Radiation Effect of X-Ray and Thermal Neutron on Robinia pseudoacacia L. and Some Other Species (아까시나무외 몇 수종(樹種)에 대(對)한 X-Ray와 Thermal Neutron의 조사효과(照射効果))

  • In an effort to improve the major tree species in Korea, the seed of Robinia pseudoacacia, Pinus rigida, Pinus densiflora, Pinus thunbergii and Larix leptolepis were treated with X-ray and thermal neutron at the Brookhaven National Laboratory, and germination rate of the seed and some characteristics of the seedlings from irradiated seed were investigated and the results were summarized as follows. 1. The germination rate of the irradiated seed of Robinia pseudoacacia, Pinus densiflora, Pinus thunbergii and Pinus rigida was decreased, when the irradiation time of thermal neutron increased from 3 hours to 9 hours. The seed of Larix leptolepis was completely died out in all range of irradiation time. 2. The seed of Pinus densiflora, Robinia pseudoacacia and Pinus rigida showed low germination rate, when the dosage of radiation increased in the range of 10,000r-30,000r X-ray. This dosage of radiation was almost lethal to the seed of Pinus thunbergii and Larix leptolepis. 3. The growth rate of radiated Robinia pseudoacacia has been decreased when the dosage of X-ray and thermal neutron increased. However, the trees treated with thermal neutron for 3 hours showed 14.9 percent-increase in seedling height and some thornless individuals appeared in this treatment. 4. Individuals with variegated leaf, rugose leaf and albino were appeared in X-ray and thermal neutron treatment. 5. Abnormal mitosis of somatic cell, cell with two nucleoli, cell with two nuclei and chromosome clump in mitosis of somatic cell were observed in Robinia pseudoacacia irradiated with thermal neutron. 6. Resistanty against pawdery mildew was decreased in Robinia pseudoacacia radiated with X-ray and thermal neutron. 7. Length of stomata did not show any difference however number of stomata per unit area decreased in Robinia pseudoacacia radiated with thermal neutron. The leaves of Robinia pseudoacacia radiated with thermal neutron were thicker than those of non-treated one, but width of palisade tissue was decreased. The most sensitive one among those species to the thermal neutron treatment was Larix leptolepis, followed by Pinus densiflora, Robinia pseudoacacia, Pinus thunbergii and Pinus rigida in the order. In X-ray treatment, the most sensitive one was Larix leptolepis, followed by Pinus densiflora, Pinus thunbergii, Pinus rigida and Robinia pseudoacacia in the order. Morphological, cytological variation of the radiated Robinia pseudoacacia seemed to indicate some possibility to be used for tree improvement.

  • Three Pinus densiflora populations as shown in location map (Fig. 1) were studied in 1977. These succeed the population numbers 10, 11 and 12 after the preceeding populations. Following the previous study methods, 20 trees were chosen from each population and the morphological characteristics such as tree forms, branching habit, needle and wood properties were investigated. The results are summerized as follows; 1. The mean stand ages were ranged from 40 to 45. The growth performances of trees of population 10 and 11 was similar, but 12 seemed to be inferior more or less. 2. The ratios of clear bole length was 0.53 in population 12 as the highest but 0.43 for population 10 as the lowest. 3. The population 12 was considered to be a stand of the coarser branching habit having the crown index (The maximum crown diameter/the crown length) 1.65 though the mean branching angle indicates almost horizontal. 4. The differences were observed in the clear bole length ratios and crown-indices between populations as shown in Fig. 3 and 4. 5. No inter-population differences in serration density of needle was shown but significant inter and intra-population and individual differences (within population) in number of stomata rows and resin duct. 6. Population 12 shown 0.119 of resin duct index as the maximum. 7. The pattern of diameter growth, analyses based on the width of 10-year-ring segment unit (for example, the 1st segment denotes the width between pith center and 10th year ring and the 2nd one is from 11th to 20th year ring and so on.), was alike among populations as shown in Fig. 9. 8. No significant differences between population in mean summer wood percentages as well as in wood specific gravity was observed. The values of wood specific gravity were increased with the increase of ages in population 10 and 11 however vice versa in population 12. 9. The fiber length was increased with the increase of age but no differences between populations as shown in Fig. 12.

The Effect of Salicylic Acid (SA) on the Photosynthetic Activity in Commelina communis L. (Salicylic acid가 닭의장풀의 광합성에 미치는 영향)

  • The effect of Salicylic acid (SA) on the leaf growth, chlorophyll content, photosynthesis, stomatal conductance in Commelina communis L. was investigated. C. communis which was grown in Hoagland solution containing 1 mM SA during 3 weeks resulted in a significant reduction of leaf length, width and area. 1 mM SA reduced about 10% of the leaf area at 1 week and 2 weeks, but inhibited 22% of the leaf area at 3 weeks. SA also showed great effect on the reduction of chlorophyll content. SA reduced 47% and 53% of chlorophyll content each at 2 weeks and 3 weeks when it was compared with the control. Chlorophyll a/b ratio in SA treated sample was increased at 3 weeks representing that SA was sensitive to chlorophyll b. The treatment of SA did not inhibit the activity of PSII+PSI, PSII and PSI. This result indicates that SA did not show direct effect oil the photosynthetic electron transport activity. However, when C. communis was grown in Hoagland solution containing I mM SA fer long period, SA showed clear inhibition of photosyhthesis in intact leaves. The treatment of SA for 3 weeks showed about 23~65% inhibition of photosynthetic activity at various light intensity (100~1000$\mu$mo1 m­$^2$$^1$). Similar effect was found on stomatal conductance. The treatment of SA caused an almost closure of stomata. Similar effects of stomatal conductance at various light intensity indicate a loss of normal control on stomatal mechanism. These results indicate that the effect of SA on photo-synthetic activity was not by SA itself but by indirect metabolic pathway.

Effects of Bagging Periods on Pericarp Characteristics and Berry Cracking in 'Kyoho' Grape (Vitis sp.) (괘대시기가 '거봉' 포도의 과피 특성과 열과에 미치는 영향)

  • The berry growth and pericarp characteristics were characterized to confirm the effects of bagging periods on berry cracking during berry development in 'Kyoho' grape. The berry weight was the highest at 13.4 g in late period of bagging treated at 7 to 9 weeks after full bloom (WAFB) as compared with the lowest of 12.3 g in total period bagging. The berry cracking rate under critical turgor pressure in the non-bagging control was 53.3%, while those of bagging treatments were decreased in the order of 42.7%, 37.3%, 33.3%, and 18.7% in bagged during 3 to 9, 3 to 5, 5 to 7, and 7 to 9 WAFB, respectively. In the results of observation on histological characteristics of pericarp, berry lenticels of whole bagging treatments had smaller and normal shape compared with non-bagging control treatment. Especially on the pericarp of late period bagged during 7 to 9 WAFB, suberization around stomata and micro-cracking were not observed and structural strength of pericarp was increased with thicker sub-epidermal layer and cell wall. Therefore, the results indicate that bagging treatment for two weeks just before the veraison when the day length and daylight is relatively longer and stronger can effectively reduce berry cracking by strengthening structure of pericarp in 'Kyoho' grape.

A Study on the Rate of Occurrence according to Watering Methods, Susceptibility and Chemical Control of Chrysanthemum Nematode (국화잎 선충에 대한 저항성 검정, 관수방법에 따른 국화잎선충의 피해율 및 화학적 방제에 관한 연구)

  • The susceptibility of 8 chrysanthemum varieties to Apelenchoides ritzemabosi, Shuhonothikara and Sagakiku were resistant by 11.7% and 14.5% each infested leaf, Sinrokusiogio, Dangonanako and Sintoa were susceptible as above 50%, Gold wave and Tenju were modertae by 24 to 26%. Percentage of infested leaf was positively correlated with the number of epidermal hair but not with leaf area, leaf thickness, stomatal size, number of stomata and length of epidermal hair. In effect of watering method, Polyethylene film cover plot was lowest by 9.4%, next conventional watering plot by 50%, Conventional watering and spray plot was the highest by 62%. The infested leaf rate was closely related with a number of rainy days than the amount of rainfall. Effect of chemical soil treatment, Temik showed the lowest infested leaf rate by 5%. There was no significant difference between the chemicals except Temik, but these are markedly effective in compare to control.

  • As a preliminary investigation to obtain useful auto- and allopolyploids a ten year old tetraploid of Pinus densiflora induced from the colchicine treated seed was observed on the cytological, morphological and physioa logical characters in the present study and the results can be summarized as follows. 1. The number of chromosome at the base of needle was 48 so that it was recognized as a tetraploid. 2. The needles were thicker and the number of them on an individual was less than the 2n plant. The needle combined to a single was appeared 2%. 3. No difference was found in the number of stomata rows on the central part of needle between the tetraploid and 2n plant, however, guard cells of tetraploid increased 36% in the long diameter compared with 2n plant. 4. Microsporangiate storbile developed normally showing the same size with 2n plant while the size of pollen grain increased about 20% larger than 2n plant. 5. Germination percentage in vitro increased until 17 hours but decreased after 26 hours compared with 2n plant. The longest length of pollen tube during the germination period appeared in the tetraploid pollen.

  • The purpose of present study is to analyze the genetic variation of natural stand of Pinus densiflora. In 1976 following after the seletion of 1974 and 1975, twenty trees from each of three natural populations of the species were selected and seeds were collected, and the locations and conditions of populations are presented in table 1, 2 and figure 1. Some morphological traits of the populations were already detailed in our fifth report of this series. The morphological traits of cone, seed and seed-wing, and also the growth performances and needle characters of the seedling were observed in the present study according to the previous methods. The results obtained are summarized as follows; 1. The meteorological data obtained by averaging the records of 30 year period(1931~1960) measured from the nearest meteorological station to each population are shown in fig. 2, 3, 4. The distributional patterns of investigated climate factors are generally considered to be similar among the locations. However, the precipitation density during growing season and the air temperature during dormant season on Samchuk area (Pop. 9) were quite different from those of the other areas. 2. The measurements of fresh cone weight, length, diameter and cone index (i.e.: length to diameter ratio) are presented in table 7. As shown in table 7, all these traits except for cone diameter seem to be not significant in population and to be highly significant in family differences within population. 3. The morohological traits of seed and seed-wing are detailed in table 8, 9, and highly significant differences are recognized among the populations and the families within population in seed weight, seed length, seed thickness but not among the populations in the other observed traits. The values of correlation between the characteristics of cone and seed are presented in table 12. As shown, the correlation between cone length and seed wing length, between seed wing width and seed width were significantly positive in population 8 and 9 but in population 7. The positive correlations between seed length and seed width were calculated in all populations studied 4. Significant statistical differences among populations and families within population are observed in the growth performances of 1-0 seedling height of these progenies. But the differences in 1-1 seedling height and root collar diameter are shown only among familes within population. As shown in table 13, the most parts of correlations are not significant statistically between the growth performances of seedling and the seed characters. 5. As shown in table 15, statistical differences are considered to be significant among the populations in stomata row on both sides of the needle but not in serration density. The correlations between progenies and parents are not generally observed in the investigated traits of needle as shown in table 16.

  • The variation of stomatal density and stomatal length of four species of oaks was studied for the purpose of examining the differences among populations and among individual trees within population. Nine populations of Quercus mongolica, four populations of Q. serrata and Q. variabilis respectively, and three populations of Q. acutissima were selected in the natural stands of oaks distributed through the whole country. Twelve leaves were sampled from each of 20 trees from each population. The length of 20 stomata and ten replications of stomatal density were measured from collodion replicas of each leaf under a microscope. Average stomatal densities and lengths ranged through $600-1000/mm^2$ and $19-26{\mu}m$ respectively in all of the species studied. The stomatal densities and lengths presented significant differences statistically at the level of 1 or 5% among populations and among individual trees within population in all the species. Quercus mongolica, especially, showed large variation among populations, while Q. variabilis did very narrow variation compared to the other species. The coefficients of variation of stomatal densities and lengths among individual trees within population exhibited small values of 3.7-12.0% and 1.4-5.3% respectively in all the populations of the species. The average stomatal densities of Q. mongolica showed statistically significant correlation of multiple correlation coefficient of $R_{df{\cdot}2.6}=0.868^*$ and multiple regression equation of $Y=0.041X_1(G.M.T.S.)+0.489X_2(G.M.H.S.)+22.37$ with the sum of growing season mean daily temperature and the sum of growing season mean daily humidity of the stand studied. However the average stomatal lengths showed no relation with the same meteological variables. The figures of frequency distribution of the measurements of leaves or the mean values of individual trees did not show normal distribution curves in some populations. The curves, as well as the results of ANOVA, exhibited the differences among populations.

  • Six natural populations of Pinus densiflora S. et Z. as shown in the location map (Flg 1) were studied during 1978. The numerial pouplation codes, 13 to 18. The results of populations 1 to 12 were reported in previous papers. Following the study methods described before, 20 trees were sampled from each population and morpological characteristics such as stem forms, branching habit needle and wood properties investigated. The results are summerized as follows; 1. The mean stand ages were ranged from 36 to 97 of years. The growth performances of trees of population 14. 15 and 18 was similar, but 13, 16, and 17 seemed to be inferior more or less. 2. The ratios of clear bole length were 0.70 in population 18 as the highest but 0.28 for population 16 as the lowest. 3. The population 17 was considered to be a stand of the coarser branching habit having the crown index (The maximum crown diameter/the crown height) 158 though the branching angles were almost horizontal. 4. The differences were observed in the clear bole length ratios and crown-indices between population as shown In Fig. 3 and 4. 5. As to the serration density, number of stomata row and resin duct; the significant differences exist between individual trees within population and also between populations. 6. Population 18 shown resin duct index 0.119 as the maximum. 7. The patterns of diameter growth, based on the width of 10-year-ring segment unit(for example, the 1st segment denotes the with between pith center and 10th year ring and the 2nd one is from 11th to 20th ring and so on.), were alike among populations as shown in Fig 9. 8.   • The variation of needle, cone and seed characteristics and of allelic frequency in isoenzyme, ADH, ME and PGI, according to different resin duct index in Pinus densiflora, Pinus thunbergii and their hybrids was analyzed. The results obtained were as follows : 1. With increase of number of resin ducts, morphological characteristics such as needle length, needle sheath length, cone size, seed size, seed wing size, 1000 seeds weight, etc. tended to he increased, while number of stomata row in needle to be decreased. 2. As the results of discriminant analysis for the morphological characteristics of needle, cone and seed, most individuals are generally coincided with number of resin duct mostly in Pinus densiflora were not. 3. According to the canonical discriminant function obtained from the morphological characteristics in Pinus densiflora, Pinus thunbergii and their hybrids including introgressive hybrid, the resin duct index, 1000 seeds weight, cone size and neelde sheath length characterized fairly their species. 4. With increase of resin duct index, hybrid index tended to be higher. The results obtained from the discriminant analysis and the hybrid index were nearly same each other. 5. With increase of number of resin duct, the allelic frequencies for isoenzyme, ADH-$B_2$, ME-$A_2$ and PGI-$B_1$. $B_2$ tended to increase but those of ADH-$B_3$, ME-$A_4$, and PGI-$B_3$, to decrease. It is thought that this increase of frequency for the former four isoenzymes was resulted in introgressive gene flow from Pinus thunbergii to Pines densiflora and accordingly the frequency of latter three isoenzymes tended to decrease.
