Artomyces microsporus의 배양적 특성과 균사 적정 배양 조건 설정 (Culture Characteristics and Optimal Conditions for Mycelial Growth of Artomyces microsporus)

  • 본 연구에서는 A. microsporus의 최적 배양 조건을 구명하고자 하였다. A. microsporus 균사체의 효율적 배양을 위한 조건은 최적 배양 온도 25℃, pH 5.0, 탄소원 soluble starch 1%, 유기질소원 malt extract 1%, 무기질소원 NH4H2PO4 0.1%, 아미노산 asparatic acid 0.1% 조건으로 선발되었고 유기산, 무기염류 첨가 시 균사생육이 저조해지는 영향을 주어 선발되지 않았다. 새로이 SMNA로 명명한 A. microsporus의 최적 배지는 기존의 PDA 배지 상에서 배양한 결과와 비교하였을 때, 균사의 생육이 균일하고 안정적이며 배양 기간을 15.1% 정도 단축하는 효과가 있었다. SMNA 배지는 추후 A. microsporus의 균사체 배양 방법에 널리 활용될 수 있을 것으로 판단된다.

국내 식품 중 밥류와 국(탕) 및 찌개류의 수용성 바이타민 B1, B2, 그리고 B3 함량 검토 (Investigation of water-soluble vitamin (B1, B2, and B3) content in various rice, soups, and stews produced in Korea)

  • 국내 식생활의 가장 기본적인 구성요소인 밥류, 국(탕)류, 그리고 찌개류에 존재하는 다양한 영양성분 중 수용성 바이타민인 $B_1$ (thiamin), $B_2$ (riboflavin), 그리고 $B_3$ (niacin)의 함량을 확인하였다. 밥류의 경우 가공방법에 따른 열처리의 조건이 다양한 수용성 바이타민의 함량을 나타내었고, 국(탕)의 경우 전체적으로 높은 수분함량으로 영양성분의 희석효과가 확인되었고, 찌개의 경우 주재료로 사용된 재료에 따라 다양한 바이타민의 함량을 나타내었다. 본 연구 결과는 국민 식사생활의 영양성분 데이터베이스의 중요한 기초자료로 활용될 것으로 판단되며, 기본 재료가 가진 영양성분의 함량이 가공조건에 따라서 다양한 잔존율을 보인다는 것을 제시하였다. 계속적인 연구를 통하여 셀 수 없이 다양하게 존재하는 한식의 영양성분에 대한 데이터베이스화를 통해 국민 식사생활 개선을 위한 노력이 필요할 것으로 판단된다.

Characterization of physiochemical and nutrient profiles in canola feedstocks and co-products from bio-oil processing: impacted by source origin

  • Objective: The objective of this study was to characterize physiochemical and nutrient profiles of feedstock and co-products from canola bio-oil processing that were impacted by source origin. The feedstocks and co-products (mash, pellet) were randomly collected from five different bio-oil processing plants with five different batches of samples in each bio-processing plant in Canada (CA) and China (CH). Methods: The detailed chemical composition, energy profile, total digestible nutrient (TDN), protein and carbohydrate subfractions, and their degradation and digestion (CNCPS6.5) were determined. Results: The results showed that TDN1x was similar in meals between CA and CH. CH meals and feedstock had higher, truly digestible crude protein (tdCP) and neutral detergent fiber (tdNDF) than CA while CA had higher truly digestible non-fiber carbohydrate (tdNFC). The metabolizable energy (ME3x), net energy (NELp3x, NEm3x, and NEg3x) were similar in meals between CA and CH. No differences were observed in energy profile of seeds between CA and CH. The protein and carbohydrate subfractions of seeds within CH were similar. The results also showed that pelleting of meals affected protein sub-fractionation of CA meals, except rapidly degradable fractions (PB1), rumen degradable (RDPB1) and undegrdable PB1 (RUPB1), and intestinal digestible PB1 (DIGPB1). Canola meals were different in the soluble (PA2) and slowly degradable fractions (PB2) between CA and CH. The carbohydrate fractions of intermediately degradable fraction (CB2), slowly degradable fraction (CB3), and undegradable fraction (CC) were different among CH meals. CH presented higher soluble carbohydrate (CA4) and lower CB2, and CC than CA meals. Conclusion: The results indicated that although the seeds were similar within and between CA and CH, either oil-extraction process or meal pelleting seemed to have generated significantly different aspects in physiochemical and nutrient profiles in the meals. Nutritionists and producers need to regularly check nutritional value of meal mash and pellets for precision feeding.

용해성 지반혼합재의 입자 용해 영향 (Particle Dissolution Effects on Soluble Geo-Mixtures)

  • 본 연구에서는 용해성 입자를 포함한 혼합재가 용해과정에서 발생하는 입자구조의 미시적 및 거시적 거동을 수치해석기법을 이용하여 조사하였다. 혼합재는 다양한 부피비의 용해성 입자와 비용해성 입자를 혼합하여 조성하였다. 용해성 입자의 용해과정은 입자의 크기를 감소시킴으로써 모사하였다. 수치해석 결과 일정한 시간까지 시료의 수직변형은 진행되며, 수직변형이 평형에 도달한 후에도 용해작용이 진행되어 간극률은 변화한다. 또한, 간극률과 수직변형은 용해성 입자의 함유량이 증가함에 따라 증가한다. 본 연구에서는 수치해석을 통하여 용해성 입자를 포함한 시료의 용해과정에서 입자구조의 변화는 특정 시간까지 진행되며, 용해성 입자의 부피가 클수록 구조 변화가 큰 결론을 얻었다.

수확 후 고농도 CO2 단기처리가 '매향' 딸기의 저장에 미치는 영향 (Effects of short-term treatment of high pressure CO2 on the changes in fruit quality during the storage of 'Maehyang' strawberries)

  • This experiment was conducted to examine the effects of a short-term treatment of high pressure $CO_2$ on shelf-life of strawberry fruit. A short-term treatment (12 hr) of 100% $CO_2$ resulted in the increase of fruit firmness up to 71.9% compared to that at harvest. The firmness of $CO_2$ treated fruit remained a significantly higher than that of control (air) up to 15 days. The alteration of pectic polymers was observed by $CO_2$ treatment such as an increase of EDTA soluble pectins and decrease of water soluble ones. The $CO_2$ treatment resulted in the increase of total amount of wall bound calcium. Pectate lyase activity, an important agent of strawberry fruit softening, was also significantly reduced by $CO_2$ treatment. Contents of soluble solids and acids of $CO_2$ treated fruit were higher than those of control fruit. Short-term treatment of high pressure $CO_2$ affected shelf-life through firmness increase whereas the visual quality and decay incidence of strawberry fruit were not affected.

한라산 1,100 m 지역의 대기 중 PM2.5에 함유된 이온 및 원소 성분의 조성특성 (Ionic and Elemental Compositions of PM2.5 at the 1,100 m-Highland of Mt. Hallasan in Jeju Island)

  • In this study, mass concentrations and chemical compositions of $PM_{2.5}$, including water-soluble ions and elements were determined at the 1,100 m-highland of Mt. Hallasan in Jeju Island across four seasons from August 2013 to August 2014. The average mass concentration of $PM_{2.5}$ was $12.5{\pm}8.41{\mu}g/m^3$ with 45.8% of the contribution from eight water-soluble ionic species. Three ionic species ($SO{_4}^{2-}$, $NH{_4}^+$, and $NO{_3}^-$) comprised 96.2% of the total concentration of ions contained in $PM_{2.5}$ and were the dominant ions, accounting for 43.5% of the $PM_{2.5}$ mass at Mt. Hallasan. On the basis of the mass concentration level, seasonal variation, enrichment factor, and relationship among elements, we can presume that Mg, K, Ca, Mn, Fe, Co, Sr, Ba, Nd, and Dy originated mainly from crust or soil and that V, Cr, Ni, Cu, Zn, As, Cd, and Pb were significantly enriched in $PM_{2.5}$ owing to the effects of the anthropogenic emissions. These results and the local distribution of emission sources and topographic characteristics near this sampling site suggest that the compositions of $PM_{2.5}$ collected at the 1100 m-highland of Mt. Hallasan were largely influenced by inflow from outside of Jeju Island.

서울시 PM10 내의 수용성 유기탄소와 수분함량과의 상관성 분석 (The Relationship between the Estimated Water Content and Water Soluble Organic Carbon in PM10 at Seoul, Korea)

  • In this study, we have analyzed relationship between the measured Water Soluble Organic Carbon (WSOC) concentrations and the estimated aerosol water content of $PM_{10}$ (particulate matter with an aerodynamic diameter of less than or equal to $10{\mu}m$) for the period between September 2006 and August 2007 at Seoul, Korea. Water content of $PM_{10}$ was estimated by using a gas/particle equilibrium model, Simulating composition of Atmospheric Particles at Equilibrium 2 (SCAPE2). The WSOC concentrations showed low correlation with Elemental Carbon (EC), but Water Insoluble Organic Carbon (WISOC) were highly correlated with EC. It seemed that hydrophilic groups were produced by secondary formation rather than primary formation. As with the previous studies, WSOC showed good correlation with secondary ions ($NO_3{^-}$, $SO_4{^{2-}}$, $NH_4{^+}$), especially WSOC was highly correlated with $NO_3{^-}$ that is a secondary ion formed by photochemical oxidation from more local sources than $SO_4{^{2-}}$. No apparent correlation between the measured WSOC and estimated water content was observed. However, WSOC showed good correlation with estimated water content when it was assumed that relative humidity was higher than the deliquescence relative humidity of the system. In conclusion, WSOC is correlated with water content by hygroscopic ions and it is expected that nitrate play an important role among the water content and WSOC.

Shelf-life prediction of fresh ginseng packaged with plastic films based on a kinetic model and multivariate accelerated shelf-life testing

  • The purpose of this study was to monitor changes in the quality of ginseng and predict its shelf-life. As the storage period of ginseng increased, some quality indicators, such as water-soluble pectin (WSP), CDTA-soluble pectin (CSP), cellulose, weight loss, and microbial growth increased, while others (Na2CO3-soluble pectin/NSP, hemicellulose, starch, and firmness) decreased. Principal component analysis (PCA) was performed using the quality attribute data and the principal component 1 (PC1) scores extracted from the PCA results were applied to the multivariate analysis. The reaction rate at different temperatures and the temperature dependence of the reaction rate were determined using kinetic and Arrhenius models, respectively. Among the kinetic models, zeroth-order models with cellulose and a PC1 score provided an adequate fit for reaction rate estimation. Hence, the prediction model was constructed by applying the cellulose and PC1 scores to the zeroth-order kinetic and Arrhenius models. The prediction model with PC1 score showed higher R2 values (0.877-0.919) than those of cellulose (0.797-0.863), indicating that multivariate analysis using PC1 score is more accurate for the shelf-life prediction of ginseng. The predicted shelf-life using the multivariate accelerated shelf-life test at 5, 20, and 35℃ was 40, 16, and 7 days, respectively.

펄라이트 재배에서 급액량이 토마토의 생육과 수량에 미치는 영향 (Effects of Drip Irrigation Volumes on Plant Growth and Yield of Tomato Grown in Perlite)

  • 본 실험은 펄라이트 수경재배에서 급액량이 토마토(Solanum lycopersicum L.)의 생육과 수량에 미치는 영향을 조사하기 위해 수행되었다. 급액 처리는 대조구인 I0와 대조구보다 급액량이 각 25%, 50% 많은 I25와 I50로 세수준 처리하였다. 처리기간 중 대조구 I0 처리구는 배액이 없었다. 조사항목으로 토마토 생육특성, 수량 및 수분이용효율을 측정하였다 토마토의 초장, 엽장 및 엽폭이. I0 처리 가장 짧았고 I25와 I50 처리에서는 통계적으로 차이가 없었다. 처리 후 111일, 132일 및 143일째에 측정한 토마토 과실의 당도, 산도 및 건물율은 관수량이 가장 적은 I0가 관수량이 많은 I25와 I50 보다 높았다. 그러나 처리 후 111일과 132일째 과실 내 수분함량은 I0 처리에서 가장 낮았다. 대과(>180g)의 수는 I25에서 유의하게 더 많았으나 총 수확과 수는 처리 간 유의한 차이가 없었다. 과실의 평균 과중(g·plant-1)과 총 수확량(g·plant-1)은 I0 처리된 토마토에서 가장 낮았다. 수분 소비량은 처리 간 유의한 차이가 없었으나 수분이용효율은 I0 처리에서 가장 낮았다. 주성분 분석 결과 과실의 당도와 산도는 양의 상관관계가 있는 것으로 나타났다. 본 실험에서 토마토 생육, 과실 특성 및 수분이용효율을 토대로 얻은 결과는 I25가 토마토 펄라이트 수경재배에서 다른 처리 보다 적절한 급액량임을 보여준다.
