The Effect of $Cd^{2+}$ on Stomatal Apertures of Epidermal Strips in Commelina communis L. ($Cd^{2+}$이 닭의장풀의 기공개폐에 미치는 영향)

  • The effect of $Cd^{2+}$ on stomatal apertures of epidermal strips and intact leaves in Commelina communis L. was investigated. Cadmium stimulated stomatal opening. The stomata, treated with 100 $\mu$M $Cd^{2+}$ opened to a degree of about 8.38 fm, but the stomata, treated with no cadmium opened to 3.74 ${\mu}{\textrm}{m}$. In order to show that there was no mechanical or osmotic impediment preventing the stomata in the epidermal strips, salicylic acid (SA) and abscisic acid (ABA) were used. The treatment of 100 $\mu$M SA and 1 $\mu$M ABA inhibited 14% and 28% of stomatal opening, respectively. Other heavy metals such as $Al^{3+} and Ag^{2+}$ were also used to investigate the effect of the stomatal apertures. The treatment of $Al^{3+} and Ag^{2+}$ also stimulated 19% and 41% of stomatal opening. To understand how cadmium open stomata, the effect of cadmium on the K+ influx into the epidermal strips was investigated. $Cd^{2+}$, SA, ABA inhibited 98%, 28%, 34% of K+ uptake respectively. 3-weeks old Commelina was transferred to and grown in Hoagland solution (0,5 mM $Cd^{2+}$, 10 mM $Cd^{2+}$) for 4 days and stomatal conductance were measured. The treatment of 5 mM $Cd^{2+}$ and 10 mM $Cd^{2+}$ showed about 51% and 70% inhibition of stomatal conductance, respectively Therefore, it could be concluded that stomata in epidermal strips and intact leaves behaved differently and cadmium- stimulated stomatal opening was due to the result of cadmium uptake into the epidermal strips instead of K+.   • This experiment was carried out to investigate the stomatal aperture of wheat variety 'Chokwang' grown in the two different nitrogen level. Stomatal aperture was measured at the maximal tillering, shooting, booting, flowering and grain filling stages. Stomatal aperture in leaf blades gradually increased in accordance with the successive leaf growth. The maximal opening of stomata was observed at flowering stage and around noon in a day, but stomata closed around six o'clock in the afternoon. Stomata opened wider in the high nitrogen application than in the low nitrogen and their effects were the highest at the booting and flowering stage. Diurnal course at stomatal aperture of upper leaves was wider than that of lower leaves regardless of growth stages. Positive correlation (r=0.66$^{**}$) appeared between nitrogen content in leaf blades and stomatal aperture. The leaves of low position and the developing leaves showed smaller stomatal aperture than the full expanded top leaves irrespective of leaf stages. Water content of leaf, root weight and root activity were increased by the nitrogen application and thus considered as factors increase the stomatal aperture.ication and thus considered as factors increase the stomatal aperture.

Effect of salicylic acid and its analogues on stomatal closing in Commelina communis L. (닭의장풀의 기공닫힘에 미치는 살리실릭산과 그 유사물의 효과)

  • ABA and SA showed different effect on stomatal closing on same condition. The addition of 1 M salicylic acid to fully opened stomata resulted in a significant reductionn of 22 % in stomatal aperture. However, 1 mM ABA reduced 73 % of stomatal aperture. The light absorption spectra of the salicylic acid solution showed that SA was degraded within 1 hour. Therefore, SA solution was resupplied % the detached epidermis every 30 min. during incubation and it was found that even at 10 $mu$M SA induced stomatal closing significantly. Its effect was also greatly pH dependent. The reduction of stomatal aperture caused by 1 mM SA was most effective at lower pH (pH 7.2, 5 %: pH 6.2, 40 %; pH 5.2, 78 %). Therefore, if SA was properly treated to the epidermal strips in the medium, the effects of SA on stomatal closing were similar with those of ABA.

  • Stomatal pore is an important physiological trait that is closely linked to photosynthesis and transpiration as carbon dioxide and water vapor move through it between the atmosphere and plants. The present study investigated stomatal traits, such as stomatal density, index and size, of herbaceous native and alien plant species living in a riparian park on the Nakdong River to understand how those traits vary and to know if successful settlement of alien plants is attributed to those traits. There was no difference in stomatal density, index and size between native and alien plants with kidney-shaped stomata, suggesting that an empty ecological niche is not an essential prerequisite for the successful settlement of alien plants. Stomatal density showed a negative correlation with leaf thickness and leaf dry weight content (LMDC), but there was no correlation with Specific leaf area (SLA). All plants with kidney-shaped stomata had amphistomatous leaves, and the density and size of dumbell-shaped stomata were lower than those of kidney-shaped stomata.

Phosphorylation, 14-3-3 protein and photoreceptor in blue light response of stomatal guard cells

  • Blue light (BL) induces stomatal opening through activation of H$^{+}$ pump, which creates electrical gradient across the plasma membrane for $K^{+}$ uptake into guard cells. The pump is the plasma membrane H$^{+}$ -ATPase and is activated via phosphorylation of the C-terminus with concomitant binding of the 14-3-3 protein. The opening is initiated by the perception of BL through phototropin (phot), which are recently identified as BL receptors in stomatal guard cells. In this study, we provide the biochemical evidence for phots as BL receptors in stomatal guard cells. vfphot was phosphorylated reversibly by BL, and phosphorylation levels of vfphot increased earlier than those of the plasma membrane W-ATPase. BL-dependent phosphorylations of vfphot and H$^{+}$-ATPase showed similar fluence dependency. Staurosporin, an inhibitor of serine/threonine protein kinase, and diphenyleneiodonium chloride (DPI), an inhibitor of flavoprotein, inhibited BL-dependent phosphorylations of vfphot and H$^{+}$ -ATPase. These results indicate that vfphot acts as a BL-receptor mediating stomatal opening.l opening.

  • This study was conducted to investigate the influence of soil moisture on the photosynthetic rate, transpiration rate, stomatal conductance, leaf water content and its any correlation in soybean. Followings were achieved as a conclusion. At the soil moisture was moved from drought condition to saturation condition, the photosynthetic rate, transpiration rate, stomatal conductance and leaf water content were gradually increased on the period of re-watering treatment. The recovery of photosynthetic rate was faster than others. There were positive correlation with the photosynthetic rate and leaf water content, the stomatal conductance and leaf water content, the photosynthetic rate and stomatal conductance respectively, but the correlation coefficient of photosynthetic rate and leaf water content was high.

  • Stomatal morphological characteristics (shape, density and length) were investigated for 8 ornamental woody species growing in the campus of Sangji College on Usandong, Wonju, Kangwondo. Stomata of Forsythia koreana, Syringa dialata, Zedkova serrata and Chaenomeles sinensis were classified as irregular-celled type (anomocytic), those of Vibrunum sargentii, Liriodendron tulipifera and Magnolia kobus as parallel-celled type (paracytic). For Forsythia koreana, stomatal length was not greatly changed as the leaf blade, growing larger. Number of stomata per unit leaf area was decreased, but number of stomata per single leaf was increased as the leaf blade, growing larger. For Taxus cuspidata, stomatal length was increased, but number of stomata per unit needle area was decreased as the needle, growing larger.

The Characteristics of Diurnal Changes in the Tissue-Water Relations of Pueraria thunbergiana (칡(Pueraria thunbergiana) 조직수분관계의 일변화 특성)

  • The diurnal changes of the stomatal conductance, transpiration and leaf water potential were measured in order to assess the water relations characteristics of Pueraria thunbergiana in August of 1995 and 1996. The results showed two different responses depending on the duration of rainless days. The microclimatic conditions were highly stressful on 2 August. Daily maximum temperature reached to $39.0{\circ}C$ and vapor pressure deficit was 3.55 KPa. During this time the leaf water potential decreased to -1.02 MPa and a marked reduction of stomatal conductance was shown. However, on 15 August the stomatal conductance increased with increment of photon flux density, and transpiration was highly maintained during the day time. Minimum leaf water potential was only -0.47 MPa in spite of high transpiration rate. Furthermore, on 15 August reduced leaf water potential during the day time was recovered rapidly with decrease of photon flux density, whereas recovery of leaf water potential on 2 August was delayed. However, reduced leaf water potential on 2 August was recovered untile the next dawn. Osmotic potential at turgor loss point of Pueraria thunbergiana on 2, 3 and 15 August was -1.79, -1.70 and -1.60 MPa, respectively. The vapor pressure deficit is more contributive to the regulation of stomatal conductance than leaf water potential.

Comparative Water Relations of Two Vitis vinifera Cultivars, Riesling and Chardonnay

  • The leaf water relations and photosynthetic rate during acute soil drying were compared in potgrown grapevine cultivars, Vitis vinifera cv. Chardonnay and V. vinifera cv. Riesling. Leaf water potential in Riesling decreased significantly from day 2 after water had been withheld, while in Chardonnay leaf water potential for the water-stressed plants was almost identical with that in well watered plants during the first 4 days. Higher stomatal conductance and photosynthetic rate in Chardonnay than Riesling were observed until day 3 after withholding water. Photosynthetic rate in water-stressed Chardonnay was not different from that in control plants until day 3 after withholding water, while that in water-stressed Riesling was reduced markedly from day 2. In Riesling, osmotic potential at turgor loss point was not changed irrespective of watering conditions. However, in Chardonnay osmotic potential at turgor loss point decreased more in the water stressed conditions than in well watered conditions. The osmotic adjustment in Chardonnay under water stress conditions must contribute to the maintenance of higher stomatal conductance and photosynthetic rate than those in Riesling for a significant period of the drying process. Though difference in stomatal conductance between the two cultivars was shown in the process of soil drying, stomatal conductance of both cultivars responded to vapor pressure difference between leaf and ambient air, rather than soil water status and leaf water potential.
