Measurement of Quality Parameters of Honey by Reflectance Spectra

  • The objectives of this study were to develop models to predict quality parameters of Korean bee-honeys by visible and NIR spectroscopic technique. Two kinds of bee-honey fronl acacia and polyflower sources were tested in this study. The honeys were harvested in the spring of 2000 and stored in the storage facility at 20$^{\circ}C$ during experiments. Total of 394 samples of honey were analyzed. Reflectance spectra, moisture contents, ash, invert sugar, sucrose, F/G (fructose/glucose) ratio, HMF (hydroxymethyl furfural), and C12/C13 ratio of honeys were measured. The average values for the tested honeys were 19.9% of moisture contents, 0.12% of ash, 68.4% of invert sugar, 5.7% of sucrose, 1.27 of F/G(fructose/glucose) ratio, 14.4 mg/kg of HMF, and -19.1 of C12/C13 ratio. A spectrophotometer, equipped with a single-beam scanning monochromator (NIR Systems, Model 6500, USA) and a horizontal setup module, was used to collect reflectance data from honey. The reflectance spectra were measured in wavelength ranges of 400∼2,498 nm. with 2 nm of interval. Thirty-two repetitive scans were averaged, transformed to log(1/Reflectance), and then were stored in a microcomputer file, forming one spectrum per measurement. A sample cell and reflectance plate were made to hold honey samples constantly. Spectra of honey samples were divided into a calibration set and a validation set. The calibration set was used during model development, and the validation set was used to predict quality parameters from unknown spectra. The PLS(Partial Least Square) models were developed to predict the quality parameters of honeys. The first and the second derivatives of raw spectra were also used to develop the models with proper smoothing gap. The MSC (multiplicative scatter correction) and the SNV & Dtr.(standard normal variate and detranding) preprocessing were applied to all spectra to minimize sample-to-sample light scatter differences. The PLS models showed good relationships between predicted and measured quality parameters of honeys in the wavelength range of 1100∼2200 nm. However, the PLS analysis was not good enough to predict HMF of honeys.

팽이버섯으로부터 Lectin의 정제와 특성 (Purification and Characterization of the Lectins from Mushroom Flammulina velutipes)

  • 식용버섯인 팽이버섯(Flammulina velutipes)으로부터 적혈구 응집능력을 갖는 렉틴(flammulina velutipes lectin, FVL) FVL-1과 FVL-2을 정제하였다. FVL-1과 FVL-2의 분자량은 각각 10.6 kDa과 37 kDa으로 추정되었다. FVL-2는 사람적혈구의 경우 모든 혈액형에서 응집현상을 나타내었으나, FVL-1은 O형의 적혈구에 대하여 높은 상대활성도를 나타내었다. 렉틴의 hemagglutination inhibition test 결과 FVL-1과 FVL-2는 fetuin, bovine submaxillary mucin, asialofetuin, porcine stomach mucin에 의해 적혈구 응집력이 저해되었다. FVL-1은 1.56 mM $Cu^{2+}$에서 적혈구 응집력이 저해되었으며, $Mg^{2+}$, $Mn^{2+}$, $Ca^{2+}$, $Fe^{2+}$ 등에서도 저해되었다. 반면 FVL-2는 금속 이온에 대한 저해가 나타나지 않았다. pH 안정성에 있어서 FVL-1은 pH $5{\sim}11$에서 안정하였으며, FVL-2는 pH 4까지의 범위에서 안정하였다. 온도 안정성에 있어서는 FVL-1과 FVL-2 각각 $55^{\circ}C$, $45^{\circ}C$까지 안정하였다. FVL-1은 단백질이었으며 FVL-2는 당 단백질이었다.

능이버섯의 영양성분에 관한 연구 (Studies on the Nutritonal Components of Mushroom(Sarcodon aspratus))

  • 본 연구는 능이버섯의 식품영양학적 및 기능성을 평가하기 위한 기초적인 연구로서 영양 성분의 함량을 조사하였다. 능이버섯의 일반성분은 수분 89.93%, 회분 1.18%, 조단백질 3.67%, 조지방 0.96%, 탄수화물 4.26%로 나타내었다. 능이버섯의 주요 유리당은 glucose, sucrose, trehalose, xylose 로 구성되어 있었고, trehalose의 함량이 가장 높았다. 능이버섯의 총아미노산의 함량은 796.85mg/100g-fr.wt.이었고, 필수 아미노산의 함량은 300.77mg/100g-fr.wt.으로 valine, leucine, threonine, Iysine의 함량이 비교적 높았고, 비필수아미노산은 486.08mg/100g-fr.wt.으로 alanine, glycine, aspartic acid, glutamic acid 등의 함량이 비교적 높았다. 총 유리 아미노산중에 필수아미노산과 비필수아미노산의 함량은 각각 124.95, 138.52mg/100g-fr.wt.이며, methionine, Iysine, valine, arginine, Aspartic acid, tyrosine 등의 함량이 높았다. 능이버섯에 존재하는 유리아미노산 유도체의 함량은 46.81 mg/100g-fr.wt.이었고, omithine, sarcosine, $\beta$-alanine, phosphoserine의 함량이 비교적 높았다. 능이버섯의 총 비타민 C 함량은 5.43 mg/100g이었고, 환원형과 산화형 비타민 C는 각각 3.50과1.93mg/100g이었고, 무기질 함량은 K, Na이 매우 높았고, 이들 함량은 각각 375.73, 61.82mg/100g이었다.

민자주방망이버섯(L. nuda)의 영양성분 분석 (Analysis of Nutritional Components of Lepista nuda)

  • 민자주방망이버섯의 식품 영양학적 기능적 평가를 위한 기초연구로서 영양 성분의 함량을 조사하였다. 일반성분은 수분 $90.90{\pm}0.09%$, 탄수화물 $4.38{\pm}0.07%$, 조단백질 $3.70{\pm}0.03%$, 조회분 $0.55{\pm}0.04%$ 그리고 조지방이 $0.47{\pm}0.01%$로 분석되었다. 환원당의 함량은 636.17 mg이었고 유리당은 galactose(51.98 mg), trehalose(22.48 mg), maltose(8.48 mg), mannose(7.96 mg), 그리고 arabinose(6.52 mg) 등의 순으로 모두 97.42mg이 함유되어 있었다. 무기질은 K와 Mg가 각각 103.10 mg과 56.01 mg으로 다른 것에 비하여 함유량이 높았다. 민자주방망이버섯의 구성아미노산은 총 310.39 mg이었고, 이 중 필수아미노산이 111.50 mg이었으며, valine(25.40 mg), leucine(22.52 mg), lysine(21.34 mg) 등의 함량이 상대적으로 높았다. 비필수아미노산은 총 198.89 mg이었으며, glutamic acid(60.72 mg), aspartic acid(30.17 mg), alanine(29.72 mg) 등의 함량이 높았다. 유리아미노산은 총 220.86 mg이었으며, 필수아미노산이 36.21 mg으로 isoleucine(21.57 mg), threonine(11.20 mg)의 함량이 높았으며, 비필수아미노산은 184.65 mg으로 glutamic acid(42.78 mg)와 aspartic acid(32.44 mg) 등의 함량이 높았다. 아미노산 유도체의 함량은 총 187.85 mg으로서 hydroxyproline이 121.50 mg으로 가장 높았으며 그 외 citrulline(15.58 mg), ornithine(15.41 mg), $\alpha$-aminoisobutyric acid(11.23 mg) 등의 함량이 높은 것으로 분석되었다.

싸리(Lespedeza bicolor) 줄기의 성분 분석 (Analysis on the Components in Stem of the Lespedeza bicolor)

  • 싸리의 생리적 기능과 이용가능성에 대한 연구의 일환으로 싸리 줄기의 일반성분과 환원당, 유리당, 무기질 그리고 아미노산의 함량 등에 대하여 분석하였다. 일반성분은 탄수화물이 82.35$\pm$ 0.17$\%$로 가장 높았으며, 수분 13.38$\pm$ 0.29$\%$, 조단백질 2.80$\pm$ 0.04$\%$, 조회분 0.86$\pm$ 0.02$\%$ 그리고 조지방이 0.61$\pm$0.02$\%$로 나타났다. 환원당의 함량은 147.58 mg/100 g이었고, 유리당의 총 함량은 52.4 mg/100g으로서 fruc-tose 31.0 mg/100 g, glucose 16.7 mg/100 g, sucrose 4.7mg/100 g으로 구성되어 있는 것으로 나타났다. 무기질 함량은 Ca가 472.97 mg/100 g으로 가장 높았고 그 다음으로 K가 205.70 mg/100 g 함유되 어 있었으며 , 그 외 Zn(25.59 mg/100g), Mg(10.28 mg/100 g), Mn(4.04 mg/100 g), Na(3.97mg/100g), Fe(2.21 mg/100g) 등이 검 출되었다. 구성아미노산은 필수아미노산 31.74 mg/100 g과 비필수아미노산 117.24 mg/100 g으로 총 148.95 mg/100 g이 함유되어 있었다. 유리아미노산은 필수아미노산이 8.41 mg/100 g, 비필수아미노산 97.98 mg/100 g으로 총 106.39 mg/100 g으로 측정되었으며, 이 중에 proline의 함량이 63.92 mg/100 g으로서 전체 유리아미노산의 59.8$\%$를 차지하는 것으로 분석되었다. 아미노산 유도체의 함량은 30.01 mg/100 g으로서 $\gamma$-aminoisobutyric acid의 함량이 12.57 mg/100 g으로 가장 높았고, $\beta$-aminoisobutyric acid가 6.72 mg/100 g, phos-phoserine이 6.12 mg/100 g 함유되어 있는 것으로 나타났다.

  • Nitrogen compounds of Panax ginseng and their biological activities in plant and animal were reviewed. Major nitrogen compounds found in P. ginseng are free amino acids. Water solilble proteins, indouble proteins and peptides. Minor nitrogen compounds are dencichine. Glycolyroteins, amines, alkaloides, methoxy or alkyl pyrazine derivatives, free nucleosides and nucleic acid bases. 4-methyl-i-thiazoltethanol and pyroglutamic acid the contents of total nitrogen and protein in root Increased until 13 years old which was the highest age tinder investigation. Soluble protein content increased with the root weight and was higher in xylem pith than cortex-epidermis indicating the close relation with root growth. Arginine, which covered 58% of total free amino acids, may serve as storage nitrogen. Arginine seems to be changed into proline in rhizome. threonine in stem and again threonine and arginine in leaf. The greater the root weight the higher the polyamine stimulated Polyamine stimlllated the growth of root callus. Physiological roles of other minor nitrogen compounds are unknown although content is relatively high ((1.if) 6.w). Biochemical and pharmacological activities of some nitrogen compounds for animal were more investigated than physiological role there plant itself. Radiation and U.V protective function (heat stable protein). insulin-like activity in lipogenesis and livolysis (adenosine and pyroglutamic acid), depression of blood sugar content (glycopevtide). htmostatic and nellrotoxic activity (dencichine) and, sedative and hypnotic activity (4-methyl-i-thiazoleethanol) are reported. Heat stable protein increased with root age. The traditional quality criteria appear to be well in accordance with biological activities of nitrogen compounds. Chemical studies of nitrogen compounds seem relatively rare, probably due to difficulty of isolation, subsequently the investigations of biological activities are little.

Alumina Column Chromatography와 HPLC에 의한 토마토의 Dehydrotomatine 및 ${\alpha}$-Tomatine 단리방법 연구 (Analytical Methods for the Isolation of Dehydrotomatine and ${\alpha}$-Tomatine in Tomato Fruits by Use of Alumina Column Chromatography and High-Performance Liquid Chromatography)

  • Tomato fruits(Lycoperisicon esculentum) synthesize the glycoalkaloids dehydrotomatine and ${\alpha}$-tomatine, possibly as defense against bacteria, fungi and insects. We developed a new effective method to prepare and purify dehydrotomatine and ${\alpha}$-tomatine that exists in tomato fruits using alumina column chromatography and high performance liquid chromatography (HPLC). The tomato glycoalkaloids(TGA) in tomato was extracted with 2% acetic acid, and then precipitated with ammonium hydroxide(pH=10.5). The dry precipitate substance was applied on alumina column, and then fractionated with water saturated n-butylalcohol. The TGA(Fr. No. 26~36) were collected and dried under reduced pressure. The TGA was performed on a reverse phase HPLC(Inertsil ODS-2, $5\;{\mu}m$), eluted with acetonitrile/20mM $KH_2PO_4$(24:76, v/v) at 208 nm. Two peaks were detected on HPLC, and individual peak was collected by repeating HPLC. Furthermore, to confirm the identity dehydrotomatine and ${\alpha}$-tomatine, each peak isolated was hydrolyzed with 1N HCl into sugar and aglycone tomatidine. The sugars were converted to trimethylsilyl ester derivatives. The nature and molar ratios of sugars were identified by gas-liquid chromatography(GLC) and the aglycone by high-performance liquid chromatography(HPLC). The first peak (Rt=17.5 min) eluted from HPLC was identified as dehydrotomatine, and second peak(Rt=21.0 min) was as ${\alpha}$-tomatine. This technique has been used effectively to prepare and isolate dehydrotomatine and ${\alpha}$-tomatine from tomato fruits.

H-Y 항원의 정제 및 특성규명에 관한 연구 (Studies on the Purification and Characterization of H-Y Antigen)

  • These studies were carried out to investigate the properties of H-Y antigen purified by immunoaffinity chromatography using monoclonal H-Y antibody. Immunoaffinity column was prepared by the coupling of monoclonal antibody to the Aminolink Coupling Gel. Murine testis supernatant was applied onto the column and eluted by O.lM glycine-HCl buffer and 31${\mu}g$ of H-Y Ag was eluted from one testis. Purified H-Y Ag strongly reacted with Con A and lentil from 6 different kinds of lectins tested, which may indicate that sugar moiety of H-Y Ag is composed of glucose, mannose and their derivatives. Con A-sepharose affinity column was used to purified H-Y Ag based on that H-Y Ag is glycoprotein. The fraction eluted by 0.2M Me-${\alpha}$-D-mannoside from the column loaded with murine testis supernatant was identified to be H-Y Ag by dot blot test. Molecular weight of the purified H-Y Ag was estimated by Sepharose G-75 gel filtration and SDS-PAGE, and showing that it was about 67,000 dalton. In fluorescence test, the ratio of XY embryos and XX embryos was 1:1.
