산불로 인한 해양환경오염원의 생물학적 방어반응 스트레스 유전자 규명

  • 해조류를 대상으로 한 해양환경 오염원의 영향에 대한 연구는 지금까지 주로 자연환경 스트레스에 대한 연구가 주로 진행되어져 왔으나 (Davison and Pearson, 1996), 해양오염원의 영향 (진 등, 1999)등은 아직 연구가 미진한 실정이다. 따라서 본 연구에서는 산불로 인하여 야기될 수 있는 오염원들을 포함한 첫물 (소나무 잿물, KOH), 토사 (황토, 화강암 마사토), 중금속 (CdCl$_2$, CuSO$_4$) 등를 재료로 하여 방사무늬김의 viability 정도 측정과 이들 stress에 따른 반응 유전자들을 분리하고자 한다. Expression of the cAMP Phosphodiesterase 7A1 Gene by Endoplasmic Reticulum Stress (소포체스트레스에 의한 cAMP phosphodiesterase 7A1 유전자의 발현)

  • This study demonstrated that upregulation of gene expression of endoplasmic reticulum (ER) stress chaperones (Bip, ERp29, calnexin, and PDI), ER stress sensors (PERK, ATF6, and Ire1), and cAMP phosphodiesterase 7A1 (cAMP PDE7A1) was induced by ER stresses in FRTL5 cells. While removing A23187 from the culture medium restored upregulation of cAMP PDE7A1 gene expression, removal of thapsigargin did not recover its expression. In addition, cAMP PDE7A1 gene expression was strongly inhibited by treatment with A23187 combined with thyroid stimulating hormone (TSH). The results are the first to show that ER stress induces cAMP PDE7A1 gene expression.

Charaterization of GroEL Homolog from Streptococcus pneumoniae (폐렴구균 열충격 단백질 GroEL의 특성)

  • Induction and purification of the GroEL homolog from Streptococcus pneumolliae were characterized. The stress conditions were determined by temperature, ethanol, NaCI, $H_2O_2$ methyl methanesulfonate, and ethyl methanesulfonate. And stress induced proteins were analyzed using [$^{35}S$]-methionine labeling method. Heat shock induced the synthesis of a set of about 3 heat shock proteins (hsps) (65, 73, and 84-kDa). Of those 3 hsps, a 65 kDa protein, hsp65, was purified by DEAE-Sephacel and ATP-agarose column chromatography, and used for antibody preparation. Immunoblot analysis employing antisera raised against pneumococcus hsp65 demonstrated cross-reactivity with a 60 kDa protein in Eschericha coli. Also cross reaction of the purified p65 with anti-Escherichia coli GroEL monoclonal antibody demonstrated that pneumococcal hsp65 is the GroEL homolog.

Anti-stress Effect by the Treatment of Vitamin C (Vitamin C 투여에 의한 항스트레스 효과)

  • The anti-stress effect by the treatment of vitamin C was investigated in this study. The treatment of ascorbic acid in the presence of $Cu^{2+}$ion induced strong time- and dose-dependent degradation of hitamine, and also the addition of histamine accelerated time-dependent decomposition of ascorbic acid in vitro. The treatment of ascorbic acid in $ODS^{od}/_{od}$rats, which cannot synthesize ascorbic acid, significantly decreased the urinary histamine. The protreatment of ascorbic acid, dexamethasone and promethazine inhibited the lethal effect induced by immobilization stress, but that of dimethylsulfoxide did not. The addition of ascorbic and to a culture of spleen cells of $ODS^{od}/_{od}$rats significantly increased the Con A-dependent T lymphocyte proliferation.

  • The change of amino acids was analysed with cotyledons and roots of young spring radish by the treatment of $Cd^{2+}$ ion and $Cd^{2+}$ ion plus PA. The concentration of Asp was decreased by the treatment of $Cd^{2+}$ ion, but Ala, Phe, Val, Ile, Typ, Lys and Arg was increased in the cotyledons. The concentration of Lys and Arg was decreased by the treatment of $Cd^{2+}$ ion plus PA at the same time. The concentration of basic amino acids, His, Lys and Arg was increased by the treatment of $Cd^{2+}$ ion, and decreased by the treatment of $Cd^{2+}$ ion and $Cd^{2+}$ ion plus PA in the roots. Only the concentration of Pro was increased by the treatment of $Cd^{2+}$ ion plus PA. This results showed that Pro was induced against stress of PA, and assumed that the change of other amino acids concentration may be relation to the metabolism against stress.

  • Simultaneous removal of phosphorus and nitrogen from wastewater was studied by the anaerobic-aerobic system of activated sludge. In the anaerobic stage, most of the influent glucose was removed and orthophosphate was released, when the nitrate and/or nitrite concentration in the wastewater was almost zero. The amount of the released phosphorus was found to be directly proportional to the amount of the removed glucose. When the ratio of phosphorus to glucose in the influent was less than 0.04, the phosphorus in the wastewater was almost completely removed during the aerobic state. Under the anaerobic condition, activated sludge released phosphate and excess removal of phosphate occurred during the aerobic condition. Namely, the stress received in anaerobic period stimulated the uptake of phosphorus in aerobic period. The amounts of phosphorus release in the anaerobic and uptake in the aerobic stage were less in proportional to the concentration of $NO_x-N$. Further, if the initial ratio of $NO_2-N$/glucose was less than 0.37, the inorganic nitrogen in the influent could be completely removed.

  • Plants are altered not only in the outward appearance but also in their physiological and biochemical properties with reaction to the environmental stresses; particularly, the biosynthetic system of protein in situ rapidly responds to this. In order to investigate the change of properties of polypeptides in rice plants induced by several stresses, the seedlings were subjected to exposure to NaCl, drought, and low and high temperatures, respectively, and then some aspects of polypeptide variations were compared. Without exception, the rice plant, which is somewhat tolerant to environmental change, shows the alteration in several polypeptides. Moreover, newly synthesized polypeptides were observed in response to stresses. The existing proteins for the primary metabolic pathways were markedly decreased as each treatment progressed.

Research on Correlation between Stress and Menstruation through Analyzing Menstruation State of Highschool Senior Female Students (고3 수험생의 월경실태분석을 통한 Stress와 원경의 상관관계 조사(Stress와 월경의 상관관계))

  • Objective: Stress is assumed to have many effects on menstruation of highschool senior female students, and it is expected to cause many problems and low efficiency during their studies. Therefore, through analyzing the menstruation state of highschool senior female students, the effects of stress on their menstruation and their education were studied. Methods: We made a survey of 583 senior female students of 'N' highschool located in Taegu. The survey was composed of the degrees of stress they were undergoing, menstrual problems caused by stress. Results: The survey results showed that 96.7% of the candidates were experiencing stress. 51.6% of them experienced an altered menstrual cycle, and menstrual irregularity was brought out in 32.1 % among them. 18.1 % of them were experiencing dysfunctional uterine bleeding. In sustaining period of menstruation, 17.5% of them had a shortened menstruation period, and 8.9% had a prolonged period. In case of menstrual quantity, less than normal quantity was reported by 26.9% of respondents and more than normal quantity was reported by 8.8%. 26.6% of respondents reported their menstrual blood was impure and the color was dark. Blood clots were increased in 27.5% of respondents. 86.6% went through premenstrual syndrome, and 34.3% had worsened menstrual pain. Studying was affected by menstrual pain in 83.9% of respondents. Of treating the disorder, 66.2% of them said they just waited till the pain went away, and 26.9% of them said they went to a pharmacy, and 2.4% of them said they went to an oriental medical clinic. Conclusions: From the above results, it is concluded that stress has a close connection with menstrual disorders. And it affects senior highschool female students' education, while they don't take proper treatments. Hence we think that we must have a lot of concern about senior female students and they must receive proper treatments.

Changes of Salmon Meat Texture During Semi-Drying Process (조미 반건조 제품 가공 공정에 따른 연어육 Texture의 변화)

  • To obtain basic data for processing semi-dried salmon meat product, the results that were measured the textural properties of salmon meat during salting, sugaring and drying process followed. Drying time and temperature were longer, the moisture amount of salmon meat were reduced. Hardness of salmon meat was direct proportion to shear stress, but hardness was inverse proportion to cohesiveness during drying process. After sugaring and salting salmon meat, drying time was longer, hardness and shear stress of salmon meat were increased. The sensory evaluation of the textures of sugaring salmon meat dried for 3 hrs showed slightly good. In the changes of texture of salmon meat during steaming hardness and shear stress of salmon meat dried for 4 hrs were higher than that dried for 10 hrs. And steaming time was longer, hardness of salmon meat dried for 4 hrs was decreased and cohesiveness was not changed.
