Back Flushing Behavior of Microfiltration Membrane Fouled by Alumna Colloidal Suspensions (알루미나 현탁액에 의해 오염된 정밀여과막의 역세척 거동)

  • Effect of backflushing on the membrane fouling for polyethylene capillary membranes was examined by measuring the flux of $Al_2O_3$ colloidal suspensions through the cross flow microfiltration. In the comparison of with and without the application of backflushing, the hydraulic resistance to permeate flow of the suspension was less with backfluslng, but the Increasing rate in permeate resistance was higher. Regardless of backflushing, the cake filtration was dominant at the initial period of filtration with backflushing, being followed by the pore blocking. And at steady state, the fouling mechanism is almost governed by the cake filtration model. On the contrary, the pore blocking preceded the cake filtration in the initial stage of the original membrane before backflushing. And irrespective of backflushing, the ratio of cake filtration to total fouling increased, compared with that fur before backflushing. For the membrane with $0.24{\mu}m$ pore size, the permeate resistance was higher than that of $0.34{\mu}m$ pore size membrane. but the ratio of cake filtration was smaller than that of large pore membrane. In comparing the ratio of each fouling component to the total fouling for the case with backflushing pore blocking was 7.8% and cake filtration was 92.2%. for the case without backflushing, total fouling was composed of 9.6% pore blocking and 90.4% cake filtration.

  • An automotive ball joint connects the suspension system to the steering system and helps to enable rotational and linear motion between the two elements for steering. This study examines a ball joint used in medium and large-sized pickup trucks. Ball joints consist of a stud, socket, bearing, and plug. The main structural performance metrics of ball joints are the pull-out strength and push-out strength. These structural parameters must meet certain criteria to avoid serious accidents. Test and simulation methods are used to investigate the design requirements, but tests are time-consuming and costly. In this study, we modeled ball joints in SolidWorks and performed a finite element analysis in Abaqus to predict structural performance. The analysis was used to obtain the structural performance required for the static analysis of a 2D axisymmetric model. The uncertainties in the manufacturing of the ball joint were assumed to be the manufacturing tolerances, and the dimensional design variables were identified through case studies. The manufacturing tolerances at each level were defined, and the results were compared with experimental results.

Experimental Investigation of Thermal Conductivities of EG-based ZnO Nanofluids Manufactured Using Pulsed Wire Evaporation Method (전기선 폭발법에 의해 제작된 에틸렌 글리콜 기반 ZnO 나노유체의 열전도도)

  • In this paper, the thermal conductivities of ethylene glycol. based ZnO nanofluids manufactured using the pulsed wire evaporation method are experimentally measured using the transient hot wire method at temperatures in the range of $10^{\circ}C$ to $50^{\circ}C$. For this purpose, ethylene glycol.based ZnO nanofluids with 1%, 3%, and 5.5% volume fractions were manufactured using the pulsed wire evaporation method. Transmission electron microscopy (TEM) was performed to investigate the suspension stability of the ethylene glycol.based ZnO nanofluids. Based on the experimental results, the thermal conductivities of ethylene-glycol-based ZnO nanofluids increase with increasing volume fractions of ZnO nanofluids. The maximum enhancement of the thermal conductivity is 26.5% for a volume fraction of 5.5% at $22^{\circ}C$. Finally, the experimental results are compared with conventional models such as the Maxwell and Hasselman & Johnson models.

Development of Techniques for Testicular Germ Cell Transplantation in Pigs (돼지에 있어서 정소 생식세포의 이식 기법 개발)

  • The current study was designed to extend the technique of spermatogonial transplantation to economically important pig model We evaluated the efficiency of pig to pig transplantation. Donor testis cells were harvested from testes obtained at castration of 10- to 14-day-old boars and were labeled with fluorescent marker(PKH26) before transplantation. The presence of infused dye or labeled pig testicular cells was confirmed in the seminiferous tubules from recipient pig. The most effective procedure of intratubular germ cell transfer was to insert an fine needle ($21{\sim}25$ gauge) through the cauda epididymis and testis into the rete testis under ultrasound guidance. Infusion of $5{\sim}7ml$ of dye solution or cell suspension could fill the rete and up to 50% of seminiferous tubules of 14-week-old boars. Testis were examined for the presence and localization of labeled donor cells immediately after transplantation and labeled donor cells were found in numerous seminiferous tubules from recipient pig testes. These results indicate that germ cell transplantation is feasible in recipient pig testis. This study represents successful spermatogonial transplantation between individual animals in a livestock species.

Effects of pH and Temperature on the Adsorption of Cationic Dyes from Aqueous Suspension by Maghnia Montmorillonite (수용액으로부터 양이온 염료 흡수에 대한 pH 및 온도 효과)

  • The effects of pH and temperature on the removal of two dyes (neutral red; NR and malachite green oxalates; MG) from aqueous effluents using Maghnia montmorillonite clay in a batch adsorption process were investigated. The results showed the stability of the optical properties of MG in aqueous solution and adsorbed onto clay under wide range of pH 3-9. However, the interaction of NR dye with clay is accompanied by a red shift of the main absorption bands of monomer cations under pH range of 3-5, whereas, those of neutral form remains nearly constant over the pH range of 8-12. The optimal pH for favorable adsorption of the dyes, i.e. ${\geq}$90% has been achieved in aqueous solutions at 6 and 7 for NR and VM respectively. The most suitable adsorption temperatures were 298 and 318 K with maximum adsorption capacities of 465.13mg/g for NR and 459.89 mg/g for MG. The adsorption equilibrium results for both dyes follow Langmuir, Freundlich isotherms. The numerical values of the mean free energy $E_a$ of 4.472-5.559 kj/mol and 2.000-2.886 kj/mol for NR and MG respectively indicated physical adsorption. Various thermodynamic parameters, such as ${\Delta}H^{\circ}$, ${\Delta}S^{\circ}$, ${\Delta}G^{\circ}$ and Ea have been calculated. The data showed that the adsorption process is spontaneous and endothermic. The sticking probability model was further used to assess the potential feasibility of the clay mineral as an alternative adsorbent for organic ion pollutants in aqueous solution.

Characteristics of Flux Decline in Microfiltration Capillary Membrane of Bentonite Colloidal Suspensions (정밀여과 모세관 막을 이용한 벤토나이트 콜로이드 현탁액의 투과유속 감소특성)

  • Permeate flux decline in a microfiltration was analyzed by measuring the permeability of bentonite colloidal solution through polyethylene capillary membranes. The flux decline with time was due to the growth of cake layer on the membrane surface and to the pore blocking by particles. As the time approaches to steady state, the permeate flux is almost controlled by the cake filtration model. Faster flux decline at high trans-membrane pressure was attributed to the formation of denser packed cake layer and pore blocking. The ratio of permeate flux to the initial permeate flux, J/J₁, decreased with increasing the trans-membrane pressure, from 45% for 0.5 kg/sub f//㎠ to 38% for 2.0 kg/sub f//㎠. In comparing the ratio of each fouling component to the total fouling for the 0.5 kg/sub f//㎠ TMP condition, complete blocking was 23.4%, standard blocking was about 14.6% and cake filtration was 62.0%, respectively. Permeate flux through the membrane increases with cross flow velocity, and the effect of the variation of velocity is more significant at 1.0 kg/sub f//㎠ rather than at 2.0 kg/sub f//㎠ of the operation pressure. Permeate flux for the membrane having the average pore diameter of 0.34 ㎛ was higher than that for the membrane of 0.24 ㎛ pore size, with the higher flux with the low concentration of feed. On the operation using the membrane of 0.34 ㎛ pore, the pore blocking in the low concentration of 200 ppm is negligible relative to the pore blocking in the 1000 ppm feed.

Quantitative Analysis of Feline Calicivirus Inactivation using Real-time RT-PCR (Real-time RT-PCR을 이용한 Feline Calicivirus 불활성화의 정량적 분석)

  • Norovirus causes acute gastroenteritis in all age groups and its food poisoning outbreaks are rapidly increasing in Korea. Reverse transcription-polymerase chain reaction (RT-PCR) is most widely used for the rapid detection of foodborne viruses due to high sensitivity. However, the false positive results of RT-PCR obtained against already inactivated viruses could be a serious drawbacks in food safety area. In this study, we investigated a method to yield true positive RT-PCR results only with alive viruses. To decompose the RNA genes from dead viruses, the enzymatic treatments composed of proteinse K and Ribonuclease A were applied to the sanitized and inactivated virus particles. Another aim of this study was to quantify the efficiencies of several major sanitizing treatments using real-time RT-PCR. Feline calicivirus (FCV) that belongs to the same Caliciviridae family with norovirus was used as a surrogate model for norovirus. The initial level of virus in control suspension was approximately $10^4$ PFU/mL. Most of inactivated viruses treated with the enzymatic treatment for 30 min at $37^{\circ}C$ were not detected in RT-PCR, Quantification results to verify the inactivation efficiencies of sanitizing treatments using real-time RT-PCR showed no false positive in most cases. We could successfully develope a numerical quantification process for the inactivated viruses after major sanitizing treatments using real-time RT-PCR. The results obtained in this study could provide a novel basis of rapid virus quantification in food safety area.

Evaluation of Running Performance of the Composite Bogie under Different Side Beam Stiffness (사이드 빔 강성에 따른 복합소재 대차의 주행성능 평가)

  • In this study, a running performance evaluation and roller rig test was conducted to evaluate the applicability of a composite bogie frame, which has the role of the primary suspension. The composite bogie frame was made of a GEP224 glass/epoxy prepreg. Vehicle dynamic analysis was carried out on the composite bogie with three different kinds of side beam thicknesses (50 mm, 80 mm, and 150 mm). From the results, the composite bogie with a side beam thickness of 80 mm satisfied all the dynamic design requirements. Although the composite bogie with the side beam thickness of 50mm also met the design requirements, its critical speed was just a 2% margin to the requirement. In contrast, the model of the side beam thickness of 150mm did not meet the ride comfort. In addition, a composite bogie frame with the side beam thickness of 80 mm was fabricated and installed on a complete bogie. Moreover, the roller rig test using the fully equipped bogie was performed to evaluate the critical speed. During the test, the lateral excitation was imposed on the wheelsets to realize the rail irregularity. There was no divergence of the lateral displacement of the wheelsets while increasing the speed. The measured critical speed was similar to the predicted result.

  • We developed insect-resistant GM rice(Bt transgenic rice) by inserting the mCry1Ac1 a modified gene from the soil bacterium, Bacillus thuringiensis. The Bt transgenic rice expressing the Bttoxin mCry1Ac1 was tested for the effects on survival of Misgurnus anguillicaudatus and Cyprinus carpio, commonly used as a model organism in ecotoxicological studies. M. anguillicaudatus and C. carpio fed 100% ground rice in suspension, using either Bt rice or non-GM counterpart rice(Nakdong). The Bt rice used for the test were confirmed to have the mCry1Ac1 gene expression by the immuno-strip and ELISA analysis. Feeding test showed that no significant differences in cumulative immobility and abnormal response of M. anguillicaudatus and C. carpio fed on between Bt rice and non-GM counterpart rice. The 96hr-LC50 values showed no difference between Bt rice(>1,000mg/L) and non-GM rice(>1,000mg/L). We concluded that there was no significant difference in toxicity for non-target organisms(M. anguillicaudatus and C. carpio) between Bt rice and non-GM counterparts.

Transport Paths of Surface Sediment on the Tidal Flat of Garolim Bay, West Coast of Korea (황해 가로림만 조간대 표층퇴적물의 이동경로)

  • Two-dimensional trend-vector model of sediment transport is first tested in the tidal flat of Garolim Bay, mid-western coast of the Korean Peninsula. Three major parameters of surface sediment, i.e., mean grain size, sorting and skewness, are used for defining the best-fitting transport trend-vector on the sand ridge and muddy sand flat. These trend vectors are compared with the real transport directions determined from morphology, field observation and bedforms. The 15 possible cases of trend vectors are calculated from total sediments. In order to find the role of coarse sediments, trend vectors from sediments coarser than < 4.5 ${\phi}$, (sand size) are separately calculated from those of total sediments. As compared with the real directions, the best-fitting transport-vector model is the "case M" of coarse sediments which is the combined trend vectors of two cases: (1) finer, better sorted and more negatively skewed and (2) coarser, better sorted and more positively skewed. This indicates sand-size grains are formed by simpler hydrodynamic processes than total sediments. Transported sediment grains are better sorted than the source sediment grains. This indicates that consistent hydrodynamic energy can make sediment grains better sorted, regardless of complicated mechanisms of sediment transport. Consequently, both transported vector model and real transported direction show that the source of sediments are located outside of bay (offshore Yellow Sea) and in the baymouth. These source sediments are transported through the East Main Tidal Channel adjacent the baymouth. Some are transported from the subtidal zone to the upper tidal flat, but others are transported farther to the south, reaching the south tidal channel in the study area. Also, coarse sediment grains on the sand ridge are originally from the baymouth, and transported through the subtidal zone to the south tidal channel. These coarse sediments are moved to the northeast, but could not pass the small north tidal channel. It is interpreted that the great amount of coarse sediments is returned back to the outside of the bay (Yellow Sea) again through the baymouth during the ebb tide. The distribution of muddy sand in the northeastern part of study area may result from the mixing of two sediment transport mechanisms, i.e., suspension and bedload processes. The landward movement of sand ridge and the formation of the north tidal channel are formed either by the supply of coarse sediments originating from the baymouth and outside of the bay (subaqueous sand ridges including Jang-An-Tae) or by the recent relative sea-level rise.
