The Effect of Chonhwasan and Okchunsan in Streptozotocin induced Hyperglycemic Rats (천화산(天花散)과 옥천산(玉泉散)이 고혈당(高血糖) 백서(白鼠)에 미치는 영향(影響))

  • This present study was carried out to investigate the effect of Chonhwasan and Okchunsan in streptozotocin induced hyperglycemic rats. Body weight, serum levels of glucose, insulin, total cholesterol triglyceride and urine levels of volume, glucose, protein were measured in streptozotocin induced hyperglycemic rats orally receiving extracts of Chonhwasan and Okchunsan plus Radix Trichosandis for 4 days. According to the result, Chonhwasan and Okchunsan have the effect of decreased body weight delate and decreased blood sugar, serum lipid metabolism and urine volume, glucose, protein in streptozotocine induced hyperglycemic rats, and they were expected to be applied to the treatment of Diabetes Mellitus.

Antidiabetic and Lipid Metabolism Effect of Paediomyces Japonica in Streptozotocin-Induced Diabetic Rats (눈꽃동충하초(paecilomyces japonica)추출물의 지질대사 및 당대사에 미치는 영향)

  • This study was carried to investigate the antidiabetic and lipid metabolism of water extract paecilomyces japonica(PJ) in Streptozotocin (STZ) induced diabetic rats. Diabetes were induced by intravenous injection of STZ at a dose of 42mg/kg dissolved in citrate buffer. The water extract of paecilomyces japonica were orally administrated once a day for 7 days at a dose of 500mg/kg or 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in PJ treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(G-6-PDH), glucokinase(GK) were significantly increased, but activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in PJ treated group compared to the those of STZ-control group. These results indicated that water extract of paecilomyces japonica would have antidiabetic and lipid metabolism effect in STZ-induced diabetic rats.

Antidiabetic Effect of Glechoma hederacea LINNAEUS in Streptozotocin-Induced Diabetic Rats (Streptozotocin 유발 당뇨 흰쥐에 대한 금전초 추출물의 혈당 강하 효과)

  • This study was done to investigate the antidiabetic effect of Glechoma hederacea LINNAEUS in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45 mg/kg dissolved in citrate buffer. The methanol extract of Glechoma hederacea was orally administrated once a day for 6 days. The contents of serum glucose, triglyceride (TG), total cholesterol and Atherogenic Index (Al) were significantly decreased, but high density lipoprotein (HDL)-cholesterol and HDL-cholesterol/total cholesterol ratio (HTR) were significantly increased in Glechoma hederacea treated STZ-sample group compared to the those of STZ-control group. The content of hepatic lipid peroxide and activity of catalase were decreased, but content of glutathione as well as activities of glutathione-S-transferase and superoxide dismutase were increased in Glechoma hederacea treated STZ-sample group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase, glucokinase were significantly increased, but activity of glucose-6-phosphatase was decreased in Glechoma hederacea treated STZ-sample group compared to the those of STZ-control group. These results indicated that methanol extract of Glechoma hederacea would have antidiabetic effect in STZ-induced diabetic rats.

Antidiabetic Effect of Yacon(Smallanthus sonchifolius) of Root and Leaf in Streptozotocin-Induced Diabetic Rats (Yacon의 뿌리와 잎의 추출물이 streptozotocin으로 유발된 흰쥐의 항당뇨 효과)

  • This study was carried to investigate the antidiabetic effect of ethanol extract of Smallanthus sonchifolius(S.S) in Streptozotocin(STZ)- induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 43mg/kg dissolved in citrate buffer. The ethanol extract of S,S in leaves and root were orally administrated once a day for 7 days at a dose of 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in S.S treated group compared to the those of STZ-control group. The content of hepatic glycogen and activity of glucokinase(GK) were significantly increased,and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in S.S treated group compared to the those of STZ-control group, but activity of glucose-6-phosphate dehydrogenase(G-6-PDH) was not significantly increased, These results indicated that ethanol extract of S.S would have antidiabetic effect in STZ-induced diabetic rats.

Antidiabetic and Antioxidative Effects of Lycii fructus in Streptozotocin-Induced Diabetic Rats (Streptozotocin으로 유발된 흰쥐에 대한 구기자 추출물의 혈당강하 및 항산화 작용에 미치는 효과)

  • This study was done to investigate the antidiabetic and antioxidant effects of Lycii fructus in Streptozotocin (STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose 45mg/kg.b.w. dissolved in citrate buffer(pH4.5). The ethanol extract of Lycii fructus was orally administrated once a day for 7 days. The contents of serum glucose, total cholesterol were significantly decreaed (p<0.05) in Lycii fructus treated group compared to the those of STZ-control group, also content of Triglyceride (TG), Altherogenic index (AI) were decreased, but not statistically significant. But high density lipoprotein (HDL)-cholesterol and HDL-cholesterol/total cholesterol ratio (HTR) were increased in Lycii fructus treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(G-6PDH), glutathione-s-transferase(GST), superoxide dismutase(SOD) were significantly increased(p<0.05), glucose-6-phos-phatase(G-6pase)was significantly decreased(p<0.05) and The glutathione(GSH), glucokinase(GK) were increased. But not statistically significant In Lycii fructus treated group compared to the those of STZ-control group These results indicated that ethanol extract of Lycii fructus would have antidiabetic and antioxidant effects in STZ-induced diabetic rats.

Effectsof Garlic Vinegar Supplementation on Body Weight, Blood glucose, and Serum Lipid Profile in Streptozotocin-induced diabetic Rats-fed High Cholesterol Diet

  • The effectsof garlic vinegar supplementation on body weight, blood blucose, and plasma triglyceride and cholesterol were investigated in streptozotocin -induced diabetic rats fed high-cholesterol(1%) diet for 4 wk. The garlic vinegar was made by fermenting 20% frsh garlic juice. There was no effect of garlic vinegar on body weight, plasma glucose or triglyceride concentrations in the diabetic rats. Plasma total -cholesterol concentrations were unaffected by garlic vinegarr supplementation. However, plasma LDL-cholesterol concentrations and atherogenic index were significantly lower in the diabetic rats supplemented with garlic vinegar diet than tin the cotnrol rats. The diabetic-rats supplemented with garlic vinegar not only had increased HDL-cholesterol levels but had decreased LDL-cholesterol. This alteration in the HDL/LDL-cholesterol ratio in the diabetic rats may decrease the risk of atheroscelrosis. Therefore, the over-all effect of garlic vinegar supplement may contribute to the antiatherogenic role in streptozotocin-induced diabetic rats.

Effects of Gentiana scabra Bunge var.buergeri Max. Extract on Blood Glucose and Lipid Composition in Streptozotcin induced Hyperglycemic Rats (용담(Gentiana scabra Bunge var.buergeri Max.) 추출액이 Streptozotocin 유발 당뇨성 흰주의 혈당 및 지질 성분에 미치는 영향)

  • This study was designed to observe the effects of the feeding Gentiana scabra Bunge var.buergeri Max. extract on the improvement of the blood glucose, lipids in the serum of streptozotcin(55mg/kg of body weight, I.P. injection) induced hyperglycemic rats (S.D. strain, ) fed the experimental diets for 4 weeks. Concentrations of blood glucose were significantly higher in the streptozotocin administration(I.P.) groups (group 2(streptozotcin+water), 3(streptozocin+Gentiana scabra Bunge extract), 4(streptozotocin+cholesterol+water), 5(streptozotocin+cholesterol+Gentiana scabra Bunge extract) than those in the control group(group 1(water)). Bolld glucose concentrations were rather lower in the group 3(streptozotocin+Gentiana scabra Bunge extract)than in the group 2(streptozotocin+water), and then those of the group 5 were rather lower than in the group 4. There was almost little significance among the groups. concentrations of total cholesterol in serum were lower in the group 5 than those in the group 4, and the group 3 were rather lower than in the group 2. Concentrations of LDL-cholesterol in serum were significantly lower in the group 3 than those 2. In the ratio of HDL-cholesterol concentrations to total cholesterol concentrations, Gentiana scabra Bunge extract administration groups were higher percentage than the other goups. Concentrations of free cholesterol and cholesteryl ester in serum were rather lower in the groups 3,5 than the other groups, and then those of the group were higher percentage than the other groups. concentrations of free cholesterol and cholesteryl ester in serum were rather lower in the groups 3,5 than the groups, and then those of the group 3 were significantly lower than in the group 4. Concentrations of triglyceride and phospholipid in serum were decreased in the group 3 than in the other groups. The activities of aspartate aminotransferase and alanine aminotransferase in serum were increased in the all experimental groups than in the control group. From the above research, the Gentiana scabra Bunge var.buergeri Max. extracts were effective on the improvement of the lipid compositions in serum of streptozotcin induced hyperglycemic and hypercholosterolemic rats.

Hypoglycemic Effect of Fractions of Cassia tora Extract in Streptozotocin-Induced Diabetic Rats (결명자 분획물이 당뇨 유발 흰쥐의 혈당에 미치는 영향)

  • The hypoglycemic effect of fractions of methanol extract of Cassia tora was investigated in diabetic rats. Sprague-Dawley male rats weighing 180∼230 g were divided into 6 groups. Diabetes was induced in the male rats by intravenous injection of streptozotocin at a dose of 45 mg/kg dissolved in citrate buffer (pH 4.5). The diabetic animals then had plasma glucose concentration of above 300 mg/㎗. Fractions of methanol extract of Cassia tora were administered orally into the diabetic rats for 14 days after streptozotocin injection. The food intake and body weight gain were monitored and plasma levels of glucose, triglyceride, free fatty acid, HDL-cholesterol, cholesterol and ALT activity were determined. Levels of glycogen, cholesterol and triglyceride in liver were analysed. Weights of liver and kidney were lighter in all the fraction-administered groups than streptozotocin-control group. Plasma glucose level was significantly decreased by the administration of butanol fraction at 14 days. Plasma triglyceride levels were lower in chloroform, butanol and H$_2$O fraction-administered groups compared to streptozotocin-control group. Plasma cholesterol and HDL-cholesterol levels were not significantly different in all groups. Administrations of each of the four fractions have decreased plasma free fatty acid level, ALT activity and liver triglyceride levels in the diabetic rats. Liver glycogen levels in chloroform, butanol and H$_2$O fraction-administered groups were higher than streptozotocin-control group. It is suggested from the results that butanol fraction of methanol extract of Cassia tora may contain the antihyperglycemic compounds.

Beneficial Effect of Lespedeza cuneata (G. Don) Water Extract on Streptozotocin-induced Type 1 Diabetes and Cytokine-induced Beta-cell Damage

  • The aim of this study was to evaluate the anti-diabetic effects of the water extract of Lespedeza cuneata (LCW) using rat insulinoma (RIN) m5F cells and streptozotocin (STZ)-induced diabetic rats. The effect of LCW on the protection of pancreatic beta cells was assessed using MTT assay, and nitric oxide production was assessed using Griess reagent. STZ-induced diabetic rats were treated with 100 and 400 mg/kg body weight of LCW for 5 weeks. In results, LCW significantly protected cytokine-induced toxicity and NO production, and increased insulin secretion in RINm5F cells. LCW significantly decreased serum blood glucose, thiobarbituric acid reactive substances (TBARS), blood urea nitrogen (BUN) and advanced glycation end products (AGEs) levels, and renal fibronectin expression in STZ-induced diabetic rats. Also, LCW effectively improved BW loss in STZ-induced diabetic rats. Thus, our results suggest that LCW has a beneficial effect on cytokine-induced pancreatic beta cell damage and biomarkers of diabetic complication in hyperglycemic rats.

  • The studies were conducted to investigate the anti-diabetic activities on the hyperglycemia induced by streptozotocin in rats, anti-hypertensive activities in SHR, anti-fatigue and decrease of body weight activities in mice by Kyung Ok-Ko water extract and drink, which is a traditional preparation in Korea. 1. The blood glucose levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(300, 600, 1200 mg/kg) of Kyung Ok-Ko water extract. Particularly, administration of 600, 1200 mg/kg were significantly shown to decrease glucose levels comparing with control group. 2. The serum total cholesterol levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(300, 600, 1200 mg/kg) of Kyung Ok-Ko water extract. Particularly, administration of 600, 1200 mg/kg were significantly shown to decrease total cholesterol levels comparing with control group. 3. The serum triglyceride levels of streptozotocin-induced hyperglycemic rats were dose-dependently decreased by administrations of various doses(300, 600, 1200 mg/kg) of Kyung Ok-Ko water extract. Particularly, administration of 600, 1200 mg/kg were significantly shown to decrease triglyceride levels comparing with control group. 4. The blood pressure in SHRs were dose-dependently lower descended by administration of Kyung Ok-Ko water extract 300, 600, and 1200 mg/kg, respectively. 5. The swimming time of Kyung Ok-Ko water extract(100, 200, 400 mg/kg) and drink(0.7, 1.4, 2.1 ml/kg) were significantly extended to all experimental group dose-dependantly. 6. The decrease of body weight of Kyung Ok-Ko water extract(100, 200, 400 mg/kg) and drink(0.7, 1.4, 2.1 ml/kg) were significantly evaluated dose-dependently in all experimental group.
