  • This study analyzed the punk style shown in the movie Sid and Nancy (1986), based on a true story. The purpose of this study was to prove that the styles of the main characters had some discriminatory mode as an anti-fashion with not only the value system of the subculture but also the diverse cultural codes applied, revealing their identity. The scope of this study covered the 1970s when Sid died while a member of the Sex Pistols, but was limited to the analysis of Sid and Nancy's costumes. As theoretical background, this study reviewed the preceding studies, specialty books, movie-related sites and their postings, and on-line news reports. To analyze the movie costumes, relevant scenes on the DVD were captured, and thereby, the scenes were categorized per character and style but a performance scene was categorized Sid and Nancy as same punky wedding style because of intention spirituality to marriage by director, and thereupon, 17-cut images were used for the analysis of main characters' styles. Sid's style consisted primarily of black leather jackets, symbolic T-shirts, and jeans, whereas Nancy's style implied a punk style with added fetish styles. Even in the same punk style, their identities were symbolized through bricolage. Through this, subculture styles, were able to confirm that in addition to the value system of subculture, discriminatory modes as anti-fashion with various cultural codes played a role in revealing their identity.

  • We investigated factors affecting primary cultures of Pacific abalone Haliotis discus hannai ovary-dissociated cells to identify general aspects of their early-phase culture. Ninety-seven cell populations derived from 30 individuals were cultured in different media with varying compositions of medium supplements, and initial attachment, subculture, and survival for ${\geq}10$ weeks were assessed according to medium composition and individual. We also examined the time required for subculture and the rate of cell death according to both culturing period and passage number within 10 weeks. A lack of fetal bovine serum (FBS) and hemolymph significantly inhibited the growth of cultured cells, while we detected no significant effect of medium composition on initial cell attachment. Through data reallocation, with the omission of data from cell populations cultured in FBS-free and hemolymph-free media, we showed that growth inhibition was also affected by individual differences among the abalones used. During the culture, we observed four different types of cell morphology. Moreover, considerable time was required for subculture-18.4 and 19.5 days for first and second subcultures, respectively-and cell death did not occur within 30 days or for passage 0. Our results will provide valuable information for developing universal cell culturing guidelines in abalone species and suggest the feasibility of culturing abalone ovary-dissociated cells.

  • Among diverse cultural types which can be shown in movie, 'Subculture' is young culture that resist to major culture. It shows its belief and identity via its own style. Their 'Subcultural Style' became popular with mass media and has been affected to major cultures. Therefore, in this research, with times background as postmodernism in end of 20th century and mainly with movie which is about sexual minorities, I analyzed categories of costumes in the movie. Hedwig's rock costume shows combined fashion with hippie, punk, glam, kitsch, goth styles from the impact of postmodernism in mid 20th century. Tommy, showed his relief and defiant intention against old generations with grunge casual and graffiti styles. After he realized Hedwig's identity and showed defiant goth rock style as a famous rocker. The band, "Angry Inch", showed dissatisfaction and cynical attitude against the American society through subculture styles. Through this research, the fact that movie costume used a tool to show human's identity and value. Also, I can see that movie costume shows times and cultural phenomenon in the end of 20th century and through this, it was confirmed that this contributes to crease new fashion style through them.

  • To improve the productivity of peroxidase (POD) of cell line SP-47 derived from cell suspension cultures of sweet potato (Ipomoea batatas (L) Lam.cv White Star), we optimized culture conditions including the composition and concentration of plant growth regulators and carbon source, and the cell inoculum size. When one g (fr wt) of cells was inoculated into 50 mL TL medium supplemented with l mg/L 2,4-D and 30g/L sucrose in 300 mL Erlenmeyer flask at 25$^{\circ}C$ in the dark (100rpm), the POD activity per g cell dry wt was maximized to be about 6,800 units after 25 days of subculture, which was about 30 times higher than that of intact roots of horseradish plants grown in the greenhouse, but the cell growth was maximum after 15 days of subculture. The protein content per g cell dry wt maintained almost plateau and after 25 days of subculture decreased as culture Proceeded further whereas the POD specific activity (unit/mg protein) was about two times higher after subculture and continuously increased from 12 days to the end of cultures (40 days). The POD isozyme patterns showed almost the same regardless of cell growth stage, but some acidic isozymes were slightly increased after 25 days of subculture. These results indicate that POD activity in suspension cultures of sweet potato is closely associated with cell growth and stresses derived from cell culture renditions and medium depletion. Due to its high POD activity the SPL47cell line seems to be suitable for the mass production of POD.

  • This study was conducted to investigate the characteristics of subculture times in the early, middle, and late passages by measuring the time under subculture until it was judged that the supernumerary tooth-derived pulp stem cells (sDPSCS) were no longer proliferating. Three supernumerary teeth from two healthy six-years old boys were extracted and stem cells were obtained from the pulp tissue. This was called SNT1 (supernumerary tooth 1), SNT2, and the supernumerary tooth from another child was named SNT3. SNT1 and 2 were subcultured at the same time and SNT3 was subcultured a little faster. The mean time of complete subculture was $3.6{\pm}1.1$ days. Total passages were cultured up to $23.3{\pm}0.6$ and took 83 days. These were divided into three groups based on the passage. The increase rate of time taken in subculture between group I and group II was 11.9%, but the rate between group II and group III was 28.6%, which was 2.4 times increased. The time taken between passages during long-term subculture up to 22 passages shows a regressive pattern y = 0.1169x + 2.25 and y = 0.1169x + 2.0. In conclusion, the passage time of SPSCs increased in late passages, and it shows a similar pattern.

  • The purpose of this study was to investigate the odontoblast gene expression related to the subculture speed of supernumerary dental pulp stem cells (sDPSCs). The stem cell is undifferentiated cells which has the ability to differentiate into various cells. Specific stimulation or environment induces cell differentiation, and these differentiation leads to bone or muscle formation. 20 sDPSCs were obtained from 20 children under aseptic condition. During the culture through the 10th passage, the third passage cells which showed short subculture period and 10th passage cells which showed long subculture period were earned. Each cell was divided into differentiated group and non-differentiated group. Quantitative real-time polychain reaction (q-RT-PCR) was performed for each group. The genes related to odontoblast differentiation, Alkaline Phosphatase (ALP), Osteocalcin (OCN), Osteonectin (ONT), Dentin sialophosphoprotein (DSPP) and Dentin matrix acidic phosphoprotein 1 (DMP-1), were measured. Differentiated cells showed more gene expression levels. Undifferentiated cells showed higher gene expression level in 10th passages but differentiated cells showed higher gene expression level in 3rd passages. Cells that showed faster subculture period showed relatively lower gene expression level except for OCN and DSPP.

  • The repeated monthly or weekly subculture of plant callus is labor intensive and increases the risk of somaclonal variation from the parental callus line. The most effective method for preserving plant callus is cryopreservation, which involves storage in liquid nitrogen. However, this method cannot be applied to the callus of different plant species in the same manner, so it is difficult to develop a standardized cryopreservation method. In addition, the survival rate of the frozen callus after thawing and the regeneration rate after survival are uncertain. Therefore, it is necessary to develop a method to extend the subculture interval of plant callus in an active state. In this study, active plant calli of various species without freezing was incubated at 15℃ for 4 to 12 weeks without subculture. After 12 weeks, 8 lines of plant callus grew less than 2-fold when cultured at 25℃, but at least 2 times as much when cultured at 15℃. Moreover, total antioxidant activity did not differ significantly between plant callus recovered at 25℃ after culturing at 15℃ or at 25℃. These results show that the subculture interval can be extended at a temperature of 15℃ without need for modified medium composition or additional processes. In addition, positive results in all calli of several plant species are expected to reduce labor as well as somaclonal variation by increasing the subculture.

  • The main purpose of this exploratory research was to examine the characteristics of consumers who patronize natural-dyed clothes and their perceptions regarding natural-dyed clothes. Thirty three participants who have worn natural-dyed clothes were interviewed for the study. They were asked about the styles and price of natural-dyed clothes they owned, their evaluation on them, and the lifestyles of themselves. Existence of subculture among the interviewes and its characteristics were also probed. The results indicated that natural-dyed clothes are relatively high-priced, mostly of modified hanbok style, and became popular in recent years. Interviewees frequently mentioned uniqueness and comfort as the main benefits of natural-dyed clothes, and expressed dissatisfaction regarding color fastness, easy care and problem of coordination. The consumers of natural-dyed clothes appeared to have strong preferences for environment conservation and Korean traditional culture. They also seemed to form a subcultural group who have commonality in their involvement in Korean cultural activities, mainly tea ceremony.

  • Classifying the diverse and complex clothing category of nationality subcultures in China is not a simple task. However, summing up, it can be largely classified that clothing of the northern area is long trousers and short skirt. Among the clothing of Nationality in China, especially, Pao(robe: 袍) has its diversity and colorfulness. whose elements forming its each characteristic feature has enough value to be investigated fully in the historical point of view. The conclusions of this study reveals that Pao in nationality subculture in China can possibly be divided into the four types as Qipao(旗袍) in Dongpei area, Mongopao(蒙古袍) in Inner Mongolia, Qiapan in Xinjiang area, and Zangpao in Tibet area. The modes of Qipao and Mongopao are mainly similar in that they have diagonally on the right, stand collar in composition, but Qipao shows its diversity in collar and slashes, and Mongopao also shows diversity in its colare and waistband. Since western culture flowed into China along with its open door and reform policy, the splendid color and distinctive pattern, decoration, diversified method of compositions and clothing categories of Nationality cultures are disappearing in its their originality and nationality.

  • 'Superflat' is a hugely influential contemporary art movement founded by Takashi Murakami. The concept of Superflat art is based on the notion that there exists in contemporary Japanese culture an inherent inclination for two-dimensionality devoid of perspective and hierarchy with all elements existing equally and simultaneously. The theory is defined in broad terms and asserts that this inclination for flat aesthetic has its roots in the traditional Japanese art and the development of post-war Japanese subculture. As such, Superflat as a theory possesses a capacity to engage and explain wide-ranging conditions in contemporary Japan. Taro Igarashi has made such a point and argues that the generations of leading Japanese architects practicing today possess Superflat 'tendency' for flat aesthetics and are inclined to focus on the expressive possibilities of the building's skin. While such sensibility could simply be interpreted as a stylistic trend that has emerged over the years, when examined against the characteristics of the art movement, there exist unmistakable similarities in terms of the design sensibility and techniques applied. Furthermore, the theory has become a force in how Japanese architecture is defined and understood internationally in the past decade.
