Serum Cholesterol Lowering Effects of the Phytosterol Derivative (LPSS) in Rats (랫드에서 식물성스테롤 유도체(LPSS)의 혈중 콜레스테롤 저하효과에 관한 연구)

  • The present study was designed to investigate the serum cholesterol lowering effect oj the phytosterol derivative (LPSS) on high cholesterol (HC) diet-induced hypercholesterolemia in male weaning Sprague-Dawley (SD) rats. Rats were fed with HC diet containing 1 % cholesterol and 0.5% cholic acid for 1 week. After 1 week, the LPSS oil suspension (0.32 g/kg B. W.) was orally administered to the rats fed with either basal diet or HC diet groups for 7 days. In addition, the LPSS powder (0.14%) mixed with basal diet or HC diet was Jed to the rats for 7 days. Serum total cholesterol and LDL-cholesterol contents were not altered by administration of the LPSS oil suspension with basal diet. However, they were significantly decreased by administration of the LPSS oil suspension with HC diet at day 14. Also, they were significantly decreased by the LPSS powder mixed with basal diet or HC diet at day 9, 11, 14. HDL-cholesterol contents were not altered by the LPSS oil suspension or LPSS powder. These results indicated that the phytosterol derivative(LPSS) might decrease serum total cholesterol and LDL-cholesterol contents in rats.

Effect of Exercise Training following Hypokinesia on the Length and Circumference of Atrophied Soleus and Plantaris Muscle in Rats (활동저하후 회복기의 운동훈련이 쥐의 위축근의 길이와 둘레에 미치는 영향)

  • The purpose of this study was to determine the effect of exercise training on the length and circumference of atrophied muscles to see whether improvement in restoring the atrophied muscle mass, of either length or circumference growth could be induced through exercise training. Adult female Wistar rats were maintained for 14 days with hindlimb suspension. Rats were then assigned randomly to a sedentary group or training group. The length and circumference of both atrophied soleus and plantaris muscle for the training and sedentary groups following hindlimb suspension were compared with those of a control group. Length and circumference of both atrophied soleus and plantaris muscle in trained group was compared with those in the sedentary group. The results can be summarized as follows ; 1) Atrophy of soleus and plantaris muscle was induced with hindlimb suspension. 2) The circumference of both the soleus and plantaris muscle following hindlimb suspension was reduced to 51. 72%, 66.67% respectively compared to the control group. The length of both the soleus and plantaris muscle following hindlimb suspension did not show any difference compared to the control group. 3) There was no difference in length for either the atrophied soleus or plantaris muscle between the trained and the sedentary group during recovery from hindlimb suspension. 4) The circumference of both the atrophied soleus and plantaris muscle did not return to normal in the sedentary group, however, the circumference of both the atrophied soleus and plantaris muscle were significantly greater than of the control in the trained group, 14.22% and 9.38% respectively. 5) The circumference of both the atrophied soleus and plantaris muscle in the trained group was significantly larger than that of both muscles in the sedentary group at day 28 of recovery. From these results, it can be suggested that improvement in restoring the atrophied muscle mass through exercise training might be induced by growth of the muscle circumference.

  • The purpose of this study was to determine the amount of soleus muscle atrophy in rats due to 28 days of decreased activity induced by hindlimb suspension, to observe the restoration of mass and relative muscle weight of the atrophied soleus muscle at day 28 of recovery to control value, and to compare the effect of run training on the mass and relative muscle weight of the atrophied soleus muscle at day 28 of recovery with that of sedentary rats. Adult female Wistar rats were maintained for 28 days with hindlimb suspension. Rats were then assigned randomly to a cage sedentary or running group. Soleus muscle mass and relative muscle weight following hindlimb suspension were compared with a control value. The soleus muscle mass and relative muscle weight of the running and cage sedentary groups following hindlimb suspension were compared with those of a control group. Soleus muscle mass and relative muscle weight of the run training group were compared with those of cage sedentary group. The results obtained were as follows : 1. Soleus muscle mass and relative muscle weight was reduced to 53.28% and 51.11% respectively by hindlimb suspension. 2. Soleus muscle mass and relative muscle weight of the training group t day 28 of recovery was restored to the control value. 3. Soleus muscle mass of the training group was greater than that of the cage sedentary group by 6.6% without statistical significance at day 28 of recovery. 4. Relative soleus muscle weight of the traning group was significantly greater than that of the cage sedentary group by 15.79% From these results, it may be concluded that run training during the posthypokinetic period facilitates the recovery of the atrophied soleus muscle mass of rats.

The Effects of Preoperative Sprayed 10% Lidocaine on the Hemodynamic Response during Suspension Microlaryngeal Surgery (술 전 분무한 10% lidocaine이 현미경 하의 후두 미세 수술 시 혈역학적 반응에 미치는 영향)

  • Purpose : It is well known that suspension microlaryngeal surgery produces marked increases in arterial blood pressure and heart rate. In this study, we evaluated the effects of 10% lidocaine preoperatively sprayed for attenuation of the perioperative hemodynamic response during suspension microlaryngeal surgery. Materials and Methods : Fifty American Society of Anesthesiologists (ASA) class 1 patients scheduled for excision of a vocal polyp by suspension laryngoscopy were randomly divided into two groups (n=25 for each group). They were intubated without 10% lidocaine spray (control group) or given 1.5 mg/kg of 10% lidocaine sprayed onto the pharyngolaryngeal and intratracheal sites 90 sec prior to intubation (10% lidocaine group). Anesthesia was maintained using desflurane in $O_2/N_2O$ 50%. The arterial blood pressure and heart rate were measured at preinduction (T0), 1 min (T1), 3 min (T2), 5 min (T3) after tracheal intubation, and 1 min (T4), 3 min (T5), 5 min (T6) and 10 min (T7) after the suspension laryngoscopy. Results : In the 10% lidocaine group, the arterial blood pressure and heart rate at 1 (T1), 3 (T2) min after tracheal intubation and 1 (T4), and 3 (T5) min after suspension laryngoscopy were lower than the same measurements in the control group. Conclusion : 10% lidocaine sprayed onto the pharyngolaryngeal and intratracheal sites before intubation was an effective method for attenuation of the perioperative hemodynamic response during suspension microlaryngeal surgery.

Effects of Intermittent Sciatic Nerve Stimulation on the Soleus and Medial Gastrocnemius Muscle Atrophy in Hindlimb Suspended Rats

  • The present study was designed to evaluate effects of intermittent electrical stimulation of the sciatic nerve on the atrophic response of antigravity muscles, such as the soleus (slow m.) and medial gastrocnemius (fast m.) muscles. Rats (Sprague-Dawley, 245-255g) were subjected to a hindlimb suspension and divided into three groups : one was with hindlimb suspension (MS) and another with hindlimb suspension plus intermittent electrical stimulation of the sciatic nerve (HS ES). Control group (CONT) was kept free without strain of the hindlimb. After 7 days of hindlimb suspension, the soleus and medial gastrocnemius muscles were cut at their insertion sites, and were then connected to the force transducer to observe their mechanical properties. Optimal pulse width and frequency of electrical stimulation were 0.2ms, 20Hz for the soleus muscle and 0.3ms, 40Hz for the medial gastrocnemius muscle under supramaximal stimulation. Body weight and circumference of the hindlimb were significantly decreased in HS and HS-ES groups compared with the control group. In HS-ES group, however, the weight of the soleus muscle was not different from that in the control group while the weight of the medial gastrocnemius muscle was lower than that in the control group. In HS group, mechanical properties of muscle contraction including contraction time, half relaxation time, twitch tension, tetanic tension, and fatigue index of both muscles were significantly decreased compared with the control group except for twitch tension and tetanic tension of medial gastrocnemius muscle. The degree of atrophy of the soleus muscle in HS group was more prominent than that of the medial gastrocnemius muscle. Twitch tension and fatigue index of the soleus muscle and fatigue index of the medial gastrocnemius muscle in HS-ES group were not different from those of the control group. While mechanical properties of the soleus muscle examined were all significantly increased in HS-ES group compared with HS group, only contraction time and fatigue index of the medial gastrocnemius muscle were significantly increased in HS-ES group. These data indicate that intermittent electrical stimulation may be useful in prevention of muscle atrophy.

The Clinical Study on the Effect of the Sulguanjul-bang No.1 According to Formulation (퇴행성 슬관절염 환자에 대한 슬관절방(膝關節方) 1호(號)의 제형변화 유효성 임상연구)

  • Objective: This study is designed to compare the effects of Sulguanjul-Bang No.1 on osteoarthritis in knee joint according to formulation. Methods: 47 patients with symptomatic osteoarthritis of the knee joint were enrolled in a open clinical trial. The 47 patients were devided into two groups at random and taken Sulguanjul-bang No.1 with decoction type, suspension type respectively for8 weeks. After 8-week treatment period, the following parameters were analyzed ; Visual analogue Scale(VAS), Lysholm index score, patients' global assessment. Results: Sulguanjul-bang No.1 treatment led to significant improvement in the pain and symptoms of osteoarthritis as determined by all efficacy measures. After 8 weeks of therapy, there was significant improvement in VAS, Lysholm index and patients' global assessment. But there is no significant difference between two groups. Conclusions: Sulguanjul-bang No.1 was very beneficial pain relief effect and functional improvement irrespect of formulation, decoction type or suspension type.

Experimental study of antigenicity test of Sweet Bee Venom in Guinea Pigs (기니픽을 이용한 Sweet Bee Venom의 항원성 평가)

  • Objectives: This study was performed to examine the antigenic potential of pure melittin (Sweet Bee Venom - SBV) extracted from the bee venom by utilizing protein isolation method of gel filtration. Methods: All experiments were conducted at Biotoxtech (Chungwon, Korea), authorized a non-clinical studies institution, under the regulations of Good Laboratory Practice (GLP). Antigenic potential of SBV was examined by active systemic anaphylaxis (ASA) and passive cutaneous anaphylaxis (PCA) in guinea pigs. SBV was subcutaneously administered at 0.07 and 0.28mg/kg and also as a suspension with adjuvant (Freund's complete adjuvant: FCA). Ovalbumin (OVA) as a suspension with adjuvant was used to induce positive control response ($5mg/m{\ell}$-FCA). Results: 1. In the ASA test, experimental groups showed some symptoms of anaphylaxis like piloerection, hyperpnea and staggering gait. 2. In the PCA test, low dosage group did not show any antibody responses, whereas high dosage group showed positive responses. 3. In the weight measurement and clinical observation, experimental groups didn't show any significant changes compared with control group. 4. In the autopsy of body, the abnormalities of lung were detected in the corpse. This means that the cause of death may induced anaphylactic shock. Conclusions: Above findings suggested that SBV had antigenic potential in guinea pig. Further studies on the subject should be conducted to yield more concrete evidences.

  • Purpose: The purpose of this study is to compare the adhesion of Candida albicans according to build angle in 3D printing denture base resin. Methods: The 3D printing was performed by setting the build angle of the disk type specimen designed by CAD program at 0 degree, 30 degrees, 60 degrees, and 90 degrees. Surface roughness was measured using a non-contact 3D microsurface profiler. The specimens were incubated in Candida albicans suspension for 24 hours. The attached Candida albicans were detached by cell scraper. The suspension of detached C. albicans was serially diluted and plated on Trypticase soy broth. After 48 hours of incubation, total colony forming unit was counted. Results: There was no significant difference in surface roughness(Sa) between the test groups, but the interlayer boundary was observed. There was no statistically significant difference in total colony forming units of Candida albicans between the test groups. Conclusion: There was no difference in the average surface roughness and adhesion of Candida albicans between the specimens. It is considered that the setting of the build angle should be set considering the accuracy or strength rather than the roughness of the surface.

  • In order to clarify the morphological changes in the mammary glands of cows affected with coliform (Escherichia coli) mastitis, histopathological observations were undertaken on the mastitis of the lactating rabbits which was experimentally induced with E. coli or its endotoxin isolated from cases of acute and chronic matitis in dairy cattle. In the bacterial suspension-infused groups the affected quarters of udder showed cloudy swelling, hyperemia and hemorrhage to local necrosis and firmness. The microscopic findings of early stage of the mastitis were appearance of large numbers of heterophils in the glandular lumina and ducts accompanied by degeneration, necrosis and desquamation of epithelial cells, and also infiltration of heterophils, hemorrhage and edema in the interstitial tissue, and destruction of alveoli. Later, proliferation of firoblasts, plasma cells, lymphocytes, eosinophils and histiocytes appeared in the glandular tissue and necrotic foci of glandular tissue were surrounded by highly proliferated connective tissue. Granuloma-like inflammatory changes could be observed in the glandular tissue on the 7th days after infusion. The inflammatory response in the group infused with E. coli strain isolated from the natural case of acute mastitis was rapid and severe as compared with that of chronic mastitis. In the endotoxin-infused group the morphological changes were similiar to those of the bacterial suspension-infused groups.

A study on the current status and perioperative management of antithrombotic in a general hospital

  • The purpose of this study is to investigate the actual status of antithrombotic management before and after the procedure or surgery, the difference between the duration of medication suspension by clinical and demographic characteristics, and the patient's understanding and satisfaction after medication management by a dedicated nurse. The results were as follows. The most commonly used antithrombotic agents were aspirin and flavitol. The drug discontinuation period according to antithrombotic, procedures, and underlying diseases, there was a significant difference in duration for each variables(p<.000). In the case of antiplatelet drugs, 5-day suspension was the most frequent, and anticoagulants 2-day suspension was the most frequent. Depending on the procedure,colonoscope,nucleoplasty,rotator cuff repair,and total knee arthroplasty commonly showed more than 80% of 5-day discontinuation. The differences according to underlying diseases are as follows. 64.7% of all diseases discontinued on the 5th. The patient's understanding of the nurse's medication management performed before and after the procedure was found to be lower in Angina patients than those with other diseases. In terms of age, those in their 50s showed higher understanding than other age groups. There were no differences in understanding and satisfaction with the remaining characteristics.
