Effective Extraction of Sea Mustard with Supercritical Carbon Dioxide (초임계 이산화탄소에 의한 미역의 효과적 추출)

  • The extraction characteristics of lipids from powdered sea mustard have been investigated by the use of supercritical carbon dioxide($SC-CO_2$) and cosolvents. The extraction rate was increased as the particle size of the sea mustard is smaller, the pressure is higher, the temperature is lower, and the quantities of the fluid is more. However, the extraction yield of lipids from sea mustard was almost constant at a given condition. The optimum extraction condition was determined with the extraction yield of $1.45wt\%$ at 300um of particle size, 313K of $SC-CO_2$ temperature, 13.8MPa of pressure, and 30L/min of flow rate. Ethanol was the most efficient cosolvent among ethanol, methanol, and hexane. The extraction yield was increased at about 2.21times by the addition of ethanol as a cosolvent to $SC-CO_2$. As the residence time and the average concentration of lipids were decreased, the mass transfer parameter($k_fa$) was increased. But the opposite result was obtained when the ethanol was used as cosolvent.

Antimicrobial Activity of Ginger(Zingiber officinale Roscoe) Oleoresin by Supercritical Fluid Extraction (초임계유체 추출에 의한 생강(Zingiber officinale Roscoe) Oleoresin의 항균활성)

  • The study indicated that antimicrobial activity about gram positive and gram negative bacteria of ginger-oleoresin(GO) extract with the condition of ethanol and supercritical fluid extractions. As the concentration of extraction increases, the clear zone of GO ethanol extract also increased dependently. This led the antimicrobial activity of gram positive bacteria to take bigger place than gram negative bacteria especially in Listeria monocytogenes. There was a high antimicrobial activity in E-III treatment where the ratio of the ginger powder extract to ethanol extraction was 1:6. It was quite effective to treat the antimicrobial activity of GO ethanol extract under $80^{\circ}C$ and there was not big difference in the intervals which were the extraction time - 1 to 7 hours. The antimicrobial activity of supercritical fluid extract seemed to take the biggest place in Listeria monocytogenes. From the supercritical fluid extract, it was shown the strong ability of antimicrobial activity in the condition with 100 bar $35^{\circ}C$, 250 bar $35^{\circ}C$ and 250 bar $65^{\circ}C$. Furthermore, according to the case of solvent extract, there was not any significant difference in the antimicrobial activity with condition of extraction. However, there was significant antimicrobial activity in E-III treatment of 100 bar and 500 bar of extraction pressure, and $35^{\circ}C$ and $65^{\circ}C$ of extraction temperature.

The activation of PPAR-α and Wnt/β-catenin by Paeonia lactiflora root supercritical carbon dioxide extract

  • The root of Paeonia lactiflora has been used in Chinese medicine. We conducted to check the comparative qualities of ethanol solvent extraction (PLE) and supercritical carbon dioxide extraction (PLS) of P. lactiflora root. PLE had higher antioxidant and polyphenol contents than PLS. But, PLS were significantly increased peroxisome proliferator-activated receptor (PPAR)-α. In addition, PLS inhibited the adipocyte differentiation of 3T3-L1 cells. When treated with the extract at a concentration of 100 ㎍/mL, the Wnt/β-catenin pathway reporter luciferase activity of HEK 293-TOP cells increased approximately by 3-folds compared to that of the untreated control group. These results indicate that P. lactiflora supercritical carbon dioxide extract may serve as a cosmeceutical for improving skin barrier function and the treatment of obesity.

The Activation of PPAR-α and Wnt/β-catenin by Luffa cylindrica Supercritical Carbon Dioxide Extract

  • Luffa cylindrica (LC) is a very fast-growing climber and its fruit have been considered as agricultural wastes. We conducted to check the comparative qualities of ethanol solvent extraction (LCE) and supercritical carbon dioxide extraction (LCS) of L. cylindrica fruit and seed. LCS had higher antioxidant and polyphenol contents than LCE. LCS were significantly increased peroxisome proliferator-activated receptor (PPAR)-a and involucrin expression as epidermal differentiation marker in 3D skin equivalent model. LCS also showed antimicrobial activity against Staphylococcus aureus, a causative bacteria in atopic dermatitis. In addition, LCS inhibited the adipocyte differentiation of 3T3-L1 cells. When treated with the extract at a concentration of 100 ㎍/mL, the Wnt/β-catenin pathway reporter luciferase activity of HEK 293-TOP cells was increased approximately by 2-folds compared to that of the untreated control group. Fabrication of Ultra-Fine TiO$_2$ Powders Using Supercritical Fluid (초임계 유체를 이용한 초미립 TiO$_2$ 제조)

  • In order to fabricate ideal powders new processing is necessary in which the solute atoms in solution ra-pidly move to mix each other to the degree of molecular level the viscosity of solution should be low not to effect the moving of solute atoms and the powders could be directly obtained as crystalline. Supercritical fluid is defined as condensed gas sated up to its critical pressure and temperature. In this paper su-percritical fluid methods were studied as a new ceramic processing of powder preparation. The crystalline powders of TiO2 which are useful for electronic ceramic materials were fabricated by hydrolysis of titanium (IV) ethoxide using ethanol as a supercritical fluid at the condition of 270$\pm$3$^{\circ}C$, 7.3 MPa for 2hr. The cry stalline anatase powders could be directly obtained and its primary particle size was 20 min.

The Anti-inflammatory Effect of Angelica gigas Nakai Supercritical Carbon Dioxide Selective Extracts (참당귀 초임계 이산화탄소 선택 추출물의 항염 효능)

  • In this study, the anti-inflammatory effect of supercritical carbon dioxide selective extracts, which extract decursin and decursinol angelate, the vital active ingredients of Angelica gigas Nakai, in high yield, was measured compared to that of ethanol extracts. To measure the anti-inflammatory effect, the production of nitric oxide(NO), an inflammatory mediator, and interleukin(IL)-6 and IL-8, inflammatory cytokines, was measured. NO production was measured by Griess assay on Raw 264.7 cells induced inflammatory response by lipopolysaccharide(LPS), and IL-6 and IL-8 production was measured by enzyme-linked immunoadsorbent assay(ELISA) on human keratinocyte cell line(HaCaT) cells induced inflammatory response by tumor necrosis factor(TNF)-α. The amount of NO production was suppressed outstandingly by the supercritical carbon dioxide extracts compared to the ethanol extracts. The amount of IL-6 and IL-8 production was increased by the ethanol extracts, whereas statistically significantly inhibited by supercritical carbon dioxide extracts at the concentration of 6.25 ㎍/mL(P<0.01). Through these results, we confirmed that the supercritical carbon dioxide selective extracts of Angelica gigas Nakai could be used as an anti-inflammatory cosmeceutical material to alleviate atopic dermatitis.

Extraction of Genistein from Sophora flavescens with Supercritical Carbon Dioxide (초임계유체를 이용한 고삼으로부터 Genistein의 추출)

  • This study was directed to finding an optimum extraction condition of genistein from the S. flavescens with supercritical carbon dioxide as a solvent. In this effort, effects of the extraction conditions including pressure, temperature and a co-solvent on the extraction efficiency were investigated. The aqueous ethanol and methanol solutions were used as co-solvents while the tested operating pressure and temperature ranges were from 200 bar to 300 bar and from 308.15 K to 323.15 K, respectively. The concentration of genistein was determined by means of HPLC equipped with a UV detector. From the results, it was observed that an increase in pressure led to the higher extraction efficiency. Further, methanol showed better performance as a co-slovent than ethanol. The DPPH radical scavenging activities were measured to compare antioxidant activities of S. flavescens extracts.

Extraction of${\beta}-carotene$ from Carrot by Supercritical Carbon Dioxide (초임계이산화탄소에 의한 당근 중의 ${\beta}-carotene$ 추출)

  • ${\beta}-carotene$ was extracted from freeze-dried carrot by supercritical carbon dioxide $(SC-CO_2)$ and mixtures of $CO_2$ doped with ethanol or methanol as a cosolvent at temperatures of 40 to $60^{\circ}C$ and pressures of 138 to 276 bar. Solubility of ${\beta}-carotene$ in $SC-CO_2$ increased with the increase of extraction pressure and the decrease of extraction temperature. The highest solubility observed was $4.90\;{\mu}g/g\;CO_2\;for\;{\alpha}-carotene\;and\;0.604\;{\mu}g/g\;CO_2$ for ${\alpha}-carotene\;at\;40^{\circ}C$ and 276 bar. Addition of ethanol increased the solubility being the largest increase of 82% using a mixture of $CO_2$ and 17.4% ethanol. $SC-CO_2$ extraction can be used to selectively obtain natural carotenoids, free of solvent residuals, which can be used as food additives.

Preparation of L-PLA Microparticles using Pure and Cosolvent-modified Supercritical Carbon Dioxide (순수 초임계 이산화탄소와 극성 공용매로 변형된 초임계 이산화탄소를 이용한 L-PLA 미세입자 제조)

  • Biodegradable poly (L-lactide) (L-PLA) solution in methylene chloride was precipitated into microparticles by using supercritical carbon dioxide modified with polar cosolvents. The effects of the amount of polar cosolvents, solution concentration, temperature, and solution flow rate on the formation of microparticles were investigated. The mean particle size was found to increase with the increase of solution concentration and flow rate. It was also observed that the particle size not only increases but the size distribution also becomes less uniform as the temperature increases. The percent recovery of microparticles was found to be 30∼40% at all experimental conditions. The supercritical carbon dioxide modified with methanol and ethanol was employed to enhance the recovery, resulting in significant improvement up to about 80 and 70%, for methanol and ethanol, respectively. Furthermore, the mean diameter of L-PLA microparticles was found to be less than 1 $\mu\textrm{m}$ for both cosolvents.

Recovery of High Unsaturated Fatty Acid from Squid Processing Wastes using Supercritical Carbon Dioxide Extraction Method (초임계 이산화탄소 추출법을 이용한 오징어 가공 부산물로부터 고도불포화 지방산 회수)

  • A squid viscera oil contains a high content of EPA, DHA, and other valuable polyunsaturated fatty acids. The extractions of squid viscera oil by supercritical carbon dioxide both with/without $3\%$ (v/v) ethanol were performed in a semicontinuous flow extractor at 8.3 to 13.8 MPa and 25 to $50^{\circ}C$. When ethanol was added to $SC-CO_2$, the extraction ratio of lipid increased. The extracts contained high content of unsaturated oils like DHA and EPA. The highest extraction yield of lipid from squid viscera oil extracted by supercritical carbon dioxide was obtained at 12.4 MPa and $40^{\circ}C$ with/without entrainer. The main fatty acids of squid viscera oil extracted by supercritical carbon dioxide were myristric acid (14:0), palmitic acid (16:0), palmitoleic acid (16:1), oleic acid (18:1), arachidic acid (20:0), eicosapentaenoic acid (20:5), and docosahaxaenoic acid (22:6).
