전자정부 구현을 위한 GIS 연계방안 연구 (A Study on Integration Strategies for e-Government and GIS in Korea)

  • 급변하는 정보기술의 발달로 국제적인 이슈로 떠오르고 있는 전자정부는 국내 정보 통신 분야에서도 많은 관심을 보이고 있다. GIS 기술에서도 전자정부는 고려하지 않으면 안될 주요 사항이지만, 국외의 경우와 달리 현재 국내 전자정부를 추진하는데 있어 공간정보와 관련한 내용은 언급되고 있지 않다. 이러한 미래의 숙제를 GIS 측면에서 풀어보고자 국내 전자정부와 GIS의 연계방안을 제시하였다. GIS 연계방안은 추진모델, 추진전략, 추진과제로 이루어진다. GIS 연계방안의 목적은 전자정부에서의 지리정보와 행정정보 연계를 통한 시너지 효과 증대와 GIS Portal 등의 구축으로 기관간 정보 공유 및 활용을 확대하여 행정업무의 효율성을 높이고, 다양한 정보제공 채널을 통하여 공간정보에 쉽게 접근할 수 있도록 하여 대국민 GIS 서비스를 향상시키는 것이다. 더불어 GIS 연계 추진모델이 구현될 수 있도록 행정부처간 협력 강화와 전자정부와의 연계를 위한 GIS 관련 법/제도 마련 등의 기반이 구축되어야 할 것이다. 이러한 GIS 연계 추진모델을 국내 전자정부 성숙모형 특성에 적용함으로써 효율적인 전자정부 구현을 위한 "서비스," "정보연계활용," "기술," "조직" 및 "기반구축"에서의 GIS 연계 추진과제를 도출하였다. 본 연구결과를 통해 전자정부와 GIS 연계는 필수 사항임을 다시 한번 강조하고, 향후 수립하게 될 제3차 국가GIS 기본계획에서는 전자정부와의 연계를 위한 지원 사업이 추진되어야 할 것이다.

대장균과 녹농균에 대항하는 황금과 황련 및 살리실산 조성물의 항균상승효과 (Synergic Antimicrobial Activity of Scutellariae Radix, Coptidis Rhizoma and Salicylic Acid Combination against Escherichia coli and Pseudomonas aeruginosa)

  • Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, Listeria. monocytogenes and Bacillus cereus are pathogenic bacteria that should not be detected in cosmetics and foodstuffs. Therefore, we first investigated the antimicrobial activities of extracts of Scutellariae Radix(SR), Coptidis Rhizoma (CR) and salicylic acid(SA) in these pathogenic microorganisms. Although SA has been known to exhibit anti-inflammation and antimicrobial activity against pathogenic microorganisms, a high concentration of SA may cause serious side effects such as skin redness, skin burning, peeling or tissue damage. Hence, we focused on diminishing side effects followed by treatment of a high concentration of SA and investigated whether the combinations of SA with various concentrations(25-400 mg/mL), SR and CR with a concentration(100 mg/mL) which did not show antimicrobial activity against E. coli and P. aeruginosa exhibited meaningful antimicrobial effect against both strains. In our results, the combinations of SA with the lowest concentration(25 mg/mL), SR(100 mg/mL) and CR(100 mg/mL) exhibited significant antimicrobial activity against E.coli in comparison to SA alone(25 mg/mL) showing no antimicrobial activity. Moreover, the combinations of SA (100 mg/mL), SR and CR showed seven times higher antimicrobial activity against E. coli than SA alone(100 mg/mL) and exhibited a significant antimicrobial activity in comparison to ampicilin (p<0.05). The combinations of SA(100 mg/mL), SR and CR showed two times higher antimicrobial activity against P. aeruginosa than SA alone. Therefore, these results indicated that the combinations of SR, CR and SA with low concentration expressed the synergistic antimicrobial effect against E. coli and P. aeruginosa and showed great potential as an antimicrobial agent.

Ethanolic Extract of Pancake Mixture Powder Supplemented with Helianthus tuberosus Enhances Antidiabetic Effects via Inhibiting Inflammatory Mediator NO Production

  • Helianthus tuberosus is perennial plant as Compositae family and is shown various physiological activities such as analgesic, antipyretic, anti-inflammatory, anti-fungal, anti-spasmodic, aperient, cholagogue, diuretic, spermatogenic, stomachic, and tonic effects. In this study, we investigated the antidiabetic and anti-inflammatory effects of pancake mixture powder (PM) supplemented with H. tuberosus (PMH) in rat skeletal muscle L6 cells and murine macrophage RAW 264.7 cells, respectively. PM and PMH inhibited in vitro α-glucosidase activity. Glucose consumption was increased by PM and PMH without cytotoxicity in rat myoblast L6 cells. Western blot analysis revealed that PM and PMH down-regulated glycogen synthase kinase (GSK)-3β activation in L6 cells. PM and PMH inhibited inflammatory mediator, nitric oxide (NO) production without cytotoxicity in LPS-induced RAW 264.7 cells. The anti-diabetic and anti-inflammatory effects of PMH was more stronger than those of PM. Anti-diabetic and anti-inflammatory effects of PMH would be due to functional characteristics of the supplemented H. tuberosus and the presence of garlic and onion used as ingredients of PM. Taken together, our results that addition of functional materials such as H. tuberosus in product has synergic effects and PMH is potential candidate for treatment of diabetes through inhibiting inflammation.

Tranquilizer-like Effects of Sanjoinine A: Possible GABA/Benzodiazepine Receptors Complex Involvement

  • Zizyphi Spinosi Semen (ZSS) has been widely used for the treatment of anxiety and insomnia in Korea and China. This experiment was performed to know whether sanjoinine A, one of major alkaloid compounds of ZSS has anxiolytic and hypnotic effects through the GABAergic systems. Our results showed that administration of sanjoinine A increased open arm entries and spent time in open arm in the elevated plus-maze and increased head dips in hole board test. Different from traditional anxiolytic, diazepam, sanjoinine A itself did not decrease locomotor activity and strength level in mice. Furthermore, Sanjoinine A (0.5-2.0 mg/kg) prolonged sleeping time and reduced sleeping latency induced by pentobarbital in a dose-dependent manner similar to muscimol, a $GABA_A$ receptor agonist. Sanjoinine A (0.25-1.0 mg/kg) also increased sleeping rate and sleeping time in the combined administration at the sub-hypnotic dose of pentobarbital and showed synergic effects with muscimol in potentiating sleeping onset and enhancing sleeping time induced by pentobarbital. However, sanjoinine A itself did not induce sleeping at the higher dose. In addition, both of sanjoinine A and pentobarbital increased chloride influx in primary cultured cerebellar granule cells. Sanjoinine A decreased the $GABA_A$ receptor ${\alpha}$-subunit expression and increased ${\gamma}$-subunit expression, and had no effects on abundance of ${\beta}$-subunit in primary cultured cerebellar granule cells, showing different expression of subunits from pentobarbital. In conclusion, sanjoinine A shows anxiolytic-like effects and augments pentabarbital-induced sleeping behaviors through the modification of GABAergic systems. 유즙내에서 메티실린 내성을 지닌 젖소 유방염 주요 원인균에 대한 항생제와 EDTA-Tris의 병합의 효과 (The Effect of Antibiotics in Combination with EDTA-Tris on the Methicillin-Resistant Major Pathogens of Bovine Mastitis in Milk)

  • The combined effects of EDTA-Tris and eighteen antimicrobial agents have been evaluated in eight clinically isolated methicillin-resistant bacteria (Staphylococcus aureus, Escherichia coli, Streptococcus uberis and Streptococcus agalactiae) from bovine mastitis. The antimicrobial activity was evaluated by measuring the minimal bactericidal concentration (MBC) for the antibiotics alone or in combination with EDTA-Tris in Mueller-Hilton broth and milk. Combined use of EDTA-Tris and antibiotics potentiated or antagonized activity of antibiotics against mastitic pathogens. Milk increased the antibiotic potency of erythromycin and spiramycin on S. aureus. Culture in milk changed patterns of EDTA-Tris combinational effects compared with that in standard Mueller-Hilton broth. Combined with EDTA-Tris in milk, synergic effects were observed in colistin, dihydrostreptomycin, kanamycin, erythromycin, gentamycin, oxytetracycline, streptomycin to E. coli, Str. uberis, and Str. agalactiae. However, significant antagonistic effects of milk on antibiotic susceptibility in combination with EDTA-Tris were noted in neomycin, streptomycin, penicillin, roxithromycin, and amoxicillin. This study indicates that combination therapy of EDTA-Tris with antibiotics in bovine mastitis should be used with caution because of the possible antagonistic effects of antibiotic combination with EDTA-Tris on mastitic pathogens. In addition, antibiotic susceptibility test in combination with EDTA-Tris in milk culture condition can be benefit in search of effective treatment regimen for some antibiotic-resistant bacteria of mastitis.

수종의 한약재 열수추출물이 멜라토닌 수용체 발현에 미치는 영향 (Effects of several herbs on the expression of MT1 and MT2 melatonin receptors in C6 glial cells)

  • Objective : This study was to investigate the effects of several herbs on the levels of MT1 and MT2 melatonin receptors Methods: It was investigated the effects of several herbs such as WEDL, WEZV, WEFO, WEOC on the levels of MT1 and MT2 melatonin receptors using C6 glial cell model. ${\beta}-estradiol$ treatment, as a positive control group, under non-cytotoxic condition. Results : 1. The water extracts of Dimocarpus long (WEDL) induced the levels of MT2 melatonin receptor expression in a concentration-dependent manner without altering the levels of MT1 melatonin receptor expression. 2. The treatment with the water extract of Zizyphus vulgaris (WEZV) induced the levels of MT1 melatonin receptor expression and the levels of MT2 melatonin receptor expression was not affected. 3. The levels of MT1 as well as MT2 melatonin receptor expression were markedly up-regulated in the water extract of Fossilia ossis (WEFO) and the water extract of Ostreae caro (WEOC)-treated C6 cells. 4. The combination treatment with WEDL and WEZV induced not only the levels of MT1 melatonin receptor expression but also MT2 melatonin receptor expression, but the synergic effects of the combination treatment with WEFO and WEOC were not detected in C6 cells. Conclusion : The study provides important new insights into the possible mechanisms on the regulation of melatonin receptor synthesis by WEDL, WEZV, WEFO and WEOC.

3T3-L1 지방세포에서 황백 추출물의 Glucose Uptake 촉진 및 인슐린 저항성 개선 활성 (Enhancing Effect of Extracts of Phellodendri Cortex on Glucose Uptake in Normal and Insulin-resistant 3T3-L1 Adipocytes)

  • Anti-hyperglycemic effects of 17 medicinal plants that have been used for ameliorating diabetes in oriental medicine were evaluated using glucose transport assay in 3T3-L1 adipocytes. Higher activities were obtained by treating water or alcohol extract of Phellodendri Cortex (PC), which showed enhancing effects both on basal and insulin-stimulated glucose uptake. The latter effect of PC was completely inhibited by wortmannin, a specific inhibitor for phosphatidyl inositol 3-kinase (PI 3-kinase), but not affected by SB203580, A specific inhibitor for p38 mitogen-activatedprotein kinase(MAPK). Genistein, an inhibitor for tyrosine kinases, abolished the PC effects completely. Treatment of vanadate, an inhibitor for tyrosine phosphatases, together with PC showed no significant synergic enhancement in glucose uptake. The results of inhibitors associated with insulin signaling pathway indicated that extracts of PC enhance glucose uptake by PI-3 kinase activation which is an upstream event for GLUT4 translocation. Antidiabetic effects of PC extract might be also due to enhanced tyrosine phosphorylation and reduced tyrosine dephosphorylation. In addition, PC accelerated insulin-stimulated glucose uptake in insulin-resistant cells, recovering the uptake level close to that of normal cells. These findings may offer a new way to utilize extracts of PC as novel anti-hyperglycemic agents.

Saponin-SOD가 SNUC-4 Cell에 의해 유발편 대장암 Nude Mice의 암 성장 억제 효과 (Tumor-Suppressing Effects of Saponin-SOD in Nude Mice)

  • This study was performed to measure the antioxidant effects of red ginseng extracts which antioxidation had been promoted through enzyme hydrolysis. In order to observe their tumor-suppressing effects, an anti-cancer medicine and Saponin-SOD, which was a highly antioxidant beverage made from red ginseng saponin adding SOD-like rice (with embryo buds) extracts, were administered to nude mice with large intestine cancer induced. There was a significant increase in the content of phenolic compounds as the enzyme was added. The red ginseng extracts showed a high electron-donating ability with the passage of time. The electron-donating ability was particularly high in the enzyme-treated red ginseng extract, and also observed as high in Saponin-SOD. The lipid-peroxide generation was inhibited depending on the concentration of Saponin-SOD added; the addition of 0.625% Saponin-SOD served to decrease the inhibition level up to 65% compared with the case of no addition (100%). As a result, it could be assumed that Saponin-SOD would strongly inhibit the oxidation of ghost membrane. After the cancer was induced in nude mice through the injection of SNUC-4 cell, there was a significant inhibition in the growth of tumors in nude mice into which Saponin-SOD were injected; the growth of tumors was gradually decreasing with the passage of time after the cancer induction. In particular, when Saponin-SOD was administered together with an anti-cancer medicine, the synergic effect was observed. In conclusion, Saponin-SOD, when used with an anti-cancer medicine, is expected to reduce the amount of free radical and lipid peroxide, which are known to cause harmful effects occurring from the internal application of medicine.

The expression and secretion of vimentin in the progression of non-alcoholic steatohepatitis

  • The pathogenesis of non-alcoholic steatohepatitis (NASH) is not fully understood. In the present study, both in vitro and in vivo vimentin expression and secretion in NASH were investigated. The exposure of palmitate and lipopolysaccharide (LPS) to HepG2 cells enhanced caspase-3 activity and vimentin expression, respectively. The combined effects of both treatments on vimentin expression and caspase-3 activation appeared to be synergic. In contrast, blockade of caspase-3 activity by zVADfmk resulted in a significant reduction of cleaved vimentin and secreted vimentin into the culture supernatant. Similarly, lipid accumulation and inflammation occurred in mice fed a methionine-choline-deficient diet; thus, vimentin expression and serum cleaved vimentin levels were increased. However, vimentin was not significantly upregulated, and no cleavage occurred in mice fed a high-fat diet. It was conclusively determined that lipid accumulation in hepatocytes induces apoptosis through a caspase-3 dependent pathway; whereas, LPS stimulates vimentin expression, leading to its cleavage and secretion. Increased vimentin fragment levels indicated the existence of substantial hepatocellular death via an apoptotic mechanism.

Up-regulation of Asiaticoside Biosynthesis by Methyl Jasmonate and Thidiazuron in Centella asiatica L. Urban

  • Centella asiatica accumulates large amounts of triterpene saponin, such as centellasaponin, asiaticoside, madecassoside. We examined the effect of two candidates, MJ (Methyl jasmonate) and TDZ (thidiazuron), on asiaticoside production and the accumulation of bAS mRNA associated with asiaticoside biosynthesis in leaves of cultured whole plants. The growth of whole plants treated with 0.1 mM MJ was found to decrease significantly, however, the growth of whole plants treated with 0.1 mM MJ plus 0.025 mg/l TDZ was better than that treated with MJ alone. When MJ alone was added to culture medium, asiaticoside contents in leaves were higher than that of control after 7 days of treatments. The maximum level of bAS $({\beta}-amyrin\;synthsae)$ mRNA in leaves of whole plant treated TDZ and MJ was transiently observed after exposure to 5 days. These results showed the up-regulation of bAS gene by adding TDZ and MJ at the molecular level, however, synergic effects of TDZ and MJ on asiaticoside biosynthesis were not testified.
