An Improved Selective Isolation of Rare Actinomycetes from Forest Soil

  • Various pretreatment procedures and selective media were applied to assess the optimal conditions for the isolation of rare actinomycetes from soil. Pretreatment of wet-heating for 15 min at 70$^{\circ}C$ and phenol treatment of soil suspension were the most effective methods for the isolation of these microorganisms. Hair hydrolysate vitamin agar (HHVA) was the most suitable medium for the recovery of rare actinomycetes. Thirty-five rare actinomycete strains were chosen using selective isolation approaches, then morphological and chemical properties of the isolates were determined. The isolates belonged to one of the following genus, Micromonospora, Microbispora, Actinoplanes and Streptosporangium.

Cryopreservation of Suspension Cultures of Camptotheca acuminata

  • Supended cells of Camptotheca acuminata were observed to lose their ability to synthesize camptothecin and its derivatives as a result of repeated cultures. Accordingly, the maintenance of high-yield cells by cryopreservation was sudied to overcome this stability problem, and various factors involved were optimized. Pregrowing the cells in 8% myoinositol for 4 days was found to be the most effective in improving survival. The highest survival was obtained when the pregrown cells were cryoprotected with a mixture of 10% DMSO, 0.6M mannitol, and 10% glycerol. When the cryopreserved cells were maintained in a freezer at $-70^{\circ}C$, 94% survival was obtained after 4 months. The survivals after 5 and 8 months of storage decreased to 52% and 45%, respectively. No loss of biosynthetic capacility of camptothecin was observed after short to medium term cryopreservation of C. acuminata.

  • The Light Rail Transit (LRT) System which has medium transport capacity between subway and bus(5,000-25,000 persons per hour) is the most advanced transportation system. It has many benefits, cheap construction, operational costs through driverless and flexible route planning. Also, the rubber tired AGT (K-AGT) of various LRT has a rubber wheels and side guide. The side guide type has an many advantages. but occur a vibration and friction noise through contact between guide rail and wheel. Most of point that decreased comport is vibration thorough the guide contact. In this paper, It is purpose to improve the maximum running speed of rubber tired AGT localization bogie which is currently developed from 70km/h to 80km/h. To satisfy comport index of railway vehicle that is required in performance test. we examined coefficient of bogie suspension which is designed.

  • In this study, the sterilization property of E. coli was established using a plasma generator with a flexible electrode structure. The bacterial suspension was prepared based on the McFarland standard 0.50 (1.5×108 CFU/mL) concentration and a specific amount was inoculated on the plate medium. After the plasma was discharged 3 cm away from the plasma generator in the range of 30s to 5 min and the results compared to the control group, the observed colonies that were formed decreased significantly as the plasma discharge time increased.

  • We report on the development of ferronematic liquid crystals, new materials that consist of a dilute suspension of ferroelectric particles in a nematic liquid crystal host. The particles share their ferroelectric properties with the nematic liquid crystal and impose a spontaneous dielectric polarization of about 10 nC/$cm^2$ to the entire medium, typical for many ferroelectric liquid crystals. As expected, these new materials have enhanced dielectric anisotropy and are sensitive to the sign of an applied electric field. The potential of their use in a number of devices are discussed.

Effect of associating polymer on the dispersion stability and rheology of suspensions

  • Associating polymers are hydrophilic long-chain molecules to which a small amount of hydrophobic groups (hydrophobes) is incorporated. In aqueous solution, the association interactions result in the formation of three-dimensional network through flowerlike micelles at high concentrations. In colloidal suspensions, the associating polymers act as flocculated by bridging mechanism. The rheological properties of suspensions flocculated by associating polymers end-capped with hydrophobes are studied in relation to the bridging conformation. At low polymer concentrations, the polymer chains effectively form bridges between particles by multichain association. The suspensions are highly flocculated and show typical viscoelastic responses. When the polymer concentration is increased above the absorbance at saturation, the excess polymer chains remaining in the solution phase build up three-dimensional network by associating interactions. Since the presence of particles does not significantly influence the network structures in the medium, the relative viscosity, which gives a measure of the degree of flocculation is decreased with increasing polymer concentration. The bridging conformation and flocculation level vary strongly depending on the polymer concentrations.

Effect of Pluronic F-68 and Oxygen Vectors on Postthaw Growth of Cryopreserved Transgenic Nicotiana tabacum

  • Effects of supplementing various 0.1-1.0%(w/v) surfactants and 0.1-1.0%(v/v) oxygen vectors on postthaw growth following cryopreservation of transgenic Nicotiana tabacum(GUS) in suspension culture were investigated. The postthaw cell growth was increased 20.68% and 23.66% compared to control when Pluronic F-68 and polyethylene glycol(PEG) were added in recovery medium respectively. Addition of n-hexadecane and FC-40 also enhanced the recovered growth by 14.89% and 20.68%. These results demonstrate that the marked Protection could be achieved by using surfactants and oxygen vectors for plant cells recovered from cryostorage.

Biodegradation of Hydrocarbons by an Organic Solvent-Tolerant Fungus, Cladosporium resinae NK-1

  • A kerosene fungus of Cladosporium resinae NK-1 was examined for its ability to degrade individual n-alkanes and aromatic hydrocarbons by gas chromatography-mass spectrometry, and its organic solvent-tolerance was investigated by making use of the water-organic solvent suspension culture method. It grew on a wide range of solvents of varying hydrophobicities and it was found to have tolerance to various kinds of toxic organic solvents (10%, v/v) such as n-alkanes, cyclohexane, xylene, styrene, and toluene. A hydrocarbon degradation experiment indicated that NK-1 had a greater n-alkane degrading ability compared to that of the other selected strains. C. resinae NK-1, which could utilize 8-16 carbon chain-length n-alkanes of medium chain-length as a carbon source, could not assimilate the shorter chain-length n-alkanes and aromatic hydrocarbons tested so far. The n-alkane degrading enzyme activity was found in the mycelial extract of the organism.

  • Production of a cardiac glycoside, digoxin, by 12$\beta$-hydroxylation from digitoxin was studied in plant cell suspension cultures of Digitalis lanata. In order to increase the conversion yield, various culture conditions including immobilization were investigated and optimized. Since digoxin was released in the medium temporarily and converted further into a glucosylated product, deacetyllanatoside C, in situ adsorption of digoxin was employed to recover the product continuously. Amberlite resin XAD-8 showed the best adsorption characteristics for digoxin among the examined resins, and an integrated process was developed to increase the productivity. In addition, it was found that the utilization of $\beta$-cyclodextrin to entrap digoxin during the culture enhanced the biotransformation yield significantly.

  • In this paper, the semi active mount featuring Magneto rheological fluid(MR Fluid) is proposed. MR fluid is suspension of micro sized magnetizable particles in a fluid medium, and its apparent viscosity can be varied by the applied strength of magnetic field. When the controllable MR fluid is applied to mechanical devices, the devices provide simple, rapid response interfaces between electronic controls and mechanical systems. The MR fluid is applied in the conventional fluid mount for improving its performance of the mount's isolation effect. A appropriate size of the MR mount is designed and manufactured on the basis of Bingham model of MR fluid. In addition, the field dependent damping forces of MR mount are evaluated with respect to the input frequency variation.
