Effect of Aeration and Agitation Conditions on the Production of Glucoamylase with Aspergillus niger No. PFST-38

  • Aspergillus niger No. PFST-38 was grown on complex media in 30L agitated fermentors at various aeration rates and stirrer speeds. We could correlate the mixing time as a function of the Reynolds number and the apparent viscosity, as follows. ${\theta}_M=2.95\;\NRe^{-0.52},\;{\theta}_M=1.88\;{\eta_a}^{0.57}$ Also, the effects of the apparent viscosity (${\theta}_a$), the impeller rotational speed (N), the air flow rate ($V_s$), and the mixing time (${\theta}_M$) on the oxygen transfer coefficient, $K_L a$ were determined experimentally, and equated as follows. $K_La=12.04N^{0.88}Vs^{0.71}{n_a}^{-0.83},\;K_La=30.2N^{0.88}Vs^{0.71}{\theta_M}^{-1.45}$ $K_La$ increased as the agitation speed and the air flow rate increased. The rate of $K_La$ increase was dependent more on the rotational speed of impeller than on the air flow rate. The glucoamylase production increased with the increase of the agitation speed upto at 500 rpm and increased with the increase of air flow rate upto at 1.0 vvm. The values calculated from the above equation confirmed that the experimental maximum production of glucoamylase was achieved when the $K_La$ and the apparent viscosity of the broth were $260\;hr^{-1}$ and 1800 cps, respectively.

Characterizations of Modified Silica Nanoparticles(I)

  • (3-mercaptopropyl)trimethoxysilane (MPTMS) was used as a silylation agent, and modified silica nanoparticles were prepared by solution polymerization. 2.0 g of silica nanoparticles, 150 ml of toluene, and 20 ml of MPTMS were put into a 300 ml flask, and these mixtures were dispersed with ultrasonic vibration for 60 min. 0.2 g of hydroquinone as an inhibitor and 1 to 2 drops of 2,6-dimethylpyridine as a catalyst were added into the mixture. The mixture was then stirred with a magnetic stirrer for 8 hrs. at room temperature. After the reaction, the mixture was centrifuged for 1 hr. at 6000rpm. After precipitation, 150 ml of ethanol was added, and ultrasonic vibration was applied for 30 min. After the ultrasonic vibration, centrifugation was carried out again for 1 hr. at 6000rpm. Organo-modification of silica nanoparticles with a ${\gamma}$-methacryloxypropyl functional group was successfully achieved by solution polymerization in the ethanol solution. The characteristics of the ${\gamma}$-mercaptopropyl modified silica nanoparticles (MPSN) were examined using X-ray photoelectron spectroscopy (XPS, THERMO VG SCIENTIFIC, MultiLab 2000), a laser scattering system (LSS, TOPCON Co., GLS-1000), Fourier transform infrared spectroscopy (FTIR, JASCO INTERNATIONL CO., FT/IR-4200), scanning electron microscopy (SEM, HITACHI, S-2400), an elemental analysis (EA, Elementar, Vario macro/micro) and a thermogravimetric analysis (TGA, Perkin Elmer, TGA 7, Pyris 1). From the analysis results, the content of the methacryloxypropyl group was 0.98 mmol/g and the conversion rate of acrylamide monomer was 93%. SEM analysis results showed that the organo-modification of ultra-fine particles effectively prevented their agglomeration and improved their dispensability.

Effects of Carbon and Nitrogen Sources on the Shoot Formation in bioreator culture of Scrophularia buergeriana Miquel (현삼에서 탄소원과 질소원의 종류와 농도가 기내 식물체 분화에 미치는 영향)

  • To determine the proper carbon and nitrogen sources and their proper levels for mass micro propagation of Scrophularia buergeriana Miquel, tonic and curing cough experiment were applied and a method for mass cultivation by using bioreactors (2.5 L) was expinined. Proper ratio of $NH_4NO_3\;:\;$KNO_3$ was 413 mg/L : 1900 mg/L for multiple shoot production. Sucrose was more effective than glucose or fractose as carbon source and 3% concentration was good for shoot formation. Total nitrogen was not detected after six weeks both in 500 ml flask and bioreactor culture. Sucrose was decreased sharply after two weeks and there was no sucrose left after three weeks both in 500 ml flask and bioreactor culture. The stirrer in bioreactor caused shear stress to shoots severely. The sphere type bioreactor was better than the cylinder type and removal of inner loop in sphere type was more effective to avoid shear stress.

  • The particulates reinforced metal matrix composites(PMMC) have a number of interesting mechanical properties. including high strength and good resistance to wear at high temperature and low thermal expansion. The equipment structure to obtain the homogeneous distribution in composites are proposed for the continuous pouring of reinforcement at the desired temperature. The particulates reinforced metal matrix composites(A357/SiCp) were fabricated by the process of the combined stirring method with the various fabrication process. The combined stirring method to niform distribution of particle is consisted of two stirring force both electro-magnetic stirring generated from induction heating and mechanical stirring with graphite stirrer. PMMC billets were fabricated with the volume fractions ranged from 0% to 20% and particle sizes ranged from 14${\mu}{\textrm}{m}$ to 25${\mu}{\textrm}{m}$. It is important to cont the size of primary $\alpha$-Al solid particles because it could become the cause of the particle pushing or capture phenomena from the fact that secondary dendrite arm spacing size depends on the cooling rate during the solidification in hypoeutectic Al-Si alloy. Therefore, the effect of primary $\alpha$-Al on the reinforcement distribution in matrix alloys has been investigated. The microstructure of PMMC fabracated with various volume fractions(0%, 10%, and 20%) and particle size were observed.

Analysis on ESD Properties of the PANI added PU/MWNT Films (PANI 첨가 PU/MWNT 필름의 정전방전특성)

  • This paper surveys the ESD characteristics of the PANI added PU/MWNT film according to the manufacturing conditions such as variation of the loading contents of PANI and the mixture ratio of 2 dispersion solutions. For this purpose, PANI added PU/MWNT ground films were made with IPA/MWNT 3wt% dispersion solution and PANI/DMF dispersion solutions(5, 10, 15, 20, 25, and 30wt% contents of the PANI) by the mixture ratio of dispersion solution(10/50, 20/40, 30/30, 40/20, and 50/10part) in the PU (972DF) 100g, which was treated with 500rpm for 30min in the stirrer with condition of the dry temperature $120^{\circ}C$ for 2min. Totally, 36 kinds of PANI added PU/MWNT film specimens were prepared. The physical properties of the PANI added PU/MWNT films such as electrical resistivity, absorbancy by UV-Vis spectrometer, and triboelectricity were measured and discussed with surface characteristics of the PANI added PU/MWNT films by SEM. The dispersion property of PANI to the DMF showed best dispersion at the 25% of PANI content. The surface electrical resistivity of the PANI added PU/MWNT films was decreased with increasing the weight content of PANI/DMF dispersion solution, and it showed the lowest value $10^6{\Omega}$ at the mixing condition of PANI/DMF 20part and MWNT/IPA 40part with 30% PANI. Furthermore, it was shown that the electrical and physical properties of the PANI added PU/MWNT film such as electrical resistivity, and triboelectricity were better than those of PU/MWNT film prepared with no PANI, which was result obtained in previous paper.

Optimization of Precipitation Process for the Recovery of Lactic Acid (Lactic acid 회수를 위한 침전공정 최적화)

  • In this study, precipitation process was developed for the recovery of the lactic acid from calcium lactate fermentation broth. Calcium lactate yield was improved by decreasing the solubility of calcium lactate through the addition of ethanol (25%, v/v) as a co-precipitant. The optimal lime type, lime concentration, stirrer speed, precipitation time, temperature, and solvent amount for $Ca(LA)_2$ precipitation were CaO, 0.0175 g/mL, 220 rpm, 24 h, $5^{\circ}C$, ethanol 25% (v/v), respectively. Lactic acid was easily and efficiently recovered from precipitated $Ca(LA)_2$ by adding sulfuric acid ($Ca(LA)_2/H_2SO_4$ molar ratio=1:1). In the model solution of organic acids and fermentation broth, the overall yields of recovered lactic acid were 62% and 55%, respectively, under the aforementioned optimal conditions.

Antioxidative Activity of Kalopanax pictus Shoot Extracted Using Different Extraction Methods (추출 방법이 개두릅 추출물의 항산화 활성에 미치는 영향)

  • The antioxidative activity of Kalopanax pictus shoots extracted using five different methods, stirrer extraction (SE), ultrasonification extraction (USE), vacuum extraction (VE), reflux extraction (RE), and reflux extraction after ultrasonification extraction (RUE), was evaluated to determine the most effective extraction method. RUE showed the highest yield of extract, and total polyphenol and flavonoid content. Additionally, the RUE extract showed higher antioxidative activity than that of the other extracts based on DPPH and ABTS free radical scavenging (90.85%, 99.83%), reducing power activity (OD 0.95, 700 nm), nitrite scavenging activity (55.46%), and ferrous ion chelating effect (45.12%).

Effect of Concentrate Level on the Formation of Conjugated Linoleic Acid and Trans-octadecenoic Acid by Ruminal Bacteria when Incubated with Oilseeds In Vitro

  • An in vitro study was conducted to examine the effect of addition level of concentrate on fermentation characteristics and long-chain unsaturated fatty acids composition, especially conjugated linoleic acid (CLA) and trans-octadecenoic acid (t-FA) by mixed ruminal bacteria when incubated with linseed or rapeseed. Four levels (0.83, 1.25, 1.67 and 2.08%, w/v) of concentrate and ground oilseeds (linseed or rapeseed; 0.83%, w/v) were added to mixed solution of strained rumen fluid with artificial saliva (1:1, v/v) in the glass jar with a glass lid equipped with stirrer, and was incubated anaerobically for 24 h at $39^{\circ}C$. Addition level of concentrate slightly reflect on pH and ammonia concentration of the culture solution at the various incubation times when incubated with both linseed and rapeseed. Total VFA concentration slightly increased with incubation times and concentrate levels for incubations with oilseeds. While CLA composition had a clearly increasing trend with incubation time when incubated with linseed, percent CLA was relatively stable when incubated with rapeseed. Percent CLA, however, had a clearly decreasing trend with concentrate level throughout incubation times with significances at 3 h incubations when incubated with linseed (p<0.038) and rapeseed (p<0.0009). The differences in compositions of t-FA were relatively small among concentrate levels for both incubations with linseed and rapeseed. The ratios of t-FA to CLA were lower for linseed with increased proportion of CLA than for rapeseed.

A Study on the Solidification and Purification of High Purity Aluminium Alternate Stirring Method (정역 회전법에 의한 고순도 알루미늄의 응고 및 정련에 관한 연구)

  • The degree of purification and the macrostructure of high purity aluminium were studied through the alternate stirring method in order to improve the nonuniformity of solute concentration in the unidirectional stirring method. The $2^3$ factorial design was done to examine the effects of experimental factors more qualitatively. In the relatively low stirring speed of 1500 rpm with alternate stirring mode, the uniform solute profile and refined grain structure were obtained due to strong washing effect and turbulent fluid flow. It was induced by the transition of the momentum boundary layer by alternation of the stirrer. It was concluded from this study that the alternate stirring mode was more effective to obtain the uniformity of solute even in the stirring speed of 1500 rpm. But the degree of purification decreased below the critical alternating period. When 2N(99.8wt.%) aluminium was used as the starting material the morphology of solid-liquid showed the cellular shape and the columnar grains were inclined to the direction of rotation. This inclined grain growth resulted from the difference of relative velocities of solid and liquid. The inclined angle was increased as the stirring speed increased and solidification proceeded. In the case of 4N aluminium, there was no inclined grain growth and it was confirmed from the macrostructure and SEM work that the morphology of solid-liquid interface was planar. From the factorial design, it was found that the alternate stirring mode showed poorer purification effect than that of unidirectional stirring mode at low speed(500 rpm). In addition, the factor that had the most significant effect on the degree of purification was the stirring speed.

Production of Mycelia and Water Soluble Polysaccharides from Submerged Culture of Lentinus lepideus in Bioreactor (생물반응기를 이용한 잣버섯(Lentinus lepideus)의 균사체 및 수용성 다당체 생산특성)

  • The mushroom Lentinus lepideus was used to produce mycelial as well as soluble polysaccharides in bioreactor cultures. To determine optimal submerged culture conditions, both growth characteristics and water soluble polysaccharides production were compared among four different types of bioreactor and culture conditions. For the production of mycelial biomass, the following bioreactors were proven to be effective in decreasing order: an external-loop type air-lift bioreactor (ETAB; 7g/l), a balloon type air bubble bioreactor (BTBB; 6.2g/l), a stirrer type bioreactor (STB; 6g/l), and a column type air bubble bioreactor (CTBB; 5g/l). Maxiaml production of water soluble exopolysaccharides (EPS; 0.62g/l) and endopolysaccharides (PPS; 7.7%) could also be obtained from BTBB. The mycelial biomass increased with increase in glucose concentration from 15g/l to 75g/l in the media. In contrast, PPS contents in the cells decreased with increase in glucose concentration in the media, showing the highest PPS content (7%) at 15g/l. Among different medium feeding types, fed-batch culture based on concentration control in media (10g/l) produced higher mycelia than fed-batch culture based on volume control of media (5.8g/l) or batch culture (3.4g/l). EPS production was also higher in fed-batch culture based on medium concentration control than that in other feeding types.
