Effective Screening of Antagonist for the Biological Control of Soilborne Infectious Disease (Damping-Off)

  • An efficient method of selecting an antagonistic strain for use as a biological control agent strain was developed. In this improved method, the surface tension reduction potential of an isolate was included in the 'decision factor,' in addition to two other factors; the growth rate and pathogen inhibition. By using a statistically designed method, an isolate from the soil was selected and identified as Bacillus sp. GB 16. In the pot test, this strain showed the best performance among the isolated strains. The lowest disease incidence rate and fastest seed growth were observed when the Bacillus sp. GB 16 was used. The action of the surface tension reducing component was assumed to enhance the wetting, spreading, and residing of the antagonistic strain in the rhizosphere. This result showed that the improved selection method was quite effective in selecting the best antagonistic strain for the biological control of soilborne infectious plant pathogens.

Selection of Superior Strains of Cordyceps militaris with Enhanced Fruiting Body Productivity

  • In vitro fruiting bodies were produced from ten different isolates of Condyceps militaris EFCC C-5736, EFCC C-5941, EFCC C-5976, EFCC C-6040, EFCC C-6849, EFCC C-7268, EFCC C-7342, EFCC C-7992, EFCC C-8027 and EFCC C-8549. Single ascospores were isolated from in vitro grown fruiting bodies and used for fruiting body production in brown rice medium by both intra-strain crossing and out-crossing. Length and dry wt. of stromata grown in vitro were measured. Strains producing highest dry wt. of stromata were selected. Both intra-strain crossings and inter-strain crossings of single ascospore strains were found to produce profuse fruiting bodies of C. militaris.

  • In order to develop probiotic strain for pigs, Lactobacillus spp. (527 isolates), Streptococcus spp. (95 isolates) and Bifidobacterium spp. (25 isolates) were isolated from the feces of 35 pigs. These isolates were tested through in vitro experiment such as acid tolerance at pH 2.0 (Lactobacillus spp. and Streptococcus spp.) or pH 3.0 (Bifidobacterium spp.), bile tolerance in MRS broth containing 0.3% (w/v) Oxgall, heat resistance at $70^{\circ}C$ and $80^{\circ}C$ for 5 min, antibiotic resistance, antimicrobial activity against pathogenic bacteria and Caco-2 cell adherence assay. Finally ten most superior strain (5 Lactobacillus spp. strain, 3 Bifidobacterium spp. strain and 2 Streptococcus spp. strain) were selected as potential candidate for probiotic use in pig industry. It could be used as an alternative to antibiotics in feed additives.

  • These studles were conducted to investigate the development of chem~cal resistance and cross.resista. nce m dlarnondback moth (Piuteiia xylostella L). The resistance development of diamondback moth greatly vaned under single selection of five insectlades. The tnflumuron and lambda cyhalothnn strains at 8th selected generation showed 374- and 29.l.fold resistant levels. respectively, as compared with the susceptible strain. However, the Baciilus thuringiensis-seleded at 8th selected generatLon exhibited 240-fold resistant level. and the prothiophos-selected at 8th generation revealed 14.3-fold resistant !eve1 while the cartap hydrochloride-selected at 8th generailon showed 9.1-fold resistant level Prothiophos- selected strain showed low cross-resistance level to cartap hydrochloride, while this strain exhibited no cross-resistance of 1.3 to 2.8-fold to other msectlcides. Cartap hydrochlonde- seleded strain showed 19.9-fold. a high cross-resistance to lambda cyhalothrin, but this strain showed 2.2-34 fold, no cross resistance to other insecticide. Lambda cyhalothnn-selected strain exhibited cross-resistance to cartap hydrochlolide and prothiophas. Triflumuron-seleded strain showed 1.3-4.9 fold. no cross-resistance to other ~nsectic~dTe he B. ihuring~ensis-selededs train showed no cross-resistance to other insecticides.

  • Overproduction of L-ornithine by mutant strains isolated from Brevibacterium detoglutamicum BK1046 was investigated. The strain was a L-ornithine auxotroph and exhibited culture instability during fermentation. Through a sequential screening effort, a highly stable strain with lless revertant formation was finally selected and designated B. ketoglutamicum BK52 (KCTC0141BP). It prouduced L-ornithine at a high concentration (above 9 g/L) independent of subculture or cultivation time, and also had a very low tendency of revertant formation. In a long-term storage, this strain maintained its cell stability and productivity of L-ornithine to a reasonable range.

Production of Gentamicin by Micromonospora purpurea (Micromonospora purpurea에 의한 gentamicin생성)

  • Using Micromonospora strain, gentamicin was produced by fermentation. The increase in gentamicin productivity was studied by strain improvement and systematic optimization of fermentation process variables. The productivity of parent strain of M.purpurea (ATCC15835) was improved by selection of superior mutant after U.V. irradiation and induction of genetamicin resistance. Potato starch and soy bean meal were the best carbon and nitrogen sources for gentamicin fermentation, respectively. The optimum stimulating concentration of Co ion for gentamicin production was 0.006g $CoCl_2$ per liter of broth. Oxygen ws found to be an important factor for gentamicin yield. The optimum pH for the cell growth and gentamicin production were 7.2 and 6.8 respectively. By controlling the pH, oxygen, and other conditions found in this study at the optimal conditions for cell growth and gentamicin production, the total productivity of gentamicin was increased significantly.

  • To isolate a mutant strain which overproduces tryptophan, mutants of Candida rugosa were screened after EMS (ethyl methane sulfonate) mutagenesis. Fluorotryptophan, a tryptophan analogue, was used for selection of a tryptophan-overproducing mutant after mutagenesis. Among 50 mutants, several candidates were selected based on intracellular tryptophan content. Amino acid analysis results showed that C3 was the best strain because it had the highest amount of tryptophan among the mutants.

Selection of High Laccase-Producing Coriolopsis gallica Strain T906: Mutation Breeding, Strain Characterization, and Features of the Extracellular Laccases

  • Commercial application of laccase is often hampered by insufficient enzyme stocks, with very low yields obtained from natural sources. This study aimed to improve laccase production by mutation of a Coriolopsis gallica strain and to determine the biological properties of the mutant. The high-yield laccase strain C. gallica TCK was treated with N-methyl-N-nitro-N-nitrosoguanidine and ultraviolet light. Among the mutants isolated, T906 was found to be a high-production strain of laccases. The mutant strain T906 was stabilized via dozens of passages, and the selected ones were further processed for optimization of metallic ion, inducers, and nutritional requirements, which resulted in the optimized liquid fermentation medium MF9. The incubation temperature and pH were optimized to be 30℃ and 4.5, respectively. The mutant strain T906 showed 3-times higher laccase activity than the original strain TCK under optimized conditions, and the maximum laccase production (303 U/ml) was accomplished after 13 days. The extracellular laccase isoenzyme 1 was purified and characterized from the two strains, respectively, and their cDNA sequence was determined. Of note, the laccase isoenzyme 1 transcription levels were overtly increased in T906 mycelia compared with values obtained for strain TCK. These findings provide a basis for C. gallica modification for the production of high laccase amounts.

Selection for Duration of Fertility and Mule Duck White Plumage Colour in a Synthetic Strain of Ducks (Anas platyrhynchos)

  • A synthetic strain of ducks (Anas platyrhynchos) was developed by introducing genes for long duration of fertility to be used as mother of mule ducklings and a seven-generation selection experiment was conducted to increase the number of fertile eggs after a single artificial insemination (AI) with pooled Muscovy semen. Reciprocal crossbreeding between Brown Tsaiya LRI-2 (with long duration of fertility) and Pekin L-201 (with white plumage mule ducklings) ducks produced the G0. Then G1 were intercrossed to produce G2 and so on for the following generations. Each female duck was inseminated 3 times, at 26, 29, and 32 weeks of age. The eggs were collected for 14 days from day 2 after AI. Individual data regarding the number of incubated eggs (Ie), the number of fertile eggs at candling at day 7 of incubation (F), the total number of dead embryos (M), the maximum duration of fertility (Dm) and the number of hatched mule ducklings (H) with plumage colour were recorded. The selection criterion was the breeding values of the best linear unbiased prediction animal model for F. The results show high percentage of exhibited heterosis in G2 for traits to improve (19.1% for F and 12.9% for H); F with a value of 5.92 (vs 3.74 in the Pekin L-201) was improved in the G2. Heritabilities were found to be low for Ie ($h^2=0.07{\pm}0.03$) and M ($h^2=0.07{\pm}0.01$), moderately low for Dm ($h^2=0.13{\pm}0.02$), of medium values for H ($h^2=0.20{\pm}0.03$) and F ($h^2=0.23{\pm}0.03$). High and favourable genetic correlations existed between F and Dm ($r_g=0.93$), between F and H ($r_g=0.97$) and between Dm and H ($r_g=0.90$). The selection experiment showed a positive trend for phenotypic values of F (6.38 fertile eggs in G10 of synthetic strain vs 5.59 eggs in G4, and 3.74 eggs in Pekin L-201), with correlated response for increasing H (5.73 ducklings in G10 vs 4.86 in G4, and 3.09 ducklings in Pekin L-201) and maximum duration of the fertile period without increasing the embryo mortality rate. The average predicted genetic response for F was 40% of genetic standard deviation per generation of selection. The mule ducklings' feather colour also was improved. It was concluded that this study provided results for a better understanding of the genetics of the duration of fertility traits in the common female duck bred for mule and that the selection of a synthetic strain was effective method of improvement.

Resting Egg Production of Six Strains of Korean Rotifer, Brachionus plicatilis (S-type) (염분에 따른 한국산 Rotifer, Brachionus plicatilis (S-type) 6 strains의 내구란 생산)

  • Six strains (OKK, MOK, CHA, WON, 505 & PUA) of Korean rotifer, Brachionus plicatilis (S-type) were isolated from salt pond, and the resting egg production of these strains was investigated with the different salinities (10, 20, 30 put), Rotifer were cultured at $28^{\circ}C$ and 2,000 lux in 20 ml test tube with feeding Nannoohloris oculata. The maximum rotifer density was 2,050 inds./ml at 10 ppt for OKK strain, and MOK strain showed the highest specific growth rate (1.028) at 10 ppt. Mixis rate of CHA and WON strains increased with the lower salinity, while MOK strain increased the rate with salinity. PUA strain did not show the mixis rate even at the high rotifer density, and OKK strain showed the very low mixis rate ranging from 1.1 to $3.0\%$. Fertilization rate of CHA, MOK, SOS and WON strains was ranged from 10.4 to $68.8\%$, and OKK strain did not showed any fertilization rate. The highest production of resting egg in 20 ml test tube was 4,065 eggs at 10 ppt for WON strain. The results may suggest that the selection of rotifer strain and salinity are important factors for the mass production of resting egg.
