A Study on Storage of Major Herbal Medicine Materials, Zingiberis Rhizoma

  • To develop optimal storage method of root of Zingiberis Rhizoma, which has been grown as major cultural herbal medicine materials, root of Zingiberis Rhizoma, was stored for 10 months with different packing materials and sealing methods. The loss in dry weight as influenced by packing materials and sealing methods was the lowest at vacuum packing and followed by complete sealing methods with transparent polyethylene film. The ratio of root rot during the storage period was not significantly different between packing materials but was significantly different between sealing conditions. Conclusionally, vacuum packing and complete sealing with polyethylene film appears to be optimal for storage of Zingiberis Rhizoma.

The loss in dry weight as influenced by packing materials and sealing methods was the lowest at vacuum packing and followed by complete sealing methods with transparent polyethylene film. The ratio of root rot during the storage period was not significantly different between packing materials but was significantly different between sealing conditions. Evaluation of physiological functionalities of Codonopsis lanceolata root extracts on the storage temperatures and durations

  • The effects of bioactivity, binding polyphenolic contents, DPPH radical scavenging activity, ABTS radical scavenging activity, xanthine oxidase and immune activity on the storage temperature and durations of Codonopsis lanceolata root were evaluated in vitro. The contents of total polyphenol and flavonoid content by storage temperature and storage period was no significant difference according to the storage period, but it was found that the content was higher at lower storage temperature. The DPPH free radical scavenging activity at six different concentrations, 500, 1000, 2500, 5000, 10000 and 20000 mg/L are measured, the scavenging activity according to different storage temperature and storage period showed relatively the higher the activity in the shorter storage period or the lower storage temperature. The ABTS radical scavenging activity did not show a significant difference under various storage temperature and storage period conditions. The xanthine oxidase inhibitory activity of C. lanceolata extract tended to decrease more, depending on the longer storage period or the higher storage temperature. In different storage period and storage temperature conditions, the immune cell growth of C. lanceolata extract promoted a concentration-dependent manner in both human T cell and B cell, and did not show a significant difference. These results of this study suggested that the root of C. lanceolata may assist in the potential biological activities, and can be used as a source of human health products.

Evaluation of the periodontal and pulpal healing of replanted rat molars with doxycycline root conditioning

  • Purpose: This study aimed to evaluate periodontal and pulpal healing in replanted rat teeth, preserved under different storage conditions, with or without root conditioning using doxycycline. Methods: A total of 40 maxillary first molars extracted from 20 Sprague-Dawley rats were stored for different durations under different conditions (5 minutes in dry storage and 60 minutes in Hank's balanced salt solution [HBSS]) and subsequently replanted. The rats were divided into 2 groups based on the use of root surface treatment: the doxycycline group (root surface treated with doxycycline) and the control group (no doxycycline treatment). Eight weeks after replantation, the animals were sacrificed, and the teeth were evaluated using micro-computed tomography (micro-CT) and histomorphometric analysis. Results: In the micro-CT analysis, the doxycycline group showed the same rate of occurrence of periapical radiolucency as was observed in the control group, but a lower degree of root resorption in teeth replanted after 60 minutes of storage in HBSS (P<0.05). In the histomorphometric analysis, the doxycycline group exhibited no improvement in either pulpal or periodontal healing of the replanted tooth after 5 minutes of dry storage, but showed a lower grade of surface root resorption ($1.37{\pm}0.77$) and inflammatory resorption in the teeth stored for 60 minutes in HBSS ($1.33{\pm}0.71$). Conclusions: In conclusion, doxycycline improved the periodontal healing of replanted teeth stored for 60 minutes in HBSS, whereas doxycycline did not improve periodontal healing of replanted tooth after 5 minutes of dry storage. Within the limits of this study, doxycycline showed more favorable periodontal healing despite delayed replantation.

Growth characteristics and variation in component of sweet potato (Ipomoea batatas) cultivars according to cultivation period

  • Cultivated varieties of sweet potato were from dry texture type to tender texture type on the basis of consumer preferences. There are many differences in the quantity of sweet potato, starch content, pigment, and sugar content depending on the cultivation season and area, even in the same variety. Therefore, in this study, we attempted to establish optimum time of harvesting through growth characteristics and variation in component like starch, sugar, polyphenol and flavonoid. Four sweet potato varieties were used in this experiment. Among them, Jinhongmi (JHM) & Yulmi (YM) were as dry texture type and Pungwonmi (PWM) & Hogammi (HGM) were as tender texture type. Sweet potatoes were transplanted on 23 May, 2016 and were investigated storage root weight and component contents every 20 days from 60 days to 120 days and surveyed yield at 110, 120, 130 days after transplantation. Result revealed that storage root weight of YM, JHM, and HGM were 30.1, 38.9, 20.8 g respectively in 60 days after transplanting. Storage roots of PWM gerw faster with the weight of 88.2 g. In 120 days after transplanting, storage root weight varied from 88.3 to 118.7 g, HGM was the smallest, and PWM was the largest. Sugar contents of sweet potato ranged from 21.0 to $23.8Brix^{\circ}$ in 60 days after transplanting and from 27.5 to $30.78Brix^{\circ}$ in 120 days after transplanting. In particular, the sugar content of HGM was the highest over $30Brix^{\circ}$ after 80 days. The starch content of dry texture type (YM, JHM) increased from 15.5% to 20.4% and tender texture type (PWM, HGM) increased from 11.0% to 17.3%. Starch content tended to be high in dry type sweet potatoes. The content of polyphenol and flavonoid were highest in 60 days after transplanting and was reduced according to cultivation period. The total yield of PWM was high as 3,154 kg/10a and large storage root of over 250 g accounted for 47.4% in 110 days after transplanting. Storage root (YM, JHM, HGM) of 81~150 g accounted for 34.9% ~ 43.2% in 120 days after transplanting. These are the most marketable. Because consumer in Korea prefers small, round and about 100g size sweet potato. The ratio of large storage root (over 250 g) were increased in all varieties at 130 days after transplanting. Therefore, it is considered appropriate to harvest PWM at 110 days and YM, JHM, HGM at 120 days after transplanting, which planted in late May.

Cytokinin signaling promotes root secondary growth and bud formation in Panax ginseng

  • Background: Panax ginseng, one of the valuable perennial medicinal plants, stores numerous pharmacological substrates in its storage roots. Given its perennial growth habit, organ regeneration occurs each year, and cambium stem cell activity is necessary for secondary growth and storage root formation. Cytokinin (CK) is a phytohormone involved in the maintenance of meristematic cells for the development of storage organs; however, its physiological role in storage-root secondary growth remains unknown. Methods: Exogenous CK was repeatedly applied to P. ginseng, and morphological and histological changes were observed. RNA-seq analysis was used to elucidate the transcriptional network of CK that regulates P. ginseng growth and development. The HISTIDINE KINASE 3 (PgHK3) and RESPONSE REGULATOR 2 (PgRR2) genes were cloned in P. ginseng and functionally analyzed in Arabidopsis as a two-component system involved in CK signaling. Results: Phenotypic and histological analyses showed that CK increased cambium activity and dormant axillary bud formation in P. ginseng, thus promoting storage-root secondary growth and bud formation. The evolutionarily conserved two-component signaling pathways in P. ginseng were sufficient to restore CK signaling in the Arabidopsis ahk2/3 double mutant and rescue its growth defects. Finally, RNA-seq analysis of CK-treated P. ginseng roots revealed that plant-type cell wall biogenesis-related genes are tightly connected with mitotic cell division, cytokinesis, and auxin signaling to regulate CK-mediated P. ginseng development. Conclusion: Overall, we identified the CK signaling-related two-component systems and their physiological role in P. ginseng. This scientific information has the potential to significantly improve the field-cultivation and biotechnology-based breeding of ginseng.

오미자 첨가 도라지 정과의 제조와 저장 중 품질 변화 (Development and Quality Characteristics of Bellflower Root Jeonggwa Added Omija (Schizandra chinensis Baillon) during Storage)

  • The purpose of this research is to develop bellflower (Platycodon grandiforum) root Jeonggwa as a health food. The Omija (Schizandra chinensis Baillon) water extract with 0-8% (w/w) Jeonggwa added was stored at $25^{\circ}C$ for 12 weeks. The quality characteristics of Jeonggwa during storage were investigated. The Jeonggwa showed a lower pH and a higher acdity as the amount of Omija water extract that was added increased. The moisture content of Jeonggwa were rose from 15-18% to 16-22% within 2 weeks of storage, and remained at that level up to the end of storage. The total viable cells of bellflower root Jeonggwa were 2.1~2.3 log CFU/g and increased in number during storage but never exceeded 4 log CFU/g. The shelf life of the Jeonggwa was extended when the Omija extract was added. The lightness (L), redness (a) and yellowness (b) of the Jeonggwa during its storage at $25^{\circ}C$ were highest in control samples, and the values decreased with the increase in the Omija extract concentration (p<0.001). The mechanical evaluation of the Jeonggwa showed that various tested parameters fell during the storage at $25^{\circ}C$. The hardness and strength of the Jeonggwa significantly decreased as the Omija extract concentration rose (p<0.05). In the ensory evaluation tests, the acceptability of the Jeonggwa was optimal when 4~6% of Omija extract was added to it.

방사선조사와 저온저장이 국화 삽수의 발근 및 생육에 미치는 영향 (Effects of Irradiation and Cold Storage on Rooting and Growth of Chrysanthemum Cuttings)

  • 본 연구는 국화의 삽수 번식을 촉진할 수 있는 방사선의 적정선량 범위를 확인하고 저온저장과 방사선의 복합처리에 의한 영향을 알아보기 위해 수행하였다 삽수를 전개엽 3매, 길이 6cm로 균일하게 잘라 실험에 사용하였다. 네 주간의 저온저장 전과 후로 나누어 0∼70 Gy감마선을 조사하여 삽아하고 15일째와 20일째 발근 및 생육을 비교하였다. 그 결과 방사선 단일 처리에서는 30Gy 조사군이 높은 발근율과 빠른 근생장을 나타냈으며, 지하부 생육도 삽수 20일째 20, 30Gy 조사군에서 양호하게 나타났다. 저온저장 후 방사선을 조사한 경우 30 Gy 조사군에서 삽수의 생존율이 80%로 가장 높았다. 반면 방사선 조사후 저온저장을 시행한 경우 50Gy 조사군에서 지하부의 생육이 가장 높았으며 저온저장 후 방사선 조사를 한 실험군에 비해 지하부 생육이 우수하였다.

가열 돈육 patty의 품질특성에 미치는 연근 및 연잎분말 첨가 효과 (The Effect of Adding Lotus Root and Leaf Powder on the Quality Characteristics of Cooked Pork Patties)

  • This study was carried out to investigate the effects of adding lotus root and leaf powder on the quality characteristics of cooked pork patties. The patties were of four types: unadulterated (control, T0), 0.5% lotus root powder (T1), 0.25% lotus root and 0.25% lotus leaf powder (T2), and 0.5% lotus leaf powder (T3). We found that the moisture content was highest in T1 group, and that the protein and fat contents were lowest in T1 (p<0.05). The ash content was lowest in T0 (p<0.05). The cooking yield and moisture retention level were not significantly different among the samples, but fat retention was the highest in T3 (p<0.05). The hardness, springiness, cohesiveness and chewiness were also highest in T3, and the gumminess was lowest in T0 (p<0.05). The $L^*$ values tended to increase with longer storage periods, and that of T0 was the highest after nine days of the storage (p<0.05). By contrast, the $a^*$ values decreased with longer storage periods (p<0.05), with that of T3 being the lowest (p<0.05) during storage periods. The water holding capacity decreased with longer storage periods, and that of T1 was the highest (p<0.05) during storage periods. The pH of T1 decreased, and those of T2 and T3 increased with a longer storage period (p<0.05). TBARS values increased with a longer storage period, and those of T0, T1, T2 and T3 were 8.69, 2.95, 0.88 and 0.55 mg/kg, respectively, after nine days of storage (p<0.05).
