In order to improve wine quality, the selection of yeast strain and of additives in the manufacture of Bokbunja (Rubus coreanus Miq.) wine was investigated. The chemical composition of the edible portions of Bokbunja fruits was 86.5% moisture, 0.2% crude protein, 0.9% crude fat, 6.6% crude fiber, 0.5% ash and $10^{\circ}Brix$ sugar, and was 2.99% fructose, 2.53% glucose and 0.07% sucrose in fruit extract. The predominant organic acids in the fruit were citric acid (14.57 mg/mL) and malic acid (2.24 mg/mL) with smaller amounts of shikimic, pyroglutamic and oxalic acid. During fermentation, citric and malic acid levels decreased, while formic and acetic acid were released. Saccharomyces cerevisiae KCCM 12224 (Sc-24) was more favorable for alcoholic fermentation of Bokbunja and the addition of 200 ppm of potassium metabisulphite to must was more efficient than other $SO_2$ sources with a higher overall acceptability score. Sc-24 increased alcohol production from 9.8 to 14.8% in a sugar concentration dependent manner $(18-28^{\circ}Brix)$. The color value of early stage Bokbunja must was improved by supplementing with Japanese apricot extract, but this did not influence the color value of Bokbunja wine after primary fermentation. The astringent taste of Bokbunja wine was reduced by removing the seed from the fruit. Sugar solution (50%, w/v) was used instead of sugar power to prevent the possibility of undissolved sugar due to insufficient mixing. This substitution did not influence sensory evaluation.
We have previously established a system for plant regeneration through somatic embryogenesis and Agrobacterium-mediated transformation of Korean ginseng. In this study to produce a fungus-resistant plant, we introduced a bean chitinase gene into ginseng using the transformation system. A binary vector pChi/748 was constructed by introducing the bean basic chitinase gene into EcoRI site of pGA748 which carries the CaMV 35S promoter governing the introduced gene and neomycin phosphotransferase II(NPT-II)gene as a positive selection marker. Cotyledonary explants were cocultured with A. tumefaciens strain LBA4404 harboring the binary vertor pChi/748 for 48 h, and transferred to MS medium supplemented with l mg/L2,4-D,0.1mg/L kinetin, 100 mg/L kanamycin, and 500mg/L carbenicillin. Kanamycin-resistant calli were formed on the cut surface of cotyledonary explants after one month of culture, and subsequently they gave rise to somatic embryos. Upon transfer onto medium containing 1 mg/L each of BA and GA$_3$, most of them converted to plantlets after 5 weeks of culture. The genomic DNA of eight kanamycin-resistant regenerants was subjected to polymerase chain reaction (PCR) using two specific 21-mer oligonucleotides derived from the chitinase gene. This study aims to experimentally investigate ground settlement and ground movement around the square pipe by its penetration in sandy ground. A series of laboratory model tests were carried out with a small-scale auger equipment for penetration of a square pipe as well as a newly designed test box with a sand raining equipment. From the experiments, it is shown that a square pipe induces ground movement evenly around it in a low overburden condition. However, as the overburden becomes higher, ground movement by a square pipe is concentrated mainly above it. Especially, horizontal strain above the square pipe was mainly dominated by its penetration. In addition, sand surface movement is the smallest in case of the dimensionless penetration rate equal to 0.2. When its penetration rate of the square pipe is fixed, the rotation speed of auger controls surface movement whether it is settlement or heaving. Therefore, the selection of an optimal dimensionless rate for the square pipe is a key design factor to minimize ground settlement in a trenchless construction.
Pinus rigida ${\times}$ P. taeda seedlings in a nursery was inoculated with basidiospores of Pisolithus tinctorius (Pt) either collected from Suweon, Korea or introduced from U.S.A. to compare the effectiveness of the spores from two different origins as mycorrhizal inocula. Nursery beds were fumigated with methyl bromide and 1g of spores was used to inoculate $1m^2$ of soil surface just before seed sowing. Seedlings inoculated with American Pt (#250 strain from Georgia, U.S.A.) were 15% taller than Korean Pt at the end of the first growing season. The seedlings from fumigation treatment only (no inoculation involved) was slightly taller (statistically unsignificant) than those with Korean Pt, but slightly smaller than those with American Pt. In a subsequent year experiment, the seedlings inoculated with American and Korean Pt after soil fumigation were 66% and 60% taller, respectively, than seedlings infected by natural fungi without soil fumigation, suggesting the dual effects of Pt and fumigation on the seedling growth. Therefore potential of Pt spores for an effective inoculum exists and selection of Pt strains which have adapted to specific local environments is needed to develop better sources of mycorrhizal inocula.

Pest population density models are very important to monitor the initial occurrence and to understand the continuous fluctuation pattern of pest in pest management. This is one of the major issues in agriculture because these predictions make pesticides more effective and environmental impact of pesticides less. In this study, we combined and predicted the mortality change of pest caused by pesticides with temperature change and population dynamic model. Sensitive strain of two-spotted spider mite (Tetranychus urticae Koch) with kidney bean leaf as host was exposed to mixed acaricide, Acrinathrin-Spiromesifen and organotin acaricide, Azocyclotin, at 20, 25, 30, and $35^{\circ}C$, respectively. There was significant difference in mortality of T. urticae among pesticides and temperatures. We used DYMEX to simulate population density of T. urticae and predicted that the initial management time and number of chemical control would be changed in the future with climate change. There would be implications for strategies for pest management and selection process of pesticide in the future corresponding climate change.
In order to obtain a method of scouring of raw silk and degumming of cocoon by the appling of bacterial enzyme of sericinase, a strain of proper bacteria was isolated and some properties of the enzyme produced by the isolated bacteria were studied and the following results were obtained. 1) Optimum pH and temperature were indicating 7.5 and $50^{\circ}C$. and on these conditions, the silk fibroin will get no modification at all. 2) Sericinase activity was inhibited by calcium ion in the free incubation but same ions reacted as an activator in the reaction with substrate. So, degumming of Tussah cocoon which contains calcium oxalate in the cocoon layer will be possible by the treatment of this enzyme. 3) This bacterial sericinase never gives any affection to the fibroin layer of the silk. 4) The factors of smooth-surface condition, damage of fibroin layer, touching, luster and degumming effect of the silk resulting by the enzyme treatment were more appropriate than the results of other scouring methods, saponin, alkali and saponin-alkali using methods. We expect to get the same scouring result that compared with the papain or pancreatin were applied.

Stenotrophomonas maltophilia BW-13 is a potent biocontrol agent to control crisphead lettuce bottom rot caused by Rhizoctonia solani. To define the optimum conditions for the mass production of the S. maltophilia BW-13, we have investigated optimum culture conditions and effects of various carbon sources on the bacterial growth. The optimum initial pH and temperature were determined as pH $6.0\~7.0 and $35^{\circ}C$, respectively. For the selection of effective carbon source for the mass production, we tested the low molecular carbon sources such as sucrose, glucose, lactose, maltose, manose and the high molecular carbon source such as dough conditioner, rice bran, corn starch, sweet potato starch. As the results, the addition of dough conditioner in a basal medium ($1.25\%\;K_{2}HPO_4,\;0.38\%\;KH_{2}PO_4,\;0.01\%\;MgSO_4{\cdot}7H_{2}O,\;0.5\%\;Yeast extract$) was able to achieve higher cell density and the antifungal activity than others. Therefore, the basal medium containing $3\%$ dough conditioner (named as dough conditioner medium) was finally selected the optimized media for the mass production of BW-13 strain.
Microorganisms, having the lower decarboxylase activity, among the isolated strains from $cheonggukjang$ and rice-straw in this study were selected by using biogenic amine (BA) media. The selected strains were identified as $Bacillus$$subtilis$ HH12, $B.$$subtilis$ HR254, and $Paenibacillus$$barcinonensis$ KR97, by using 16S rRNA analysis. PCR analysis showed that the histidine decarboxylase ($hdc$) gene was absent in the HH12, HR254, and KR97 strains. However, PCR analysis showed that the tyrosine decarboxylase ($tdc$) gene was present in the HH12, HR254, and KR97 strains. Quantitative analysis of the selected strains by using high-performance liquid chromatography showed that histamine was absent in the HH12, HR254, and KR97 strains. However, these 3 strains showed tyramine concentrations of 6.09, 3.68, and 6.30 mg/L, respectively. These strains produced lower concentrations of amines (approximately 7.9, 0, and 9.3% amines in the HH12, HR254, and KR97 strains, respectively) than the $B.$$subtilis$ MC138 strain, which showed the higher protease activity.
In oder to select an antifungal substance-producing antagonistic bacterium against Fusarium oxysporum casuing fusarium wilt on tomato, strain BK4 was isolated from local soil of Gyeoungbuk and was identified as Bacillus thuringiensis by 16s rDNA analysis, biochemical test, and Mcirolog TM 3.0 System. The antibiotic of B. thuringiensis BK 4 was highly produced at $30^{\circ}C$ in nutrient broth (pH 9.0). The crude antibiotic was even stable at $121^{\circ}C$ and more stable at slight alkalic condition than acid condition. It was also remained $50{\%}$ activity at pH 3.0. B. thuringiensis BK4 showed the inhibition of spore germination and the biocontrol ability against F. oxysporum causing fusarium wilt of tomato in vivo test. According to these results, B. thuringiensis BK4 was enough to use with a microbial agent for biocontrol against fusarium wilt.

The thirteen commercial insecticides to Lycoriella mali were investigated on their insecticidal activities and mycelial growth of two Letinula edodes strains. For the adults, insecticides showing over 95% insecticidal activity were chlorpyrifos-methyl, fenthion, fenitrothion, benfuracarb, furathiocarb and deltamethrin. For tile larvae, diflubenzuron and cyromazine showed over 90% insecticidal activity. Fenthion, benfuracarb, furathiocarb, deltamethrin, diflubenzuron and cyromazine did not affect the mycelial growth of L. edodes strain, in Imhyup 1 variety. And deltamethrin, diflubenzuron and cyromazine did not affect that in Sanlim-5. Insecticides showing over 80% residual effect for 14 days were benfuracarb to the adults and diflubenzuron and cyromazine to the larvae. Control effect of furathiocarb, fenthion, benfuracarb, deltamethrin, diflubenzuron and cyromazine against larvae of Lycoriella mali showed 90% ten days after application under sawdust cultivation. Among them, benfuracarb, fenthion and furathiocarb revealed excellent control effect against adults. These results indicate that benfuracarb, fenthion, and furathiocarb can be used for the control of Lycoriella mali in the field. However, further studies are needed on the effect of insecticides treatment oil fruit-body yield and chemical residue in the mushroom tissues.
