PURPOSES : The objective of this study is to evaluate the SDAR (solvent deasphaltene residue), which is obtained from the solvent deasphalting (SDA) process, as a pavement material. METHODS : The physical properties of the SDAR were evaluated based on its chemical composition, and asphalt mixtures with the SDAR were fabricated and used for the evaluation of mechanical properties. Firstly, the chemical composition of SARA (saturate, aromatic, resin and asphaltene) was analyzed using the TLC-FID (thin-layer chromatography-flame ionization detector). Moreover, the basic material properties of the asphalt binder with the SDAR were evaluated by the penetration test, softening point test, ductility test, and PG (performance grade) grade test. The rheological properties of the asphalt binder with the SDAR were evaluated by the dynamic shear modulus ($G^*$) obtained using the time-temperature superposition (TTS) principle. Secondly, the mechanical properties of the asphalt mixtures with the SDAR were evaluated. The compactibility was evaluated using the gyratory compacter. Moreover, the tensile strength ratio (TSR) was used for evaluating the moisture susceptibility of the asphalt mixtures (i.e., susceptibility to pothole damage). The dynamic modulus $E^*$, which is a fundamental property of the asphalt mixture, obtained at different temperatures and loading cycles, was used to evaluate the mechanical properties of the asphalt mixtures. RESULTS AND CONCLUSION : The SDAR shows stiffer and more brittle behavior than the conventional asphalt binder. As the application of the SDAR directly in the field may cause early failures, such as cracks on pavements, it should be applied with modifiers that can favorably modify the brittleness property of the SDAR. Therefore, if appropriate additives are applied on the SDAR, it can be used as a pavement material because of its low cost and strong resistance to rutting.
대두(大豆)중에 존제하는 장내(腸內) 가스 발생인자인 raffinose와 stachyose의 제거에 이용될 수 있는 Aspergillus niger의 ${\alpha}$-galactosidase는 Czapeck-Dox 액체배지와 밀기울 배지에서 5일간 배양하였을 때 효소생성이 제일 높았다. Czapeck-Dox 액체배양에서 raffinose와 $NaNO_3$가 효소생산에 가장 효과적인 탄소원과 질소원으로 나타났으며 밀기울 배양에서는 질소원 또는 탄소원의 첨가효과가 없었다. 생성된 조효소(組酵素)용액의 작용 최적 pH는 4.0-5.0, 안정도 최적 pH는 3.5-6.5, 최적온도는 $40-50^{\circ}C$ 였다. 합성기질인 p-nitrophenyl-${\alpha}$-D-galactoside에 대한 Michaelis상수는 0.42mM, 최대반응속도는 152${\mu}moles$ substrate/min/kg (고체배지)이었다. $HgCl_2$는 강력한 비경쟁 저해제로 작용하였고 p-chloromercuribenzoate는 낮은 농도에서 경쟁 저해제로 작용하였다. 조효소 용액은 raffinose와 stachyose를 완전 분해하여 단당류 만을 생성하였다.
볶은 들깨박에 존재하는 항암효소계 유도물질을 분리하기 위해 용매분획과 preparative TLC를 실시하여 이들에 대한 암예방지표효소인 quinone reductase와 AHH 유도활성을 조사하였다. 볶은 들깨박의 메탄올 추출물을 용매분획하여 QR을 측정한 결과 chloroform층에서 가장 높은 활성이 나타났다. 들깨박 메탄올 추출물을 TLC로 분리한 분획가운데 QR과 AHH유도활성은 F1$(R_{f}=0.8)$에서 가장 높았으며, 항산화능은 F1$(R_{f}=0.8)$과 F2$(R_{f}=0.7)$에서 가장 강한 것으로 나타나 QR유도성분과 항산화성분이 동일성분일 가능성이 높은 것으로 사료 된다.
본 연구에서는 비교적 성장이 빠르고 배양이 용이한 황갈편조 식물문 중 규조강에 속하는 P. tricornutum으로부터 선정하여 항 산화성 물질을 분리$\cdot$정제하여 물질의 구조를 밝혔다. P. tricornutum의 각 유기용매 추출물의 항산화성은 chloroform획분이 가장 좋았으며, 이 획분을 시료로 하여 silica gel column chromatography, PTLC 및 HPLC를 사용하여 항산화성 물질을 분리$\cdot$정제하였다. Silica gel column을 사용하여 분리된 획분의 항산화성은 dichloromethane: methanol (5: 1)로 용출시킨 획분에서 가장 높았으며, 이 획분의 PTLC 획분은 항산화성이 $20\%$ 증가하였다. 최종 정제된 물질의 $IC_50$값은 $8.3{\mu}g/mL$으로 화학적으로 합성된 항산화제인 BHT 및 BHA보다는 낮았지만 천연 항산화제인 $\alpha-tocopherol$보다는 다소 높게 나타났으며, 이 물질의 화학적 구조는 carotenoid류인 zeaxanthin으로 동정되었다.
The SCO0284 gene of Streptomyces coelicolor A3(2) is predicted to encode an α-galactosidase (680 amino acids) belonging to glycoside hydrolase family 27. In this study, the SCO0284 coding region was cloned and overexpressed in Streptomyces lividans TK24. The mature form of SCO0284 (641 amino acids, 68 kDa) was purified from culture broth by gel filtration chromatography, with 83.3-fold purification and a yield of 11.2%. Purified SCO0284 showed strong activity against p-nitrophenyl-α-D-galactopyranoside, melibiose, raffinose, and stachyose, and no activity toward lactose, agar (galactan), and neoagarooligosaccharides, indicating that it is an α-galactosidase. Optimal enzyme activity was observed at 40℃ and pH 7.0. The addition of metal ions or EDTA did not affect the enzyme activity, indicating that no metal cofactor is required. The kinetic parameters Vmax and Km for p-nitrophenyl-α-D-galactopyranoside were 1.6 mg/ml (0.0053 M) and 71.4 U/mg, respectively. Thin-layer chromatography and mass spectrometry analysis of the hydrolyzed products of melibiose, raffinose, and stachyose showed perfect matches with the masses of the sodium adducts of the hydrolyzed products, galactose (M+Na, 203), melibiose (M+Na, 365), and raffinose (M+Na, 527), respectively, indicating that it specifically cleaves the α-1,6-glycosidic bond of the substrate, releasing the terminal D-galactose.
A novel ${\beta}$-agarase, AgaJ5, was identified from an agar-degrading marine bacterium, Gayadomonas joobiniege G7. It belongs to the glycoside hydrolase family 86 and is composed of 805 amino acids with a 30-amino-acid signal peptide. Zymogram analysis showed that purified AgaJ5 has agarase activity. The optimum temperature and pH for AgaJ5 activity were determined to be $30^{\circ}C$ and 4.5, respectively. AgaJ5 was an acidic ${\beta}$-agarase that had strong activity at a narrow pH range of 4.5-5.5, and was a cold-adapted enzyme, retaining 40% of enzymatic activity at $10^{\circ}C$. AgaJ5 required monovalent ions such as $Na^+$ and $K^+$ for its maximum activity, but its activity was severely inhibited by several metal ions. The $K_m$ and $V_{max}$ of AgaJ5 for agarose were 8.9 mg/ml and 188.6 U/mg, respectively. Notably, thin-layer chromatography, mass spectrometry, and agarose-liquefication analyses revealed that AgaJ5 was an endo-type ${\beta}$-agarase producing neoagarohexaose as the final main product of agarose hydrolysis. Therefore, these results suggest that AgaJ5 from G. joobiniege G7 is a novel endo-type neoagarohexaose-producing ${\beta}$-agarase having specific biochemical features that may be useful for industrial applications.
하이브리드 로켓 연소에서 발생하는 산화제 난류 유동과 연료의 기화로 인한 분출유동 사이의 상호 간섭은 매우 복잡하고 특별한 유동 간섭을 일으킨다. 이를 연구하기 위하여 연소반응을 제외하고, 산화제의 난류 유동과 연료 벽면에서의 분출 유동을 모사한 채널 유동에 대한 LES 해석을 수행하였다. 고체추진 로켓의 연소 과정에서 관찰되는 현상과 매우 흡사하게 벽면 근처에서 특정주파수로 진동하는 유동 현상이 존재한다는 것을 확인하였고, 산화제와 분출 유동의 간섭에 기인한 유동의 진동현상은 벽면 근처의 매우 얇은 영역에서만 존재하였다. 큰 길이 스케일의 유동현상을 보여주는 압력 섭동장으로부터 채널 내 주유동이 특정 주파수 특성을 갖고 하류로 진행해 가는데, 이는 산화제 유동이 분출유동과 상호작용을 하면서 발생된 전단유동의 특성을 나타낸 것이다. 그러나 하이브리드 로켓 연소실 유동의 진동 특성은 고체 추진 로켓에서 관찰되는 유동 특성과는 달리, 진동의 강도가 벽면에서 온도 구배를 변화시켜 열전달의 향상을 발생시키기에는 충분하지 못한 것으로 보인다. 그러나 벽면 근처에서 특정 주파수 특성을 갖는 유동현상이 존재한다는 사실은 비슷한 크기의 주파수를 갖는 음향 가진과 같은 외부교란이 작용한다면 공진으로 발전할 수 있는 가능성을 의미한다.

A biosurfactant-producing bacterial strain was selected from oil-contaminated soil because of its ability to degrade crude oil and tributyrin $(C_{4:0})$. The strain was identified as Bacillus subtilis A8-8 based on its morphological, biochemical, and physiological characteristics. When B. subtilis A8-8 was grown with crude oil as the sole carbon source, the biosurfactant from the strain emulsified crude oil, vegetable oil, and hydrocarbons. Soybean oil was the optimum substrate for the emulsifying activity and emulsion stability of the biosurfactant, both of which were superior to those of several commercially available surfactants. The biosurfactant was purified by a procedure including HCl precipitation, methanol treatment, and silica-gel chromatography. The partially purified biosurfactant was analyzed by TLC (thin-layer chromatography), SDS-PAGE, and HPLC and it reduced the surface tension of water from 72 mN/m to 26 mN/m at a concentration of 30 mg/l. Therefore, the purified lipopeptide biosurfactant has strong properties as an emulsifying agent and acts as an emulsion-stabilizing agent.
In the original plant of Arisaematis Rhizomas, the features of external and internal shape of Arisaema amurense var. serratum $N_{AKAI}$ and A. amurense $_{MAXIM}$. are as follows. 1. In the external shape of original plant, Arisaema amurense var. serratum $N_{AKAI}$ has high stem, an oval-elliptic leaflet and a serrate leaf margin. But A. amurense $M_{AXIM}$ has a relatively low stem, an upside oval leaflet and no serrate leaf margin. 2. On the herbs character of original plant, Ansaema amurense var. serratum $N_{AKAI}$ is small in height and diameter, but A. amurense $M_{AXIM}$ is relatively large in height and diameter. 3. On the gathered herbs character of original plant, the section of Arisaema amurense var. serratum $N_{AKAI}$ is brightly white, easily spilt and strong scent. But the section of A. amurense $M_{AXIM}$ is thin yellowish brown, not easily spilt and weak scent. Also the grade of gathered herbs, both can be classified by diameter and external scent. 4. In the current herbs character, Korean and Sichuan Province products are irregullar shape and have no scent, Jilin Province products are regular shape. Also, Guangxi Province products are small size and concave in one side, Hebei Province products are irregular size and shape. 5. In internal shape of original plant, epidermal cell of Arisaema amurense var. serratum is very tight. With the except of micro difference in parenchyma cell of cortex, on the whole there are nearly no differences with A. amurense $M_{AXIM}$. Also, samely in the internal shape, according to collecting sites, epidermis exhibits a lot layer in curving state and secreting duct is developed and vascular bundle and exists between parenchyma cell of cortex. In the future, additional study is needed to distinguish herb and effect between same genus-degree of relatedness.
Constitutive cyclooxygenase (COX-I) is present in cells under physiological conditions, whereas inducible cyclooxygenase (COX-II) is induced by some cytokines, mitogens, and endotoxin presumably in pathological conditions such as inflammation. We have evaluated the inhibitory effects of solvent fractionated extracts of natural products on the activities of COX-I and COX-II. Oxygen uptake COX assay was performed, as a primary screening from the tissue extracts of bovine seminal vesicles (BSV), by monitoring the initial rate of oxygen uptake using an oxygen electrode. Additionally, we evaluated plant extracts for the inhibitory effects of COX-I (in HEL cells) and COX-II (in lipopolysaccharide activated J774A.1 macrophages) using thin layer chromatography of prostanoids produced from $^{14}C-labelled$ arachidonic acid (AA). The use of such models of COX-I and COX-II assay will lead to the identification of specific inhibitors of cyclooxygenases with presumably less side effects than present therapies. Inhibitory effects of 50 kinds of plant extracts on the COX-I and COX-II activities were determined and the active fractions were found in the ethyl acetate fractions of Dryopteris crassirhizoma (roots), Amomum cardamomum (roots), Triticum aestivum (seeds), Perilla sikokiana (leaves), Anemarrhena asphodeloides (roots). Especially, the ethyl acetate fraction of Dryopteris crassirhizoma (roots), which exhibited the strong inhibition against BSV COX $(IC_{50},\;65.4\;{\mu}g/ml)$, COX-I $(IC_{50},\;8.5\;{\mu}g/ml)$, and COX-II $(IC_{50},\;17.2\;{\mu}g/ml)$, is under investigation to isolate active principles using activity-guided fractionation method.
