The Degradation of Phenolic Compounds by Lignolytic Streptomyces strains (Lignin을 분해하는 Streptomyces strainsdop 의한 페놀화합물의 분해)

  • The purpose of this was to investigate the degradation efficiency of phenol compounds(catechol, ferulic acid, protocatechuic acid, syringic acid, vanillic acid) by Streptomyces halstedii scabies SAI-36, Streptomyces avendulas SA2-14, and Strptomyces badius(ATCC 39117, control group). The results were as follows: Catechol showed the degradation efficiency that is lower than 50% in three strains. Ferulic acid and vanillic acid showed high degradation efficiency of 98.8% and 94.5% respectively by Streptomyces lavendulas SA2-14. protocatechuic acid and syringicacid showed high degradation efficiency of 89.6% and 77.9%. The degradation efficiency of catechol by Streptomyces halstedii scabies SAI-36, Streptomyces lavendulas SA2-14 and Streptomyces badius(ATCC 39117) was low as 49.2%, 40.2% and 20.2% respectively. But the degradation of other phenolic compoumds except catechol by Streptomyces laven-dulas SA2-36 and Streptomyces badius(ATCC 39117). The results demonstrated that two experimental strains are superior ability to control group in degradation of phenol compounds and Streptomyces lavendulas SA2-14 was superior of two experimental strain. This results were consistent with previous research results that Streptomyces lavendulas SA2-14 was the best strain in degradation ability for lignin, decoloration abilities for variousdyes, and various enzyme production abilities. Therefore, it is suggested that lignin can be used as a indicator when selecting Actinomycetes for degradation of non-degradable materials such as phenol compounds.

The Suitable Condition for Mycelial Growth of Volvariella volvacea strains and Selection of the Superior strain (풀버섯의 균사배양 적합조건 및 우량균주 선발)

  • This study was carried out to obtain basic data on mycelial growth characteristics for an artificial cultivation of Volvariella volvacea. Twelve URP primers were used to assess genetic relationship of V. volvacea and its result was divided into two groups. But there was nothing different in morphological characteristics in V. volvacea. Among five kinds of mushroom media, MCM medium was selected as the favorable culture medium. The optimal range of temperature and pH for mycelial growth on V. volvacea were 35oC and pH 6~8, respectively. Carbon sources had not an effect on 10 strains of V. volvacea, and nitrogen source for the optimum mycelial growth was yeast extract. Also, we selected GMVV 79004 as a superior strain of 10 strains V. volvacea based on the mycelial growth and yield.

Tensile strain-hardening behaviors and crack patterns of slag-based fiber-reinforced composites

  • A strain-hardening highly ductile composite based on an alkali-activated slag binder and synthetic fibers is a promising construction material due to its excellent tensile behavior and owing to the ecofriendly characteristics of its binder. This study investigated the effect of different types of synthetic fibers and water-to-binder ratios on the compressive strength and tensile behavior of slag-based cementless composites. Alkali-activated slag was used as a binder and water-to-binder ratios of 0.35, 0.45, and 0.55 were considered. Three types of fibers, polypropylene fiber, polyethylene (PE) fiber, and polyparaphenylene-benzobisethiazole (PBO) fiber, were used as reinforcing fibers, and compression and uniaxial tension tests were performed. The test results showed that the PE fiber series composites exhibited superior tensile behavior in terms of the tensile strain capacity and crack patterns while PBO fiber series composites had high tensile strength levels and tight crack widths and spacing distances.

Production of Gentamicin by Micromonospora purpurea (Micromonospora purpurea에 의한 gentamicin생성)

  • Using Micromonospora strain, gentamicin was produced by fermentation. The increase in gentamicin productivity was studied by strain improvement and systematic optimization of fermentation process variables. The productivity of parent strain of M.purpurea (ATCC15835) was improved by selection of superior mutant after U.V. irradiation and induction of genetamicin resistance. Potato starch and soy bean meal were the best carbon and nitrogen sources for gentamicin fermentation, respectively. The optimum stimulating concentration of Co ion for gentamicin production was 0.006g $CoCl_2$ per liter of broth. Oxygen ws found to be an important factor for gentamicin yield. The optimum pH for the cell growth and gentamicin production were 7.2 and 6.8 respectively. By controlling the pH, oxygen, and other conditions found in this study at the optimal conditions for cell growth and gentamicin production, the total productivity of gentamicin was increased significantly.

  • To study the effect of welding methods on the Stress Corrosion Cracking (SCC) behavior of welded AISI type 316L and 304 austenitic stainless steel, the Slow Strain Rate Technique(SSRT) has been adopted in the boiling 45 wt% $MgCl_2$ solution. The results are as follows. 1) Welded sections are more susceptible than base metal in SCC, and the rank of SCC, and the rasistance in welding method is TIG, MIG, $CO_2$ and ARC. 2) The Ultimate tensile strength(UTS) and the strain of both base metal and welded joint are reduced as decreasing extension rate. 3) The SCC resistance of 316L base metal and welded sections are superior than that of 304. 4) The tendency of pitting and the SCC suseptibility are agreed well, and the SCC site is welded deposit section in 316L whereas HAZ in 304.

  • Polymer-Impregnated Concrete(PIC) can be considered composite material of concrete and polymer and has superior properties compared to conventional cement concrete, such as strength, stiffness, toughness, durability, water-proofing, chemical resistance. However, so far, the usage of PIC has been limited to repairing materials and non-structural applications, due to the lack of the design criteria and the analytical model to determine structural behavior. The objective of this study is to define the stress-strain response and strength characteristics of PIC in uniaxial and various biaxial compressive loading. On the bases of experimental results, general stress-strain relation, biaxial failure envelope and strength evaluation formular of PIC made with normal aggregate and methylmethacrylate(MMA) are proposed.

Tensile Mean Strain Effects on the Fatigue Life of SiC-Particulate-Reinforced Al-Si Cast Alloy Composites (SiC입자강화 주조Al-Si복합재의 피로수명에 대한 인장평균변형률의 영향)

  • The low-cycle fatigue behaviour of a SiC-particulate-reinforced Al-Si cast alloy with two different volume fractions has been investigated from a series of strain-control led fatigue tests with zero and nonzero tensile mean strains. The composites including the unreinforced matrix alloy, exhibited cyclic hardening behaviour, with more pronounced strain-hardening for the composites with a higher volume fraction of the SiC particles. For the tensile mean strain tests, the initial high tensile mean stress relaxed to zero for the ductile Al-Si alloy, resulting in no influence of the tensile mean strain on the fatigue life of the matrix alloy. However, tensile mean strain for the composite caused tensile mean stresses and reduced fatigue life. The pronounced effects of mean strain on the low-cycle fatigue life of the composite compared to the unreinforced matrix alloy were attributed to the initial large prestrain and non-relaxing high tensile mean stress in the composite with very limited ductility and Cyclic plasticity. Fatigue damage parameter using strain energy, density efficiently accounted for the mean stress effects. Predicted fatigue life using the damage parameter correlated fairly well with the experimental life within a factor of 3. Also, the fatigue damage parameter indicated the inferior life in the low-cycle regime and superior life in the high-cycle regime for the composite, compared to the unreinforced matrix alloy.

Effect of Interaction between Nutritional Level and Breed on Performance of Broiler Production (BROILER 사료의 영양수준이 육용계종의 산육능력에 미치는 영향)

  • This experiment was carrid out to investigate the interaction between boilelr strains and nutrition levels, and the performances of four broiler strains such as Han Hyup 603, Hubbard, Anak and Filch when they were fed by four different nutrition levels (High Protein and energy; HP. HE., Medium Protein and energy; MP. ME., Low Protein ana energy; LP. LE., and low protein and energy; LLP. LLE.). The data used in this study were obtained from a total of 1200 broiler type chicks in Poultry Testing Station, Korean Poultry Association from June 16, to August 11, 1978. Differences of all characters among four nutrition levels were significant except viability and carcass rate. HP. HE and MP. ME treatments showed nearly the same performances in body weight, feed efficiency and point, spread but they were significantly superior to those of LP. LE and LLP. LLE. There were not significant differences among four strains in feed efficiency and viability but other characters, body weight, point spread and carcass rate were observed that the performance of the best strain B was significantly superior to strain D but it was not recognized significance compared with strain A, C in tile result of statisticel analysis. In the interaction between strains and nutrition levels, body weight at high and levels showed significantly differences but at low and low nutrition levels were nearly same among four strains. Therefore this study demonstrated that comparision of body weights between strains should be performed at medium nutrition level or above. Also point spread calculated as index of body weight and feed efficiency was observed that strain B at low nutrition level is excellently higher than other strains and there were little differences at low nutrition level among all strains. It was found that ]it tie differences between performances of high arid medium levels seemed to be as the reason of high fat addition for energy source to high mutrition feed, and in general superior strain showed good performance at all the nutrition levels in$.$all characters but in body weight and point spread there were significantly different responses with different nutrition level, The most superior strain B among four strains earned the most profit per bird, Although performances of high and medium nutrition levels were nearly the same, medium nutrition level also showed the most profit because the feed cost of high nutrition level was higher than that of medium nutrition level.

Strain Improvement of Yeast for Ethanol Production Using a Combined Treatment of Electric Field and Chemical Mutagen N-Methyl-N'-nitro-N- nitrosoguanidine

  • The feasibility of using combined treatments of electric field and chemical mutagen N-methyl-N'-nitro-N-nitroso-guanidine (NTG) for the strain improvement of Saccharomyces sp. in ethanol production was examined. The treatment of electric field alone resulted in no effect on the lethality of yeast cells under the conditions of this study. However, when the electric field was applied together to the treatment of yeast cells with NTG, the electric field increased the lethal effect and auxotrophic mutation rate of NTG. The combined treatment of electric field and NTG also increased the chances of. obtaining superior yeast strains for the ethanol production from tapioca. A higher number of improved clones was obtained by the combined treatments of electric field and NTG than by the NTG treatment alone. The best clone, NF 30-9, which was also obtained by the combined treatment, produced $11.07\%$ (w/v) ethanol from tapioca slurry containing 25% (w/v) reducing sugar while the parental strain produced 9.77% (w/v).

  • The knowledge of material stress-strain behavior is an essential requirement for design and analysis of deformation processes. Empirical stress-strain relationship and constitutive equations describing material behavior during deformation are being widely used, despite suffering some drawbacks in terms of ease of development, accuracy and speed. In the present study, back-propagation neural networks are used to model and predict the flow stresses of a HSLA steel under conditions of constant strain, strain rate and temperature. The performance of the network model is comparedto those of statistical models on rate equations. Well-trained network model provides fast and accurate results, making it superior to statistical models.
