The virulence of five Verticillium lecanii strains against greenhouse whiteflies, Trialeurodes vaporariorum, was tested at various temperatures as a major environmental factor. Strain CS-626 was found to be the most durable strain at a broad temperature range, and highly virulent against greenhouse whiteflies in a detached-leaf bioassay. In a tomato plant pot experiment, the $LT_{50}\;and\;LC_{50}$ of the CS-626 strain were 6.2 days and $2.3{\times}10^6$ conidia/ml, respectively. The optimal concentration of CS-626 for successful infection was $1{\times}10^8$conidia/ml. These results indicate that the CS-626 strain of Verticillium lecanii has a strong potential for effectively controlling greenhouse whiteflies.
Practical superconducting wires are designed with a composite structure to meet the desired engineering characteristics by expert selection of materials and design of the architecture. In practice, the local strain exerted on the superconducting component influences the electromagnetic properties. Here, recent progress in methods used to measure the local strain in practical superconducting wires and conductors using quantum beam techniques is introduced. Recent topics on the strain dependence of critical current are reviewed for three major practical wires: $Nb_3Sn$, BSCCO-2223 and REBCO tapes.
In order to elucidate the mechanism which regulates the production of cyclodextrin glucanotransferase (CGTase) and to achieve overproduction of CGTase by releasing catabolite (glucose) repression, several catabolite repression resistant mutants were selected from newly screened Bacillus firmus var. alkalophilus H609, after NTG (N-methyl-N -nitro-N-nitrosoguanidine) treatment, using 2-deoxyglucose as a nonmetabolizable analog of catabolite glucose and as a selection marker. Five catabolite repression resistant mutants were selected from about 30, 000 2-deoxyglucose resistant colonies. Relative catabolite repression indices of the selected mutants were in the range of 8~80% assuming 100% for parent strain. The amount of CGTase produced by the mutant strain CR41, which was 250 units/ml, was three times larger than that produced by its parent strain. The mutation seems to have occurred in the regulatory region of CGTase gene and not in the structural region or the glucose transporting system in cell membrane. The enzymatic properties of CGTase excreted from parent and mutant strains were also compared.
Currently, the genetic modification of Aspergillus oryzae is mainly dependent on protoplast-mediated transformation (PMT). In this study, we established a dual selection marker system in an industrial A. oryzae 3.042 strain by using Agrobacterium tumefaciens-mediated transformation (ATMT). We first constructed a uridine/uracil auxotrophic A. oryzae 3.042 strain and a pyrithiamine (PT)-resistance binary vector. Then, we established the ATMT system by using uridine/uracil auxotrophy and PT-resistance genes as selection markers. Finally, a dual selection marker ATMT system was developed. This study demonstrates a useful dual selection marker transformation system for genetic manipulations of A. oryzae 3.042.

An improved strain of the red seaweed Porphyra suborbiculata containing an increased amount of the essential amino acid L-lysine was obtained through mutation and analog enrichment. Mutagenesis using a 10% lethal dose of ultraviolet irradiation and an enrichment culture with the L-lysine analog aminoethyl-L-cysteine (AEC) was repeated to select the most productive strain using monospores of P. suborbiculata. The concentrations of AEC required to produce 50 and 100% inhibition of survival were 60 and 115 mM in the parent strain, and 72 and 135 mM in the selected AEC-resistant strain, respectively. The AEC-resistant strain, L130, produced 1.74-fold more lysine compared to its parent strain. Thus, mutagenesis with analog enrichment shows promise for selecting seaweed strains that can overproduce this essential amino acid.
A new bacterial strain, was isolated from activated sludge, identified and named P. aeruginosa EMS1. The new strain produced surface-active rhamnolipids by batch cultivation in mineral salts medium with waste flying oils. The mutant strain KH7, designated P. aeruginosa EMS1, derived by random mutagenesis with N-methyl-N-nitro-N-nitrosogoanidine treatment producing high levels of the biosurfactants was selected by an ion-pair plate assay. The mutant strain KH7 showed 4-5 times more hydrocarbon emulsification as compared to the parent when grown on waste frying oils and various hydrocarbons. Furthermore, P. aeruginosa EMS1 and mutant strain KH7 was also able to use whey as a co-substrate for growth and biosurfactant production. As results of this study, mutant strain KH7 is a very efficient biosurfactant producer, and its culture conditions are relatively inexpensive and economical. Rhamnolipid is synthesized by the rhlAB-encoded rhamnosyltransferase. To be convinced of these genes, we performed PCR based on P. aeruginosa PAO1 whole-genome database. rhl gene cluster nucleotide and amino acid sequences were compared for both parent and mutant. Comparison of nucleotide sequence of rhlAB, there were usually terminal's codons exchange.

Concrete Filled Fibre Reinforced Polymer Tube (CFFT) for new columns construction has attracted significant research attention in recent years. The CFFT acts as a formwork for new columns and a barrier to corrosion accelerating agents. It significantly increases both the strength capacity (Strength enhancement ratio) and the ductility (Strain enhancement ratio) of reinforced concrete columns. In this study, based on predefined selection criteria, experimental investigation results of 134 circular CFFT columns under axial compression have been compiled and analysed from 599 CFFT specimens available in the literature. It has been observed that actual confinement ratio (expressed as a function of material properties of fibres, diameter of CFFT and compressive strength of concrete) has significant influence on the strength and ductility of circular CFFT columns. Design oriented models have been proposed to compute the strength and strain enhancement ratios of circular CFFT columns. The proposed strength and strain enhancement ratio models have significantly reduced Average Absolute Error (AAE), Mean Square Error (MSE), Relative Standard Error of Estimate (RSEE) and Standard Deviation (SD) as compared to other available strength and strain enhancement ratios of circular CFFT column models. The predictions of the proposed strength and strain enhancement ratio models match well with the experimental strength and strain enhancement ratios investigation results in the compiled database.

Recently, dense sensor instrumentation for structural health monitoring has motivated the need for novel passive wireless sensors that do not require a portable power source, such as batteries. Using a layer-by-layer self-assembly process, nano-structured multifunctional carbon nanotube-based thin film sensors of controlled morphology are fabricated. Through judicious selection of polyelectrolytic constituents, specific sensing transduction mechanisms can be encoded within these homogenous thin films. In this study, the thin films are specifically designed to change electrical properties to strain and pH stimulus. Validation of wireless communications is performed using traditional magnetic coil antennas of various turns for passive RFID (radio frequency identification) applications. Preliminary experimental results shown in this study have identified characteristic frequency and bandwidth changes in tandem with varying strain and pH, respectively. Finally, ongoing research is presented on the use of gold nanocolloids and carbon nanotubes during layer-by-layer assembly to fabricate highly conductive coil antennas for wireless communications.
An improved method for the selective isolation of high-yielding mutant strains for the production of antifungal antibiotic KRF-001 was investigated. The mutant strain U. V 4, which produces high titer of KRF-001, was selected on the high potency agar plate after ultraviolet light irradiation. The U. V 4 strain produced 2-fold more KRF-001 than the mother strain in production media. Large scale fermentation was performed using the U. V 4 strain in 100$\ell$ fermenter. The antifungal antibiotic KRF-001 secreted into culture broth was detected by HPLC in 24hrs of fermentation.
International conferences to block the spread of Avian bird flu occurred in Beijing, 2006 and others warned of the seriousness of the H5N1 strain. The meetings succeeded in generating billions of dollars from USA, EU and World Bank. Migratory birds seem to play a major role in the spread of the aggressive strain globally from Asia to Europe and Africa. Experiences of tolerance breeding of maize (Zea mays L.) for four decades against 20 biotic stresses suggest that the prime cause of the occurrence of H5N1 strain was due to the human beings' counter-efforts against nature. Excessive use of chemicals (spray and injection) in the commercial poultry farms had created high selection pressure on virus. The new strain had mutated for survival. Attempting to eliminate the virus by chemicals for 100% control is a dangerous way to control biotic stresses. This can create more aggressive strains. A solution would be to build up tolerability of the commercial animals against the virus. Improvement of poultry cage environments and respect for nature must be integrated. Potential foes must be watched.
