  • The contents and their seasonal changes of non-volatile organic acids and sugars of various fruits in Korea, apricot(maesil), peach, grape, apple and pears were measured. The organic acid contents were determined by gas chromatography and the free sugars were detected by thin layer chromatography. The results were as follows: 1) The common non-volatile organic acids found in those fruits were oxalic, fumaric, succinic, maleic, tartaric and citric acids: though their contents varied from almost none to 3430mg/100g. 2) Malic acid was contained in all above fruits with generally the highest contents ranging $18{\sim}3430mg/100g$ among different fruits. In every fruits oxalic was the least contained among other organic acids, almost none to trace except apricot(maesil) which contained 10.8mg/100g. 3) It seemed that generally all the non-volatile acids contents decreased after ripening except maleic acid and the cases of an apple and a pear varieties where they increased. 4) Glucose and fructose were detected in all fruits both matured and unripened by thin layer chromatography. Maltose was found in apricot(maesil), peach, grape and apple. Sucrose was detected in apricot(maesil), peach, grape, apple and pear.

Effects of additives on physicochemical properties of gruels during storage (첨가 재료에 따른 저장 중 죽의 품질 특성에 미치는 영향)

  • This study examined the effects of additives on the physicochemical properties of gruels during storage. The color, a-value, and b-value of fine-nut, dried pollack, soybean, and chicken gruels increased, while the L-value and whiteness decreased. In addition, the a-value, b-value, and L-value of oyster mushroom gruel decreased upon increase in the storage period. Texture analyzer measurements of gruels revealed that the gumminess, cohesiveness, adhesiveness, and hardness decreased during the storage period of 90 days. The amylose and total sugar contents were also decreased during this storage period. Majority of the fatty acid composition of the gruels during the storage period at 25℃ was due to unsaturated fatty acid. The total unsaturated fatty acid content decreased, whereas saturated fatty acid content increased during the storage period of 45 days.

Development of Meat Seasoning Sauce Using Propolis (Propolis를 이용한 육류양념소스 개발)

  • We investigated the development of a new seasoning sauce using port, medicinal herbs and bee's propolis on the basis scientific experiment. We divided the sample manufacturing conditions of medicinal herbs into the following four formulae A (control), B (boiling water extraction, with added propolis), C (boiling water extraction, without propolis) and D (pressure extraction, without propolis). There was little difference in pH, titrable acidity, sodium chloride and sugar composition among the four formulae. The color value, was not changeable, despite being maintained in temperature storage (at $4^{\circ}C\;{\pm}2$) for 60 days. The order was formulae B, C, A and D. In particular, formulae B with added medicinal herbs and propolis showed greater change in oxidation than the other formulae, but lesser change in pH. A comparison of commercial and sample products, indicated only a small change in physicochemical characteristics. However, the sample products showed much superior sensory characteristics, than commercial products.

A Study on Texture of Jeung-pyun According the Kinds of Rice (쌀 품종에 따른 증편의 조직특성에 관한 연구)

  • The composition and polymeric structure of starch are the most important factors to determine the functional properties of rice. In an effort to relate the structural characteristics and its functional properties of starch in rice, molecular structural properties of starch from 6 rice cultivars were analysed. To relate the structure and function of starch the texture of Jeung-pyun made of rice were analyzed during retrogradation. The polymeric structure of rice starch was analyzed by debranching with isoamylase after gelatinization and fractionated with Sephadex G-75 gel filteration. The size fractionated debranched starch was categorized into four groups such as Fraction I, II, III and Intermediate Fraction depending in their max, The fractions with the max higher than 620 nm were designated as Fraction I, while those in the range of 600-620 nm and 540-600 nm as the Intermediate Fraction and Fraction II, respectively. The Fractions with the max lower than 540 were described as Fraction III. The average degree of polymerization (DP) of the Fraction I was estimated to be higher than 200, and those of other fractions, i.e. the Intermediate Fraction I, Fraction II and III were 150,45 and 25, respectively. The levels of Fraction I were varied from II to 35% of total sugar. The Fraction I showed the linear relationship with the amylse contents, and the Intermediate Fraction, which might contain the mixture of short chain of amylose and debranched long chain of amylopectin, were measured to be in the range of 2.7∼8.4%. The levels of fraction II and III, both to be considered as the branches of amylopectin, were ranged 14.5∼23.6% and 39.7∼73.0%, respectively. The ratio of Fraction III to Fraction II describing the degree of branch or compactness of amylopectin was estimated to be around 4.0 for waxy varieties and around 2.0 for high amylose cultivars. With these results, it can be suggested that the degree of branch of the amylopectin may effect on amylose contents of starch or rice versa, To invertigate processing aptitude of different rice cultivars for the preparation of Jeung-pyun, its texture was analyzed by Instron, hardness, cohesiveness, elasticity, gumminess and chewiness of Jeung-pyun made of rice showed the significant relationship with the amylose content. Hardness was increased during retrogradation of product, but the relationship between hardness and amylose content due to not only difference in amylose content but also difference in structural characteristics of starch. In analysis of relationship between structure of rice starch and mechanical properties of Jeung-pyun during retrogradation elasticity did not show any relationship before retrogradation bur during retrogradation showed significant correlation. With these result, it can be suggested that the degree of branch of the amylopectin may effect on elasticity during retrogradation. However hardness, cohesiveness, gumminess and chewiness which were significant different before retrogradation, showed some correlation with structure of rice starch during retrigradation.

  • In an attempt to study the possibility of substituting sweet potato for cereals as raw materials for Kochujang, three kinds of Kochujang were made from barley, sweet potato or 50% barley and 50% sweet potato, The results of component variation during the ripening and the effect of temperature on the aging in the harvesting season of sweet potato were as follows: 1) Kochujang is, usually, manufactured in May, but no difference was observed in aging condition when it was manufactured in September, the season of sweet potato harvest, since the inner temperature of the product was around $20^{\circ}C$. 2) The total reducing sugar content and maturity reached maximum after $40{\sim}50\;days$ of fermentation in the three treatments and crude fat content tended to be slightly decreased during fermentation. 3) Lightness in color of the three kinds of Kochujang decreased by exposure to sunlight. Sweetpotato Kochujang showed a stronger tendency to increase the redness as compared with the rest. 4) Sensory evaluation showed that sweet potato Kochujang was less acceptable than barley Kochujang due to disagreeable flavor of sweet potato but mixed Kochujang showed little difference, as compared with barley Kochujang.

Chemical Components in Different Parts of Pumpkin (늙은 호박의 부위별 화학성분)

  • The contents of free sugars, organic acids, amino acids, total carotenoids and pectin of pumpkin were analysed. Weight percentage of flesh was 84% of total weight. Rind and funicular attachment of seed were 10% and 3.5%, respectively. Major free sugars in pumpkin were fructose, glucose, sucrose. In flesh, fructose and glucose were the major free sugars, corresponding to 87% of total free sugars . Total sugar content in the flesh was three times higher than that in the funicular attachment of seed. Contents and composition of free amino acids were quite different according to the different parts of pumpkin. Non-volatile organic acids in pumpkin was malic, succinic and fumaric acid. The major acid in the flesh and funicular attachment of seed was malic acid and the contents in these parts were 85.2mg% and 226.5mg%, corresponding to 63% and 70% of the total organic acids, respectively. Eigh-tyseven percent of total carotenoids in the pumpkin was in the funicular attachment of seed, amounting 65.3mg%. However flesh and rind contained 6.6mg% and 3.3mg%, respectively. Water soluble pectin consisted 58% of the total soluble pectin in different parts of pumpkin, and soluble Pectin content was higher in funicular attachment of seed than in flesh. The major neutral sugars of the pectin in flesh and funicular attachment of seed were galactose and glucose consisting 45% and 36.5% of total neutral sugars.

Anti-Inflammatory Activity of Liquid Fermentation by Phellinus linteus Mycelium (상황버섯(Phellinus linteus) 균사체 액체발효물의 항염증 활성)

  • To investigate the industrial availability of liquid fermentation (PL-ferment) by Phellinus linteus mycelium as a postbiotics for the inhibition of inflammation, PL-ferment was fractionated into culture supernatant (CS), hot-water extract (HW) from PL-ferment, EtOH-precipitate (CP) fractionated from HW, and the dialysate (DCP) of CP. Compared to the other fractions, DCP which is expected to contain exopolysaccharide (EPS) as the major component, significantly decreased the production of NO, IL-6, and MCP-1 in LPS-induced RAW 264.7 cells, and IL-6 and IL-8 in TNF-α and IFN-γ-induced HaCaT cells. The general component analysis results showed that no significant difference in components was observed between the fractions, whereas sugar composition analysis revealed that DCP had decreased glucose and increased mannose contents compared to the other fractions. This suggests that mannose played an important role in the anti-inflammatory activity of the active fraction, DCP. Molecular weight distribution analysis revealed that DCP was mainly composed of low-molecular-weight material-removed high-molecular-weight polysaccharides of 18-638 kDa, suggesting that EPS originated from P. linteus EPS. In conclusion, our results suggest that the DCP of P. linteus mycelium fermentation using the anti-inflammatory activity could be used industrially as postbiotic material.

  • The recent surge in consumption of traditional liquor related to health, the results of analyzing the chemical characteristics and antioxidant activity by creating Kwaha-ju and Baekhwha-chun, which use Godubab as a method of treating glutinous and normal grains, are as follows. Kwaha-ju is mainly made of glutinous rice, 5.21~12.52% alcohol content, 1.07~2.47g/L in reducing sugar content, 3.22~3.77% pH, Baikhwha-chun alcohol content was 3~5%, except for some experimental groups, and pH was low. As a result of the spectrum analysis of traditional liquor, 50% of the experimental group in Baikhwa-chun is located in the negative area of PC1 such as overload, and some amino acids such as lactic acid, isoleucine, and choline are the main ingredients of glucose, xylose, and maltose. Therefore, the chemical properties and antioxidant activity of Kwaha-ju and Baikhwa-chun, which show similar composition ratio of traditional liquor materials, and use them as basic data to identify and manufacture traditional liquor fermentation through mutual analysis.

A Study on the Method of Manufacturing Lactic Acid from Seaweed Biomass (해조류 바이오매스로부터 Lactic acid를 제조하는 방법에 관한 연구)

  • With the spread of COVID-19 worldwide, non-face-to-face services have grown rapidly, but at the same time, the problem of plastic waste is getting worse. Accordingly, eco-friendly policies such as carbon neutrality and sustainable circular economy are being promoted worldwide. Due to the high demand for eco-friendly products, the packaging industry is trying to develop eco-friendly packaging materials using PLA and PBAT and create new business models. On the other hand, Ulva australis occurs in large quantities in the southern seas of Korea and off the coast of Jeju Island, causing marine environmental problems. In this study, lactic acid was produced through dilute acid pretreatment, enzymatic saccharification, and fermentation processes to utilize Ulva australis as a new alternative energy raw material. In general, seaweeds vary in carbohydrate content and sugar composition depending on the species, harvest location, and time. Seaweed is mainly composed of polysaccharides such as cellulose, alginate, mannan, and xylan, but does not contain lignin. It is difficult to expect high extraction yield of the complex polysaccharide constituting Ulva australis with only one process. However, the fusion process of dilute acid and enzymatic saccharification presented in this study can extract most of the sugars contained in Ulva australis. Therefore, the fusion process is considered to be able to expect high lactic acid production yield when a commercial-scale production process is established.

Comparative Physicochemical Characteristics and Microbial Communities in Commercial Kimchi and Mukeunji Products (국내 시판 김치와 묵은지의 이화학적 특성 및 미생물 군집 구조 비교)

  • This study compared and analyzed the microbial composition and physicochemical characteristics of kimchi (gimjang kimchi) and mukeunji (aged kimchi). Commercial kimchi and mukeunji products were purchased through an online market. After an analysis of physicochemical characteristics, the pH of the mukeunji samples was found to be lower and the acidity higher than in the kimchi samples. There was no significant difference in salinity between kimchi and mukeunji, but the sugar content was higher in the kimchi samples. The phylogenetic diversity index, which incorporates phylogenetic difference between species, was significantly higher in mukeunji than in the kimchi. The most dominant order in both groups was Lactobacillales, but several lactic acid bacteria, such as the Pediococcus and Lactobacillus species, which may be more acid tolerant or more competitive, are relatively predominant in mukeunji. Beta set-significance analysis based on two different distance metric results revealed that microbial distributions of population were different at the statistical confidence level (p<0.001). We investigated the effect of respective species on total microbial community using the LEfSe (linear discriminant analysis effect size) mechanism. According to the results of LEfSe testing, a relatively higher abundance of Weissella kandleri in kimchi and a higher abundance of Pediococcus inopinatus in mukeunji have the greatest influence on the differences in microbial structure between the two groups.
