고콜레스테롤 식이 섭취 흰쥐에 대한 적송잎 추출물의 항산화 효과 (Antioxidative Activity of Pine (Pinus densiflora) Needle Extracts in Rats Fed High-Cholesterol Diet)

  • 본 연구는 선행연구에서 적송잎 열수추출물의 항산화 활성이 높았으며 아임계 추출물 중에 항산화 활성이 높은 proanthocyanidin 함량이 높은 것으로 나타났으므로, 이러한 in vitro 실험의 결과를 in vivo 실험을 통하여 입증하고자 하였다. 적송잎의 열수추출물과 아임계 추출물이 콜레스테롤 식이를 투여한 흰쥐의 체내 항산화능에 미치는 영향을 검토하기 위하여 혈청 중의 AST, ALT, ALP 및 간장중의 항산화 효소인 SOD, CAT, GPx의 활성을 측정하였다. 체중 증가량은 콜레스테롤 식이를 투여한 CHOL군이 일반 대조식이군인 CON군보다 유의적으로 증가하는 결과가 나타났다. 콜레스테롤 식이 투여 시 식이섭취량은 일반식이군에 비하여 감소하였으나 식이효율은 유의적으로 증가하였다(p<0.05). 적송잎 추출물을 투여하였을 경우 체중증가량은 감소하였으나 유의차가 없었으므로 적송잎 추출물이 콜레스테롤 식이로 인한 체중증가에는 별다른 영향을 주지 않는것으로 나타났다. 혈청 중의 ALT 및 AST 활성은 콜레스테롤 식이를 투여한 CHOL군이 CON군에 비하여 활성이 증가되었으나 적송잎 추출물의 투여로 인하여 감소하는 경향을 타나내었다. 특히 열수 추출물보다 아임계 추출물에서 AST의 활성은 유의적으로 낮게 나타났으며, ALT는 유의성은 없었으나 CHOL군보다 활성이 낮게 나타났다. 간 조직 중의 항산화 효소 결과에서 SOD 유사활성은 CHOL군에서 대조군에 비해 유의적으로 감소하였으나 적송잎 아임계추출물 투여에 의해 증가하는 결과가 나타났다. GPx활성은 CON군에 비해 CHOL군에서 유의적으로 감소하였으나 적송잎 추출물 투여에 의해 CHOL군에 비해 현저하게 높은 결과로 나타나, 콜레스테롤식이 투여로 인한 효소활성 저하가 적송잎에 의해 회복됨을 알 수 있었다. 이상의 결과, 고콜레스테롤 유발 흰쥐에게 적송잎 추출물의 급여는 체내 항산화효소의 활성에 영향을 미치는 것으로 나타나 적송잎 추출물이 체내 항산화능 개선에 효과가 있을 것으로 기대된다.

딸기의 휘발성 향기성분의 초임계 유체 추출 (Supercritical Fluid Extraction of Volatile Components from Strawberry)

  • SFE법을 이용하여 딸기의 휘발성 향기성분을 추출하기 위한 시료의 적정 전처리 및 SFE 작동조건을 검토하였으며, SDE법 및 SE법 등의 추출방법과 휘발성 향기성분 조성을 비교하였다. 딸기 생시료, 동결건조시료, 그리고 생시료에 celite를 30, 70%를 혼합하여 제조한 시료를 이용하여 SFE 처리후 향기성분을 분석한 결과, 생시료에 celite를 70% 첨가하는 방법이 향기성분을 추출하는데 가장 효과적인 것으로 나타났다. 온도(40, $55^{\circ}C$)와 압력(3,000, 6,000 psi)을 달리하여 SFE 처리한 결과, 3000 psi, $55^{\circ}C$ 조건에서 alcohol류와 acid류의 추출효율이 증가하였으며, $\gamma$-dodecalactone의 함량도 3배 정도 높은 수준으로 나타났다. SDE법, SFE법 및 SE법 등의 추출방법별 딸기의 휘발성 향기성분을 분석한 결과, SFE법은 SDE법에 비해 추출한 휘발성 향기성분의 종류는 적지만 추출량이 비슷하면서 열분해 및 열변성이 발생하지 않아 딸기 고유의 향기에 가까운 향기패턴을 나타내었으며, 기호적으로 우수한 acid류가 다량 함유된 향기성분을 추출하기에 적합한 것으로 나타났다.

저콜레스테롤 계란제품의 생산기술과 부산물의 재활용 (Production Technology of Low-cholesterol Egg Prodecuts and Recycling of By-Products)

  • Hurdle technique was used to remove cholesterol efficiently from liquid egg yolk. The quality of the low cholesterol egg products from the process were evaluated. From the 75 % cholesterol reduced egg yolk through $\beta$-cyclodextrin treatment. 2 times weight of soy bean oil was added to the egg yolk and homogenized followed by centrifuged to be maximized to remove cholesterol. When the pH of the yolk was adjusted to 9, 92 % of cholesterol was removed while 95.4 % of cholesterol was removed when 3 times weight of soy bean oil was added to the egg yolk. As the results of application of supercritical carbon dioxide extraction to the 75 % cholesterol reduced egg yolk through ${\beta}$-cyclodextrin treatment, 92.5 % of the cholesterol was removed from the egg yolk at $35^{\circ}C$, 4,500 psi, for 4 hours under co-solvent. The quality characteristics of the produced low cholesterol egg products were analysed. The cholesterol reduced egg yolk produced from ${\beta}$-cyclodextrin and soy bean oil treatment showed the lower emulsion capacity compared with control. The fatty acid composition of the cholesterol reduced egg yolk produced from ${\bet}a$-cyclodextrin and soy bean oil treatment showed increased C18:2 and C18:3 compared with control while decreased C16:1 and C18: 1 compared with control. The saponification method with extracting solvent of hexane showed that cholesterol concentration was 28.1 %. The quantity of hydrolysis solution(95 % ethanol : 33 % KOH = 94 : 6) was varied from 40 to 80 times of sample weights and the cholesterol concentration of 35.7 % was the highest result in the 60 times(v/w) hydrolysis solution. Cholesterol concentration of 35.7 % was recovered at the first trial with saponification method. but it could be improved up to 95.7 % after 4 times repetitive purification.

Subacute oral toxicity and bacterial mutagenicity study of Korean Red Ginseng oil

  • Background: Red ginseng oil (RGO) is produced by supercritical $CO_2$ extraction of secondary products derived from Korean Red Ginseng extract. As the use of RGO has increased, product safety concerns have become more important. Methods: In the present study, the subacute oral toxicity and bacterial reverse mutagenicity of RGO were evaluated. Sprague-Dawley rats were orally administered with RGO for 28 d by gavage. Daily RGO dose concentrations were 0 mg/kg body weight (bw), 500 mg/kg bw, 1,000 mg/kg bw, or 2,000 mg/kg bw per day. Bacterial reverse mutation tests included five bacterial strains (Escherichia coli WP2 and Salmonella typhimurium TA98, TA100, TA1535, and TA1537), which were used in the presence or absence of metabolic activation. The plated incorporation method for mutation test was used with RGO concentrations ranging from $312.5{\mu}g$ to $5,000{\mu}g$ per plate. Results: The subacute oral toxicity test results did not reveal any marked changes in clinical characteristics. There were no toxicological changes related to RGO administration in hematological and serum biochemical characteristics in either control or treatment animals. Furthermore, no gross or histopathological changes related to RGO treatment were observed. The bacterial reverse mutation test results did not reveal, at any RGO concentration level and in all bacterial strains, any increase in the number of revertant colonies in the RGO treatment group compared to that in the negative control group. Conclusion: The no-observed-adverse-effect level of RGO is greater than 2,000 mg/kg bw and RGO did not induce genotoxicity related to bacterial reverse mutations.

박하(Mentha arvensis) 향료의 향기성분이 정신적 스트레스 완화에 미치는 효과 (Fragrance Chemicals in the Essential Oil of Mentha arvensis Reduce Levels of Mental Stress)

  • 본 연구는 박하(Mentha arvensis) 식물 유래 향료의 향기성분을 구명하고, 향기성분들이 인간의 뇌파에 어떠한 영향을 미치는지를 이해하고자 수행하였다. 초임계추출기를 이용하여 박하 식물(Mentha arvensis L. f. piperascens)로부터 에센셜오일을 얻었으며, 최적 회수율은 $70^{\circ}C$, 200 bar 조건에서 2.38%이었다. 박하 에센셜오일에 함유되어 있는 향기 화합물을HS-SPME/GC-MS로 분석한 결과, 총 32종의 화합물이 검출되었는데 alcohol 류가 6종(67.11%), hydrocarbon 류가 13종(17.05%), ester 류가 9종(11.50%), ketone 류가 2종(7.16%), oxide가 1종(2.77%) 그리고 aldehyde가 1종(0.56%)이었다. 박하 에센셜오일에 함유된 주된 향기 화합물은 (Z,Z,Z)-9,12,15-octadecatrien-1-ol (50.06%), 2-hydroxy-4-methoxyacetophenone (7.50%)과 3,4-dihydro-8-hydroxy-3-methyl-1H-2-benzopyran-1-one (6.60%) 이었다. 총 20명의 피험자(남녀 각 10명)를 대상으로 박하 에센셜오일 향기 흡입 전과 흡입 중에 뇌파분석을 수행한 결과, 향기를 흡입 중에는, 흡입 전과 비교하여, relative fast alpha power spectrum이 유의성 있게 증가하는 반면(p<0.05), gamma power spectrum과 spectral edge frequency 90% 지표는 유의성 있게 감소하는 결과를 얻었다(p<0.05). 본 연구의 결과들은 박하의 향기성분이 정신적 긴장을 완화시킨다는 것을 시사하여 준다.

Effect of replacement feed ingredients of Micropterus salmoides in exotic species

  • Background: Largemouth bass (Micropterus salmoides) is introduced species that has caused aquatic ecology activity both in vitro and in vivo were investigated for the possibility of application of the bass extract as an alternative feed ingredient. Methods: The bass oil was extracted using a 1-L supercritical extractor, while the protein was extracted from 250 g of bass dry matter, which was dissolved in 1 mL of H2O at 50℃. Both oil and protein extracts were evaluated antioxidant activities and the level of DPPH radical scavenging assay and nitric oxide (NO) production assay with lipopolysaccharide response. Oral administration of 6.6 µL/g bass protein and 5.38 µL/g bass oil conducted for investigating serological and physiological effect. Results: DPPH radical scavenging showed similar radical scavenging ability of 50 µM of ascorbic acid at 200 ㎍ of protein and 10% of oil treatment. NO concentration was diminished by the treatment of bass oil. Oral administration of both bass oil and proteins to mice showed that the body weight increase rate of the bass oil treated group was significantly reduced by 1.55 g compared to the other groups. The number of white blood cells (WBC) was increased by 4.52 k/µL in the bass protein-treated group and 4.44 k/µL in the bass oil-treated group compared to the control group. However, the serum IgG level did not show a significant difference between the bass extract-treated groups and the control group. Conclusions: These studies demonstrate that both bass oil and proteins extracted from the bass not only provide excellent effects of antioxidant and immune activity but can also be used as functional food supplements.

Induction of Nrf2/ARE-mediated cytoprotective genes by red ginseng oil through ASK1-MKK4/7-JNK and p38 MAPK signaling pathways in HepG2 cells

  • Background: The induction of cellular defensive genes such as phase II detoxifying and antioxidant enzymes is a highly effective strategy for protection against carcinogenesis as well as slowing cancer development. Transcription factor Nrf2 (nuclear factor E2-related factor 2) is responsible for activation of phase II enzymes induced by natural chemopreventive compounds. Methods: Red ginseng oil (RGO) was extracted using a supercritical $CO_2$ extraction system and chemical profile of RGO was investigated by GC/MS. Effects of RGO on regulation of the Nrf2/antioxidant response element (ARE) pathway were determined by ARE-luciferase assay, western blotting, and confocal microscopy. Results: The predominant components of RGO were 9,12-octadecadienoic acid (31.48%), bicyclo[10.1.0] tridec-1-ene (22.54%), and 22,23-dihydrostigmasterol (16.90%). RGO treatment significantly increased nuclear translocation of Nrf2 as well as ARE reporter gene activity, leading to upregulation of heme oxygenase-1 and NAD(P)H:quinone oxidoreductase 1. Phosphorylation of the upstream kinases such as apoptosis signal-regulating kinase (ASK)1, mitogen-activated protein kinase (MAPK) kinase (MKK)4/7, c-Jun N-terminal kinase (JNK), and p38 MAPK were enhanced by treatment with RGO. In addition, RGO-mediated Nrf2 expression and nuclear translocation was attenuated by JNK inhibitor SP600125 and p38 MAPK inhibitor SB202190. Conclusion: RGO could be used as a potential chemopreventive agent, possibly by induction of Nrf2/ARE-mediated phase II enzymes via ASK1-MKK4/7-JNK and p38 MAPK signaling pathways.
