Influence of Nutrient Supply on Growth, Mineral Nutrients and Carbohydrates in Cucumber (Cucumis sativus L.) (무기영양액 농도 차이가 오이 생육, 무기성분 흡수 및 탄수화물 합성에 미치는 영향)

  • We investigated the growth of cucumber plants, the uptake and use of mineral nutrients, such as $NO_3$-N, $NH_4$-N, $K^+$, $Ca^{+}^{+}$, $Mg^{+}^{+}$ and $Na^+$, absorbed from media solution, and the synthesis and distribution of soluble sugars under nutrient-deficient condition. Difference in plant growth revealed after 20 days of treatment. Nitrate uptake in nutrient-deficient condition was significantly reduced compared with nutrient-normal treatment, and its distribution was primarily in petioles, stem, roots and less in leaves. In contrast, ammonium content was markedly predominated in fast growing organs, and it was significantly different in growing leaves, expanded leaves, and roots under similar growth conditions. $K^+$, lack by deficient nutrient condition, was found in growing leaves. The $Ca^{+}^{+}$ content did not show significant difference between treatments and a substantial portion of $Ca^{+}^{+}$ remained in petioles. The $Mg^{+}^{+}$ content was significantly higher in the leaves of nutrient-normal condition compared with nutrient-deficient condition while significantly lower in stem and roots. The behavior of $Na^+$ in plant was similar to $K^+$ although its content was relatively little. The highest $CO_2$ assimilation was observed in fully expanded leaves of nutrient-normal condition, which was 1.7 times higher compared with nutrient-deficient condition. The instantaneous water use efficiency (A/E) and the A/gsratio, which is an index of leaf intrinsic water use efficiency for individual leaves, was 1.2 and 1.1 times higher, respectively. The total soluble sugar (TSS) contents were highest in leaves followed by petioles, stems and roots, and in younger leaves. The growing leaves contained about 7,200 mg $kg^{-1}$ of TSS in nutrient-normal condition whereas the TSS contents in nutrient-deficient condition were not significantly different between leaves. The $Mg^{+}^{+}$ and $NH_4$- N were positively correlated with the TSS whereas $NO_3$ - N was negatively correlated.

Mycelial Growth Using the Natural Product and Angiotensin Converting Enzyme Inhibition Activity of Pleurotus eryngii (천연물을 이용한 큰느타리 균사배양 및 Angiotensin Converting Enzyme 저해활성)

  • To develop the health/functional food materials, we investigated the cultural condition of mycelial growth on the solid state fermentation using the brown rise, Acanthopanax sp. and Artemisia sp., and also evaluated inhibitory activity of angiotensin converting enzyme (ACE) of hot water extracts from cultured media of Pleurotus eryngii. As the amount of Acanthopanax nnd Artemisia In the cultural media increased, the mycelial growth rate decreased. Especially, addition of Aeantopanax showed marked effect than Artemisia. Moisture contents in three kinds of cultured media were in the range of $10.9{\sim}12.0%$. Crude protein fat and crude fiber content were the highest value in cultured brown rice medium, whereas the mineral contents (Ca, K and P) were higher in the Acanthopanax supplemented (5%) medium than the other media, The extraction yield of the Artemisia supplemented (5%) medium was the highest value of 4.80%, and the pH of hot water extract from cultured brown rice medium showed the lowest value of 6.1. Lightness (L) values in three kinds of extracts from cultured media were in the range of $85.8{\sim}87.1$. Redness (a) value was the highest In the brown rice and Acanthopanax supplemented media, however cultured Artemisia supplemented medium showed the highest value in yellowness (b). In comparison of sugar components analyzed by the thin layer chromatography with three kinds of samples, two spots were detected to be glucose and maltose, respectively. The ACE inhibitory activity of hot water extract from the cultured Acanthopanax supplemented medium showed the highest value at the concentration of $0.2{\sim}1.0\;mg/ml$. These results suggest that the Pleurotus eryngii grew in natural media using brown rice and Acanthopanax can be supplemented to the brown rice medium to enhance its ACE inhibitory activity as health/functional food materials.

Quality characteristics of fried fish paste with squeezed Aronia melanocarpa juice (아로니아 착즙액 첨가 튀김어묵의 품질특성)

  • This study was carried out to investigate the quality characteristics of fried fish paste prepared with different amounts of squeezed Aronia melanocarpa juice. Squeezed Aronia melanocarpa juice (AMJ) was incorporated into fish paste at different levels (containing 2, 7, and 12 g of Aronia melanocarpa juice in 2 AMJ, 7 AMJ, and 12 AMJ, respectively) based on the total weight of water. Sugar contents and total acidity increased with increasing AMJ content. With increasing amounts of AMJ in fried fish paste, L value inside and on the surface decreased, a value increased, and b value inside decreased, whereas b value on the surface increased. pH decreased with increasing levels of AMJ. As the result of textural properties, folding test in all samples showed that AA means good flexibility. The strength, hardness, and chewiness of fried fish paste with AMJ increased while cohesiveness was not significantly different. Total polyphenol contents increased with higher levels of AMJ. DPPH radical scavenging activity was significantly higher than those of the control. In the sensory evaluation, fried fish paste containing 7 AMJ received the highest score than both the control and other samples.

Changes of Ginsenosides and Physiochemical Properties in Ginseng by New 9 Repetitive Steaming and Drying Process (새로운 자동 구증구포방법에 의한 인삼사포닌의 변환 및 이화학적 특성)

  • This study was conducted to investigate the contents of ginsenosides and physiochemical properties of Panax ginseng after 9 times steaming and drying treatment by using the new auto steamer which is more fast and simple than previous report. In the process of steaming and drying, the content of six major ginsenosides such as Rg1, Re, Rb1, Rc, Rb2 and Rd were gradually decreased. On the other hand, the content of seven minor ginsenosides includes Rh1, 20(S)-Rg2, 20(R)-Rg2, 20(S)-Rg3, 20(R)-Rg3, Rk1 and Rg5 were gradually increased. We observed the protopanxadiol ginsenosides such as Rb1, Rb2, Rc and Rd were converted into 20(S)-Rg3, 20(R)-Rg3, Rk1 and Rg5; similarly protopanxatriol ginsenosides of Rg1 and Re were converted into Rh1, 20(S)-Rg2 and 20(R)-Rg2. Based on the result of fresh ginseng, the contents of reducing sugar, acidic polysaccharide and total phenolic compounds were gradually increased and reached to maximum at 7 times repetitive steaming process of the fresh ginseng. Whereas DPPH radical scavenging activities were gradually decreased to 68% at 7 times steaming. New auto 9 repetitive steaming and drying process has similar production with original methods, but content of benzo(a)pyrene were not almost detected comparatively taking less time. The present results suggested that this method is best for the development of value-added ginseng industry related products.

Effect of Particle Size on Physico-Chemical Properties and Antioxidant Activity of Corn Silk Powder (옥수수수염 분말의 입자크기별 이화학적 특성과 항산화활성)

  • The study was carried out to analyze the relationship between analysis of antioxidant activity and the level of functional components according to particle size of corn silk. Particle size was classified into 5 groups. By particle size distribution and color difference, the total phenol content and DPPH radical scavenging activity were observed. The particle sizes of corn silk were $199.17{\mu}m$, $178.27{\mu}m$, $85.48{\mu}m$, $27.4{\mu}m$ and $20.97{\mu}m$, respectively. The lightness of colored pigments was increased when the particle size was decreased. The contents of free sugar (fructose, glucose, galactose, sucrose, and maltose) of corn silk were analyzed using a HPLC. The total phenol contents by the particle sizes of corn silk were 2.01 mg/g, 2.02 mg/g, 2.06 mg/g, 2.26 mg/g and 2.26 mg/g, respectively. DPPH radical scavenging activities of samples were 21.00%, 21.75%, 22.90%, 24.35% and 23.67%, respectively. Antioxidative activities of Trolox and Fe(II) in corn silk were measured by ferric reducing antioxidant power (FRAP) assay and Trolox equivalent antioxidant capacity (TEAC) assay. TEAC values of samples were $2.36{\mu}mol$ TE / g dw, $2.81{\mu}mol$ TE / g dw, $3.20{\mu}mol$ TE / g dw, $3.36{\mu}mol$ TE / g dw, and $3.44{\mu}mol$ TE / g dw, respectively. FRAP values of samples were $11.67{\mu}mol$ Fe(II) / g dw, $12.80{\mu}mol$ Fe(II) / g dw, $13.43{\mu}mol$ Fe(II) / g dw, $13.85{\mu}mol$ Fe(II) / g dw and $15.95{\mu}mol$ Fe(II) / g dw, respectively. Total phenolic content and antioxidantive activities based on FRAP assay and TEAC assay were increased with decreasing particle size. In addition, DPPH radical scavenging activity was also increased. A significant correlation was also noted between DPPH radical scavenging activities and the content of phenolic compounds.

Variation in Quality and Preference of Sogokju (Korean Traditional Rice Wine) from Waxy Rice Varieties (찰벼 품종에 따른 소곡주의 품질 및 기호도 변이)

  • This study was carried out to compare the physicochemical characteristics and preference as a sensory quality of Sogokju (Korean traditional rice wine) from waxy rice varieties. The protein and moisture contents of milled waxy rice varieties were ranged 6.9~7.2% and 12.1~ 12.6%, respectively. Nunbora had the largest grain size. In pasting properties, Hangangchalbyeo had the highest peak, trough and final viscosities, and Dongjinchalbyeo had the lowest viscosity curve. These differences suppose to be caused by the amylopectin(AP) structure: Dongjinchalbyeo has the largest short AP chains (degree of polymerization (DP) 6-12) and the smallest middle AP chains (DP 13-24) in 9 waxy rice varieties, while Hangangchalbyeo has the smallest short AP chains and the largest middle AP chains. The alcohol contents of Sogokju brewed from 9 waxy rice varieties were 17.6~19.9%. The brix degree were ranged $20.5{\sim}23.9^{\circ}Bx$. The organic acid of Sogokju consisted mainly of succinic acid, and the free sugar of it consisted mostly of glucose. The sensory evaluation showed the highest palatability at the Sogokju from Baegseolchalbyeo. The palatability was positively correlated with the brix degree, the glucose content, and the turbidity, and negatively correlated with the production yield of Sogokju.

Components and Antimicrobial Activity of Veiled Lady Mushroom, Dictyophora echinovolvata (흰돌기망태버섯(가칭; Dictyophora echinovolvata)의 일반성분 및 항미생물활성)

  • A strain of Dictyophora echinovolvata ASI 32002 showing good fruiting body formation was selected. Analyses of chemical and nutritional components as well as antimicrobial activity of different parts of the mushroom such as mycelium, egg, and fruiting body were carried out. There were differences in the chemical compositions and the quantities depending on developmental stages of veiled lady mushroom, D. echinovolvata ASI 32002. Nitrogen, phosphate, magnesium, and calcium in inorganic chemicals were abundant in mycelium, and potassium and mineral elements were abundant in the egg and fruiting body. Mannitol and trehalose were abundant in free sugar contents. Glutamic acid and arginine in mycelium and aspartic acid and glutamic acid in egg and fruiting body were abundant in free amino acid contents. Linoleic acid, an polyunsaturated fatty acid, was abundant in all parts of the Dictyophora species, but compositions and quantities of other fatty acids varied depending on the different parts of the mushroom. It was detected that malic acid, lactic acid and acetic acid in mycelium, formic acid, acetic acid and fumaric acid in egg, and malic acid, citric acid, lactic acid, fumaric acid in fruiting body were abundant. The methanol extracts of D. echinovolvata ASI 32002 mycelium showed antifungal activity with minimal inhibition concentration (MIC) of $62{\sim}125\;{\mu}g/ml$ that was similar levels of cyclohexamide against Aspergillus awamori, Hypocrea nigricance and Trichoderma virens. The MIC of extracts from mycelium and fruiting body against Candida albicans was $250\;{\mu}g/ml$, similar to that of tetracycline. Chemical Constituents of Korean Chopi (Zanthoxylum piperitum) and Sancho (Zanthoxylum schinifolium) (한국산 초피와 산초의 화학성분)

  • Zanthoxylum piperitum and Z. schinifolium have been utilized not only as food stuffs, but also as medicinal plants in Korea. In this study, lipids, sugar, amino acids and other components of Zanthoxylum piperitum and Z. schinifolium peels and seeds were analyzed by HPLC and GLC. Four samples contained common fatty acids such as linoleic, linolenic, palmitic, oleic and stearic acid. The contents of unsaturated fatty acids were 87.1% and 64.8% in Z. schinifolium peels and seeds, 73.6% and 62.9% in Z. piperitum peels and seeds, respectively. Z. schinifolium peels contained only beta-sitosterol, whereas other three samples contained campesterol, stigmasterol and beta-sitosterol. In case of free amino acids, peels of both species showed higher contents of acids than seeds of both species. Glutamic acid, aspartic acid, arginine, valine, and leucine were found in all four samples. Essential oils consisted of limonene (30.1-66.8%), beta-phellandrene (4.8-13.3%), citronellal (1.5-22%) and cineol (1.6-3.9%). It is worthwhile to note that the content of citronellal in Z. schinifolium seeds was higher than that of the others.

Effects of Additives on Quality Attributes of Minced Ginger During Refrigerated Storage (첨가물이 냉장 중 생강 다대기의 품질특성에 미치는 영향)

  • Quality of fresh ginger deteriorates rapidly during low temperature storage, and its storage life is short due to sprouting and microbial spoilage. The objectives of this research were to develop, using additives, a minced ginger product, which could maintain acceptable quality for over 30 days, and to investigate its quality changes during the cold storage. Storage stability of minced ginger product was investigated from the standpoint of the inhibition of brown discoloration, gas formation and liquid-solid separation. Fresh ginger was peeled and ground to produce minced ginger (control). Sodium bisulfite, L-cysteine, NaCl, sodium benzoate, modified starch, and/or xanthan gum were added to the control to minimize quality loss during storage, and to develop an optimum formula (A) of minced ginger. Samples were packed in Nylon/PE films, stored at $5^{\circ}C$, sampled at a 30-day interval, and subjected to quality evaluations. Changes in pH, surface color, gas formation, liquid-solid separation, contents of free amino acids, free sugars, organic acids, and fatty acids were determined. Gas formation was effectively inhibited in samples with sodium benzoate and/or NaCl. Samples with xanthan gum did not result in liquid-solid separation. L-Cysteine and sodium bisulfite were effective in controlling discoloration. pH decreased during storage in all samples, except sample A. Organic acid contents of all samples increased during storage, with lactic acid content showing the highest increase. Free amino acid content decreased with increasing storage time. Free sugar content of all samples decreased during storage. Sensory results showed sample A maintained acceptable quality until 90 days of storage. These results suggest that quality of minced ginger could be successfully maintained with the additions of selected additives for up to 90 days.

Physicochemical characteristics and physiological activities of mixture extracts from Liriope platyphylla, Schizandra chinensis, and Panax ginseng C.A. Meyer (맥문동, 오미자 및 인삼 혼합추출물의 이화학적 특성 및 생리활성)

  • This study was conducted to examine the antioxidant activities and physiological activities of mixture extracts (Liriope platyphylla, Schizandra chinensis and Panax ginseng C.A. Meyer) with different extraction mixing ratios (MEC, 2:1:1; ME1, 1:2:1; ME2, 1:1:2; ME3, 1.34:1.33:1.33). The yield of extracts ranged from 25.33 to 33.87%. The total polyphenol and total flavonoid contents of ME1 extracts were 1.01 g/100 g, 0.07 g/100 g, respectively. The total sugar contents of MEC extract was 22.83 g/100 g, respectively. The DPPH and ABTS radical scavenging activities of ME1 extracts at $1,000{\mu}g/mL$ were 26.79% and 21.08%. The superoxide radical scavenging and ferric-reducing antioxidant power of ME1 extracts at $1,000{\mu}g/mL$ were 67.83% and $295.47{\mu}M$, respectively. The functionalities of extracts were investigated with L-132 and RAW264.7 cell lines. The extracts on different mixing ratios did not show the toxicity on L-132 and RAW264.7 cell line in $100-2,500{\mu}g/mL$. The ME1 extract of $1,000{\mu}g/mL$ performed better than other extracts protective effects against oxidative stess in L-132 cells (81.22%) and the ME2 extract at $1,000{\mu}g/mL$ decreased nitric oxide production by $7.48{\mu}M$ which was more potent than other extracts. There results suggest that the ME1 extracts may be a useful functional food material in the food industry.
