Prevalence of Antibiotics in Nectar and Honey in South Tamilnadu, India

  • Reverse-Phase High-performance Liquid Chromatography (RP-HPLC) based technique is one of the most sensitive techniques to detect the antibiotics present in honey. In the southern part of Tamilnadu, India, majority of the farmlands are occupied by plantations such as coconut, banana and rubber. A variety of antimicrobial compounds and antibiotics, which have been reported in pollen, nectar and other floral parts of the plant, gets accumulated in honey through honeybees (Apis mellifera). We have collected the nectar samples from banana (Musa paridasiaca) and rubber (Ficus elastica) flowers and the honey from honey hives of banana and rubber cultivated areas. The extracted nectar and honey samples are subjected to RP-HPLC analysis with authentic antibiotic standards. Nectar and honey samples showed 4-17, 11-29 ${\mu}g/kg$ of streptomycin, 2-29, 3-44 ${\mu}g/kg$ of ampicillin and 17-34, 26-48 ${\mu}g/kg$ of kanamycin respectively.

  • One hundred and sixteen strains of E. coli isolated from drainage in Seoul city in 1987 and 104 strains of E. coli isolated from stools of domestic animals in suburb of Seoul in 1987 were examined for susceptibilities to 10 antimicrobial agents by the agar dilution method. The susceptibilities of the two groups to each antimicrobial agent were compared and correlations in the antimicrobial susceptibility of the 220 strains of E. coli among the 10 antimicrobial agents were also analyzed. The frequency of resistance strains was highest to tetracycline with 77%, and followed by chloramphenicol, streptomycin, ampicillin, kanamycin and cephalosporin in the descreasing order, tanging from 62% to 10%. Strains resistant to tobramycin, nalidixic acid, gentamicin and amikacin occupied 3 strains, 2 strains, 2 strains and 1 strain respectively.

  • About thirty bacterial strains of actinomycete isolated from the soil were examined for the presence of restriction endonuclease activity. Streptomyces diastatochromogenes, which was identified previously, was found to contain restriction endonuclease activity. The purification of this enzyme, SdiI, was carried out via streptomycin sulfate precipitation and ammonium sulfate fractionation followed by hydroxylapatite column chromatography. Sephacryl S-200 HR column chromatography and second hydroxylapatite column chromatography. SDS-polyacrylamide gel electrophoresis of the active protein (purified from various column chromatography) resulted in 35,000 Da protein.

  • Ganodema lucidum protoplasts were formed by the treatment of Novozym 234. The osmotic stabilizers such as mannitol were effective enough to produce protoplasts up to 10$^{6}$ $m\ell$. For regeneration, however, MgSO$_4$.7$H_2O$ was suitable. When inositol and sucrose were employed as osmotic stablizers, the regeneration ratio reached to 0.26%. Overlay of Streptomycin sulfate added agar was required to prevent bacterial contamination.

  • A new technique for single spore isolation was developed for predacious fungi forming constricting rings directly on the spores using Dactylaria brochopaga and Arthrobotrys dactyloides. Constricting rings were induced directly on the spores by transferring the spores in 25 ppm solution of DL-Valine in sterile distilled water. Freshly hatched and thoroughly washed second stage juveniles of Meloidogyne incognita were transferred into cavity blocks containing induced rings for trapping and killing of nematodes. The killed nematodes were surface sterilized with streptomycin and inoculated into petri dishes containing maize meal agar media with 100 ppm streptomycin. The petri dishes were incubated at $29{\pm}1^{\circ}C$ for few days which yielded axenic culture of these fungi.

Comparative Analysis of Korean and Japanese Strains of Pseudomonas syringae pv. actinidiae Causing Bacterial Canker of Kiwifruit

  • Genomic and phenotypic characteristics of the bacterial strains of Pseudomonas syringae pv. actinidiae and P. syringae pv. syringae collected from several kiwifruit orchards of Korea were investigated and compared with those from Japan to elucidate their phylogenic relationships. All the strains of P. syringae pv. actinidiae and pv. syringae tested were sensitive to copper sulfate but Korean and Japanese strains showed quite different responses to streptomycin. Korean strains were sensitive to streptomycin, but most of the Japanese strains of P. syringae pv. actinidiae were highly resistant to streptomycin. Japanese strains were also relatively more resistant to oxytetracycline than Korean strains. Plasmid profiles were not valuable to distinguish Korean strains of P. syringae pv. actinidiae frombJapanese strains. One or more indigenous plasmids with more than 15 kb in size were detected in all strains of P. syringae pv. actinidiae, but the number and sizes of plasmids harbored in P. syringae pv. actinidiae were variable among the strains regardless of their geographic origins. There also observed no significant relationship among resistance levels of the strains of P. syringae pv. actinidiae to antibiotics, their pathogenicity and plasmid profiles. RAPD profiles were useful to analyze the strains of P. syringae pv. actinidiae and pv. syringae. All the strains of P. syringae pv. actinidiae fell into a wide cluster separated from the strains of P. syringae pv. syringae, but Korean strains of P. syringae pv. actinidiae were separated from Japanese strains. The results support that Korean and Japanese strains of P. syringae pv. actinidiae may have different phylogenic origins.

  • The rational approach to antimicrobial therapy of infected root canals is based on accurate identification of the infecting organism and on the organism's susceptibility to antimicrobial agents as measured by standardized techniques. In establishing criteria for the selection of antibiotics, a susceptibility test should be performed. The purpose of this study was to investigate the susceptibility of 224 aerobic and anaerobic microbial strains isolated from infected root canals to various antibiotics. This was performed by using 7 antibiotic sensi-disc: Penicillin (10 units), Ampicillin (10 mcg), Tetracycline (30 mcg), Streptomycin (10 mcg), Kanamycin (30 mcg), Lincomycin (2 mcg), and Clindamycin (2 mcg). The results were as follows; 1. Strains isolated from infected root canals was shown to be most susceptible to Clindamycin, while Streptomycin exhibited least antibacterial properties. 2. Anaerobes were found to be susceptible to Penicillin, Ampicillin, and Clindamycin. 3. Streptomycin and Kanamycin were shown to be effective against Staphylococcus aureus and Staphylococcus epidermis, however, unidentified G(+) cocci organisms were found to be resistant to these agents. 4. Bifidobacterium sp. was susceptible to Lincomycin while G(+) rods were resistant to it. 5. Staphylococcus aureus, Micrococcus sp., and anaerobes were highly susceptible to Clindamycin. 6. All of the antibiotics tested were shown to be very effective against Eubacterium sp.

Influence of Commercial Antibiotics on Biocontrol of Soft Rot and Plant Growth Promotion in Chinese Cabbages by Bacillus vallismortis EXTN-1 and BS07M

  • We investigated influence of three commercial antibiotics viz., oxolinic acid, streptomycin, and validamycin A, on biocontrol and plant growth promoting activities of Bacillus vallismortis EXTN-1 and BS07M in Chinese cabbage. Plants were pre-drenched with these strains followed by antibiotics application at recommended and ten-fold diluted concentration to test the effect on biocontrol ability against soft rot caused by Pectobacterium carotovorum SCC1. The viability of the two biocontrol strains and bacterial pathogen SCC1 was significantly reduced by oxolinic acid and streptomycin in vitro assay, but not by validamycin A. In plant trials, strains EXTN-1 and BS07M controlled soft rot in Chinese cabbage, and there was a significant difference in disease severity when the antibiotics were applied to the plants drenched with the two biocontrol agents. Additional foliar applications of oxolinic acid and streptomycin reduced the disease irrespective of pre-drench treatment of the PGPRs. However, when the plants were pre-drenched with EXTN-1 followed by spray of validamycin A at recommended concentration, soft rot significantly reduced compared to untreated control. Similarly, strains EXTN-1 and BS07M significantly enhanced plant growth, but it did not show synergistic effect with additional spray of antibiotics. Populations of the EXTN-1 or BS07M in the rhizosphere of plants sprayed with antibiotics were significantly affected as compared to control. Taken together, our results suggest that the three antibiotics used for soft rot control in Chinese cabbage could affect bacterial mediated biocontrol and plant growth promoting activities. Therefore, combined treatment of the PGPRs and the commercial antibiotics should be carefully applied to sustain environmental friendly disease management.

Studies on variation of somatic cell in milk after administration of staphylococcus aureus vaccine and immunostimulant and antibiotics resistance of isolated staphylococcus spp in milk from dairy cow (유우에서 포도상구균 백신과 면역증강제 투여후 우유의 체세포수 변화 및 분리된 포도상구균의 항생제 내성에 관한 조사)

  • Thirty-one dairy cow from two farm(more than 500,000 cells/ml of bulk milk) in Kyongbuk northern province were selected because of their high somatic cell(more than 500,000 cells/ml of milk In individual cow). Each cow received. staphylococcus aureus vaccine(Labac Staph) and immunostimulant(Ultracon) by intramuscular injection to be repeated every fifteen days for S times. The present study was investigated variation of somatic cell after administration of Labac Staph and Ultracon, and antibiotics resistance of isolated staphylococcus spp from milk in selected cow. The results obtained through the survey were summarized as follows ; 1. Ten dairy cow was injected in A farm. Chronic mastitic two cow after 2rd injection was weeded out the herd. Decrease rate of somatic cell after 1st, 2nd, ,3rd, 4th and 5th administration were 41.4%, 35.6%, 56.4%, 65.4% and 36.7%, respectively. Twenty-one cow was injected in B farm. Chronic mastitic five cow after ,3rd injection was weeded out the herd. Decrease rate of somatic cell after 1st, 2nd, 3rd, 4th and 5th administration were 36.9%, 59.9%, 24.5%, 62.6% and 78.4%, respectively. 2. In A farm, isolated staphylococcus spp were identified as S hyicus 2 strains(11.8%), coagulase negative staphylococcus 15 stains(89.2%) and S epidermidis 6strain(35.3%). In B farm, isolated staphylococcus spp were identified as S aureus 19 strains(55.98%) and coagulase negative staphylococcus 15 strains (44.2%). 3. In A fm, antibiotics resistant rate of isolated staphylococcus spp was high at ampicillin, penicillin and kanamycin, and middle at neomycin, streptomycin and erythromycin. in B farm, antibiotics resistant rate was moderate at ampicillin, penicillin, gentamicin, ka-namycin, neomycin, streptomycin, erythromycin and tetracycline, and coagulase negative staphylococcus spp was moderate at streptomycin.

Somatic Hybrids by Electro-Protoplast Fusion between N. tabacum and N. glutinosa (담배(N. tabacum)와 N. glutinosa 종간 원형질체 융합식물의 생성)

  • Protoplasts, isolated from leaf of N. tabacum NR-/SR+ and N. glottnosa were electrofused and divided with a plating efficiency of 30∼35% in AAPI 9M medium. Green callus lines were selected in protoplast-derived colonies on MSNO3 selection medium with 1.2mg/ml streptomycin sulfate on the basis of nitrate reductase proficiency and streptomycin resistance. Four putative hybrid plant lines regenerated from the green callus lines had intermediate morphology between that of parents with respect to floral shape, corolla length and ovate leaf blade. Zymograms of leaf peroxidase and esterase from these putative hybrid plant lines showed isozyme profiles derived from both parents and also, they exhibited additional and lost bands. Cytological analysis of two putative hybrid plant lines gave chromosome counts of 2n=66 in L22 and 2n=54 in L44 which were less than the expected number of N. tabacum(2n=48) and N. glutinosa(2n=24).
