Development of Bacteria for Removal of the Nitrogen in Wastewater (하ㆍ폐수 고도처리를 위한 다기능의 질소원 분해능 균주의 분리)

  • Ninety strains of photosynthetic bacteria were isolated from a local stream at Kyonggi-do, Korea and were further screened. Using these isolated strains, experiments were performed under various light and oxygen conditions in order to select strains with high nitrogen $(NH_3-N,\; NO_3^--N)$ removal efficiencies. Results showed that all the strains screened removed $NH_3-N$, the light had no effect on nitrogen removal, and the nitrogen removal rate was higher aerobically than anaerobically. The removal of $NO_3^--N$ was showed up to 35.3% in some specific strains. Results of batch experiments using Rhodocyclus gelatinosus, an isolated strain with a superior removal rate of $NH_3--N$ and $NH_3-N$, under the anaerobic condition, showed that the removal rate of organics and $NH_3-N$ was the highest (98.2 and 89.0%, respectively) at the CODcr (mg/L)/biomass (mg/L) ratio of 0.2, and the $NH_3-N$ concentration did not increase with the decreasing $NH_3-N$ concentration. Experimental results from various C/N ratios confirmed that the effective removal rate (75.8%) of $NH_3-N$ occurred even at the low (5:1) C/N ratio as well as high ratios, and the simulataneous removal of $NO_3^--N$ (96.0%).

Characteristics of Red Wines Fermented with Different Strains of Dry (건조 효모와 포도 품종에 따른 적포도주의 발효 특성 변화)

  • This study investigated the effects of 3 different strains of my yeast (Saccharomyces cerevisiae Montrachet (UCD #522), S. bayanus Pasteur Champagne (UCD #595), and S. cerevisiae Epernay (CEG) and two grape varieties (Tamnara and M.B.A) on the quality of red wines. During fermentation, sugar content decreased and alcohol content increased Addition of my yeast increased fermentation speed for both grape varieties. Sensory quality tests and tests with an electronic tongue showed that red wine made from M.B.A grapes was superior to that made from Tamnara grapes and that red wine made without the addition of my yeast was better than that made by using dry yeast.

Changes in Quality Characteristics of Cheongkukjang added with Deodeok (더덕이 첨가된 청국장의 품질특성 변화)

  • In order to demonstrate the usefulness of Deodeok. the functions of Deodeok and the qualities of Cheongkukjang were investigated. DPPH (1.1-diphenyl-2-picryl-hydrazyl) radical scavenging activity of Deodeok extracted with 70% ethanol was higher than that of water extract. The direct antimutagenic effect of ethanol extract of Deodeok was examined by Ames test using Salmonella typimurium TA 98. The inhibition rates on ethanol extract at concentrations of 200, 1.000, 2.000, 3.000 and 4.000 ${\mu}g$/plate were 5.75, 31.38, 34.75, 53.50 and 83.75%, respectively. The inhibition rates of ethanol extract was higher over 2 times than that of water extract. The qualities including physicochemical and sensory properties of Deodeok Cheongkukjang were investigated over the following range of Deodeok levels; 5, 10, 15 and 20% (w/w). The strain used in Cheongkukjang manufacture was Bacillus sp. B-3 with the highest enzyme activities such as amylase and protease. During fermentation at $40^{\circ}C$ for 6 days. amino-type nitrogen content of Cheongkukjang containing 10% (w/w) Deodeok was more than others. When Deodeok content exceeded 15% (w/w), higher contents resulted in lesser amino-type nitrogen production. The results showed that Deodeok had influenced the growth of Bacillus sp. B-3. The L-value of Deodeok Cheongkukjang was decreased according to increasing the Deodeok contents. Sensory evaluation showed that Deodeok Cheongkukjang containing 10% (w/w) Deodeok was superior to other tested.

Preparation and Properties of Coimmobilized Glucose Oxidase-Catalase (Glucose Oxidase와 Catalase의 동시 고정화 제품과 성질)

  • For the study of glucose oxidase(GOD) and catalase(CAT) coimmobilization system, the enzymes were obtained from Penicillium spp., PS-8, and the strain itself was used as an immobilizing matrix. To separate glucose oxidase and catalase after the ammonium sulfate fractionation of the culture broth, DEAF-cellulose column was used and its activity yield was 54 and 34%, respectively. Both enzymes were immobilized on the cell matrix, followed crosslinking with 2.5% glutaraldehyde for 12hr. In the determination of efficiencies of GOD and CAT of dual, mixed and soluble enzyme systems, the dual immobilized one w-as superior to those of the soluble or mixed ones. In the comparison of pH profiles, the dual and mixed types showed broader maximum pH ranges than the soluble type. Varying CAT/GOD ratio of the dual system, the higher the ratio showed the broader activity profile. In the comparison of apparent $K_m$ of GOD only and CAT/GOD=10, they were $7.1{\times}10^{-2}$ and $5.1{\times}10^{-2}M$. Their activation energies showed 3.98kcal/mole/deg for GOD only and 2.98kcal/mole/deg for CAT/GOD=10.

Field Performance and Morphological Characterization of Transgenic Codonopsis lanceolata Expressing $\gamma-TMT$ Gene.

  • Field performance and morphological characterization was conducted on seven transgenic lines of Codonopsis lanceolata expressing ${\gamma}-TMT$ gene. The shoots were obtained from leaf explants after co-cultivation with Agrobacterium tume-faciens strain LBA 4404 harboring a binary vector pYBI 121 that carried genes encoding ${\gamma}-Tocopherol$ methyltransferase gene (${\gamma}-TMT$) and a neomycin phosphotransferase II gene (npt II) for kanamycin resistance. The transgenic plants were transferred to a green house for acclimation. Integration of T-DNA into the $T_0\;and\;T_1$ generation of transgenic Codonopsis lanceolata genome was confirmed by the polymerase chain reaction and southern blot analysis. The progenies of transgenic plants showed phenotypic differences within the different lines and with relative to control plants. When grown in field, the transgenic plants in general exhibited increased fertility, significant improvement in the shoot weight, root weight, shoot height and rachis length with relation to the control plants. However, all seven independently derived transgenic lines produced normal flower with respect to its shape, size, color and seeds number at its maturity. Indicating that the addition of a selectable marker gene in the plant genome does not effect on seed germination and agronomic performance of transgenic Codonopsis lanceolata. $T_1$ progenies of these plants were obtained and evaluated together with control plant in a field experiment. Overall, the agronomic performance of $T_1$ progenies of transgenic Codonopsis lanceolata showed superior to that of the seed derived non-transgenic plant. In this study, we report on the morphological variation and agronomic performance of transgenic Codonopsis lanceolata developed by Agrobacterium transformation.

Effects of Composite Floor Slab on Seismic Performance of Welded Steel Moment Connections (철골모멘트 용접접합부의 내진성능에 미치는 합성슬래브의 영향)

  • Traditionally, domestic steel design and construction practice has provided extra shear studs to moment frame beams even when they are designed as non-composite beams. In the 1994 Northridge earthquake, connection damage initiated from the beam bottom flange side was prevalent. The upward moving of the neutral axis due to the composite action between steel beam and floor deck was speculated to be one of the critical causes. In this study, full-scale seismic testing was conducted to investigate the side effects of the composite action in steel seismic moment frames. The specimen PN700-C, designed following the domestic connection and floor deck details, exhibited significant upward shift of the neutral axis under sagging (or positive) moment, thus producing high strain demand on the bottom flange, and showed a poor seismic performance because of brittle fracture of the beam bottom flange at 3% story drift. The specimen DB700-C, designed by using RBS connection and with the details of minimized floor composite action, exhibited superior seismic performance, without experiencing any fracture or concrete crushing, almost identical to the bare steel counterpart (specimen DB700-NC). The results of this study clearly indicate that the beams and connections in seismic steel moment frames should be constructed to minimize the composite action of a floor deck if possible.

Verification of Antibacterial Activities of Oriental Herbal Medicine Extracts (한약재 추출물의 항균활성 검증)

  • The antibacterial activities of nine oriental herbal medicine extracts were measured against seven kinds of bacteria known to cause food poisoning and disease. The extracts were prepared with distilled water using an autoclave ($121^{\circ}C$ for 30 min). The extraction yields of nine oriental herbal medicines were in the rage of 0.5%-33.4%. The antibacterial activities were evaluated using the paper-disc method. Extracts of Hwangryeon, Jiyu, and Ohbaeja showed antibacterial activities while the others did not. Extracts of Coptis japonica, Hwangryeon, and Sanguisorba officinalis, Jiyu showed antibacterial activities only against Clostridium perfringens and Listeria monocytogenes. Extracts of Rhus javanica, Ohbaeja showed the strongest antibacterial activities against all the bacteria tested. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of R. javanica extracts were in the range of 2.4~20 mg/ml for the bacteria tested. In conclusion, the extract of R. javanica showed superior antibacterial activities against the seven bacterial strains tested. The MIC and MBC values for each bacterium were represented as strain names (MIC, MBC), which were Bacillus cereus (11.2, 20), Clostridium perfringens (2.4, 4.7), Escherichia coli (9.1, 9.1), Listeria monocytogenes (11.1, 20), Salmonella typhi (4.7, 9.1), Staphylococcus aureus (2.4, 4.7), and Vibrio parahaemolyticus (2.4, 5.3). Further purification and study of this antibacterial material would be helpful for developing antibiotics or promoting synergistic effects with known antibiotics, producing antibacterial material for lengthening the shelf-life of food, and so on.

Probiotic Properties and Immunomodulator Evaluation of the Potential Feed Additive Pediococcus acidilactici SRCM102607 (잠재적 사료첨가제로서 Pediococcus acidilactici SRCM102607의 생균제 특성 및 면역활성 효과)

  • The purpose of this study was to investigate the probiotic characteristics and immune activities of selected lactic acid bacterial (LAB) strains as feed additives in livestock. 301 LAB strains isolated from traditional fermented foods were first assessed for their antibacterial activity potential. Of the 301 isolates, five showed antibacterial activity against five livestock pathogens (Esherichia coli KCCM11234, Listeria monocytogens KCTC3710, Salmonella Typhimurium KCTC1926, Staphylococcus aureus KCCM11593, and Shigella flexneri KCTC2517). The probiotic characteristics of the five selected strains were also investigated by antioxidative activity, hemolysis, bile salt hydrolase, acid resistance and bile tolerance. The SRCM102607 strain was found to have superior probiotic properties and was selected for further experimentation. 16S rRNA gene sequencing showed that SRCM102607 is Pediococcus acidilactici, which was labeled as P. acidilactici SRCM102607 (KCCM 12246P). The survival characteristics of P. acidilactici SRCM102607 in artificial gastrointestinal conditions were assessed under exposed acidic (pH 2.0) and bile (0.5% and 1.0%) conditions. P. acidilactici SRCM102607 was also confirmed to have resistance to various antibiotics, including amikacin, gentamicin, vancomycin, and etc. The TNF-α production by P. acidilactici SRCM102607 was 171.86±4.00 ng/ml. These results show that P. acidilactici RCM102607 has excellent potential for use as a probiotic livestock feed additive.

Mucosal Mast Cell Responses in the Small Intestine of C3H/HeN and BALB/c Mice Infected with Echinostoma hortense

  • In the intestinal mucosa, mast cells are thought to be responsible for the expulsion of parasites. We investigated the relationship of worm expulsion and mast cells in C3H/HeN and BALB/c mice infected with Echinostoma hortense. In addition, we examined whether the worm recovery rate was associated with the strain of mice, and whether a toluidine stain and immunohistochemistry using the c-kit antibody was effective in the detection of mast cells. In order to investigate the mucosal immune response of C3H/HeN and BALB/c mice, each mouse was infected orally with 30 E. hortense metacercariae. Then, the number of mucosal mast cells and worm recovery rates was observed in experimentally infected mouse strains between 1 week and 8 weeks post infection (PI). Mucosal mast cells were increased in 3 weeks P.I. in C3H/HeN and BALB/c mice. On the other hand, only mucosal goblet cells and worm recovery rates correlated in C3H/HeN mice (P=0.0482). Worm recoveries in C3H/HeN mice were 65.7$\pm$5.6, 53.3$\pm$5.4 and 6.7$\pm$0.6 in week 1, 2, and 3 P.I. and strongly decreased in week 3 P.I. Worm recoveries in BALB/c mice were 23.0$\pm$2.5, 10.0$\pm$1.0, and 6.7$\pm$0.6% in week 1, 2, and 3 P.I. and gradually decreased from week 1 P.I. to week 3 P.I. Worm recoveries in C3H/HeN mice were significantly higher than in BALB/c mice (P<0.00l). The number of mast cells in C3H/HeN and BALB/c mice using the anti-c-kit antibody reached to a peak in week 3 P.I. and recovered as normal level in week 5 P.I. and 6 P.I. The number in E. hortense-infected C3H/HeN mice (P=0.0015) was higher than in E. hortense-infected BALB/c mice (P=0.01) compared with the control group. There were significant differences in the number of mast cells among regions of the intestine in in C3H/HeN mice (P<0.05) but not in BALB/c mice (P>0.05). Immunohistochemistry using the anti-c-kit antibody was significant method as an examination of the number of mast cells (P=0.0002). In conclusion, the present study demonstrated that mast cells play an important role in worm recovery, and immunohistochemistry using the anti-c-kit antibody was superior to toluidine stain as an examination of mast cells.

Studies on Formation of Organic Acid and Saccharifing Amylase in Koji Culture by Asp. usamii shirousamii $U_2$ (Asp. usamii shirousamii $U_2$ 균의 국식배양에 의한 유기산 및 당화효소 생성에 관한 연구)

  • This experiment was carried out to investigate the producing conditions of organic acid and saccarifing amylase in Koji culture by Asp. usamii shirousamii $U_2$. The results were as follows. 1. When the strain $U_2$ was incubated at $30^{\circ}C$, for 3 days in wheat flour and wheat bran media, the organic acid production was maximum. In the case of incubation at $35^{\circ}C$, for 3 days in wheat flour medium and at $35^{\circ}C$, for 2 days in wheat bran medium the activity of saccharifing amylase was highest. 2. When water was added 60% to wheat flour and 50% to wheat bran in the case of 3 days incubation, the organic acid production was superior. Both in wheat flour and wheat bran media, the saccharifing amylase production was most highly, when water was added 90-100%. 3. Comparatively speaking, the organic acid production was better in wheat flour medium than wheat bran medium, but the activity of saccharifing amylase was higher in wheat bran medium. 4. When the sweet potato starch waste and the wheat flour were mixed with same amount, the organic acid and saccharifing amylase production were higher than in simple wheat flour medium. 5. In the medium of sweet potato starch waste the organic acid and saccharifing amylase production were low extremely. 6. In the case of incubation at $30^{\circ}C$, 3 days in wheat flour medium admixed with 60% water, the amount of citric acid in the organic acid formed was about 91%.
