Classical Swine Fever Virus: Discrimination Between Vaccine Strains and Korean Field Viruses by Real-time RT-PCR

  • Classical swine fever (CSF) is a contagious disease of swine with serious economic losses in pig industry [1]. The disease is caused by CSFV which belongs to the viruses of bovine viral diarrhea (BVDV) and border disease virus (BDV) make up the Pestivirus genus within the family Flaviviridae [2]. Attenuated Korean LOM strains were used in Korea. For these reasons a practical approach for discrimination between vaccine and field strains is needed. Here, we described the deveopment of real-time RT-PCR to discriminate between vaccine strains and Korean field viruses of CSFV. (omitted)

2009년 신종 인플루엔자로 영남대학교 의료원에 입원한 환아의 실태 조사 (Experience for S-OIV of Admission Pediatric Patient with S-OIV at YUMC, 2009)

  • The clinical picture in severe cases of pandemic (H1N1) 2009 influenza is markedly different from the disease pattern seen during the epidemics of seasonal influenza as many of those affected were previously healthy young people. Current predictions estimate that during a pandemic wave, 12~30% of the population will develop clinical influenza (compared with 5~15% for seasonal influenza) with 4% of those patients requiring hospital admissions and one in five requiring critical care. Until July 6, 94,512 people have been infected in 122 countries, of whom 429 have died with an overall case-fatality rate of <0.5%. Most of the confirmed cases of S-OIV (Swine-Origin Influenza A Virus) infection have been characterized by a self-limited, uncomplicated febrile respiratory illness and 38% of the cases have also included vomiting or diarrhea. Efforts to control these outbreaks are based on our understanding of novel S-OIV (Swine-Origin Influenza A Virus) and the previous influenza pandemics. So, this review covers the experience with S-OIV (Swine-Origin Influenza A Virus) for the admission and background data and the clinical presentation, diagnosis and treatment of H1N1 in pediatric patient with S-OIV (Swine-Origin Influenza A Virus) at YUMC, 2009.

Assessing the risk of recurrence of porcine epidemic diarrhea virus in affected farms on Jeju Island, South Korea

  • Background: Porcine epidemic diarrhea virus (PEDV) is a swine enteropathogenic coronavirus that has devastated the swine industry in South Korea over the last 30 years. The lack of an effective method to control the endemics has led to a surge in PEDV recurrences in affected farms throughout the country. Objectives: In the first step toward establishing systematic monitoring of and active control measures over the swine populations, we constructed an assessment model that evaluates the status of (1) biosecurity, (2) herd immunity, and (3) virus circulation in each of the PEDV-infected farms. Methods: A total of 13 farrow-to-finish pig farms with a history of acute PEDV infection on Jeju Island were chosen for this study. The potential risk of the recurrence in these farms was estimated through on-site data collection and laboratory examination. Results: Overall, the data indicated that a considerable number of the PEDV-infected farms had lax biosecurity, achieved incomplete protective immunity in the sows despite multi-dose vaccination, and served as incubators of the circulating virus; thus, they face an increased risk of recurrent outbreaks. Intriguingly, our results suggest that after an outbreak, a farm requires proactive tasks, including reinforcing biosecurity, conducting serological and virus monitoring to check the sows' immunity and to identify the animals exposed to PEDV, and improving the vaccination scheme and disinfection practices if needed. Conclusions: The present study highlights the significance of coordinated PEDV management in infected farms to reduce the risk of recurrence and further contribute towards the national eradication of PEDV.

Growth Performance and Post-Weaning Diarrhea in Piglets Fed a Diet Supplemented with Probiotic Complexes

  • Weaning stress can affect the growth performance and intestinal health of piglets. Dietary alternatives to antibiotics, such as dietary probiotics, especially those containing multiple microbial species, are a preventive strategy for effectively controlling post-weaning diarrhea. In this study, we investigated forty-eight crossbred piglets in three treatment groups for 21 days: the control and experimental groups were supplemented with Enterococcus faecium DSM 7134, Bacillus subtilis AS1.836 plus Saccharomyces cerevisiae ATCC 28338 (EBS) or Lactobacillus paracasei L9 CGMCC No. 9800 (EBL). On day 21, weaned piglets supplemented with two kinds of probiotic complexes showed increased growth performance and significantly reduced post-weaning diarrhea (p < 0.05). The EBS treatment increased acetic acid and propionic acid in the feces (p < 0.05), and the EBL treatment increased fecal acetic acid, propionic acid, butyrate and valerate (p < 0.05). Moreover, the fecal microbiota of the piglets changed markedly in EBL treatment. The addition of EBS and EBL may have similar effects on the prevention of diarrhea by improving the intestinal morphology and regulating the microbiota during the weaning period.

  • This study examined the prevalence of adherence factors, toxin genes, antimicrobial resistance phenotypes, and resistance genes in Escherichia coli (E. coli) isolated from piglets with diarrhea before and after the ban on antibiotic growth promoters (AGPs) in Korea from 2007 to 2018. In this period, pathogenic 474 E. coli isolates were obtained from diarrheic piglets. The virulence factors and antimicrobial resistance genes were assayed using a polymerase chain reaction, and the susceptibility to antibiotics was tested according to the Clinical and Laboratory Standards Institute guidelines. After the ban on AGPs, the frequency of F4 (12.5% to 32.7%) increased significantly, and LT (31.9% to 20.3%) and EAST-I (46.5% to 35.2%) decreased significantly. In addition, the resistance to streptomycin (45.8% to 67.9%), cephalothin (34.0% to 59.4%), and cefazlin (10.4% to 28.8%) increased significantly. Colistin resistance plasmid-mediated genes, mcr-1 and mcr-3, were detected after the ban on AGPs. The results of this study can provide useful data for analyzing the impact of the ban on AGPs on the virulence profiles and antimicrobial resistance of E. coli isolated from piglets with diarrhea in Korea.

전북지역 돼지유행성설사 바이러스 Spike 유전자분석 (Sequence analysis of spike genes of porcine epidemic diarrhea virus (PEDV) from Jeonbuk province)

  • Porcine epidemic diarrhea (PED) is a highly contagious enteric viral disease of pigs with watery diarrhea in piglets, which ultimately results in huge economic losses in the swine industry. The spike (S) protein plays an important role in viral pathogenicity, tissue tropism, infection, dissemination and the trypsin-dependent proliferation of the PED virus (PEDV). In the present study, we determined the full-length spike (S) gene sequences of twenty PEDV field strains detected in Jeonbuk province in 2022. Phylogenetic analysis showed that the twenty PEDV field strains were classified into G2b group and shared 98.6~100% of nucleotide homology and 97.4~100% of amino acid homology with each other. Mutations of amino acid sequences on the neutralizing epitope of S protein were observed in the twenty field strains compared to the previous vaccine strain SM-98-1 (G1a group). Therefore, these amino acid mutations in the PEDV S protein may result in a new genotype of the virus and highly pathogenic virus, so continuous monitoring is required.

Prevalence and molecular characterization of novel recombinant enterovirus G isolates in Jeju Province of South Korea

  • Enterovirus species G (EV-G) is highly diverse, and is ubiquitous in pig populations, usually without diarrhea. The present study aimed to investigate the presence of novel EV-G recombinants with the torovirus papain-like cysteine protease (PLCP) in Jeju pig herds. EV-G1-PLCP mono-infections were most prevalent in diarrheic weaned piglets. The PLCP genes of the Jeju isolates varied in size and junction sequences, and were greatly heterogeneous, with 77.0-90.7% homology amongst all recombinants. Our results suggest that the exogenous PLCP gene has undergone continuous rapid mutation in the individual EV-G genomes following cross-order recombination, thereby causing clinical disease in swine.

  • Since its identification in April 2009, a swine-origin H1N1 influenza A virus (S-OIV) which is a reassortment of gene segments from both North American triple-reassortant and Eurasian swine influenza has been widely spread among humans in unexpected rapidity. To date, each gene segment of the 2009 influenza A (H1N1) outbreak viruses have shown high (99.9%) neucleotide sequence identity. As of July 6, 94,512 people have been infected in 122 countries, of whom 429 have died with an overall case-fatality rate of <0.5%. Most confirmed cases of S-OIV infection have been characterized by self-limited, uncomplicated febrile respiratory illness and 38% of cases have also included vomiting or diarrhea. Standard plus droplet precautions should be adhered to at all times. Tests on S-OIV have indicated that current new H1N1 viruses are sensitive to neuraminidase inhibitors (oseltamivir). However, current less virulent S-OIV may evolve into a pathogenic strain or acquire antiviral resistance, potentially with more severe clinical consequences. Efforts to control these outbreaks would be based on our understanding of novel S-OIV and previous influenza pandemics.

Lactobacillus plantarumCLP-1이 돼지바이러스에 미치는 효과 (The Effect of Lactobacillus plantarumCLP-1 on the Swine Viruses)

  • To isolate Lactic acid bacteria for animals, we have screened from Kim-chi, swine intestine, swine feces, and dairy products by random selection and anti-viral, antipathogenic bacteria test. Among them, CLP-1 shown that inhibitory effect against rotavirus, porcine epidemic diarrhea (PED) virus, Salmonella sp, and E.coli. By examining biological property, API-ZYM and identified Lactobacillus plantarum by 16S rDNAgene sequence. CLP-1 determined resistance to low pH and bile salt. Futhermore, the cell body of CLP-1 adhered to the intestinal epithelium tissue of swine and Caco-2 cell. CLP-1 was examined on cell immune system modulating activity in vitro. The whole cell and cell culture supernatant was increasing of interferon-${\beta}$ activity. And then, CLP-1 increased prevention effect by Salmonella enteritidis infection in SPF chickens. And we determined similar result in pigs.

HACCP시스템 적용이 양돈농장의 장·단점과 폐사두수에 미치는 영향 (Effects of HACCP System Implementation on Advantage and Disadvantage and Mortality Number of Swine Farms in Korea)

  • 본 연구는 양돈농장에 HACCP 제도 도입이 폐사두수에 미치는 영향과 HACCP 제도 적용 목적 및 장 단점을 분석하기 위한 목적으로 실시하였다. HACCP 제도 적용 전 후에 따른 돼지의 폐사두수 변화는 HACCP 제도 적용 전 호흡기와 설사로 인한 폐사두수가 각각 288.30두와 122.90두로 나타났으나 HACCP 적용 후에는 각각 261.60두와 101.10두로 감소하는 것으로 나타났다. 또한 유 사산, 사고사 및 기타 폐사두수는 HACCP 적용 전에 각각 91.08두, 18.22두, 108.10두로 나타났으나 HACCP 적용 후에는 85.91두, 16.37두, 108.60두로 조사되었다. 따라서 총 폐사두수는 HACCP 적용 전 628.70두에서 HACCP 적용 후 573.60두로 감소하였다. 양돈농장의 HACCP 적용 목적 중 1순위와 2순위는 각각 농장의 경쟁력 향상(26.92%)와 위생적이고 안전한 돼지생산(23.43%)인 것으로 나타났다. 또한 HACCP 적용에 따른 가장 큰 장점으로는 농장의 위생관리 수준 향상(20.90%)이었으며, 단점은 HACCP 기록(23.10%)으로 조사되었다.
