Identification of vitro vulnificus lrp and Its Influence on Survival Under Various Stresses

  • An lrp gene encoding a leucine-responsive regulatory protein was identified from Vitro vulnificus, and its role in the survival of the organism was assessed by analyzing the stress tolerance of the isogenic mutant, in which the lrp gene had been inactivated. The results demonstrated that Lrp contributes to the survival of V. vulnificus is dependent of the phase of growth.

  • 카드뮴은 환경과 인체 위해도에 큰 영향을 미치는 중요한 환경오염물질로 잘 알려져 있다. 본 연구에서는 토양선충인 Caenorhabditis elegans에 카드뮴을 12시간과 48시간으로 나누어 처리하여 시간에 따른 장, 단기적 독성영향을 알아보고자 하였다. 이때 생리학적 수준으로 성장 및 생식을 조사하고, 분자수준에서 스트레스 관련 유전자들의 시간에 따른 발현 정도를 관찰하였다. 생식에서는 단기노출(12시간) 시 그 영향이 대조군에 비해 크게 나타났으며, mtl-2의 스트레스 관련 유전자가 증가하였다 장기 노출(48시간) 시에는 cyp35a2, ape-1, sod-1, ctl-2 유전자가 대조군에 비해 약 $2{\sim}4$배 가량의 발현 증가 결과를 조사할 수 있었다. 본 연구결과들을 통해 스트레스 관련 유전자의 발현을 조사하는 것이 중요하고 민감한 생체지표가 된다는 것과 토양선충 C. elegans는 환경중 오염물질에 대한 장기, 단기적 영향을 평가하기 위한 좋은 생물학적 모델이 된다는 것을 알 수 있었다.

Association Analysis of Reactive Oxygen Species-Hypertension Genes Discovered by Literature Mining

  • Oxidative stress, which results in an excessive product of reactive oxygen species (ROS), is one of the fundamental mechanisms of the development of hypertension. In the vascular system, ROS have physical and pathophysiological roles in vascular remodeling and endothelial dysfunction. In this study, ROS-hypertension-related genes were collected by the biological literature-mining tools, such as SciMiner and gene2pubmed, in order to identify the genes that would cause hypertension through ROS. Further, single nucleotide polymorphisms (SNPs) located within these gene regions were examined statistically for their association with hypertension in 6,419 Korean individuals, and pathway enrichment analysis using the associated genes was performed. The 2,945 SNPs of 237 ROS-hypertension genes were analyzed, and 68 genes were significantly associated with hypertension (p < 0.05). The most significant SNP was rs2889611 within MAPK8 (p = $2.70{\times}10^{-5}$; odds ratio, 0.82; confidence interval, 0.75 to 0.90). This study demonstrates that a text mining approach combined with association analysis may be useful to identify the candidate genes that cause hypertension through ROS or oxidative stress.

  • Too little and too much water due to climatic events is a significant cause of global food insecurity. Crops are less productive under water-limited conditions and all major crops, with the exception of rice (Oryza sativa), die within a few days of complete submergence. To complement our studies on genes such as SUB1A, (an ERF-VII transcription factor that provides robust submergence tolerance) and AG1 (a TREHALOSE 6-P PHOSPHATASE that promotes establishment of young seedlings underwater), we have retooled INTACT (${\underline{I}}solation$ of ${\underline{N}}uclei$ ${\underline{TA}}gged$ in specific ${\underline{C}}ell$ ${\underline{T}}ypes$) and TRAP (${\underline{T}}ranslating$ ${\underline{R}}ibosome$ ${\underline{A}}ffinity$ ${\underline{P}}urification$) for rice. These technologies enable us to follow dynamics in chromatin, nuclear pre-mRNAs and ribosome-bound mRNAs in meristems and diverse cell types. With these technologies we can better interpret responses to stresses and reestablishment of homeostasis. These include stress acclimation strategies involving changes in metabolism and development, such as dynamics in suberin deposition in sub-epidermal layers of roots that limit water loss under drought and oxygen escape during waterlogging. Our new data uncover dynamic and reversible regulation at multiple levels of gene regulation and provide new insights into processes of stress resilience. Molecular Cloning, Phylogenetic Analysis, Expressional Profiling and In Vitro Studies of TINY2 from Arabidopsis thaliana

  • A cDNA that was rapidly induced upon abscisic acid, cold, drought, mechanical wounding and to a lesser extent, by high salinity treatment, was isolated from Arabidopsis seedlings. It was classified as DREB subfamily member based on multiple sequence alignment and phylogenetic characterization. Since it encoded a protein with a typical ERF/AP2 DNA-binding domain and was closely related to the TINY gene, we named it TINY2. Gel retardation assay revealed that TINY2 was able to form a specific complex with the previously characterized DRE element while showed only residual affinity to the GCC box. When fused to the GAL4 DNA-binding domain, either full-length or its C-terminus functioned effectively as a trans-activator in the yeast one-hybrid assay while its N-terminus was completely inactive. Our data indicate that TINY2 could be a new member of the AP2/EREBP transcription factor family involved in activation of down-stream genes in response to environmental stress.

OxyR Regulon Controls Lipid Peroxidation-mediated Oxidative Stress in Escherichia coli

  • Membrane lipid peroxidation processes yield products that may react with DNA and proteins to cause oxidative modifications. The oxyR gene product regulates the expression of enzymes and proteins that are needed for cellular protection against oxidative stress. Upon exposure to tert-butylhydroperoxide (t-BOOH) and 2,2'-azobis (2-amidinopropane) hydrochloride (AAPH), which induce lipid peroxidation in membranes, the Escherichia coli oxyR overexpression mutant was much more resistant to lipid peroxidation-mediated cellular damage, when compared to the oxyR deletion mutant in regard to growth kinetics, viability, and DNA damage. The deletion of the oxyR gene in E. coli also resulted in increased susceptibility of superoxide dismutase to lipid peroxidation-mediated inactivation. The results indicate that the peroxidation of lipid is probably one of the important intermediary events in free radical-induced cellular damage. Also, the oxyR regulon plays an important protective role in lipid peroxidation-mediated cellular damage.

OsAREB1, an ABRE-binding protein responding to ABA and glucose, has multiple functions in Arabidopsis

  • Expression patterns of OsAREB1 revealed that expression of OsAREB1 gene can be induced by ABA, PEG and heat. Yeast one-hybrid assay demonstrated it can bind to ABA-responsive element (ABRE), which was found in most stress-induced genes. Transgenic Arabidopsis over-expressing OsAREB1 had different responses to ABA and glucose compared to wild-type plants, which suggest OsAREB1 might have a crucial role in these two signaling pathways. Further analysis indicate that OsAREB1 have multiple functions in Arabidopsis. First, OsAREB1 transgenic plants had higher resistance to drought and heat, and OsAREB1 up-regulated the ABA/stress related gene such as RD29A and RD29B. Second, it delayed plant flowering time by down-regulating the expression of flowering-related genes, such as FT, SOC1, LFY and AP1. Due to the dates, OsAREB1 may function as a positive regulator in drought/heat stresses response, but a negative regulator in flowering time in Arabidopsis.

Effects of Heterologous Expression of Thioredoxin Reductase on the Level of Reactive Oxygen Species in COS-7 Cells

  • Thioredoxin reductase (TrxR), a component of the redox control system involving thioredoxin (Trx), is implicated in defense against oxidative stress, control of cell growth and proliferation, and regulation of apoptosis. In the present study a stable transfectant was made by introducing the vector pcDNA3.0 harboring the fission yeast TrxR gene into COS-7 African green monkey kidney fibroblast cells. The exogenous TrxR gene led to an increase in TrxR activity of up to 3.2-fold but did not affect glutathione (GSH) content, or glutaredoxin and caspase-3 activities. Levels of reactive oxygen species (ROS), but not those of nitric oxide (NO), were reduced. Conversely, 1-chloro-2,4-dinitrobezene (CDNB), an irreversible inhibitor of mammalian TrxR, enhanced ROS levels in the COS-7 cells. After treatment with hydrogen peroxide, the level of intracellular ROS was lower in the transfectants than in the vector control cells. These results confirm that TrxR is a crucial determinant of the level of cellular ROS during oxidative stress as well as in the normal state.

Expression of Ajuba, a Novel LIM Protein, is Regulated by Endorlasmic Reticulum Stress (소포체 스트레스가 Ajuba 발현유도)

  • Ajuba is a number of proteins containing cytosolic LIM domain. Its function may provide a new pathway whereby cell-cell adhesive events are transmitted to the nucleus to regulate cell proliferation and differentiation decisions. Here, Ajuba gene expression was investigated its molecular properties associated with endoplasmic reticulum (ER) stresses (tunicamycin, DTT, A23187 and BFA) which induced remarkable ex-pression of Ajuba mRNA. The mRNA half life of Ajuba was also determined, its half life of Ajuba mRNA in FRTL-5 cells was approximately 2 hr after the initial translation. Although the obvious bioligical function of Ajuba is not clear, on the base of the results, Ajuba gene expression is deeply associated with ER stresses.

Isolation and Characterization of Terpene Synthase Gene from Panax ginseng

  • Terpene synthase plays a key role in biosynthesis of triterpene saponins (ginsenosides) and is intermediate in the biosynthesis of a number of secondary metabolites. A terpene synthase (PgTPS) cDNA was isolated and characterized from the root of Panax ginseng c.A. Meyer. The deduced amino acid sequence of PgTPS showed a similarity with A. deliciosa (AAX16121) 61%, V. vinifera (AAS66357) 61%, L. hirsutum (AAG41891) 55%, M. truncatula (AAV36464) 52%. And the segment of a terpene synthase gene was amplified by reverse transcriptase-polymerase chain reaction (RTPCR). We studied expression of terpene synthase under stressful conditions like chilling, salt, UV, and heavy metal stress treatment. Expression of PgTPS was increased gradually after exposure to stresses such as chilling, salt, and UV illumination. But its transcription seems to be reduced by cadmium and copper treatment.
