동물의 번식 효율 증진을 위한 한약제의 활용 (Application of pure organic phyto-formulations to optimize performance results in the conventional European animal breeding and production)

  • 유럽에서 돼지 번식 효율 증진과 비육을 위한 적절한 활용방법과 몇 가지 잘 조제된 한약제가 규격화되어 나왔다. 이 자료에는 한약제의 효능의 기준을 비롯하여 위생에 대해서도 제시하고 있다. 대학 및 연구기관, 임상 수의사들과 사료업체들이 수행했던 지난 수년간의 실험과 연구 결과를 토대로 돼지 번식효율 증진을 위한 한약제를 사료첨가제로 활용하여 최적화시켰다. 유럽의 수의분야에서 사용되었던 4가지의 특정 한약제를 기본으로 하여 돼지 사양에 있어서 가장 중요한 문제들의 대부분이 성공적으로 해결되었다. 즉 수정률, 호흡기질병, 설사와 스트레스에 관한 문제들인데 면역이 억압되고 저질의 자돈이 생산됨에 따라 생산성 저하에 따른 엄청난 경제적인 손실을 대처할 수 있다. 또한 일반적인 임상증상을 치료하면서 동시에 가장 중요한 것 중의 하나는 자기방어체계를 증진시킨다는 것이다. 이로써 동물의 면역력과 활력을 증가시키고 건강을 유지시켜서 폐사율을 감소시킨다. 게다가 여기에서 제시된 자료들은 일상적으로 사용되었던 항생제나 호르몬제제들의 사용을 대체할 수 있는 한약제들인데 고효율임이 증명되었다. 이러한 한약제들은 백신 프로그램이나 성장을 촉진시키는 인자들과 상호보완 상승작용도 가져 온다.

포유 및 이유자돈의 질병발생 동향 (Investigation of diseases incident to pre- and pos-weaning piglets)

  • This experiment was carried out to investigate the tendency of incident diseases in pre- and post- weaning piglets which ages were 1 to 7 weeks old by laboratory diagnosis and in order to minimize death in preweaning piglets and of stunted growth in postweaning piglets. The result of this experiment used as the basic data for the preventive programs in pre- and post- weaning piglets and were as follows: 23 different diseases diagnosed in 331 cases were studied in relation to age, season, and etiology. The most prevelent diseases of pre- and post- weaning piglet were Colibacillosis(79 case, 23.9%) and the major diseases were Salmonellosis(44 cases, 13.3%), Anemia(37 cases, 11.2%). Unknown viral disease(20 case, 6.1%), Rota viral infection(19 case, 5.8%), Porcine reproductive & respiratory syndrome(PRRS; 15 case 4.5%), Transmissible gastroenteritis(TGE; 12 case, 3.6%). The gastrointestinal disease, such as Colibacillosis, Salmonellosis, Swine dysentery, Clostridial infection, Rotaviral infection, TGE, Porcine epidemic diarrhea(PED) and Ballantidiosis occured pro- dominently in the period of pre- and post- weaning, which were 178 cases(53.8%) and not related to occurrence according to age and season. The respiratory diseases were Atrophic rhinitis(AR), Swine enzootic pneumonia, Pneumonic pasteurellosis, Pleuropneumonia, Branchopneumonia, PRRS and which were 48 cases(14.5%) and higher prevalent in spring and summer. The viral diseases was 73 cases(22.1%) that occurred in the period of 5 weeks piglet and prevalent mainly in spring. The bacterial diseases were 188 cases(56.8%) that were not related to occurrence according to age and season. Salmonellosis was prevalent in 3 to 5 weeks piglet and mainly occurred in summer. Viral septicemia and rotaviral infection occurred after 5 weeks piglets intensively and 3 to 5 weeks, respectively. And the both occurred without relation with season. PRRS occurred after 4 weeks piglet and prevalent in summer. TGE occurred 1 to 7 days old piglets and prevalent in spring and winter. Hematologic values of anemia was decrease in number of Red Blood Cell, concentration of Hemoglobin and Hematocrit. Amikacin, cephalothin, colistin, norfloxacin were effective to E coli, and amikacin, cephalothin, nortloxacin, neomycin were effective to Salmonellra spp. but clindamycin, erythromycin, penicillin, sulfonamides were resistant to E coli and Salmonella spp.

돼지 분변 유래 PORCINE ENTEROVIRUS의 물리화학적 특성 및 병원성에 관한 연구 (Studies on Physico-chemical Properties an d Pathogenicity of Porcine Enterovirus Isolated from Feces of Pigherds)

  • 28 porcine enteroviruses were isolated from 86 pig-feces of 9 swine farms located in south region, Chung-buk, from March to September 1990. Physico-chemical properties and pathogenicity of isolates were investigated. Results obtained throughout experiments are summarized as follows. According to the age, weanlings(40-90 days), sucklings(10-30 days) and adult pigs(6 months over) showed the isolation rate of 67%. 8% and 4%, respectively. By physico-chemical tests, YD-90/22, YD-90/43 and YD-90/64 strains were found to be ether, chloroform and PH stable. Nucleic acid test suggests the virus to have a DNA genome. Most of the Isolates were not evident of hemagglutinin using erythrocytes from various mammalian & avian. 22 strains among the isolates were shown CPE type I and the remainders were CPE type II. 3 strains among isolates of CPE type I strains were neutralized with high titers to serotype 2 antiserum. In the study on virus growth curve in PK-l5 cells, YD-90/22, YD-90/43 and YD-90/64 strains showed the maximum infectivity titers($10^{6.0}-l0^{6.5} TCID({50}ml$) at 4days post inoculation(PI). When 30 day-old commercial piglets were inoculated only intraoral route with the YD-90/22 strain at $10^{6.0} TCID_{50}ml,$ piglets not showed the symptoms. But piglets inoculated by intramuscle route, intraoral and intramuscle route after pretreat with dexamethasone(2.5mg /kg) for 5 days were shown the symptoms of anorexia, diarrhea, pyrexia and ataxia at 4th-6th days PI. The viral reisolation in the virus-inoculated piglets was examined from feces. The viruses were recovered intermittently from 2nd to 16th day PI and at 4th-6th day PI, all piglets excreted viruses.

Prevalence and genotypes of pestivirus in Korean goats

  • In total, 1,142 serum samples were collected from 223 goat flocks rising in five different regions of Korea. These samples were screened for the presence of border disease virus (BDV) antibodies using an enzyme linked immunosorbent assay. Of the 1,142 samples, we found 47 bovine viral diarrhea virus (BVDV) positive cases (4.1%). These positive serum samples were also examined further by using the virus neutralization test against BDV. In addition, samples were tested for both BVDV and classical swine fever virus (CSFV). All of the samples that were seropositive for BDV also demonstrated positive antibody titers against BVDV and CSFV. Due to their common antigenicity, we also determined further the prevalence and carried out virus neutralization test against three pestiviruses: 314 of the goat samples were screened using reverse transcription polymerase chain reaction with primer pairs specific to common pestivirus genome regions. Overall, 1.6% (5/314) of the samples tested was positive for pestivirus. Based on the nucleotide sequence data and the phylogenetic analysis, three isolates were characterized as BVDV type 1 and two isolates as BVDV type 2. However, none of the isolates could be classified as BDV. These results indicate that BVDV-1 and BVDV-2 are the pestivirus strains circulating among Korean goat populations.

Functional Characterization and Proteomic Analysis of Porcine Deltacoronavirus Accessory Protein NS7

  • Porcine deltacoronavirus (PDCoV) is an emerging swine enteric coronavirus that causes diarrhea in neonatal piglets. Like other coronaviruses, PDCoV encodes at least three accessory or species-specific proteins; however, the biological roles of these proteins in PDCoV replication remain undetermined. As a first step toward understanding the biology of the PDCoV accessory proteins, we established a stable porcine cell line constitutively expressing the PDCoV NS7 protein in order to investigate the functional characteristics of NS7 for viral replication. Confocal microscopy and subcellular fractionation revealed that the NS7 protein was extensively distributed in the mitochondria. Proteomic analysis was then conducted to assess the expression dynamics of the host proteins in the PDCoV NS7-expressing cells. High-resolution two-dimensional gel electrophoresis initially identified 48 protein spots which were differentially expressed in the presence of NS7. Seven of these spots, including two up-regulated and five down-regulated protein spots, showed statistically significant alterations, and were selected for subsequent protein identification. The affected cellular proteins identified in this study were classified into functional groups involved in various cellular processes such as cytoskeleton networks and cell communication, metabolism, and protein biosynthesis. A substantial down-regulation of α-actinin-4 was confirmed in NS7-expressing and PDCoV-infected cells. These proteomic data will provide insights into the understanding of specific cellular responses to the accessory protein during PDCoV infection.

국내 분리 오제스키병 바이러스 접종자돈의 병리학적 관찰 (Pathological observation on the piglets experimentally infected with Aujeszky′s disease virus isolated in Korea)

  • Pathological studies were performed on the five piglets experimentally infected with Aujeszky's disease virus(pseudorabies), NYJ isolate, isolated from the naturally infected pigs in Korea: two piglets were inoculated intramuscularly, two piglets intranasally, and one piglet subcutaneously at the dose of 1$m\ell$ per animal with the 105.5 $TCID_50$/0.1ml titer. Clinical signs included dyspnea, high fever(>$41^{\circ}C$), anorexia, vomiting, diarrhea or constipation, ataxia, circling movement, posterior paralysis, intermittent convulsion, and coma followed by death although some variations by age and inoculated routes were observed. Gross features included multiple necrotic foci in the liver, congestion and hemorrhage in the lymph nodes and spleen, petechial hemorrhage in the kidney, hemorrhagic pneumonia, marked meningeal congestion, severe sub meningeal hemorrhage in the spinal cord, excessive cerebrospinal fluid retention, and muscular necrosis at the inoculated area. Microscopically, non suppurative meningoencephalitis with gliosis and perivascular cuffing in CNS, ganglioneuritis, necrohemorrhagic splenitis, necrotic hepatitis, tonsillitis and rhinitis, hemorrhagic or interstitial pneumonia, and non-suppurative myositis in the injected area were observed. Eosinophilic intranuclear inclusion bodies were found in a variety of tissues the including the liver, kidney, adrenal gland, spleen, lymph nodes, tonsil, and lung. Ultrastructurally, virus particles were confirmed in nucleus and cytoplasms of pneumocytes around the necrotic areas.

Evaluation of porcine intestinal organoids as an in vitro model for mammalian orthoreovirus 3 infection

  • Background: Mammalian orthoreovirus type 3 (MRV3), which is responsible for gastroenteritis in many mammalian species including pigs, has been isolated from piglets with severe diarrhea. However, the use of pig-derived cells as an infection model for swine-MRV3 has rarely been studied. Objectives: This study aims to establish porcine intestinal organoids (PIOs) and examine their susceptibility as an in vitro model for intestinal MRV3 infection. Methods: PIOs were isolated and established from the jejunum of a miniature pig. Established PIOs were characterized using polymerase chain reaction (PCR) and immunofluorescence assays (IFAs) to confirm the expression of small intestine-specific genes and proteins, such as Lgr5, LYZI, Mucin-2, ChgA, and Villin. The monolayered PIOs and three-dimensional (3D) PIOs, obtained through their distribution to expose the apical surface, were infected with MRV3 for 2 h, washed with Dulbecco's phosphate-buffered saline, and observed. Viral infection was confirmed using PCR and IFA. We performed quantitative real-time reverse transcription-PCR to assess changes in viral copy numbers and gene expressions linked to intestinal epithelial genes and antiviral activity. Results: The established PIOs have molecular characteristics of intestinal organoids. Infected PIOs showed delayed proliferation with disruption of structures. In addition, infection with MRV3 altered the gene expression linked to intestinal epithelial cells and antiviral activity, and these effects were observed in both 2D and 3D models. Furthermore, viral copy numbers in the supernatant of both models increased in a time-dependent manner. Conclusions: We suggest that PIOs can be an in vitro model to study the infection mechanism of MRV3 in detail, facilitating pharmaceutical development.

돼지 유행성 설사병 바이러스 스파크 단백질 유전자 발현 형질전환 담배 배양세포 (Transgenic tobacco culture cells expressing spike protein gene of porcine epidemic diarrhea virus)

  • Porcine epidemic diarrhea virus (PEDV)는 돼지의 급성장염을 유발하여 설사 등의 증상을 일으키는 바이러스이다. 본 연구에서는 PEDV 항원단백질을 생산하는 담배 배양세포주를 개발하고자 하였다. PEDV에서 항원성이 알려진 스파크 단백질의 일부분을 암호화하는 유전자를 PCR로 합성하여 4종류의 형질전환 벡터를 제작하였다. 담배 배양세포 BY-2를 재료로 하여 Agrobacterium tumefaciens을 매개로 형질전환하였다. 선발배지 (MS salt, $KH_2PO_4$ 370 mg/L, 2,4-D 0.18 mg/L, Thiamin HCl 1 mg/L, kanamycin 100 mg/L, 침랙무 400 mg/L)에서 캘러스를 3주 간격으로 3개월 동안 계대배양하여 카나마이신 저항성 캘러스를 선발하였다. 선발된 캘러스를 대상으로 PCR 분석한 결과 형질전환 효율은 75% 이상이었으며 벡터당 40 여개 이상의 형질전환 배양세포주를 얻었다. 형질전환 배양세포주를 대상으로 Southern blot 분석하여 PEDV 유전자가 고구마 식물체의 게놈으로 안정적으로 도입되었음을 확인하였다. Northern blot 분석 결과 PEDV 스파크 단백질 유전자가 높은 수준으로 발현함을 확인하였으며 dot blot으로 PEDV 스파크 단백질 고생산 배양세포주를 선발하였다. 형질전환 담배 배양세포로부터 생산된 PEDV 항원단백질을 BALB/c 마우스에 경구투여 하여 면역활성을 조사한 결과 형질전환 세포주인 35S::SP1-M, 35S::SP2-M, 35S::SP4-M 세포주에서 1:10의 희석배수까지 바이러스 억제효과가 관찰되었다. 제작된 형질전환 벡터는 고구마와 같은 경구용 사료작물에 활용할 수 있을 것이다.

이유자돈의 실험적 enterotoxigenic Escherichia coli K88 감염에 의한 설사증에 대한 박테리오파지의 예방과 치료에 대한 효능 평가 (Evaluation of bacteriophages for prevention and treatment of diarrhea due to experimental enterotoxigenic Escherichia coli K88 infection of weaned piglets)

  • Colibacillosis in pigs remain a major swine industry bruden worldwide. Although some progress has been made in treating collibacillosis in pigs by using biosecurity and antimicrobials, it still remain a considerable problem. The use of host-specific bateriophages as a biocontrol is one possible alternative. The purpose of this study was to evaluate the effect of bacteriophage against enterotoxigenic Escherichia coli (ETEC) K88 infection in piglets. Twenty-eight piglets were randomly divided into four groups and each group was allocated with 7 pigs. Group B, C and D were inoculated with 5 ml of ETEC K88 ($1{\times}10^8$ CFU/ml) per head of piglet via oral. Group C and D were fed with bacteriophages (Group C, $1.0{\times}10^6$ PFU/g; Group D, $1.0{\times}10^8$ PFU/g; CJ CheilJedang Corp., Korea) orally as treatment. In piglets administrated bacteriophages and challenged with ETEC K88 (Group C and D), Clinical signs and the growth performance were improved and antibody titers were maintained low level compared with piglets challenged with ETEC K88 (Group B, P<0.05). Group B were shown high pH in the alimentary tracts compared with other piglets (P<0.05). In quantitative analysis by real-time PCR, the results of Group C and D were lower than those Group B in faecal and intestinal samples (P<0.05). Severe villus atrophy and crypt hyperplasia were observed in Group B consequently V/C ratio increased, compared with other piglets. These results indicate that feeding with bacteriophage has effect to prevent ETEC K88 infection in piglets and suggest that use of bacteriophage can be considered a valid antibiotic alternative.

Effects of dietary L-glutamine and glutamic acid combination, and whey powder on the performance and nutrient digestion in weaned piglets fed grain-based diets

  • Objective: The present study aimed to evaluate the influence of including L-glutamine along with glutamic acid as a supplement in weaned piglets' diets with and without whey powder. Methods: Two assays were carried out. A total of 40 piglets ([Landrace×Large White]×Pietrain) weaned at 24 days of age with an initial body weight of 6.6±0.6 kg were used in the first assay, and the following parameters were evaluated: growth performance, the incidence of diarrhea, morphometry, intestinal integrity, and hepatic glycogen index. The animals were then blocked into four groups according to different diets: diet all-grain feeding (G); diet all-grain feeding with whey powder (GW); and with vs without 1% supplementation of the commercial product containing L-glutamine and glutamic acid (A or NA). Whey powder was added according to the stage of life, corresponding to 17%, 10%, and 5%, respectively, in order to meet the need for lactose. The animals were evaluated at 24 to 42 days and at 24 to 55 days of age. The nutrient digestibility for the second assay was carried out by using 24 animals with an average weight of 11.49±1.6 kg, and the same diets were tested. Results: The supplementation of L-glutamine + glutamic acid or the addition of whey powder in diets for weaned piglets provided (p<0.05) greater feed intake, greater weight gain and improved feed conversion in the initial period (24 to 42 days age). However, in the whole period (24 to 55 days age) only amino acid supplementation affected (p<0.05) growth performance. There was a positive interaction (p<0.05) between the type of diet and L-glutamine + glutamic acid supplementation on villus height, crypt depth and the villus:crypt ratio in the duodenum. In addition, L-glutamine + glutamic acid supplementation reduced (p<0.05) the crypt depth and improved the villus:crypt ratio in the jejunum. The inclusion of whey powder affected (p<0.05) positively the digestibility coefficients analyzed except mineral matter digestibility coeficients. The supplementation of 1% the commercial product composed of L-glutamine and glutamic acid improved (p<0.05) only the digestibility coefficient of crude protein. Conclusion: These results indicate that supplementation of 1% commercial product containing L-glutamine + glutamic acid in diets for piglets from 24 to 55 days of age, dispenses with the use of whey powder when evaluating growth performance. Amino acid supplementation alone or associated with whey powder affects (p<0.05) positively the indicators of the intestinal integrity.
