whenever each chapter is finished. learners' understanding can be evaluated by formative test. Applicability test of the developed CAI in Eumsung Senior High School. is statistically evaluated as more effective than that of the blackboard-based lesson.

Inhibitory Effect of Cephalosporin C on Growth of Cephalosporium acremonium M-113 (Cephalosporium acremonium M-113의 세팔로스포린에 의한 생장억제 효과)

  • Cephalosporin C(CPC) inhibited the growth of Cephalosporium acremonium M-113, a potent CPC producer derived from C acremonium ATCC 20339. Similar inhibitory effects of CPC were also observed in growth of C. acremonium ATCC 20339 and ATCC 14553. Minimum inhibitory concentrations (MIC) of CPC on the growth of conidia and hyphae of C. acremonium M-113 were 200-500 and 3000-4000$\mu\textrm{g}$/$m\ell$ respectively in synthetic medium. MIC values were increased in complex media. The inhibitory effect of CPC was due to CPC-exerted inhibition of amino acids uptake by the cells. 3'-Group of CPC might be important in its inhibitory action. In audition, CPC itself could be utilized by the cells as a nitrogen source under nitrogen limited condition.

Effect of Culture Medium on Results of Maerobroth Dilution Antifungal Susceptibility Testing of Candida albicans (Candida albicans의 시험관 희석법에 의한 항균력 검사시 배지가 항균력에 미치는 영향)

  • A total of 42 strains of Candida albicans were examined for susceptibility to three antifungal agents, amphotericin B(AMB), 5-fluorocytosine(5-FC), and ketoconazole(KTZ), using defined medium, synthetic amino acid medium-fungal(SAAM-F), supplemented yeast nitrogen base(SYNB) and undefined medium Sabouraud's dextrose broth(SDB) and Kimmig broth media. A tube dilution method was used with minimum inhibitory concentrations(MICs) determined after incubation for 24 hour and 48 hours. All testes were performed in duplicate. In general, MICs were more reproducible after 48 hour of incubation. Forthermore, MICs determined after incubation for 48 hours were significantly higher than those determined after 24 hours. The actural MICs obtained with the different antifungal agents were clearly influenced by the test medium used. The rank order of AMB MICs according to the test medium was as follows: SAAM-F>SYNB>SDB>Kimmig broth. With 5-FC, the following pattern was observed: SYNB>SAAM-F>SDB>Kimmig borth. For ketoconazole, the MICs according to the test medium was SAAM-F>SDB>SYNB> Kimmig broth. In amphotericin B, the MICs mean value with the test medium was as follows: SDB, 0.24 mcg/ml; Kimmig broth, 0.29 mcg/ml; SYNB, 0.21 mcg/ml and SAAM-F, 0.15mcg/ml. The actural value of 5-FC was; SDB, 37.20 mcg/ml; Kimmig broth, 67.41mcg/ml; SYNB, 21.29 mcg/ml and SAAM-F, 24.61 mcg/ml and in ketoconazole, the MICs value was; SDB, 1.83 mcg/ml; Kimmig broth, 4.08 mcg/ml; SYNB, 1.95 mcg/ml and SAAM-F, 1.41 mcg/ml. The results of this investigation suggested that broth dilution susceptibility testing of yeast and yeast-like fungi are best performed with an incubation period of 48 hours. Furthermore, medium composition can significantly influence the results of such testing.
