Physicochemical Properties of Starches Isolated from New Potato Cultivars in Korea

  • Physicochemical properties of starches isolated from 3 new potato cultivars developed by Potato Valley Ltd. were investigated and compared to those of starch isolated from 'Superior' being distributed prominently in Korea. Significant differences were observed in physicochemical properties such as granule size, amylose content, phosphorus content, water binding capacity, swelling power, solubility, and in vitro digestibility of starches from different potato cultivars. Thermal properties were evaluated using differential scanning calorimetry (DSC), and onset gelatinization temperatire ($T_o$), peak temperature ($T_p$), and enthalpies of gelatinization (${\triangle}H$) of different potato cultivars ranged as $58.0{\pm}0.3-61.7{\pm}0.4^{\circ}C,\;63.7{\pm}0.2-$ $66.5{\pm}0.0^{\circ}C$, and $15.6{\pm}0.5-17.0{\pm}0.3$ J/g, respectively. Pasting properties were evaluated using a rapid visco analyzer (RVA), and pasting temperature, peak viscosity, and final viscosity of different potato cultivars ranged as $65.0{\pm}0.1-68.9{\pm}0.1^{\circ}C$, $8,163.7{\pm}196.3-9,035.7{\pm}6.4$ cp, and $4,397.7{\pm}166.7-7,025.0{\pm}271.3$ cp, respectively. Especially, 'Gogu valley' starch showed the highest values in the amylose and phosphorus content among tested potato cultivars and higher water binding capacity, swelling power, and solubility than those of other tested starches. And it also showed high pasting temperature, peak viscosity, trough viscosity, and final viscosity as compared to other tested starches.

Effect of Post-harvest Temperature on Potato Piece Rot in Relation to Suberin and Periderm Development (감자 수확후 온도가 절단괴경의 suberin 및 periderm 형성에 관련된 부패에 미치는 영향)

  • As the important pathogens of potato storage diseases, Fusarium solani, F. reseum, F. oxysporum, and Erwinia carotovora were isolated from rot potato tubers. The cut potato pieces of the three cultivars, Epicure, Irish Cobbler, and Superior were held in moist chambers of $4^{\circ},\;14^{\circ},\;24^{\circ},\;and\; 34^{\circ}C$ and 7 days and then rated for suberin and periderm development. The cut potato pieces thus treated were inoculated with the four organisms and held at $24^{\circ}C$ for 9 days and then rated for decay. As the temperature and period of holding increased, more suberin and periderm were developed with decrease in decay. Although there were differences in pathogenicity of the organisms, varietal reaction and protective barrier development, the effect of temperature and holding period had greater importance for decay prevention. At $4^{\circ}C$ within 7 days of holding period the potato pieces developed no protective barrier with severe decay. It is required to avoid placing cut potatoes directly in cold storage of the low temperature. At $34^{\circ}C$ the pieces developed abundant protective barriers even though decay occurred in somecases. Practically no decay was found with moderate protective barrier development after 3 days and 5 days at $24^{\circ}C\;and\;14^{\circ}C$, respectively. Since the potato pieces decayed occasionally during the holding period when they were held at the higher temperature, the holding at $14^{\circ}C$ for longer than 5 days is considered to be feasible for prevention of storage rots.

Development of New Value-added Corn Dog Using Milk Powder, Egg and Potato (분유, 계란 및 감자를 이용한 새로운 Value-added Com Dog 개발에 관한 연구)

  • This study was conducted to manufacture new value-added corn dog using potato, milk powder and egg as nutritious and surplus agricultural and livestock products. A typical corn dog was manufactured with corn dog powder, potato, milk powder and egg as the same method of conventional corn dog. Corn dogs were divided into four groups; control group A [conventional corn dog], group B [corn dog manufactured with the fixed content of corn dog powder, milk powder, and egg, potato, water (3: 3: 1: 1: 2)], group C [corn dog manufactured with the fixed content of corn dog powder, milk powder, and egg, potato, water (3: 3: 2: 1: 2)], group D [corn dog manufactured with the fixed content of corn dog powder, milk powder, and egg, potato, water (3: 3: 1: 2: 2)], Viscosity of corn dog batter, pH of corn dog, rheology and sensory evaluation were measured. There were no significant differences for viscosity and pH between original corn dog and manufactured corn dog (p>0.05). However, hardness and brittleness of manufactured corn dog D were superior to the other groups (p<0.05). Also, manufactured corn dog D was superior to the other groups by the results of sensory evaluation. Therefore, these results suggest that it may be possible to manufacture new value-added corn dog which can help to stimulate the consumption of nutritious and surplus agriculture and livestock products.

Infestation of Potato Cultivars by Potato Aphid, Macrosiphum euphorbiae Thomas, and its Infestation-related Factors (감자수염진딧물(Macrosiphum euphorbiae Thomas)에 의한 감자 품종별 피해정도와 관련요인 분석)

  • Fifty potato cultivars were screened for potato aphid, Macrosiphum euphorbiae Thomas infestation degree in the field. Aphid honey dew secretion and contents of foliar substances(glycoalkaloids, total nitrogen, Ca, K and Mg) of the cultivars were measured in the laboratory to analyze the correlation between infestation degrees and those factors. The number of aphids ranged from 4.1 to 31.4 per five leaves. The lower density(less than 5 aphids) was found on cvs. Jidose, Recent, and Sebago. The aphid secreted highest(40.3 droplets) and lowest(15.7 droplets) honeydew on cvs. Jopoong and Anco, respectively. Degree of infestation is positively correlated to glycoalkaloids and total nitrogen contents of the potato leaf.

Susceptibility of Two Potato Cultivars to Various Environmental Stresses (다양한 환경스트레스에 대한 감자 2품종의 감수성 분석)

  • Environmental stress is the major limiting factor in plant productivity. In order to evaluate the stress tolerance of potato plants, leaf discs of two potato cultivars, Atlantic and Superior, were subjected to various stress conditions of high temperature, methyl viologen, H2O2, or $H_2O$$_2$. When potato leaf discs were exposed to high temperature at 37$^{\circ}C$ for 84 hr, Atlantic plants, a cultivar with high sensitivity to heat stress, showed about 20% higher membrane damage than Superior plants. When exposed to 2$\mu$M methyl violgen (MV), a superoxide generating non-selective herbicide, for 36 hr, Atlantic plants also showed about 38% higher membrane damage than Superior plants, and were more susceptible up to 10$\mu$M MV concentration tested. On treatment with 0.75M NaCl, Atlantic plants also had about 45% less chlorophyll contents in leaf discs than Superior plants. There was, however, no difference in chlorophyll content of two cultivars at higher NaCl concentrations. The effect of $H_2O$$_2$ on the two cultivars was mixed. At low $H_2O$$_2$ concentration (25 mM) , Superior plants were more susceptible to $H_2O$$_2$stress after 36 hr. However, at high $H_2O$$_2$ concentration (100 mM), Atlantic plants exhibited higher susceptibility after 36 hr. The results indicate that in vitro leaf discs reflecting the whole plants in this study will be useful for selection and characterization of elite transgenic potato plants with enhanced tolerance to environmental stress.

Growth and Yield of Potato after Transplanting of Potato Plug Seedlings Grown at Different Plug Cell Size and Photoperiod (다른 플러그 셀 크기와 일장에서 생산된 감자 플러그 묘의 정식 후 생육과 수량)

  • This experiment were conducted to investigate the response of growth and yield of potato after transplanting of plug seedlings 'Superior' and 'Dejima' produced at different plug cell size (105, 162, and 288) and photoperiod (8/16, 12/12, and 16/8, day/night) for 20 days in controlled plant growth system. Growth and relative growth rate of plug seedling 'Superior' was affected by plug cell size and photoperiod at 7weeks after transplanting. Tuber weight of 'Superior' was increased as cell size and photoperiod increased. That of 'Dejima' was highest in 105 cell and different with photoperiod. At 90 days after transplanting, tuber weight ($258.9{\sim}471.9\;g/plant) of 'Superior' was high in 105 and 162 cell size and 16/8 hr photoperiod. That ($278.2{\sim}428.0\;g/plant$) of 'Dejima' was high in 105 cell size, but was not different with photoperiod. The number of tuber per plant was $2.6{\sim}6.9$ of 'Superior' and $2.2{\sim}3.6$ 'Dejima'. Tuber number per plant was not significantly different with cell size and photoperiod. The large tuber over 80 g was $32.0{\sim}50.9%$ of 'Superior' and $41.0{\sim}56.7%$ of 'Dejima'. The large tuber in 'Superior' and 'Dejima' lowered as the cell size decreased. The large tuber of 'Superior' increased as photoperiod increased, but that of 'Dejima' was not differed. As the results, the optimal plug cell size and photoperiod of potato seedling is considered to be below 162 cell and over 12 hr of photoperiod.

Field Performance of a New Fungicide Ethaboxam Against Cucumber Downy Mildew, Potato Late Blight and Pepper Phytophthora Blight in Korea

  • Ethaboxam is the first proprietary fungicide developed in Korea, registered in 1998 and commercialized in 1999 by LG Chemical Ltd., Korea. It is a derivative of aminothiazole carboxamide and formulated into 25% wettable powder for practical application in fields. Ethaboxam effectively controlled cucumber downy mildew caused by Pseudoperonospora cubensis, potato late blight caused by Phytophthora infestans, and pepper Phytophthora blight caused by P. capsici, and was superior or comparable to the commercial standards, when foliarly sprayed 3∼5 times until dripping off at approximately 7-day intervals during the growing season. Ethaboxam was required at least 125 mg/liter and 250 mg/liter for effective control of cucumber downy mildew, and potato late blight and pepper Phytophthora blight, respectively. There was not phytotoxicity observed o leaves, stems or fruits of cucumber, potato and pepper from any trial.

Improved in vitro Regeneration of Potato (Solanum tuberosum cv. Superior) Transformed by Agrobacterium Expressing $\beta-Glucuronidase$

  • In order to enhance the system of potato transformation and further regeneration, potato was transformed using the Agrobacterium tumefaciens harboring $\beta$-glucuronidase (GUS) gene. We found that a series fo modified medium ttained 100% shoot regeneration within 5 weeks after the preincubated explants on stage I medium were infected with Agrobacterium. Callus appeared at the cut edges of stem segments on stage II medium, mainly at the basal parts. Some explants started to form shoots after two to three weeks on stage III medium containing kanamycin (50 mg/L). When transferred to MS medium containing 200 mg/L kanamycin, 81% of the transformed shoots formed roots at the cut edge of the plantlets. In contrast, untrasformed shoots never rooted and became yellowish after few weeks under the same conditions. Southern and northern analysis indicated in vitro shoot regeneration on the callus derived from the potato explants, which were incubated with Agrobacteria. The regeneration cycle was shortened after the transformatin and finally the transformation efficiency was highly enhanced.

Complementary DNA Cloning and Sequencing of the Coat Protein Gene of Potato Virus Y-Ordinary Korean Strain (감자바이러스 Y의 OK계통에 대한 외피단백질 유전자 cDNA 클로닝 및 염기서열 분석)

  • Complementary DNAs (cDNAs) to the coat protein gene of an ordinary Korean strain of potato virus Y (PVY-OK) isolated from potato (cv. Superior) were synthesized and cloned into a plasmid pUC119 and sequenced. The RNA of the virus propagated in tobacco (Nicotinaa sylvestris) was extracted by the method of phenol extraction. The first strand of cDNAs to the coat protein penomic RNA of the virus was made by Moloney murine leukemia virus reverse transcriptase. The cDNA were synthesized and amplified by the method of polymerase chain reaction (PCR) using a pair of oligonucleotide primers. PVYCP3P and PVYCP3M. The size of cDNAs inserted in pUC119 plasmid was estimated as about 840 bp upon agarose gel electrophoresis. Double stranded cDNAs were transformed into the competent cell of E. coli JM109. Sequence analysis of cDNAs was conducted by the dideoxynucleotide chain termination method. Homology of cDNAs of the PVY-OK coat protein genomic RNA with those of PVY-O (Japan), PVY-T (Japan), PVY-TH (Japan), PVYN (The Netherlands),and PVYY (France) was represented as 97.3%, 88.9%, 89.3%, 89.6% and 98.5%, respectively. Homology at the amino acid level turned out to the be 97.4%, 92.5%, 92.9%, 92.9% and 98.5%, respectively.

Properties of Low-Fat Pork Patties Formulated with Carrageenan Alone or in Combination with Pectin or Potato Starch (Carrageenan에 pectin 및 potato starch를 혼합 첨가하여 제조한 저지방 돈육 patty의 특성연구)

  • This study was conducted to investigate the quality characteristics of low- fat pork patties containing fat replacers. Carrageenan(CA) in combination with pectin(PE) or potato starch(PO) was added as fat replacers and physicochemical properties were evaluated in low-fat pork patties cooked by oven-roasting or pan-frying. Uncooked and cooked pork patties formulated with fat replacers were higher in moisture and ash content and lower in fat content than those of control. The cooking yield and reduction in diameter of pork patties were significantly improved by the addition of fat replacers(p<0.05). Pork patties formulated with CA had the highest fat retention, while CA+PE had the highest water holding capacity. Hunter s L(lightness) value was not different from among patties and a(redness) and b(yellowness) values were higher in carrageenan-based patties than those of control patties. Hardness was higher in carrageenan-based patties than that of control and cohesiveness and springiness showed no difference among patties. Sensory analysis showed that flavor, juiciness and palatability of carrageenan-based patties were superior to control patties and combined use of CA with PE or PO provided improved acceptability of low-fat pork patties over that from using single carrageenan.
