경옥고의 생리활성 (II) -고혈당, 고혈압, 지구력 및 체중감소에 미치는 영향- (The Physiological Activities of Kyung Ok-Ko(II) -Effects on the Hyperglycemia, Hypertension, Anti-Fatigue and Decrease of Body Weight-)

  • The studies were conducted to investigate the anti-diabetic activities on the hyperglycemia induced by streptozotocin in rats, anti-hypertensive activities in SHR, anti-fatigue and decrease of body weight activities in mice by Kyung Ok-Ko water extract and drink, which is a traditional preparation in Korea. 1. The blood glucose levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(300, 600, 1200 mg/kg) of Kyung Ok-Ko water extract. Particularly, administration of 600, 1200 mg/kg were significantly shown to decrease glucose levels comparing with control group. 2. The serum total cholesterol levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(300, 600, 1200 mg/kg) of Kyung Ok-Ko water extract. Particularly, administration of 600, 1200 mg/kg were significantly shown to decrease total cholesterol levels comparing with control group. 3. The serum triglyceride levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(300, 600, 1200 mg/kg) of Kyung Ok-Ko water extract. Particularly, administration of 600, 1200 mg/kg were significantly shown to decrease triglyceride levels comparing with control group. 4. The blood pressure in SHRs were dose-dependently lower descended by administration of Kyung Ok-Ko water extract 300, 600, and 1200 mg/kg, respectively. 5. The swimming time of Kyung Ok-Ko water extract(100, 200, 400 mg/kg) and drink(0.7, 1.4, 2.1 ml/kg) were significantly extended to all experimental group dose-dependantly. 6. The decrease of body weight of Kyung Ok-Ko water extract(100, 200, 400 mg/kg) and drink(0.7, 1.4, 2.1 ml/kg) were significantly evaluated dose-dependently in all experimental group.

Effects of Swimming Exercise and Joint Mobilization on HSP 70 Levels in Osteoarthritic Rats

  • Purpose: This study was performed to investigate the effect of joint mobilization on pain relief and cartilage repair in an induced osteoarthritis rat model by analyzing the expression of heat shock protein 70 in articular cartilage. Methods: MIA was injected into SD rats to induce osteoarthritis. These rats were divided into 4 groups: control group (n=30), no further treatment after the MIA injection ; experimental group I(n=30), performed swimming exercise after the MIA injection experimental group II (n=30), underwent joint mobilization after the MIA injection and experimental group III (n=30), performed swimming exercise and underwent joint mobilization after the MIA injection. For the histologic and pathophysiologic evaluation, safranin-O staining and for the immunohistochemical evaluation, the expression of HSP 70 in articular cartilage was analyzed 1, 7, 14, and 21 days after the MIA injection. Results: The inflammatory response and loss of tissue declined in experimental groups I and II over time, whereas the greatest decreases were noted in experimental group III. In the articular cartilage, low expression of HSP 70 was observed in every group on day 1, whereas HSP 70 expression was elevated on days 7 and 14 in experimental groups II and III. After 21 days, experimental group II displayed the strongest positive reaction, whereas HSP 70 was higher in experimental group III at this time point compared to that after 14 days. Conclusion: Our results showed that swimming exercise and joint mobilization had positive effects on pain relief and histologic and functional recovery in an induced osteoarthritis rat model.

작약감초탕의 추출용매에 따른 성분 분석 및 항피로 효능 비교 (Comparison of Ingredient Quantities and Anti-Fatigue Effects of Jakyakgamcho-Tang according to Extraction Solvent)

  • Objectives : Jakyakgamcho-tang has been used as analgesic and spasmolytic for muscle pain. It has reported to anti-diabetes, anti-inflammation, and neuro-protective effects. A clinical study of muscle fatigue and pain improvement effect of Jakyakgamcho-tang has been reported with increasing frequency of clinical use. However, the anti-fatigue effect of Jakyakgamcho-tang in animal model has not been studied. In this study, we compared anti-physical fatigue effects of water and 30% ethanol extract of Jakyakgamcho-tang. Methods : The amounts of components contained in water and 30% ethanol extract of Jakyakgamcho-tang were compared by ultra-performance liquid chromatography. Anti-physical fatigue effects were evaluated using weight-loaded forced swimming test. We also investigated the effects of Jakyakgamcho-tang on the change of fatigue parameters by blood biochemical analysis. Results : The relative amount of components of Jakyakgamcho-tang were about 19-53% higher in the 30% ethanol extract than in the water extract. In the swimming test, 30% ethanol extract showed a significantly greater anti-fatigue effects than conventional water extract of Jakyakgamcho-tang. The 30% ethanol extract of Jakyakgamcho-tang improved the exhausted swimming time (54%), and serum lactate dehydrogenase (48%) and lactic acid (60%) levels compared with water extract of Jakyakgamcho-tang. Conclusions : These results showed that differences in the amounts of components by different extraction were associated with differences of anti-fatigue effect of Jakyakgamcho-tang. Thus, the 30% ethanol extraction method could be applied to dose-reducing formulations of traditional herbal medicines.

서로 다른 형태의 지방산 투여가 훈련된 흰쥐의 지구성 운동수행력, 안정시기와 운동스트레스 시기의 에너지 기질, Insulin 호르몬과 Lipase 활성에 미치는 영향 (The Effects of Different Type of Triglyceride Supplements on Exercise Performance Time, Energy Substrates, Insulin Hormone and Lipase Activity in the Trained Rats)

  • 본 연구는 장기간의 수영훈련을 마친 통제 훈련군, MCT 투여 훈련군 및 LCT 투여 훈련군에서 지구력 운동수행력과 장기간의 훈련 안정시기 및 운동스트레스 부여시기(실험동물이 상당한 스트레스를 느끼는 160분간의 강제수영)에서 에너지 기질의 변화, 인슐린 호르몬, 및 Lipase 효소의 활성화를 함께 비교하여, 만약 투여 훈련군에서 높은 운동수행력이 나왔다면 이것이 어느 에너지 기질 및 피로물질과 연관되는지 혹은 인슐린 호르몬과 지방분해 효소인 Lipase 효소와는 어떠한 연관이 있는지를 규명하여 지구성 운동수행력에 훈련과 투여효과를 과학적으로 검증하는 데에 연구목적이 있었다. 본 연구에서 사용된 실험동물은 평균체중 140-155 g의 Sprague-Dawley계 횐쥐 수컷으로(7 wk), SPF (Specific Pathogen Free: 특정병원체 부재동물)를 대한바이오링크에서 분양 받은 후에 D대학교 실험동물 사육실에서 사육하였다. 실험동물은 총 54마리를 환경적응과 적응훈련을 포함하는 총 9주간의 훈련을 거친 후 크게 세 그룹으로 나누어 비 투여 그룹 18마리, MCT 투여그룹 18마리, LCT 투여그룹 18마리로 구분하였고 각 그룹에서 안정 시기 6마리와 160분간의 운동 스트레스 시기 6마리 및 지구력 운동수행력시기(survival test) 6마리로 세분화 하여 분석하였다. 본 연구의 결과로 일반적인 지방 형태인 LCT의 투여는 추가적인 체중증가를 일으켰고, 운동수행력의 증가도 가져왔다. 하지만 같은 강도의 운동 부하에 대해서 운동 시 활용되는 지방의 형태인 지방산의 동원이 상대적으로 적었고, 운동증료시기에도 많은 양이 TG의 형태로 존재함을 알 수 있었다. 그리고 운동종료 시 피로물질인 젖산의 축적량이 증가하였고, 지방산화 효소인 Lipase 활성도도 크게 증가하지 않으면서 오히려 인슐린 호르몬을 증가시켜 많은 양의 에너지를 제공하여 지구력 운동 수행에는 도움을 주지만 효율성이 떨어짐을 알 수 있었다. 하지만 MCT의 투여는 인슐린 호르몬의 증가를 일으키지 않으면서 lipase 효소의 활성도를 증가시키고, 젖산 축적을 적게 하면서 유리지방산의 동원을 원활하게 하여 운동 지속시간을 상당히 연장했음을 알 수 있었다. 그리고 TG의 축적을 적게 일으키면서 체중의 감소도 나타났다. 이러한 결과를 통하여 장기간 지구성 운동시 추가적인 에너지 공급을 위해 MCT를 투여하는 것이 운동 수행력의 증가는 물론, 에너지 기질의 활성화를 도모하고 피로물질의 축적을 적게 일으켜, 효율적인 에르고게닉에이드로 권장된다. 뿐만 아니라 에너지 효율성을 증가하여 추가적인 지방축적을 일으키지 않기 때문에, 체중증가를 우려할 필요가 없으며, 오히려 많은 지방을 에너지로 사용하기 때문에 에너지 효율성의 면에서 운동만 한다면 다이어트에도 도움이 되리라 생각한다.

우울증 유발 생쥐에서 온담탕가오약(溫膽湯加烏藥)의 항우울 효과 (Anti-Depressive Effects of OnDam-Tang with Addition of Linderae Radix (ODT-L) after Chronic Immobilization Stress in C57BL/6 Mice)

  • The purpose of this study was to examine the anti-depressive effects of OnDam-Tang with addition of Linderae Radix (ODT-L) on the animal model of depression induced by chronic immobilization stress. Depression model was made by chronic immobilization stress for 2 hours for 21 days. And we performed forced swimming test, analysis of the neurotransmitter and immunohistochemical staining, measured expression levels of serotonin in the brain. ODT-L has decreased immobilization time in forced swimming test. ODT-L has increased amount of melatonin in the brain. ODT-L has increased expression levels of serotonin in the brain. ODT-L prevented damage in the hippocampal region. ODT-L has reduced the expression level of CRF receptors in in hippocampus region. These results suggest that ODT-L may have anti-depressive effects on depression.

Effects of Lycium chinense Miller Fruit and its Constituent Betaine on Immunomodulation in Balb/c Mice

  • BACKGROUND: The aim of this study is to investigate the effects of Lycium chinense Miller fruit and its representative constituent betaine on reduction of immobility time and blood parameters in balb/c mice. METHODS AND RESULTS: We investigated the immobility time and the changes in aspects of blood biochemical parameters by the administration of L. chinense Miller fruit and its representative constituent betaine, after the forced swimming test. The immobility time was significantly reduced about 41.3% and 53.6%, respectively, in the animal of L. chinense Miller fruit and its representative constituent betaine-administrated group for 7 days, in comparison with that of the control group. The level of blood urea nitrogen was significantly decreased in L. chinense Miller fruit and its representative constituent betaine-treated group compared with the control group (P < 0.05), respectively. In addition, the interlukin-2 levels of mice in L. chinense Miller fruit and betaine treated group was increased compared with the control group. CONCLUSION: These results indicate that L. chinense Miller fruit and betaine might be helpful in the immune function improvement, enhance physical stamina, and fatigue recovery.

엘리트 수영선수들의 수중 훈련 전후의 상지 근육 특성 변화 분석 (Analysis of Upper Limb Muscles Properties In Elite Swimmers Before and After Training)

  • Objective: The aim of this study is to verify the differences in muscle characteristics of elite level swimmers before and after a 2-hour practice session. Method: The study was conducted on 15 elite swimmers. Preliminary measurements for each muscle (Anterior Deltoid, Triceps Brachii, Biceps Brachii, Flexor Carpi Ulnaris) were taken using the MyotonPRO device before training. After approximately 2 hours of training, the same muscle areas were measured again. The collected data was analyzed through descriptive statistics and two-way 2×2 RG·RM ANOVA, and all statistical significance levels were set at α=.05. Results: After analyzing the characteristics of the Flexor Carpi Ulnaris (FCU) before and after training in both proficiency level swimmers (excellent, non-excellent), it was found that the interaction effect of group X repetition in muscle tension (F), muscle stiffness (S), and body recovery time (R) was statistically significant. Secondly, in the analysis of the Biceps Brachii (BB), the main effect of repetition in muscle tension (F), muscle stiffness (S), and body recovery time (R) was statistically significant. Furthermore, the interaction effect of group X repetition in muscle stiffness (S) and body recovery time (R) was statistically significant. Conclusion: The efficient use of FCU and BB suggests that it is an important factor distinguishing the performance of excellent and non-excellent swimmers in swimming. Therefore, if we develop and apply measures to efficiently utilize FCU and BB during training, it can help improve the performance of the athletes.

Gene Expression Profiles of Long-Chain Acyl-Coenzyme A Dehydrogenase, Nuclear Distribution C-Containing Protein 3, and Receptor Tyrosine Kinase Tie-1 in Swimming Larva of Sea Cucumber Apostichopus japonicus

  • The sea cucumber, Apostichopus japonicus, is one of the most valuable aquatic species. The color of body wall and appearance are important for the value of sea cucumbers. To examine expression pattern of long-chain acyl-coenzyme A dehydrogenase (LCAD), nuclear distribution C-containing protein 3 (NUDCD3), and receptor tyrosine kinase Tie-1 (TIE1), previously reported as differently expressed genes during the pigmentation of sea cucumber, we analyzed the temporal profiles of LCAD, NUDCD3, and TIE1 mRNAs in LED-exposed and light-shielded A. japonicus. Real-time quantitative PCR revealed that the LCAD, NUDCD3, and TIE1 mRNAs from the juveniles at 40-60 days post-fertilization (dpf) exhibited increasing patterns as compared to those of an early developmental larva (6-dpf). At 60-dpf juveniles, the LCAD and TIE1 mRNA levels of LED-exposed individuals were higher than those of light-shielded ones, whereas at 40-dpf and 50-dpf juveniles, the NUDCD3 mRNA expression was higher in the light-shielded condition (p<0.05). In the pigmented juveniles (90-dpf), the LCAD and TIE1 mRNA levels tended to show higher levels in red individuals than those in green ones, but there was a conversely higher level of NUDCD3 mRNA in green larva. In situ examination of LCAD and NUDCD3 mRNAs in light-shielded 6-dpf larva revealed that both genes are mainly expressed in the internal organs compared to the body surface. Together, these results may provide insights into the differential gene expression of LCAD, NUDCD3, and TIE1 during pigmentation process of the sea cucumber.

가능성 식품으로의 활용을 위한 한약자원에 관한 연구 -인삼잎과 줄기 혼합 추출액의 항피로 효과 및 안전성 평가- (Study of Medicinal Herbs for Applications in Functional Foods -Antifatigue Effects and Safety Evaluation of Leaf and Stem Extract of the Panax Ginseng-)

  • This study was conducted to investigate the application possibility of leaf and stem extract(LSE) from the mixture of leaf and stem of ginseng radix(Panax Ginseng CA Meyer). This study measured antifatigue effects by rota-rod test and swimming test. Also we examined the cell toxicity in normal liver and kidney cells, and acute toxicity in mice. Dropping times of LSE treated group decreased about 39-83% compared to the control group according to supplementation intake levels. Swimming time in LSE treated group increased compared to the control group at short and long supplementation. Negative effects were not found from the results of the cell toxicity. Also acute toxicity not shown. These results imply that the leaf and stem of ginseng radix could be used as possible food resources and functional food material and feed stuff.

Enhancement of immune activities of Dioscorea japonica Thunberg in in vivo and ex vivo models

  • Dioscorea japonica Thunberg (DJ) has been widely used as a healthy food in Korea for the enhancement of physical stamina. Hence, the present study evaluated the immune-enhancing effect of DJ in forced swim test of mouse model. The immobility time of the group treated with DJ for 7 days was significantly reduced in comparison with that of the control group. After a forced swimming test, the changes in blood biochemical parameters and splenic T lymphocyte populations induced by the administration of DJ were assessed. Serum levels of lactic dehydrogenase, creatine phosphokinase, and aspartate aminotransferase were significantly decreased in DJ-administered group compared to the control group. However, administration of DJ did not affect the splenic T lymphocyte populations. Moreover, DJ significantly increased the production of interferon-g and interleukin-2 compared to the media control in splenocytes. Collectively, it may be concluded that DJ is useful for enhancement of physical and immune function.
