Genetic properties of R plasmids in Shigella isolates of swine origin in Korea (돈(豚)에서 분리(分離)한 Shigella균유래(菌由來) R plasmid의 유전적(遺傳的) 특성(特性)에 관한 연구(硏究))

  • This paper dealt with the distribution of Shigella spp. on 5 piggeries in Taegu and Kyungpook during the period from August to October 1987. Isolated Shigella were examined for serogrouping, antimicrobial drug resistance and detection of R plasmid. Genetic properties of R plasmid in Shigella have examined to fertility inhibition (Fi) and gel electrophoresis was performed for the isolation of plasmid DNA. The results obtained were summarised as followings; 1. Of total 2,978 samples from 5 piggeries, 82 strains (2.8%) of Shigella spp. were isolated from 82 samples. The isolated strains were identified as S dysenteriae (60 strains), S flexneri (20 strains) and S sonnei (2 strains). 2. Of the 82 strains examined 67 (95.1%) were resistant to one or more antibiotics, such as ampicillin (Am), chloramphenicol (Cm), kanamycin (Km), nalidixic acid (Na), rifampicin (Rf), streptomycin (Sm), sulfademethoxine (Su), and tetracycline (Tc) and higher resistant to Su (90.2%), Sm (63.4%) and Tc (63.4%). 3. Of the 78 resistant Shigella strains 26 (33.3%) harbored conjugative R plasmids and the transfer frequency of Sm (50.0%), Cm(33.3%) resistance was much higher than that of the other drug resistance. 4. The most common resistant patterns were SmSuTc, Su and AmSmSuTc. 5. Out of the 26 Shigella R plasm ids examined for Fi, 14(53.8%) were $Fi^+$ and the remainder were $Fi^-$. 6. The plasmid DNA profiles in Shigella spp. (9 strains) isolated from pigs were confirmed as being 2 to 9 fragments by the gel electrophoresis. Their molecular size ranged 2.17 to 87.62 kilobase (Kb). All strains of Shigella spp. consisted in 15.4 Kb plasmids.

Antimicrobial susceptibility and genetic characteristics of Streptococcus uberis isolated from bovine mastitis milk (젖소 유방염 유즙에서 분리한 Streptococcus uberis의 항생제 감수성 및 유전학적 특성)

  • Streptococcus spp. comprising Streptococcus (S.) uberis S. dysgalactiae strains is major causeof bovine mastitis from particularly well-managed or low somatic cell count herds that have successfullycontrolled contagious pathogens. In this study, antimicrobial susceptibility and genetic characteristics of S.uberis isolated from clinical or subclinical mastitis milk at 2003 were investigated. Eighty seven isolatesof Streptococus spp. were identified by the conventional biochemical methods. The antimicrobialsusceptibility by disk diffusion method was determined for 46 S. uberis, 11 S. bovis, 10 S. oralis, 6 S.uberis and 14 other Streptococcus spp.. Overall, the tested strains were susceptible to tetracycline (11.5%),amikacin (14.9%), streptomycin (16.1%), neomycin (26.4%), kanamycin (35.6%), gentamicin (65.2%),oxacillin (70.1%), ampicillin (75.9%), chloramphenicol (78.2%), and cephalothin (97.7%). Additionally, S.uberis strains were susceptible to pencillin G (97.8%), but resistant to erythromycin (76.0%) by minimalinhibitory concentration test. The multiple-drug resistance rate of isolated bacteria to 4 more thanamplification fingerprinting patterns amplifed with primer 8.6d showed that 3 to 8 number of distinguishableDNA fragments ranged from 180 bp to 1,20 bp. Thirty seven isolates of S. uberis strains were subtypedinto 8 distinct patterns. Each subtype revealed a typical pattern of antimicrobial susceptibilities. Thesefindings demonstrate that S. uberis isolates were mastitis pathogens of diverse serotypes, and oftenencountered the diverse resistant patterns.

Antimicrobial resistance of Escherichia coli isolated from healthy animals during 2010-2012 (건강한 가축에서 분리한 Escherichia coli의 항생제 내성 조사(2010~2012년))

  • The purpose of present study was to investigate the antimicrobial resistance in Escherichia coli isolated from healthy animals in all provinces of the Republic of Korea. A total of 2,085 E. coli strains isolated from 11,336 fecal samples of healthy animals during 2010-2012 were examined for antimicrobial resistance. Comparison of average resistance rate through the years revealed that tetracycline (47.0% and 76.1%) and streptomycin resistance (42.6% and 64.6%) was most frequently observed in cattle and pigs, respectively. Whereas, in chicken isolates, resistance against nalidixic acid (90.9%) was highest among the antimicrobials tested. Percentage of E. coli that showed multidrug resistance (resistance against ${\geq}$ three subclasses of antimicrobial agents) was 17.6% (151/860) in cattle, 69.4% (506/729) in pigs, and 86.1% (427/496) in chickens. Overall, the rates of resistance are apparently different between animal species and, in particular, resistance was less prevalent in cattle than in pigs and chickens. In conclusion, this study showed higher prevalence of resistance in commensal E. coli strains to antimicrobial agents in Korean livestock and highlighted the urgent need for measures to regulate the abuse of antimicrobial agents.

Studies on the Isolation of Vibrio vulnificus from Sea Water and Marine Products in the Southern Sea of Korea (한국 남해안 일대의 해수 및 해산물에서 Vibrio vulnificus의 분리연구)

  • Authors studies on the isolation of V. vulnificus from sea water, sea mud fishes, shellfishes and algae at the seasides of Pusan, Masan, Chungmu and Ulsan in Korea in 1985. Authors carried out test for isolated strains to bacteriological test, hemolysis test about erythrocytes of various animal, sensitivity test of various chemotherapeutic agents and serological test with antiserum of V. vulnificus. The resultls obtained were as follows: 1. V. vulnificus was isolated 15 strains from 399 total specimens: 110 cases of sea water, 40 cases of sea mud, 90 cases of fishes, 60 cases of shellfishes and 79 cases of various algae, respectively. 2. Nine strains were isolalted from sea water, 4 strains were isolated from sea mud and 2 strains were isolated from fishes, respectively. 3. Two strains among 15 strains isolated were lactose positive reaction. 4. All strains isolated were grown in concentration of $0.5%{\sim}7.0%$ NaCl, but were not grown 0% and 8.0% NaCl. 5. Hemolysis reaction about various erythrocytes was sensitived to guinea pig, human and rabbit erythrocytes, but was not sensitived to sheep erythrocytes. 6. Sensitivity test using with chemotherapeutic agents of "BioLab" Microbial Sensitivity Test Discs were generally sensitived to amikacin, ampicillin, clindamycin, erythromycin, gentamycin, kanamycin, streptomycin, tetracyclin and tobramycin, and were moderate to penicillin, but were resistant to methicillin and lincomycin, respectively. 7. The distribution of serotypes of V. vulnificus isolated were on antiserum of $0.1{\sim}07$ of V. vulnificus: 1 case of 01 and 2 cases of 07, respectively.

The R-Plasmid Transfer and Elimination of Shigella Cultures (Shigella균속의 항균제내성, 전달성 R-plasmid 및 제거에 관한 연구)

  • On hundred and forty stains of shigella cultures isolated from the twelve hygiene laboratories of cities and provincial general hospital laboratories in 1983 were tested for their resistance to thirteen antimicrobial drugs and their R-plasmid transfer. Antimicrobial drugs were used amikacin, ampicillin, cephalothin, chloramphenicol, gentamicin, kanamycin, nalidixic acid, rifampicin, streptamycin, tetracycline, tobramycin, cefoperazone and piperacillin. All strains were resistant to one or more of thirteen antimicrobial drugs but 94.3% were susceptible to amikacin, gentamicin and tobramycin of total isolated. The most strains commonly found resistance was to chloramphenicol (94%) followed by streptamycin (93%), tetracyline (92%) piperacillin (90%) ampicillin (83%), cefoperazone (42%), nalidixic acid (14%), cephalothin (17%), rifampicin (22%) and kanamycin (6%), sixty percent of strains among 140 were resistance to ampicillin, chloramphenicol, streptomycin, tetracycline at the same time. The transfer of drug resistance by conjugation was tested and ninety four strains (94.3%) were resistant to one or more drugs were found to transfer their drug resistance of E. coli. percentage of transfer frequency by conjugation was one strains (54%), the transfer frequency of drug resistance varied by donor strains and recipients, but not by selecting drugs. Resistance to nalidixic acid was not transferred by conjugation to recipients. Percentage of plasmid curing after the treatment of acriflavine, acridine orange was about 8%. Among strains cured two strains were tested compare original strains with them in biochemical properties in arginine dihydrolase and arabinose fermentation reaction. It was found to growth curves of No.2 shigella flexneri, serotype 1b, and its derivatives cured with acriflavine in $M{\ddot{u}}ller$ Hinton broth medium (pH 7.4, $38^{\circ}C$) by temperature Gradient Biophoto Recorder TN-1120 (Tokyo, Japan).

A report on the Salmonella cultures isolated in Korrea(1974) (1974년(年) 한국(韓國)에서 분리(分離)된 쌀모넬라 균(菌)에 관(關)한 보고(報告))

  • The authors identified 717 Salmonella cultures among 1504 suspectable cultures collected from all over the country in 1974. According to the results obtained from the physical and biochemical tests, and the antigenic formula analysis, seven cultures of S. paratyphi A, one culture of S. paratyphi B, Six cultures of S. typhimurium, one culture of S. thompson, four cultures of S. enteritidis and six hundred and ninty eight cultures of S. typhi were confirmed. The results of antibiotics-sensitivity test on Salmonella cultures revealed that chloramphenicol, gentamycin and ampicilin were considered to be the drugs of choice according to the results from the In Vitro test performed.

Physiological Characteristics and Anti-obesity Effect of Lactobacillus plantarum K10

  • This study aimed to investigate the physiological characteristics and anti-obesity effects of Lactobacillus plantarum K10. The ${\alpha}-amylase$ inhibitory activity, ${\alpha}-glucosidase$ inhibitory activity, and lipase inhibitory activity of L. plantarum K10 was $94.66{\pm}4.34%$, $99.78{\pm}0.12%$, and $87.40{\pm}1.41%$, respectively. Moreover, the strain inhibited the adipocyte differentiation of 3T3-L1 cells ($32.61{\pm}8.32%$) at a concentration of $100{\mu}g/mL$. In order to determine its potential for use as a probiotic, we investigated the physiological characteristics of L. plantarum K10. L. plantarum K10 was resistant to gentamycin, kanamycin, streptomycin, ampicillin, ciprofloxacin, tetracycline, vancomycin, and chloramphenicol. It also showed higher Leucine arylamidase, Valine arylamidase, and ${\beta}-galactosidase$ activities. Moreover, it was comparatively tolerant to bile juice and acid, exhibiting resistance to Escherichia coli, Salmonella Typhimurium, Listeria monocytogenes, and Staphylococcus aureus with rates of 90.71%, 11.86%, 14.19%, and 23.08%, respectively. The strain did not produce biogenic amines and showed higher adhesion to HT-29 cells compared to L. rhamnosus GG. As a result of the animal study, L. plantarum K10 showed significantly lower body weight compared to the high-fat diet group. The administration of L. plantarum K10 resulted in a reduction of subcutaneous fat mass and mesenteric fat mass compared to the high-fat diet (HFD) group. L. plantarum K10 also showed improvement in gut permeability compared to the HFD positive control group. These results demonstrate that L. plantarum K10 has potential as a probiotic with anti-obesity effects.

Isolation and antimicrobial susceptibility test of Mannheimia haemolytica from slaughtered cattle (도축우에서 Mannheimia haemolytica의 분리 및 항생제 감수성검사)

  • This study was carried out to identify and investigate antimicrobial susceptibility for Mannheimuia haemolytical which is responsible for shipping fever. Samples were collected from nasal and lung of 100 adult healthy cattle which are slaughtered in Samsung meat corporation located in Incheon metropolitan city. lung lesion index have been investigated within 0-5 range according to Shewen and Willkie(Can J Vet Res 52:30-36, 1988). Eighty-seven of 100 cattle were under normal condition with 0-1 ranges. A total of 129 strains were collected from blood and tryptic soy agar. Among these strains, 100 strains were identified with Staphylococcus, Streptococcus and enterobacteria containing E coli. Biochemical and fermentation assay of arabinose, trehalose, xylose, mannose, mannitol, lactose and salicin were tested to identify with Mannheimia sp. for 7 strains shown haemolytic activity on blood agar. Five strains were identified with Mannheimia haemolytica and 2 strains were untyped. In seasonal survey, Mannheimia sp recovered from fall to winter(5 of 7) have been highly isolated rather than those from spring to summer(2 of 7). Mannheimiz haemolytica were susceptible to antibacterials tested in this study but more resistant to oxytetracycline and streptomycin.

Studies on the brucellosis in Kyongbuk area (경북지역의 부루셀라병에 관한 연구)

  • The present study was carried out to investigate the prevalence of brucellosis in Kyungbuk area for the 3 years from 1966 to 1998. Collective milk samples were routinely screened to detect positive farms by using the milk ring test(MRT), and serum agglutination test was performed to detect sero-positive individuals in the MRT positive farms. Attempt were made to isolate the causative organismas from slaughtered sero-positive reactors and some biochemical and polymerase chain reation characters of the isolates were also made to identify the organisms. Seroprevalence to brucellosis in peoples who are close contact with infected dairy herds was also investigated. Brucellosis of dairy cattle was rare before 1997, but has been broken more frequently since early 1998. By the MRT for dairy herds, positive rate was gradually increased every year : 0.6% in 1996, 1.5% in 1997, 3.9% in 1998. Among 262 MRT-positive herds, only 21 herds(8.0%) showed positive brucellosis in serological test. The isolation rates of Brucella sp from tested materials were 51.2% in supramammary glands, 39.5% in milks, and 50.0% in pulmonary Iymphnode, respectively. Isolated strain and biotype were Brucella(B) arbortus biotype 1 in 26 heads, and were B suis biotype 1 in 2 heads. Isolated strain and vaccine strain were very similar in their colony morphology and staining. In drug susceptibility, isolated stains(B abortus) and vaccine strain(B abortus RB-51) were sensitive to ampicillin, gentamycin, kanamycin, neomycin, penicillin, streptomycin, and to tetracycline, but resistant to erythromycin. In the PCR, field strains reacted to BA and IS711 primers, and vaccine strain reacted to BA, IS711, and RB5l primers. In the plate agglutination test of 96 sera of human contacted with animals, serum antibody titer detected 1 : 100 in one person, 1 : 200 in one, and below 1 : 25 in the others.

Prevalence and antimicrobial susceptibility of Salmonella isolated from Korean slaughter pigs

  • During the period of December 2000 to November 2001, a total of 419(10.9%) Salmonella was isolated from 3,850 slaughtered pig samples(2,732 lymph nodes and 1,118 caecal contents) in Korea. Three hundred and seventy(13.5%) and 49(4.4%) Salmonella were isolated from lymph nodes and caecal contents, respectively. In the isolation ratio of Salmonella spp originated from each class of season, summer(15.7%), autumn(11.5%), spring(10.0%) and winter(8.3%), in order. As the result of serotyping, B group(67.5%) were the most common, in order of C$_1$(14.6%), D$_1$(6.0%), C$_2$(4.1%) and E$_1$(3.3%). 32 serovars were found, the major serotypes were as follows; S typhimurium(21.7%), S schwarzengrund(16.0%), S derby(15.8%), S typhimurium variant copenhagen(9.8%), S enteritidis (6.0%) and S mbandaka(6.0%). All of the isolates were susceptible to norfloxacin and ofloxacin. The isolates were resistant in order of doxycycline(69.2%), tetracycline(68.7%), penicliin(54.9%) and streptomycin(52.5%).
