Silybin Synergizes with Wnt3a in Activation of the Wnt/${\beta}$-catenin Signaling Pathway through Stabilization of Intracellular ${\beta}$-Catenin Protein (Silybin에 의한 Wnt/${\beta}$-catenin 신호전달체계의 활성화)

  • The Wnt/${\beta}$-catenin signaling pathway regulates diverse developmental processes and adult tissue homeostasis. Inappropriate regulation of this pathway has been associated with human diseases, such as cancers, osteoporosis, and Alzheimer's disease. Using a cell-based chemical screening with natural compounds, we discovered silybin, a plant flavonoid isolated from the Silybum marianum, which activated the Wnt/${\beta}$-catenin signaling pathway in a synergy with Wnt3a-conditioned medium (Wnt3a-CM). In the presence of Wnt3a-CM, silybin up-regulated ${\beta}$-catenin response transcription (CRT) in HEK293-FL reporter cells and 3T3-L1 preadipocytes through stabilization of intracellular ${\beta}$-catenin protein. Silybin and Wnt3a-CM synergistically reduced expression of important adipocyte marker genes including peroxisome-proliferator-activated $receptor{\gamma}$ ($PPAR{\gamma}$) and CAATT enhancer-binding protein ${\alpha}$ (C/$EBP{\alpha}$) in 3T3-L1 preadipocytes, accompanied by the activation of Wnt/${\beta}$-catenin signaling pathway. Taken together, our findings indicate that silybin is a small-molecule synergist of the Wnt/${\beta}$-catenin signaling pathway and can be used as a controllable reagent for investigating biological processes that involve the Wnt/${\beta}$-catenin signaling pathway.

Chemical Structure and Physiological Activity of Lignan Component in Sesame (참깨 리그난 성분의 화학구조와 생리활성)

  • Sesame seed are one of the oldest oil-bearing crops known to man. Not only are they nutritious, they are physicologically beneficial. Because of these properties, the promotion of sesame as a crop is set to increase from now on. The lignan component of sesame, which has an antioxidative function, varies considerably. In addition, the seeds remain viable for longer than other crops. At low temperature, it has been reported that the seed remain viable for up to thirty years. This is due to the lignan content of the seeds. In the last twenty years, the mysteries of the natural chemistry of sesame seeds have been unravelled one by one. Up to now, the lignan component has been found to have an $\alpha$-tocopherol synergist effect, an anti-aging effect, a cancer suppressing effect, a hypertension reduction effect, an effect on the promotion of liver function, an effect on the control of the ration of unsaturated fat, and an effect on the synthesis of unsaturated fatty acids. The lignan component of sesame does not simply have an anti-oxidant function, but has diverse recently discovered physiological uses which make the study of lignan highly rewarding.

Comparison of the EMG Activities of Scapular Upward Rotators and Other Scapular Muscles Among Three Lower Trapezius Strengthening Exercises (아래등세모근 근력강화 운동방법들 사이의 어깨뼈 위쪽 돌림근과 다른 어깨 근육들의 근활성도 비교)

  • The aim of this study was to compare the electromyographic (EMG) activity levels of the scapular upward rotators [upper trapezius (UT), lower trapezius (LT), and serratus anterior (SA)] and other scapular muscles [posterior deltoid (PD), levator scapulae (LS), and infraspinatus (IS)] during isometric lower trapezius exercises. Twenty males with no medical history of shoulder pain or upper extremity disorders were recruited for this study. EMG activity was recorded from the UT, LT, SA, PD, LS, and IS while subjects performed three different exercises: Prone arm lift (PAL), Backward rocking diagonal arm lift (BRDAL), Modified Prone Cobra (MPC). One-way analysis of variance (ANOVA) was used to determine any significant differences among the three exercises. A lower relative activation of UT, LT, and SA was seen with the MPC than with the other exercises (p<.05). The relatively lower activation of the UT identified, the MPC exercise as the preferred choice for preferential strengthening the LT (p<.05). However, a higher activation in the PD, LS, and IS occurred with the MPC than with the other exercises (p>.05). The recruitment pattern of synergist varied depending on the exercise posture. These findings suggest that exercise posture is an important factor in the selection of strengthening exercise for weak muscle.

Antioxidative Effects of Ethanol Extract Obtained from Rooibos Tea(Aspalathus linearis) and It's Application of Food (Rooibos Tea(Aspalathus linearis)에탄올 추출물의 항산화 효과 및 식품에 대한 응용)

  • This study was designed the antioxidative effect of the ethanol extract obtained Rooibos tea (Aspalathus linearis) on various kinds of oil, and examined the synergistic effect of Rooibos tea extract by addition of citric acid to the antioxidation activity and also investigate to antioxidation effect of the extract in food production and storage peroids. The antioxidative activity of 0.2mg of Rooibos tea extract was showed similar to same doses of $\alpha$-tocopherol, BHA and BHT in linolieic acid-ethanol system. The antioxidative effect of the mixture with 0.1% Rooibos tea extract on lard was more effective than that of the mixture with same doses of $\alpha$-tocopherol, BHA and BHT. The antioxidative effect of Rooibos tea extract was showed slightly effects on lard or soybean oil. Antioxidative effects of Rooiboe tea extract in addition of citric acid as synergist showed more effective in linoleic acid-ethanol system, but did not showed in the other oils. In the application of Rooibos tea extract to food prodctuction and storage period, the antioxidative effect was more effective in biscult, preparation added lard mixed with 0.1% Rooibos tea extract.

Effect of TIBA, PCIB and phloroglucinol on somatic embryo maturation and germination in Japanese larch (Larix leptolepis) (낙엽송의 체세포배 발생 및 발아에 미치는 TIBA, PCIB 및 phloroglucinol의 효과)

  • The effect of auxin transport inhibitor (TIBA and PCIB) or auxin synergist (phloroglucinol) on somatic embryo maturation and germination in Japanese larch (Larix leptolepis) was examined. The addition of 15.8 mg/L ABA+5.0 mg/L PCIB showed most promoted the maturation of cotyledon -staged somatic embryos (177.7/90 mg ESM). In contrast, with treatment of 5.0 mg/L PCIB or 5.0 mg/L TIBA, no somatic embryos were obtained. Considering from this result, PCIB or TIBA alone could not substitute for exogenously supplied ABA for maturation of somatic embryos. In the test of below concentration of 5.0 mg/L PCIB, the highest results were recorded in 15.8 mg/L ABA+2.0 mg/L PCIB (109.3/90 mg ESM) or 15.8 mg/L ABA+5.0 mg/L PCIB (103.7/90 mg ESM). However, 5.0 mg/L phloroglucinol (0/90 mg ESM) or no ABA addition (3/90 mg ESM) had little influence on somatic embryos maturation. In germination study, the highest frequency of plantlet regeneration obtained from the somatic embryos which had matured on 15.8 mg/L ABA+5.0 mg/L PCIB (67.9%). However, either 5.0 mg/L PCIB nor 5.0 mg/L TIBA resulted in obtained from plantlets.

Storage Stability of Refined Sardine Oil (정제정어리유의 저장안정성에 관한 연구)

  • For the utilization of polyunsaturated fatty acids in sardine (Sardinops melanosticta) oil, mainly as a source for dietary supplement of eicosapentaenoic acid(EPA) and docosahe-xaenoic acid(DHA), the effects of refining and storage, conditions, and addition of stabili-zing agents on the storage stability of EPA condensed and refined sardine oil were tested. Refined, sardine oil deacidified, decolored, and deodorized, was more stable to autoxidation than crude or partially purified oils such as deacidified or/and decolored. The refined sardine oil must be kept at $5^{\circ}C$ or lower temperture for longer than two month storage. The addition of $0.018\%\;\alpha-tocopherol$ or BHT could enhance the stability to autoxidation, and EDTA or citric acid was useful as synergist. Especially the addition of $0.02\%$ citric acid to the oil still hot after deodorizing process ($125^{\circ}C$) was of benefit to stabilize the refined oil.

Effects of Antibrowning Agents on the Browning Reaction of Enzymatic Garlic Hydrolyzate (마늘가수분해물의 갈변반응에 미치는 항갈색화제의 영향)

  • The antibrowning effcts of cysteine, citric acid and ascorbic acid on the browning reaction of enzymatic garlic hydrolyzate were investigated at 37$^{\circ}C$ for 12 days. Cysteine was the most effective antibrowning agent followed by citric acid. The antibrowning effects of cysteine and citric acid were greater as concentrations increased, and the optimal concentration of both cysteine and citric acid as antibrowning agents was 0.3%. Ascorbic acid itself contributed to the browning reaction and showed an accelerating effect as the concentration increased. The addition of 0.1% ascorbic acid as synergist either to 0.3% cysteine or 0.3% citric acid did not enhance significantly the antibrowning effect of cysteine or citric acid. When stored at 3$0^{\circ}C$, 4$0^{\circ}C$ and 5$0^{\circ}C$, the browning reaction was accelerated as the temperature increased, especially at 5$0^{\circ}C$. Even though the effects of citric acid and cysteine as inhibitors on the browning reaction decreased as temperature increased, cysteine was more effective in decreasing browning reaction than citric acid.

Effect of 1-RM Direct Measurement Method on Beginners' Back Squat (1-RM 직접측정법이 초보자의 백스쿼트에 미치는 영향 )

  • Objective: This study aims to verify effect of 1-RM direct measurement method of back squat on beginners. Method: Total of 8 healthy adults were recruited for this study (age: 29 ± 3.81 yrs., height: 174 ± 3.83 cm., body mass: 74 ± 11.63 kg., 1RM: 96 ± 19.78 kg). All participants performed the back squat with 80%, 90% and 100% of the pre-measured 1RM. A three-dimensional motion analysis was performed with 8 infrared cameras and 3 channels of EMG were used for this study. One-way ANOVA with repeated measure was used for the statistical analysis with the significant level set to α=.05. Results: The ankle joint ROM in the transverse plane was significantly increased as the weight increased during the concentric contraction phase 2 (p < .05). In addition, the erector spinae and the gluteus maximus, which are synergist for the motion, showed a significant difference according to the increased weight (p < .05). Conclusion: Our results revealed that beginners increase potential dynamic knee valgus as weight increased. Therefore, it is thought that field coaches should pay attention to this to minimize and prevent injuries when measuring 1-RM for beginners.

Analysis of the Coordination Relationship and Muscle Fatigue of Agonist and Synergist During Dumbbell Side Lateral Raise Using Biosignals (생체신호를 이용한 덤벨 사이드 레터럴 레이즈 시 주동근과 협력근의 협응 관계 및 근피로 분석)

  • This study aimed to analyze the coordination of muscle and muscle fatigue between biceps, lateral deltoid, and anterior deltoid during dumbbell side lateral raise using bio-signal. One male subject performed dumbbell side lateral raise with 3% of 1RM dumbbell at the rate of 30bpm until failure (8 minutes). While performing, ECG were recorded to observe the participant's performance. EMG were recorded from biceps, lateral deltoid, and anterior deltoid for observing coordination and fatigue. ECG were analyzed in time and frequency domain to observe Heart rate, normLF, normHF. Changes in heart rate, normLF, and normHF indicate that the sympathetic nervous system is activated, while changes in median frequency (MDF) indicate the occurrence of muscle fatigue. Moreover, the coordination relationship of muscle changed. The correlation of MDF between each muscles indicated that lateral deltoid is associated with biceps and anterior deltoid. These results showed that our study can contribute to improving understanding of muscle fatigue and muscle coordination relationships.

Natural Products for Cancer-Targeted Therapy: Citrus Flavonoids as Potent Chemopreventive Agents

  • Targeted therapy has been a very promising strategy of drug development research. Many molecular mechanims of diseases have been known to be regulated by abundance of proteins, such as receptors and hormones. Chemoprevention for treatment and prevention of diseases are continuously developed. Pre-clinical and clinical studies in chemoprevention field yielded many valuable data in preventing the onset of disease and suppressing the progress of their growth, making chemoprevention a challenging and a very rational strategy in future researches. Natural products being rich of flavonoids are those fruits belong to the genus citrus. Ethanolic extract of Citrus reticulata and Citrus aurantiifolia peels showed anticarcinogenic, antiproliferative, co-chemotherapeutic and estrogenic effects. Several examples of citrus flavonoids that are potential as chemotherapeutic agents are tangeretin, nobiletin, hesperetin, hesperidin, naringenin, and naringin. Those flavonoids have been shown to possess inhibition activity on certain cancer cells' growth through various mechanisms. Moreover, citrus flavonoids also perform promising effect in combination with several chemotherapeutic agents against the growth of cancer cells. Some mechanisms involved in those activities are through cell cycle modulation, antiangiogenic effect, and apoptosis induction.Previous studies showed that tangeretin suppressed the growth of T47D breast cancer cells by inhibiting ERK phosphorylation. While in combination with tamoxifen, doxorubicin, and 5-FU, respectively, it was proven to be synergist on several cancer cells. Hesperidin and naringenin increased cytotoxicitity of doxorubicin on MCF-7 cells and HeLa cells. Besides, citrus flavonoids also performed estrogenic effect in vivo. One example is hesperidin having the ability to decrease the concentration of serum and hepatic lipid and reduce osteoporosis of ovariectomized rats. Those studies showed the great potential of citrus fruits as natural product to be developed as not only the source of co-chemotherapeutic agents, but also phyto-estrogens. Therefore, further study needs to be conducted to explore the potential of citrus fruits in overcoming cancer.
