Determination of Methylmercury in Biological Samples Using Dithizone Extraction Method Followed by Purge & Trap GC-MS

  • In this study, a dithizone extraction technique involving purge & trap GC-MS was developed for the determination of methylmercury in biological samples, especially blood and fish. After alkaline digestion, methylmercury in biological samples was extracted into dithizone and back-extracted into aqueous sulfide solution. The extracted methylmercury was converted to the volatile ethyl derivative, purged and trapped onto a solid-phase collection medium, and then introduced into the GC-MS system. The determined MDLs of the established method were 0.9 ng·g?1 for biological samples and its accuracy and precision were found to be 93% and 3.8%, respectively. The method was validated by analysis of CRMs such as SRM 966, BCR 463 and IAEA 407 and all analytical results were within certified ranges with average RSDs of less than 6%. The analytical results of field-sampled fish also showed that the method can be successfully used as an alternative for commonly used distillation method followed by GC-CVAFS detection.

폐쇄성(閉鎖性) 황달의 진단에 관한 핵의학적(核醫學的) 고찰 (Scan Manifestation and Trace-Dose Radiogold Clearance Rates in Obstructive Jaundice)

  • The present study has been undertaken to analyze scan manifestations in obstructive jaundice. Scan changes were cprrelated with the duration of jaundice. In addition, clearance rates of trace dose of colloidal radiogold were assessed in each case. The materials were consisted of 19 cases with surgically and/or histopathlogically verified obstructive jaundice from various causes including common duct stone, empyema of the gall bladder and carcinoma of the pancreas head etc. Blood clearance rates of colloidal $^{198}Au$ were determined after the injection of 8 uCi by simple geometrical drawing of recorded strip charts. Scan was performed following additional injection of the same radiogold. In 16 out of 19 cases, the scan revealed rather typical "arborifrom" mottlings around the porta hepatis. The intensity and appearance of such mottlings on the scan was related with the duration of jaundice and level of serum bilirubin and alkaline phosphatase. The blood clearance rates remained to be within normal range in 74 percent of the patients. In the remaining 26 percent, clearance rates ranged between 4.1 and 6 minutes. It is concluded that the association of "arboriform" mottlings and normal or mildly delayed clearance rates in the patients with jaundice is characteristic of obstructive changes in the biliary system.

  • A short and efficient synthesis, solvent extraction and potentiometric measurements of new thiazole-containing naphtho-crown ethers are reported. The naphthalene moiety enhances the ammonium ion selectivity over potassium ion. The selectivity of ${NH_4}^+/K^+$ follows the trend $3\;{\approx}\;2\;>\;1$, indicating that the differences in conformational changes of 2 and 3 in forming ammonium complexes affect little on the resulting ammonium/potassium extraction selectivity ratio. The ammonium ion-selective electrodes were prepared with noctylphenyl ether plasticized poly(vinyl chloride) membranes containing 1-4 the effect of one naphthalene unit introduced on either right (2) or left (3) side of thiazolo-crown ether on their potentiometric properties (e.g., ammonium ion selectivity over other cations, response slopes, and detection limits) were not apparent. However, the ammonium ion selectivity of 1, 2 and 3 over other alkali metal and alkaline earth metal cations is 10-100 times higher than that of nonactin.

산업폐기물의 가속 탄산화법을 이용한 CO2 고용화 및 중금속 안정화 특성 연구 (Stabilization of Heavy Metal and CO2 Sequestration in Industrial Solid Waste Incineration Ash by Accelerated Carbonation)

  • In this study, an accelerated carbonation process was applied to stabilize hazardous heavy metals of industrial solid waste incineration (ISWI) bottom ash and fly ash, and to reduce $CO_2$ emissions. The most commonly used method to stabilize heavy metals is accelerated carbonation using a high water-to-solid ratio including oxidation and carbonation reactions as well as neutralization of the pH, dissolution, and precipitation and sorption. This process has been recognized as having a significant effect on the leaching of heavy metals in alkaline materials such as ISWI ash. The accelerated carbonation process with $CO_2$ absorption was investigated to confirm the leaching behavior of heavy metals contained in ISWI ash including fly and bottom ash. Only the temperature of the chamber at atmospheric pressure was varied and the $CO_2$ concentration was kept constant at 99% while the water-to-solid ratio (L/S) was set at 0.3 and $3.0dm^3/kg$. In the result, the concentration of leached heavy metals and pH value decreased with increasing carbonation reaction time whereas the bottom ash showed no effect. The mechanism of heavy metal-stabilization is supported by two findings during the carbonation reaction. First, the carbonation reaction is sufficient to decrease the pH and to form an insoluble heavy metal-material that contributes to a reduction of the leaching. Second, the adsorbent compound in the bottom ash controls the leaching of heavy metals; the calcite formed by the carbonation reaction has high affinity of heavy metals. In addition, approximately 5 kg/ton and 27 kg/ton $CO_2$ were sequestrated in ISWI bottom ash and fly ash after the carbonation reaction, respectively.

膈下逐瘀湯이 Diethylnitrosamine과 CCl4로 유발된 흰쥐의 肝癌 形成과 肝損傷에 미치는 영향 (The Effect of the Keughachukeo-tang Extract on the Hepatocellular Carcinogenesis and Acute Liver Damage Induced by Diethylnitrosamine and CCl4 in Rats)

  • This study was performed to observe the effect of Keughachukeo-tang(KH) extract on the hepatocellular carcinogenesis and acute liver damage induced by Diethylnitrosamine(DENA) and $CCl_4$ in Rats. Experimental groups were divided into four; normal group(Nor), acute liver damage and hepatocellular cancer inducing control group(Con), KH extract 350㎎/㎏/day(KHA), and 700㎎/㎏/day(KHB) administered groups to Con. The results obtained are as follows: The body weight increased in KHA and KHB than Con from 7th week to the 8th week. The activities of Alanine aminotransferase(ALT) were the most increased in the Con among experimental group. The activities of aspartate aminotransferase(AST), alkaline phosphatase(ALP), and lactacte dehydrogenase(LDH) were significantly decreased(p$<$0.05) in the KHA and KHB compared with Con. Alpha fetoprotein(AFP) were the most increased in the Con among experimental groups. The activities of superoxide dismutase(SOD) were the most increased in the Con among experimental groups. The activities of catalase were significantly increased(p$<$0.05) in the KHA and KHB compared with Con. The results of light microscopical observation, a number of hepatocytes were damaged in the Con compared with Nor and KH extract administerd groups. The number of hepatic p53 positive cells was reduced in the KH extract administered groups compared with Con. These results suggest that administration of KH extract suppress or retard on the Hepatocellular Carcinogenesis and acute liver damage induced by DENA and $CCl_4$ in Rats.

알칼리금속염으로 처리된 활성탄에 대한 CO2의 흡착특성 (Adsorption Characteristics of CO2 on Activated Carbons Treated with Alkali-metal Salts)

  • 연소배가스로부터 $CO_2$를 선택적으로 분리하기 위한 활성탄 흡착공정 사용시 흡착능 향상을 위하여 $CO_2$와 친화력이 있는 화합물을 함침시키는 방법과 KOH를 함침시킨 후 고온에서 열처리하므로서 활성화시키는 방법이 사용되었다. 알칼리금속, 알칼리토금속, 또는 전이금속의 염화물을 함침시킨 활성탄에 대한 $CO_2$의 흡착량을 측정한 결과 함침 전의 활성탄의 흡착량보다 적었다. 이것은 함침되는 물질이 $CO_2$에 대한 친화력이 없이 단지 활성탄의 미세기공만 막는 결과임을 알 수 있었다. 알칼리금속수산화물 중 KOH를 함침시킨 활성탄에 대한 $CO_2$의 파과실험 결과 유입되는 기체에 수분이 있을 경우 흡착량이 증가했는데 이것은 KOH가 $CO_2$를 흡수하는 성질 때문이었다. 그러나 이 흡착제에 함친된 KOH가 $CO_2$와 반응하여 $K_2CO_3$로 변함에 따라 재현성이 없음을 알 수 있었다. KOH를 함침시킨 후 $800^{\circ}C$에서 열처리하여 활성화시킨 활성탄의 경우 함침된 KOH의 양이 증가할수록 $CO_2$의 흡착량이 증가했으며, KOH와 활성탄의 무게비(KOH/Activated-Carbon)가 4일 때 최대였다. 이 흡착제에 대해 온도별로 측정된 $CO_2$의 흡착량으로부터 Clausius-Clapeyron식을 이용하여 등량흡착열을 구했다. 그리고 고정층 파과실험을 통해 $CO_2$농도와 유속에 따른 파과특성을 살펴보았다.

Identification and Characterization of a New Alkaline SGNH Hydrolase from a Thermophilic Bacterium Bacillus sp. K91

  • est19 is a gene from Bacillus sp. K91 that encodes a new esterase. A comparison of the amino acid sequence showed that Est19 has typical Ser-Gly-Asn-His (SGNH) family motifs and could be grouped into the SGNH hydrolase family. The Est19 protein was functionally cloned, and expressed and purified from Escherichia coli BL21(DE3). The enzyme activity was optimal at 60℃ and pH 9.0, and displayed esterase activity towards esters with short-chain acyl esters (C2-C6). A structural model of Est19 was constructed using phospholipase A1 from Streptomyces albidoflavus NA297 as a template. The structure showed an α/β-hydrolase fold and indicated the presence of the typical catalytic triad Ser49-Asp227-His230, which were further investigated by site-directed mutagenesis. To the best of our knowledge, Est19 is a new member of the SGNH hydrolase family identified from thermophiles, which may be applicable in the industrial production of semisynthetic β-lactam antibiotics after modification.

인진호탕(茵蔯蒿湯) 추출액이 Diethylnitrosamine과 CCl4로 유발된 흰쥐의 간암(肝癌) 형성과 간경변(肝硬變)에 미치는 영향 (The Effect of Injinho-tang Extract on Hepatocellular Carcinogenesis and Hepatic Cirrhosis Induced by Diethylnitrosamine and CCl4 in Rats)

  • Injinho-tang(IJ) has been used for the treatment of hepatobiliary diseases. This study was performed to observe the effect of IJ extract on the hepatocellular carcinogenesis and hepatic cirrhosis induced by Diethylnitrosamine(DENA) and $CCl_4$ in Rats. Experimental groups were divided into two ; 8th and 12th week group, and subdivided into four; normal group(Nor), hepatocellular cancer and hepatic cirrhosis inducing control group(Con), and IJ extract 260mg/kg/day(IJA) or 520mg/kg/day(IJB) administered groups to Con. The results obtained are as follows: The body weight was decreased in the Con, IJA and IJB compared with the Nor from the 2nd week to the 12th week. The weight of liver and the weight of liver/100g body weight were decreased significantly in Con, IJA and IJB compared with the Nor. The activities of aspartate aminotransferase(AST) and alanine aminotransferase(ALT) were significantly increased in the Con compared with Nor, but decreased in the IJA and IJB compared with Con from the 8th week group. The activities of alkaline phosphatase(ALP), lactacte dehydrogenase (LDH) and alpha fetoprotein(AFP) were increased significantly in the Con compared with Nor, but decreased in the IJA and IJB compared with Con. The activities of superoxide dismutase(SOD) were decreased in the IJA and IJB compared with Con, but the activities of catalase were increased in the IJA and IJB compared with Con. According to the light and electron microscopical observation, IJA and IJB improved the morphological and histopathological changes of the liver injured by DENA and $CCl_4$. The number of hepatic p53 positive cells was decreased in the IJA and IJB compared with Con. These results suggest that administration of IJ extract suppress or retard DENA and $CCl_4$-induced hepatocelluar carcinogenesis and hepatic cirrhosis in rats.

리튬이온전지용 TiO2 나노튜브 음전극 특성 (Anode Properties of TiO2 Nanotube for Lithium-Ion Batteries)

  • 리튬이온전지의 음전극으로 사용하기 위해 주로 알카리 수열합성법과 열처리에 의해 제조되는 $TiO_2$ 나노튜브의 전기화학적 특성에 관한 연구결과를 조사하여, 그 충방전 특성을 분석하였다. 현재까지 리튬과 $TiO_2$의 전기화학반응으로 생성되는 $Li_xTiO_2$의 이론용량인 $335mAh\;g^{-1}$(x=1)를 초과하는 최대방전용량 $338mAh\;g^{-1}$(x=1.01)을 $TiO_2(B)$ 상을 갖는 나노튜브가 나타내었다. 이것은 리튬의 자가확산이 활성에너지 0.48 eV 정도로 느리므로 이보다 확산거리가 짧도록 $TiO_2$ 나노튜브의 구조를 조정하여 리튬 수송이 원활하도록 하였기 때문이다. 또한 $TiO_2$ 나노튜브 구조체는 벌크상은 물론 표면에서의 뛰어난 이온저장성 때문에 리튬이온전지의 음전극 소재뿐만 아니라 고출력 특성이 필요한 커페시터 소자의 전극소재로도 활용할 수 있다.

수열합성법에 의한 BaTiO3분말합성 및 소결체의 제조 (Preparation and Characterization of Hydrothermal BaTiO3 Powders and Ceramics)

  • 공침전물을 원료로 사용한 수열합성법을 통해 저온에서 미립의 BaTiO$_3$ 분말을 합성하였다. 출발물질로는 BaCl$_2$와 TiCl$_4$ 의 수용액을 사용하였다 이들의 혼합용액을 제조한 다음 과산화물 형태의 침전을 얻기 위해 과산화수소수($H_2O$$_2$)를 첨가하였으며 이렇게 준비된 용액을 암모니아수에 적하하여 공침전물을 얻었다. 이 과산화공침전물을 수열합성의 원료로 사용하였으며 반응온도와 시간 및 pH를 달리하면서 합성하였다. pH 13 이상이 요구되어 강염기인 KOH나 NaOH를 사용하는 일반적인 수열합성법과 달리 암모나아수로도 얻을 수 있는 pH 12 이하에서 perovskite BaTiO$_3$가 합성되었으며, 11$0^{\circ}C$ 이상에서 균일하고 미세한 BaTiO$_3$ 분말을 합성할 수 있었다. 13$0^{\circ}C$ 이상에서 얻은 분말의 경우 합성시간과 관계없이 일정한 물성을 보여주었다. 이렇게 합성된 분말은 76 $m^2$/g의 높은 비표면적을 보였으며 20 nm 이하의 미세한 일차입자들이 약하게 결합된 응집체를 이루고 있었다. 수열합성분말을 이용하여 l150~120$0^{\circ}C$의 온도범위에서 소결한 시편의 소결특성 및 유전 물성을 평가하였다.
