Different parameters of subclinical mastitis were compared in 327 cattle and 493 buffaloes and the effect of immunopotentiating agents on subclinical mastitis in these animals was studied. Subclinical mastitis was detected in 8.2 percent buffalo and 24.0 percent cow quarter by modified whiteside test (MWT). In both the species there was decrease in lactose contents with increase in the degree of MWT reactivity i.e. $4.8{\pm}1.14$ to $2.31{\pm}0.82$ in cattle and $5.01{\pm}1.47$ to $2.36{\pm}1.02$ in buffaloes. While the chloride contents of the milk increased with increase in the MWT reactivity i.e. $0.19{\pm}0.4$ to $0.30{\pm}0.06$ in cattle and $0.20{\pm}0.04$ to $0.31{\pm}0.13$ in buffaloes. Micro-organisms belonging to Staphylococcus, Micrococcus, Bacillus, Streptococcus, Enterobacteria, Corynebacterium groups and yeasts were isolated from subclinical mastitis cases. Vitamin E and Levamisole cured 64.5 and 60.0 percent cases of subclinical mastitis in buffaloes but only 32.0 and 24.0 percent cases in cattle. Cure was not affected by the degree of MWT and the type of organisms involved.
The experiment was undertaken to study the effect of subclinical mastitis on the biochemical parameters of milk in crossbred (Sahiwal $\times$ Jersey) cows. Subclincial mastitis was judged using California Mastitis Test and graded on a scale of 0 to 2. Twenty six milk samples of each grade were subjected to somatic cell count. Separated milk whey was analysed for total protein, immunoglobulin and trace mineral content. Results indicate linear increases in somatic cell count, total protein and immunoglobulin concentrations in milk with increase in grade of subclinical mastitis. However, copper, zinc, manganese and iron concentration in milk was not affected by grade of mastitis.
A study was carried out to assess the therapeutic effect of ascorbic acid in mastitis of dairy cows. The herd with a population of 250-275 lactating cows was screened for clinical and subclinical mastitis for a period of 5 months. Based on inclusion and exclusion criteria, eighteen animals each with clinical and subclinical mastitis in one quarter only were selected as study population. Twelve cows (group A) with normal udder and health were also selected as a healthy control. Clinical mastitis cows were grouped as B (n=12) and C (n=6). Cows of group B were treated with ascorbic acid at 25 mg/kg, subcutaneously for 5 consecutive days and intramammary infusion (Ampicillin sodium 75 mg and Cloxacillin sodium 200 mg/infusion) based on antibiotic sensitivity test, till complete recovery. Group C cows received only intramammary infusion till the complete recovery. Eighteen subclinical mastitis cows were divided in group D (n=12) and E (n=6). Cows of group D were treated with ascorbic acid at 25 mg/kg subcutaneously for 5 consecutive days while group E did not receive any treatment. California mastitis test (CMT), somatic cell count (SCC), physical changes of udder and milk were used to diagnose and classify the mastitis. Evaluation of the therapy was based on CMT score and physical changes of udder and milk. Sample size calculation was also performed but was not followed for control groups due to scarcity of cases. Adequate blinding was done when and where required to avoid the biases. Confounding variables like herd, age of the cow, stage of the lactation, season and geographical region were duly considered and adequate blocking was followed. Ascorbic acid was administered in clinical and subclinical cases even after cure considering its immunostimulatory and healing inducing effects. The recovery rate was faster in cases of clinical mastitis treated with ascorbic acid along with an intramammary infusion (group B) than the quarters of group C cows. Quarter wise the average duration/number (3.16${\pm}$0.11 days) of antimicrobial intramammary infusion was significantly (p<0.01) less in group B than that of average duration/number (5.33${\pm}$0.20 days) of group C. Subclinical mastitis cows treated with ascorbic acid showed 83.33% recovery while 16.77% did not respond to treatment till last day of study. Cows of group E (untreated) did not recovered from the mastitis. Subjective parameters viz. swelling, pain reflex of udder and physical changes in milk from quarter of ascorbic acid treated cows (group B) disappeared earlier than that of group C cows. It is concluded from this study that the ascorbic acid might be useful as an adjunct in case of clinical mastitis to get quick recovery with less number of intramammary infusions. High recovery rate in subclinical mastitis quarters of group D cows is appreciable and opens a new avenue to conduct further trials in a larger population in various field conditions. However, the pharmacology of ascorbic acid with particular reference to health of mammary gland needs to be investigated.
Mastitis is a highly morbid disease that requires detection at the subclinical stage. Tropical countries like India mainly depend on milch buffaloes for milk. The present study was conducted to investigate whether the trace minerals viz. copper (Cu), iron (Fe), zinc (Zn), cobalt (Co) and manganese (Mn) and enzyme activity of lactate dehydrogenase (LDH), alkaline phosphatase (ALP) and aspartate aminotransferase (AST) in riverine buffalo milk can be used as an indicator of subclinical mastitis (SCM) with the aim of developing suitable diagnostic kit for SCM. Trace elements and enzyme activity in milk were estimated with Atomic absorption Spectrophotometer, GBC 932 plus and biochemical methods, respectively. Somatic cell count (SCC) was done microscopically. The cultural examination revealed Gram positive bacteria as the most prevalent etiological agent. A statistically significant (p<0.01) increase in SCC, Fe, Zn, Co and LDH occurred in SCM milk containing gram positive bacterial agents only. ALP was found to be elevated in milk infected by both gram positive and negative bacteria. The percent sensitivity, specificity and accuracy, predictive values and likelihood ratios were calculated taking bacterial culture examination and $SCC\geq2{\times}10^5$ cells/ml of milk as the benchmark. Only ALP and Zn, the former being superior, were found to be suitable for diagnosis of SCM irrespective of etiological agents. LDH, Co and Fe can be introduced in the screening programs where Gram positive bacteria are omnipresent. It is recommended that both ALP and Zn be measured together in milk to diagnose buffalo SCM, irrespective of etiology.
A total of 127 foremilk samples from dairy farms in Chonnam district was examined for the subclinical mastitis eve. six months, using a method of the N-acetyl-${\beta}$ -D-glucosaminidase (NAGase) test in relation to the California mastitis test(CMT) and the somatic tell count(SCC) and the compatibility and efficiency rating between the NAGase test and the other screening test were conducted. Fifteen subclinically mastitic cows were treated with a single oral dose of 7.5mg/kg of levamisole hydrochloride. The results are summarized as follows. 1. A linear relationship was found among the NAGase level, the CMT score and the SCC level, and it was found that NAGase activity measurements were comparable with other screening tests for diagnosing cows with mastitis. 2. Compatibilites between the NAGase and the CMT were 96.1%, and that of the NAGase and SCC were 93.7%. On the other hand, relative efficiency ratings of Postle's equation between the NAGase and CMT were 89.4%, and that of the NAGase and SCC were 84.1%. 3. Considering the result of CMT, SCC and NAGase level after treatment with levamisole hydrochloride it seemed to be of special value in the treatment of bovine subclinical mastitis.
A total of 2,024 quarters of 515 dairy cattle in Kangweon area were examined for incidence of subclinical mastitis. Milk samples from cattle infected with subclinical mastitis were studied bacteriologically and the bacterial strains isolated were further examined for sensitivity to 12 antibacterial agents. And the status of carrying out the mastitis control program in 28 dairy farms was examined. The results obtained were summerized as follows ; 1. A total of 308(59.8%) of 515 cattle and 656(32.4%) of 2,024 quarters were found to be infected with subclinical mastitis. 2. The 277 strains of etiological agents were isolated from 358 subclinical quarters. These were identified as Staphylococcus aureus(14.4%), other staphylococci(36.5%), Streptoccus agalatiae(8.7%), other streptococci(30.7%), Bacillus spp.(1.8%), Corynebacterium spp.(1.4%) and coliform(0.7%). 3. The 109 strains of streptococci and 141 strains of staphylococci were examined for sensitivity to 12 antibacterial agents. All the strains of streptococci were sensitive to penicillin, ampicillin and cephalothin, and they were also sensitive to erythromycin(88.1%), clindamycin(83.5%), enrofloxacin(75.2%), trimethoprim+sulfamethoxazole(67.9%), The strains of staphylococci were sensitive to cephalothin(97.2%), gentamicin(83.0%). enroflozacin(80.9%), trimethoprim+sulfamethoxazole(78.0%), erythromycin(71.6%) and clindamycin(71.6%). But all the strains resisted to colistin. 4. In the 28 dairy farms examined, condition of udder before washing was dirty in most of the farms (89.7%). Hygiene of milking equipment was only good in the 5 farms(17.9%). Teat preparation before milking was good in the 6 farms(21.4%). The farms in which teat dipping after milking was conducted were 46.4%. Dry cow treatment for the complete herd was carried out in most of the farms(89.3%) but mastitis checking was only carried out in the 8 farms(28.6%) irregularly.

A total of 137 strains of Staphylococcus aureus were isolated from dairy cow's milk with subclinical mastitis from 33 herds in 5 provinces and 36 strains of S aureus from clinical mastitis from 4 herds where the mastitis were severe problem. Arbitrary primed polymerase chain reactions with 10 bp oligonucleotide primer were performed and the PCR products were analysed with image analyzer, The S aureus strains were genotyped into 20 distinct DNA fingerprinting profiles. The size of PCR products ranged from 163 to 2,479 bp and PCR products of 506, 770, 784 and 2,479 bp were the most prevailing bands. Genotype 3 was founded in all 5 provinces. The various genotypes were identified in newly founded dairy herds, however, only one or two genotypes were identified in the closed herds. In clinical mastitis, only a limited number of different S aureus genotype was founded in each of the herds in comparision with subclinical mastitis. The results demonstrated that PCR-based DNA fingerprinting analysis of S aureus strain can be used to study epidemiology of mastitis, in addition, common genotype in geographic region can be useful for the development of an effective S aureus bacterin.

Importance: Bovine mastitis, predominantly associated with gram-positive Staphylococcus aureus, poses a significant threat to dairy cows, leading to a decline in milk quality and volume with substantial economic implications. Objective: This study investigated the incidence, virulence, and antibiotic resistance of S. aureus associated with mastitis in dairy cows. Methods: Fifty milk-productive cows underwent a subclinical mastitis diagnosis, and the S. aureus strains were isolated. Genomic DNA extraction, sequencing, and bioinformatic analysis were performed, supplemented by including 124 S. aureus genomes from cows with subclinical mastitis to enhance the overall analysis. Results: The results revealed a 42% prevalence of subclinical mastitis among the cows tested. Genomic analysis identified 26 sequence types (STs) for all isolates, with Mexican STs belonging primarily to CC1 and CC97. The analyzed genomes exhibited multidrug resistance to phenicol, fluoroquinolone, tetracycline, and cephalosporine, which are commonly used as the first line of treatment. Furthermore, a similar genomic virulence repertoire was observed across the genomes, encompassing the genes related to invasion, survival, pathogenesis, and iron uptake. In particular, the toxic shock syndrome toxin (tss-1) was found predominantly in the genomes isolated in this study, posing potential health risks, particularly in children. Conclusion and Relevance: These findings underscore the broad capacity for antibiotic resistance and pathogenicity by S. aureus, compromising the integrity of milk and dairy products. The study emphasizes the need to evaluate the effectiveness of antibiotics in combating S. aureus infections.
A total of 210 dairy cows of Seongwhan area were epidemiologically investigated for mastitis by Milk Quality Test(MQT) method. Milk samples from cows infected with mastitis were also bacteriologically studied. Seventy four (42.5%) of 174 dairy cows were positive for mastitis and fifty nine (79.7%) among them were infected with subclinical mastitis and fifteen (20.3%) were clinical mastitis. High infective rate were shown on August (48.7%) and July (45.7%) and it was lowered on December (35.8%). The incidence rate of subclinical mastitis for herd shown tendency of increase at the barn in which located on lower place (30.7~43.9%), whereas, the barn in which was on higher place and with bucket type milking system shown tendency of decrease causative organisms. A total of 230 strains of etiological agents were isolated from 578 infected duarters which shown MQT score (+) or more. The relevant causative organisms isolated in this study were Staphylococcus aureus (18.3%), Staphylococcus epidermidis (16.9%), fungi (16.5%), Pseudomonas aeruginosa (15.2%), Escherichia coli (7.8%) and Serratia marcescens (2.27), respectively.
