Shooting and Fruiting Characteristics of 'Hachiya' Persimmon Tree Affected by Sub-zero Temperature Treatment at Early Budding Stage (발아 초기에 영하온도 처리에 따른 감나무 '갑주백목' 의 신초 생장과 착과 특성)

  • The purpose of this study was to investigate shooting and fruiting characteristics of persimmon 'Hachiya' affected by sub-zero temperature at early budding stage. The number of developed shoots, and the number of shoots with young fruits, and the shooting and fruiting of terminal buds of one-year-old branch (1YOB) were significantly decreased with the sub-zero temperature treatment (SZT). The number of shoots developed in the top of 1YOB decreased 47.2% due to SZT. The number of shoots with fruits was significantly decreased at 50% or higher in non-budding rate of 1-YOB. Especially the number of fruits (Y) at the three terminal buds of 1YOB was significantly affected by SZT and the non-budding rate (X) as Y = -0.145X + 12.950 ($r^2=0.4672^*$). Therefore, in the early budding stage of persimmon, when tree is affected by SZT ($-2^{\circ}C$, 3 hours), shoots growth is reduced and increase of 10% in non-budding rate reduces 1.4 of fruits per the three terminal buds of 1YOB.

Study on Correlation between Dynamic Cone Resistance and Shear Strength for Frozen Sand-Silt Mixtures under Low Confining Stress (낮은 구속응력에서 모래-실트 혼합토의 동결강도 평가를 위한 동적 콘 저항력 및 전단강도 상관성 연구)

  • Investigation of in-situ ground in cold region is difficult due to low accessibility and environmental factors. In this study, correlation between dynamic cone resistance and shear strength is suggested to estimate the strength of frozen soils by using instrumented dynamic cone penetrometer. Tests were conducted in freezing chamber after preparing sand-silt mixture with 2.3% water content. Vertical stresses of 5 kPa and 10 kPa were applied during freezing, shearing, and penetration phase to compare the dynamic cone resistance and shear strength. The dynamic cone resistance, additionally, is calculated to minimize the effect of energy loss during hammer impact. Experimental results show that as the shear strength increases, the dynamic cone penetration index (DCPI) decreases nonlinearly, while the dynamic cone resistance increases linearly. This study provides the useful correlation to evaluate strength properties of the frozen soils from the dynamic cone penetration and direct shear tests.

Characteristics of Shear Strength and Elastic Waves in Artificially Frozen Specimens using Triaxial Compression Tests (삼축압축실험을 이용한 인공동결시료의 강도평가 및 탄성파 특성변화)

  • For accurate laboratory evaluations of soil deposits, it is essential that the samples are undisturbed. An artificial ground-freezing system is the one of the most effective methods for obtaining undisturbed samples from sand deposits. The objective of this study is to estimate the shear strengths and the characteristics of elastic waves of frozen-thawed and unfrozen specimens through the undrained triaxial compression test. For the experiments, Jumunjin standard sands are used to prepare frozen and unfrozen specimens with similar relative densities (60% and 80%). The water pluviation method is used to simulate the fully saturated condition under the groundwater table. When thawing the frozen specimens, the temperature is measured every minute. After the specimens are completely thawed, undrained triaxial compression tests are conducted using the same procedures as for the unfrozen specimens. During the triaxial tests (saturation, consolidation, and shear phase), compressional and shear waves are measured. The results show that the freeze-thaw process has minor effects on the peak deviatoric stress and shear strength values, and that the process does not affect the internal friction angle. The compressional wave velocity increases with increasing B-value to 1800 m/s in the saturation phase, but tends to remain constant in the process of consolidation and shearing. The shear wave velocity decreases with increasing B-value in the process of saturation, but changes velocity in accordance with the change in effective stress in the processes of consolidation and shearing. The compressional wave velocity has similar values regardless of the freeze-thaw process, but values of shear wave velocity are slighly lower in frozen-thawed specimens than in unfrozen specimens. This study is a preliminary experiment for estimating the shear strength and characteristics of elastic wave velocity in undisturbed frozen specimens that have been obtained using the artificial ground-freezing method.

Variationsin Air and Ground Temperatures During a Frozen Season in the Subalpine Zone of Mt. Halla (한라산 아고산대의 동결기 기온 및 지온변화)

  • In order to examine the temperature regime responsible for periglacial processes, air and ground temperatures were monitored from October 2010 to May 2011 at a subalpine bare patch (1,710m asl) of Mt. Halla. Four thermistor sensor probes were installed at 55 cm above a ground surface and depths of 2 cm, 10 cm, and 20 cm, respectively. A mean air temperature is $-0.1^{\circ}C$, while mean ground temperatures are $1.8^{\circ}C$ at 2 cm, $2.6^{\circ}C$ at 10 cm and $3.2^{\circ}C$ at 20 cm deep. A mean monthly ground temperature at 2 cm deep demonstrates below $0^{\circ}C$ successively from January to March, while those at 10 cm and 20 cm deep show no sub-zero temperature. A total of 72 freeze-thaw cycle was observed in air temperature. However, the numbers in ground temperature reduced into 17 at 2 cm, 8 at 10 cm, and 3 at 20 cm deep. The cycles of air temperature and ground temperature at 2 cm deep mostly fluctuated diurnally, while those of ground temperature at 10 cm and 20 cm deep exhibited a several-daily oscillation. Snow cover over 55 cm high remained from January to early April, and it seemed to disappear completely on April 16. A seasonal frost of at least 2 cm thick was formed on late December and the isotherm of $0^{\circ}C$ descended slowly into 10 cm deep on late March to early April due to the insulating snow cover. It showed the maximum freezing depth of 20 cm on April 7 to 14 and then thawed rapidly so that the frozen ground did not longer after April 17. Periglacial processes are predominant during a freezing period than a thawing period when the ground surface is still covered with snow. The periglacial mass movement in the subalpine zone of Mt. Halla is mainly generated by frost creep in terms of the occurrence depth of diurnal freeze-thaw cycle and the maximum freezing depth of ground.

Improvement of an Analytical Method for Methoprene in Livestock Products using LC-MS/MS (LC-MS/MS를 이용한 축산물 중 살충제 메토프렌의 잔류분석법 개선)

  • The research aims to develop a rapid and easy analytical method for methoprene using liquid chromatography-tandem mass spectrometry (LC-MS/MS). A simple, highly sensitive, and specific analytical method for the determination of methoprene in livestock products (beef, pork, chicken, milk, eggs, and fat) was developed. Methoprene was effectively extracted with 1% acetic acid in acetonitrile and acetone (1:1), followed by the addition of anhydrous magnesium sulfate (MgSO4) and anhydrous sodium acetate. Subsequently, the lipids in the livestock sample were extracted by freezing them at -20℃. The extracts were cleaned using MgSO4, primary secondary amine (PSA), and octadecyl (C18), which were then centrifuged to separate the supernatant. Nitrogen gas was used to evaporate the supernatant, which was then dissolved in methanol. The matrix-matched calibration curves were constructed using 8 levels (1, 2.5, 5, 10, 25, 50, 100, 150 ng/mL) and the coefficient of determination (R2) was above 0.9964. Average recoveries spiked at three levels (0.01, 0.1, and 0.5 mg/kg), and ranged from 79.5-105.1%, with relative standard deviations (RSDs) smaller than 14.2%, as required by the Codex guideline (CODEX CAC/GL 40). This study could be useful for residue safety management in livestock products.

  • High performance concrete containing silica fume for use in bridge deck overlay emerged as a viable type of bridge deck overlay that economic advantage in construction. They have gained acceptance in Europe, America and Canada in a relatively short time due to their low cost. In this study, high-performance concretes containing silica fume were tested and evaluated in the laboratory to assess their applicability for use in bridge deck overlay. It was conducted with experiments of mechanical and durability characteristics in compressive strength, flexural strength, chloride permeability, abrasion resistance, repeated freezing and thawing cycles and deicing salt scaling resistance. Laboratory test result describe that high-performance concrete containing silica fume for bridge deck overlay application shows most outstanding capacity.

Developmental Ability of Bovine Embryos Nuclear Transferred with Frozen-thawed or Cooled Donor Cells

  • This study was designed to investigate the in vitro developmental ability and apoptosis of bovine embryos nucleartransferred (NT) with frozen-thawed or cooled donor cells. Cultured adult bovine ear cells were used as donor cells after sub-culturing to confluence (CC), cooling to 4$^{\circ}C$ for 48 h, or freezing-thawing (FT). Apoptotic cells in blastocysts were evaluated for apoptosis by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling (TUNEL) method. Fusion, cleavage and blastocyst rates were 69.0 (167/242), 68.8 (115/167), and 29.9 (50/167) with CC cells, 70.4 (88/125), 69.3 (61/88), and 29.6 (26/88) with cooled cells and 66.1 (117/177), 70.1 (82/117), and 13.7 (16/117) with FT cells, respectively. Blastocyst rates of NT embryos derived from FT cells were significantly lower than those from CC or cooled cells (p<0.05). In addition, NT blastocysts produced by using FT cells showed significantly higher apoptosis rates (6.4${\pm}$4.0%) than those produced by CC (2.8${\pm}$1.7%) or cooled (2.3${\pm}$1.3%) cells. However, cooling of donor cells had no significant adverse effect on blastocyst rate as well as apoptosis rate. Therefore, our results suggest that cooled cells may be used as an alternative to freshly cultured confluent culture cells, as donor cells, for the production of Somatic nuclear cloned cattle.

A Study for Microstructure and Durability of Metakaolin Concrete (메타카올린 콘크리트의 미세 공극구조 및 내구성에 관한 연구)

  • The requirement for durability of concrete is increasing recently as a large-scale concrete structure is built. For this reason, the concern about high-durable concrete is being high. Recently, metakaoline to be profitable in economical aspect as well as to have strength and durability of level similar to silica fume is evaluated highly as new admixture. In this study, the scaling, the drying shrinkage, the chloride resistance and the air-void structure are compared for both metakaolin and silica fume concrete. According to the results, the replacement of metakaoline improved the resistance of chloride penetration, freezing and thawing in concrete. On the other hand, as metakaolin was replaced to 10%, it was similar level with OPC in the property of scaling. It was showed that replacement of only metakaoline was similar with OPC in the drying shrinkage. However, MS5 reduced the drying shrinkage about 10%. In conclusion, replacement of the metakaoline 10% is the most excellent performance in terms of durability of concrete.

Relative Microalgal Concentration in Prydz Bay, East Antarctica during Late Austral Summer, 2006

  • Microalgae using a submersible fluorescence probe in water column (up to 100 m) were measured during the austral summer of 2006 (February) in Prydz Bay, East Antarctica (triangular-shaped embayment in the Indian sector of Southern Ocean). Concurrently, environmental parameters such as temperature, salinity and nitrogen (nitrate, ammonium, urea) uptake rates were measured. The concentration of phytoplankton is relatively high due to availability of high nutrients and low sea surface temperature. Phytoplankton community is dominated by diatoms whereas cryptophytes are in low concentration. The maximum concentration of total chlorophyll is 14.87 ${\mu}g\;L^{-1}$ and is attributed to upwelled subsurface winter water due to local wind forcing, availability of micro-nutrients and increased attenuation of photosynthetically available radiation (PAR). Concentration of blue-green algae is low compared to that of green algae because of low temperature. Comparatively high concentration of yellow substances is due to the influence of Antarctic melt-water whereas cryptophytes are low due to high salinity and mixed water column. Varied concentrations of phytoplankton at different times of Fluoroprobe measurements suggest that the coastal waters of Prydz Bay are influenced by changing sub-surface water temperature and salinity due to subsurface upwelling induced by local winds as also melting/freezing processes in late summer. The productivity is high in coastal water due to the input of macro as well as micro-nutrients.

Effect of Aloe on Learming and Memory lmpaiments in Dementia Animal Model SAMP8 (치매동물모델 SAMP8에 있어서 기억. 학습장해에 미치는 알로에의 영향 III. SAMP8의 신경전달물질 및 그 대사산물에 미치는 알로에의 투여효과)

  • Aloe(Aloe arborescens M$_{ILL}$) has been used as a home medicine for the past several thousand in the world, and has been studied on anti-bacterial and anti-fungal activities, hypotension, atherosclerosis, myocardiac infartion, apoplexy, diabetes as a chronic digenerative disease, tumors, gastrointestinal tract, liver and pancreas' diseases, and genitourinary tract etc. SAMP8 as a learing and memory impairment animal model were fed basic and/or experimental diets with 1.0% freezing dried(FD)-aloe for 8 months. The passive avoidance tests such as acqusition trial and retention test were significantly higher in aloe group than in control group. Grading score of senescence resulted in a marked decreases in aloe group compared with control group. Acetylcholinesterase(AChE) activity was remarkably increased in aloe group compared with control group. Neurotransmitters such as dopamine(DA) and serotonin(5-HT) almost did not change by the feeding of aloe-added diet, but their metabolites such as homovanillic acid(HVA) and 5-hydroxy-indole acetic acid(5-HIAA) in aloe group were significantly increased compared with control group. Therefore, the ratios of HVA/DA and 5-HIAA/5-HT as a ratio of metabolite on neurotransmitter were significantly increased by the feeding of aloe-added diet. These results suggest that aloe vara may be activated acetylcholinesterase, the metabolite of neurotransmitter, and ratios of metabolite on neurotransmitter, resulting ina greater prevention of learning and memory impairments such as Alzheimertype dementia.
