환경 시료 중 설폰아미드계 항생제 및 대사체 분석방법 연구 (The study of analytical method for sulfonamide antibiotics and their metabolites in environmental samples)

  • 본 연구에서는 동물용 의약품으로 널리 사용되고 환경 중 잔류 가능성이 높은 설폰아미드계 항생제 5종 (sulfadiazine, sulfamerazine, sulfamethazine, sulfamethoxazole, sulfathiazole) 및 대사체 5종($N^4$-acetylsulfadiazine, $N^4$-acetylsulfamerazine, $N^4$--acetylsulfamethazine, N4-acetylsulfamethoxazole, sulfamethoxazole-$N^1$-glucuronide)의 분석방법을 연구하였다. 다양한 전처리 조건을 비교 검토하여 최적의 동시 추출방법과 LC/ESI-MS/MS를 이용한 기기분석 조건을 확립하였다. 카트리지별 추출효율은 C18 카트리지에서 12~94%, HLB 카트리지에서 60-95%, MCX 카트리지에서 25-123%였으며, MCX와 HLB 카트리지를 결합한 방법의 회수율은 70-90%이었다. HLB를 선택하여 확립된 분석방법의 회수율은 66~115%, 상대표준편차 5~17%이었으며, 방법검출한계는 0.001~0.187 ng/mL 였다. 이를 실제 환경시료에 적용한 결과 4종의 설폰아미드 항생제가 0.008~2.153 ng/mL 농도범위로 검출되었으며, 대사체의 경우 $N^4$-acetylsulfamethoxazole이 확인되었으나 검출한계 미만으로 관찰되었다.

도시하수 및 그 주변 하천 환경 중 항생제 내성 세균 노출 특성 (Characteristics of Antibiotic Resistant Bacteria in Urban Sewage and River)

  • 본 연구는 도시 하수처리장의 미생물 군집 내 항생제 내성 세균의 특징과 하천으로의 항생제 내성 세균 노출의 계절적 변화의 특성을 평가하였다. 일반 종속영양세균 배양을 위한 R2A agar (R2A)와 대장균군을 선택하여 배양하는 MacConkey sorbitol agar (MSA)에 항생제를 첨가 하여 제작한 배지에 하수처리장 시료를 도말하여 배양한 결과 모든 시료에서 다제 항생제 내성 세균을 검출 해 낼 수 있었다. Ampicillin과 sulfathiazole의 내성률이 다른 나라에 비해 높게 측정 되었으며 시료내 유기물의 정도, 사용된 배지에 따라 내성률이 다름을 볼 수 있었다. R2A 배지에서 분리된 다제 항생제 내성 세균은 모두 기존에 알려진 병원성 세균과 그 염기서열이 유사한 것으로 볼 때 병원성 세균의 항생제 내성 정도가 일반 세균보다 높음을 본 연구 결과로 보일 수 있었다. 또한 본 연구에서는 하수처리장이 하천에 미치는 유해성을 계절별로 관찰하여 전체 미생물중 항생제 내성 세균의 비율은 수온과 비례한다는 결과를 얻었다. 이 결과는 지구 온난화가 미생물 유해성을 증가시킬 가능성을 시사한다.

설폰아미드계 항생물질의 오존산화분해에 대한 계산화학적 해석 및 실험적 검증 (Degradation of Sulfonamide Antibiotic Substances by Ozonation: An Experimental and Computational Approach)

  • 본 연구에서는 상수원에서 문제시 되고 있는 미량오염물질의 처리에 대한 연구를 하기 위해선 많은 시간과 비용이 소요되는데, 이를 절감하기 위한 대안으로 양자화학적 기반의 범밀도함수이론(Density Functional Theory, DFT)을 활용하여 물질간의 상호 반응성 및 분해과정을 해석하였다. 본 연구에서 다루고 있는 물질은, 최근 낙동강 수계에서 빈번히 검출되고 있는 Sulfonamide 물질 3종(sulfamethazine, sulfathiazole, sulfamethoxazol)을 선정하였으며, 이론적인 연구로는 DFT모델링, 실험적 연구로는 UV-VIS 및 FT-IR 등의 분광분석을 하여 비교 및 검증을 하였다. DFT모델링을 실시한 결과 Sulfonamide물질의 HOMO(highest occupied molecular orbital)와 오존의 LUMO (lowest unoccupied molecular orbital) 사이에서 반응이 가장 유리하며, Sulfonamide 물질의 HOMO를 가시화 한 결과 Sulfanilamide기에서 전자밀도가 높게 나타나므로 Sulfanilamide기에서 반응이 활발할 것이라 예측되었다. UV-VIS 실험결과 260 nm에서 Sulfanilamide기가 검출되었으며, 오존산화시 검출된 Sulfanilamide기가 빠르게 사라짐을 알 수 있었다. FT-IR분석결과로써 Sulfanilamide기에서도 그 한 부분인 아민기(N-H)에서 가장 활발한 제거반응이 일어남을 알 수 있었으며 이러한 결과로부터 DFT모델링 방법을 통해서 정수처리 공정에 대해서 반응을 예측할 수 있음을 확인하였다.

사용량과 잠재적 환경 유출 가능성을 기준으로 한 축산용 항생제의 우선순위 결정 (Prioritizing Environmental Risks of Veterinary Antibiotics Based on the Use and the Potential to Reach Environment)

  • 가축 질병의 예방 및 치료뿐만 아니라 사료 이용효율 증대와 생장 촉진을 위하여 축산용 항생제가 쓰여져 왔으며, 이 항생제는 가축분뇨를 통해 배출되어 농경지에 유입될 가능성이 있다. 나아가 용탈과 표면 유거를 거쳐 수계에 도달할 수 있으나, 모든 항생제의 환경 유출 가능성을 검토하는 데에는 많은 시간과 노동력, 예산이 필요하다. 본 연구는 축산용 항생제의 사용량과 잠재적 환경 유출 가능성을 기준으로 우선 순위를 결정하고자 수행되었다. 2004년에 우리 나라에서 판매된 항생제는 83종으로 약 1,400 톤에 달하였으며, 이 가운데 25 톤 이상 사용된 항생제는 10종이었다. 3 톤 이상 쓰인 36 종의 항생제를 대상으로, 가축의 배출율과 토양 흡착 정도를 가지고 잠재적 환경 유출 가능성을 판단하여 17 종을 선발하였다. 축산용 항생제의 사용량과 잠재적 환경 유출 가능성을 종합하여 검토한 결과, amoxicillin과 carbadox, chlortetracycline, neomycin, oxytetracycline, sulfamethazine, sulfathiazole, tylosin 등 8종이 상대적으로 환경으로의 유출 가능성이 높아 금후 정밀한 평가가 요구되었다. 비록 항생제의 가축분뇨로의 배출율과 토양내 동태에 대한 정보가 제한적일지라도, 본 연구에서는 한국에서 쓰이고 있는 축산용 항생제의 사용량과 알려진 항생제의 배출율과 토양 흡착 정도를 바탕으로 환경으로의 유출 가능성이 큰 항생제를 결정하였다.

절박 도축우의 항생제 및 설파제 잔류 조사 (Detection of residual antibiotic and sulfonamide agents in emergency slaughtered cattle)

  • This survey was carried out to detect the residual antibiotics and sulfonamides in emergency slaughtered cattle(n=265) from slaughter houses in Kyeonggi province by EEC 4-plates method, Charm II and HPLC. The results were summarized as follows ; 1. Antimicrobial substances were detected from 24 samples(9.1%) by EEC 4-plates method and the detection ratio were highest in summer(13.8%). 2. Twenty-two of the 24 positive samples were classified as sulfonamide(34.4%), TCs(31.3%), $\beta$-lactam(23.5%) and aminoglycoside(9.3%) by Charm II test. 13(59.1%) of the 22 samples contained single agent and 9 samples(40.9%) contained 2 or more agents. 3. Oxytetracycline(27.3%), penicillin G (27.3%) and sulfathiazole(18.2%) were detected in 20 from 22 samples by the HPLC and Charm II test. 4. The residual concentration of oxytetracycline, penicillin G and sulfonamide were 0.29~9.30 ppm, 0.05~9.58 ppm and 0.04~7.59 ppm, respectively and 19 samples(7.2%) were exceeded tolerance levels.

우분 퇴비공장 주변 농경지 및 수계의 계절별 잔류 항생물질 모니터링 (Seasonal Monitoring of Residual Antibiotics in Soil, Water, and Sediment adjacent to a Cattle Manure Composting Facility)

  • Overuse of veterinary antibiotics threats public health and surrounding environment due to the occurrence of antibiotic resistant bacteria. The objective of this study was to evaluate the antibiotic's concentrations of tetracycline (TC), chlortetracycline (CTC), and oxytetracycline (OTC) in a tetracycline group (TCs), sulfamethazine (SMT), sulfamethoxazole (SMX), and sulfathiazole (STZ) in a sulfonamide group, lasalocid (LSL), monensin (MNS), and salinomycin (SLM) in a ionophore (IPs), and tylosin (TYL) in a macrolide (MLs) group from soil, water, and sediment samples adjacent to a cattle manure composting facility. For all samples of soil, water, and sediment, the highest concentrations were detected in TCs among the tested antibiotics because of its higher annual consumption in veterinary farms, Korea and its higher cohesiveness with divalent or trivalent cations in soil. Moreover, the concentrations of residual antibiotics in September were generally higher than in June because of heavier rainfall in June. We suggest that continual monitoring and developing guideline of antibiotics are needed to control residual antibiotics in the environment.

Monitoring of Antibiotics in the Soil and Sediment Near at the Animal Feeding Operation and Wastewater Treatment Plant

  • Antibiotics have been used for treating human and animal disease and enhancing growth of cattle, swine, and poultries. However, overused antibiotics can be released into the environment and produce antibiotic resistance genes (ARGs) in the environment. Main objective of this research was to monitor residual of antibiotics in solid matrix such as manure, wastewater sludge, soil and sediment. Total of six antibiotics, Chlortetracycline (CTC), Oxytetracycline (OTC), Tetracycline (TC), Sulfamethazine (SMT), Sulfamethoxazole (SMX), Sulfathiazole (STZ), used for both human and animal was monitored. Result showed that the detection frequency of 6 antibiotics was ordered SMT (100%) > TC = CTC (75%) > OTC (38%) > STZ (13%) > SMZ (0%) and the highest concentration ($309.83{\mu}g\;kg^{-1}$, SMT) was observed in manure. Comparing residual concentration of antibiotics (TC, CTC, and OTC) in soil and sediment, higher concentration was observed in sediment indicating that dissolved forms of antibiotics are released into river and sorbed into sediment particle. In conclusion, monitoring for residual of antibiotics in the environment is necessary and more research should be conducted to verify the source of antibiotic release.

잔류 Chloramphenicol 검사용 효소 면역측정법의 개발에 관한 연구 (Study on Development of Enzyme-Linked Immunosorbent Assay for the Screening of Chloramphenicol Residues)

  • The monoclonal antibody to chloramphenicol(CAP) was produced to develop an enzyme-linked immunosorbent assay(ELISA) for residual CAP. An immunogen(CAP-BSA) was prepared by immunogen, antibody titer was measured by indirect ELISA. Spleen cells form the immunized mouse were fused with SP2/OAg14 myeloma cells. Among hybridomas selected in HAT media, 6 clones shown high antibody titer to CAP were subjected to cloning by limit dilution, and all of the monoclonal antibodies(MCA1, 2, 3, 4, 5, 7 and 9) produced by each clone were identified as IgG1 by ELISA isotyping analysis. Competitive ELISA condition was established by using the purified monoclonal antibody MCA1 as primary antibody and CAP-HSA conjugate as coating antigen. Standard curve of CAP(n=28) showed that the lowest detection limit of CAP is 20ng/ml level. The cross-reactivities of the 6 monoclonal antibodies showed that CAP sodium succinate. CAP base, P-nitrophenol, and p-nitrobenzyl alcohol were 89∼178, 0.050∼2.237, 0.056∼0.794 and 0.013∼7.939%, respectively. No cross-reactivities were observed with phenylalanine, tyrosine, glutamine, thiamphenicol, neomycin, streptomycin, gentamicin, sulfamethazine, sulfathiazole, chlortetracycline and p-aminobenzoic acid.

축산식품 중에 잔류하는 Gentamicin 검사를 위한 ELISA 개발에 관한 연구 (Competitive Enzyme-Linked Immunosorbent Assay for Detection of Gentamicin Residues in Edible Animal products)

  • An enzyme-linked immunosorbent assay(ELISA) was developed for the detection of residual gentamicin(GM) in edible animal products. The immunogen(GM-KLH conjugate) and coating antigen(GM-BSA conjugate) were prepared by coupling GM sulfate to keyhole limpet hemocyanin(KLH) and bovine serum albumin(BSA) in the presence of 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride, respectively. Polyclonal antibody to GM was produced in rabbits(New Zealand White, female) by using the immunogen and the antibody titer was measured by indirect ELISA. A competitive ELISA was developed using GM-bovine serum albumin conjugate as a coating antigen, GM(as standards or sample), polyclonal antibody to GM, secondary antibody conjugated with horseradish peroxidase as an enzyme, and H2O2 and o-phenylenediamine dihydrochloride as a substrate and a chromophore, respectively. The detection limit of GM was 10 ng/ml and the standard curve of GM(n=26) was linear up to 10 $\mu\textrm{g}$/ml in this competitive ELISA system. There were no cross-reactivities of the partially purified antibody between GM and the various antibiotice such as amikacin, benzyl-penicillin, chloramphenicol, erythromycin, furazlidone, kanamycin, neomycin, oleandomycin, streptomycin, sulfathiazole and thiamphenicol(CR50<0.05%)

HPLC와 LC/MS에 의한 식육내 잔류 설파제의 동시 분석법 (Simultaneous analysis of sulfonamides in beef and pork by high performance liquid chromatography and electrospray ionization mass spectrometry)

  • A multiresidual analysis was performed to determine 12 sulfonamides(sulfacetamide, sulfadiazine, sulfisomidine, sulfathiazole, sulfapyridine, sulfamerazine, sulfamethazine, sulfamonomethoxine, sulfisoxazole, sulfamethoxazole, sulfaquinoxaline, and sulfadimethoxine) in beef and pork simultaneously. The multiresidual analysis for the sulfonamides currently used was able to analyze 5 kinds of sulfonamides at the same time. The method of this 12 sulfonamides multiresidual analysis in this study was matrix solid-phase dispersion(MSPD) by high performance liquid chromatography (HPLC) and liquid chromatography mass spectrometry (LC/MS). The recovery rate of the materials was measured by MSPD method with 3 different extraction solvents; Dichloromethane, DCM: Ethylacetate(3:1), DCM:EA(9:1). Also, samples (84 beef and 205 pork samples) which were positive by EEC-4 plate test from 2001 to 2003 were tested to investigate the kinds of sulfonamides using HPLC. The results from the study were as follows; 1. The recovery rate of the materials was measured by MSPD method with 3 different extraction solvents; Dichloromethane, DCM:Ethylacetate(3:1), DCM:EA(9:1). The method of extraction solvent with DCM:ethyl acetate(9:1) was the most excellent(87.7∼99.3%) in separation and reappearance. 2. In the LC/MS analysis. of sulfonamides, signal to noise ratio was showed relatively high in the positive mode and special ion in the quality analysis was determined via [M+H]$\^$+/ and m/z 156. A spectrum of sulfonamides was showed from all 12 sulfonamides. 3. The samples positive by the EEC-4 plate, a screening test method, were categorized by sulfonamides through Charm II and confirmed the kinds of sulfonamides through HPLC. 1) Among 84 beef samples positive by EEC-4 plate, 20 samples were positive by Charm II and identified as 7 sulfamethazine, 9 sulfadimethoxine, 1 sulfamonomethoxine and 3 unknown status. 2) Among 205 pork samples positive by EEC-4 plate, 42 samples were positive by Charm II and identified as 19 sulfamethazine, 1 sulfadimethoxine, 4 sulfamonomethoxine and 5 unknown status.
