타액의 수소이온 농도, 점도, 세균 및 면역글로블린 A에 관한 연구 (A Study on pH, Viscosity, Microorganisms and Immunoglobulin A of the Saliva)

  • The purpose of this study is to investigate the age-and sex-related changes in the pH of resting saliva, viscosity, microorganisms and immunoglobulin A of stimulated whole saliva, and to investigate their correlations. The 120 healthy subjects were included in this study and the author used cone-and plate digital viscometer for viscosity, MSB agar for Streptococcus mutans, SL Rogosa agar for lactobacilli, and single radial immunodiffusion technique for immunoglobulinA. The obtained results were as follows : 1. There was no significant difference in pH, viscosity, Streptococcus mutans lactobacilli and immunoglobulin A of the saliva between males and females. 2. The viscosity values of stimulated whole saliva showed the increasing pattern with aging. 3. DMFS (or dmfs) rate was not correlated with pH, viscosity, Streptococcus mutans, lactobacilli and immunoglobulin A of the saliva. 4. There was a significant difference in the concentration of immunoglobulin A between the group under 10 and groups above 10. 5. The viscosity values of stimulated whole saliva showed the increasing pattern with decreasing of the number of Streptococcus mutans.

  • Glycyrrhizin (18$\beta$-glycyrrhetic acid $\beta$-D-glucuronyl a-D-glucuronic acid, GL), a main component of liquore extract (Glycyrrhiza glabra), is ingested orally as a component in the oriental medicine. By human intestinal bacteria, glycyrrhizin (18$\beta$-glycyrrhetinic acid $\beta$-D-glucuronyl a-D-glucuronic acid, GL) was metabolized to glycyrrhetinic acid (GA): main pathway metabolizing GL to GA by glucuronidases of Bacteroides J-37 (Kim et al., 1997) and Eubacterium sp strain GLH (Akao et al., 1987) and minor pathway metabolizing GL to GA via 18$\beta$-glycyrrhetic acid D-glucuronic acid (GAMG) by $\beta$-glucuronidase of Streptococcus LJ-22 and glucuronidases of Bacteroides J-37 / E. coli. $\beta$-Glucuronidase from Streptococcus LJ-22 hydrolyzed GL to GAMG, not GA. $\beta$-Glucuronidase of Streptococcus LJ-22 hydrolyzed $\beta$-glucuronic acid conjugates of polysaccharides rather than aglycone-$\beta$-glucuronides Optimal pH of Streptococcus LJ-22 $\beta$-glucuronidase was 5-6 and its molecular weight was 250 kDaltons. Km for GL was 0.37mM.

Streptomyces sp. M-20 균의 대사물에 의한 Streptococcus mutans의 Glucosyltransferase 활성 억제 효과 (Inhibitory Effect of Metabolites isolated from Streptococcus mutas sp. M-20 on Glucosyltransferase Activity from Streptococcus mutans)

  • Dental caries is one of the most common oral diseases in the world. Glucosyltransferase (Gtase) of Streptococcus mutans (S. mutans) plays an important role in the develo pment of dental caries. For the purpose to develop anti- caries, we examined the effect of metabolites isolated from Streptomyces sp. M-20 on Gtase and the growth of S. mutans. Streptomyces sp. M-20 isolated from Mongolian soil showed 95~96% sequence homology with that of Streptomyces lin- colnensis. The metabolites of Streptomyces sp. M-20 were partially purified by extraction with ethyl acetate, silica gel column chromatography and preparative TLC. Partially purified metabolite, red colored component (MR-20) in ethyl acetate fraction showed potent antibacterial activitiy against S. mutans and inhibitory activity against Gtase purified from S. mutans, while another isolated yellow component (MY-20) showed no activity against S. mutans. The inhibitory activity of MR-20 against Gtase was confirmed by activity staining on SDS-polyacrylamide gel electrophoresis. The concentration of MR-20 for 50% inhibition $(IC_{50})$ against Gtase activity was $60{\mu}g/ml$. These results suggest that MR-20 can be developed for antibacterial agent and anticaries.

  • Streptococcus gordonii는 그람 양성이면서, 통성 혐기성, 및 비운동성 구균이다. S. gordonii는 사람의 구강 내 정상세균 총의 하나이고, 치면 생체막 형성의 선구적 세균 종이다. S. gordonii는 감염성 심내막염과 패혈성관절염 뿐만 아니라 유치의 치수염에 연관이 있다. S. gordonii KCOM 1506 (= ChDC B679) 균주가 사람 급성치수염 병소에서 분리되었으며 그 유전체 염기서열을 해독하여 보고한다.

  • Streptococcus constellatus는 그람 염색에 양성을 띄며, 조건 혐기성이면서 아포를 생성하지 않는 구균이다. S. constellatus는 사람 구강의 정상 세균 총에 속한다. 수술후 상악낭종 병소에서 S. constellatus KCOM 1039 균주가 분리되었다. 여기에서 S. constellatus KCOM 1039 균주의 유전체 염기서열을 결정하여 보고한다.

  • Firmicutes에 속하는 Streptococcus sp. strain NM을 두경부암 환자로부터 분리하였다. 본 연구에서는 약1.90 Mb의 크기와 39.3%의 평균 G+C 함량을 가진 NM 균주의 비완전한 유전체를 보고한다. 유전체는 1,845개의 코딩서열, 12개의 리보솜 RNA 및 58개의 전사 RNA유전자를 포함하였다. 본 유전체로 부터, 항생제내성, 용혈 및 방어시스템과 관련된 유전자들이 확인되었다.

  • The methanol extract of Cinnamoni Cortex showed antibacterial action against cariogenic bacterium, Streptococcus mutans OMZ 176. The active principle of the extract was identified to be trans-cinnamaldehyde, which was bactericidal in the minimal inhibitory concentration (MIC) of $100\;\mu$g/ml against the strain. From the results of antibacterial activity of cinnamaldehyde and its derivatives, the acrolein group in the cinnamaldehyde was elucidated to be an essential element for the activity.

  • A combination of magnolol or honokiol with lysozyme isolated from the egg white of the Korean Ogol fowl (Korean natural monument No.265) exhibited synergistic effect of bactericidal activity against a typical cariogenic bacterium, Streptococcus mutans OMZ 176. The synergistic ratio increased with time dependence.

  • In this case, Grant's Zebra which had been bred and exhibited in Farmland Zoological Garden for 4 years, died after treated dystocia. Result in autopsy, she had large hepatic abscess, and blood chemistry were increased G07, GPT and total billirubin. The microbiological culture with the hepatic abscess idetified the Streptococcus equi. It is diagnosed of hepatic abscess which was caused by Streptococcus equi infection.

  • Here, we have cloned and sequenced Streptococcus parauberis pgm gene, encoding the enzyme phosphoglucomutase (PGM), which is known to be in association with virulence in other streptococcal species. The PGM of S. parauberis is the most closely related to that of S. iniae based on their amino acid sequences.
