RENIN 분필(分泌)에 미치는 온도의 영향(影響) (Effect of Temperature Changes on the Renin Release in Vitro Experiments)

  • It has been well known that beta-adrenoceptor is responsible for the renin release stimulatory and alpha-adrenoceptor may be inhibitory. It has been observed accidently that alpha-adrenergic agonist can inhibit renin release by just changing the medium temperature in Vitro experiment in this laboratory. A series of experiments were performed to clarify this interesting phenomena in Vitro experiment. Rat renal slices were incubated in PSS medium under gas phase at $37^{\circ}C$. The following results were observed. 1) Isoproterenol and norepinephrine resulted in renin release stimulatory in dose-dependent by the concentrations of $10^{-9}$ to $10^{-5}\;M/L$ at $37^{\circ}C$. 2) Norepinephrine resulted in renin release inhibitory in dose dependent by the concentrations of $10^{-7}$ to $10^{-5}\;M/L$, and almost no effect by isoproterenol $10^{-6}\;M/L$ at $20^{\circ}C$. 3) Phenoxybenzamine pretreatment at $37^{\circ}C$ accentuated isoproterenol stimulatory effect at $37^{\circ}C$. 4) Phenoxybenzamine pretreatment at $20^{\circ}C$ attenuated isoproterenol stimulatory effect at $37^{\circ}C$. These data suggest that the renal adrenoceptor(s) related to renin release maybe a single entity, and can be interconverted different forms in certain conditions.

Biological effect of recombinant IL-1$\beta$ on the expression of antiviral genes in the gill of rainbow trout, Oncorhynchus mykiss

  • We have investigated the biological effects of recombinant IL-1$\beta$ (rIL-1$\beta$) on the expression of antiviral genes such as Myxovirus-3 (MX-3) and Interferon regulating factor-1 (IRF-1), which are related to type I interferon. When ten micrograms of rIL-1$\beta$ were treated, we observed the stimulatory effect on the expression of these antiviral genes. Interestingly, at the early stage of stimulation, these genes were down-regulated and then up-regulated by the results obtained that the expressions of these genes were decreased at day 1 post-injection and gradually increased at day 3 post-injection. Thus, the stimulatory effect of rIL-1$\beta$ on the expression of MX-3 and IRF-3 gene might be an indirect stimulatory effect because significant up-regulation was delayed until day 3 post-injection.

  • Ginsenoside-Rg$_{1}$(G-Rg$_{1}$) has been shown to stimulate DNA synthetic activity in SK-HEP-1 cells. This study was therefore designed to determine in SK-HEP-1 cells whether the stimulatory effect of G-Rg$_{1}$ may be mediated by protein kinase C (PKC) which is known to play a key role in the signal transduction pathway leading to the cell proliferation. Using the tn situ PKC assay method, the PKC enzyme activity was determined in SK-HEP-1 cell cultures in response to G-Rg$_{1}$ at 3*10$^{-5}$ M or phorbol 12-myristate 13-acetate(PMA) at 10$^{-6}$ M which in the enzyme activity by 1.5- and 7-fold, respectively. Furthermore, G-Rg$_{1}$, was also able to synergistically increase the enzyme activity by 11-fold m the cell cultures in the presence of PMA. These stimulatory effects of G-Rg$_{1}$ or PMA on the DNA synthetic activity and the PKC activity were ablished by a specific PKC inhibitor, GF109203X. These results suggest that the stimulatory effect of G-Rg$_{1}$ on the DNA synthetic activity may be partly due to stimulation of PKC-mediated signal transduction pathway leading to the proliferation of SK-HEP-1 cells.

  • Ginsenosides present in the roots of panax ginseng C.A. Meyer were shown to induce a stimulatory effect on the overexpressed cellular chicken c-src protein tyrossine kinase in NH3T3 cells. Among 4 ginsenosides studied $(G-Rb_2,\;G-Rc,\;G-Re\;and\;G-Rg_1),\;G-Rg_1$ showed the most stimulatory effect at $16.7\mu{g/ml}$ ginsenoside concentration increasing the activity by 2-4 times. Inhibitors of either protein synthesis or RNA synthesis blocked the activation of c-src proein tyrosine kinase. These results suggest that the csrc kinase activation apprars to involve an increase in the amount of protein of the kinase by transcriptional control mechanism rather than an increase in the kinase activity.

  • Adjuvant is an immune enhancer commonly used during vaccination to enhance the host immune response. In the present study, we produced the several recombinant protein from immune related gene of olive flounder (Paralichthys olivaceus). Especially, to produce the soluble type of recombinant protein, we constructed the MBP (Maltose binding protein) fusion G-CSF (Granulocyte colony stimulating factor) recombinant protein among the flounder immune related genes. To verify the immune stimulatory effect and safety of this recombinant protein (rPoGCSF), expression changes of several immune genes were tested using quantitative real-time PCR method with gene specific primer from flounder head kidney leukocytes. As a result, we confirmed that the rPoGCSF has an ability of immune stimulatory effect, also it has broad range of pH and temperature.

Albumin Fractions from Different Species Stimulate In Vitro Progesterone Production by Granulosa Cells in Buffalo

  • The ovarian follicular fluid was found to contain steroidogenesis stimulatory protein similar to albumin from human and buffalo. Therefore, the albumins from various species, commercial and purified, were studied for their steroidogenic effect on progesterone secretion by granulosa cells from buffalo ovaries, during culture. A dose of $20{\mu}g$ of bovine serum albumin was optimum to exhibit maximum progesterone secretion on day 6 of culture, in medium ($350{\mu}l$) containing $10^5$ cells. Among commercial albumins, chicken albumin showed highest effect on progesterone secretion, which was followed by albumins from goat, bovine, human, sheep and rat, respectively at day 6 of culture. The albumins were also purified from blood serum of buffalo, goat and rat using salt fractionation, ion-exchange chromatography, gel filtration and SDS-PAGE. The highest stimulatory effect on progesterone secretion was shown by albumin purified from buffalo blood serum and lowest by that from rat blood. Comparatively the buffalo and goat albumins were more biologically active than commercial albumins. The presence of some active molecules conjugated with freshly purified albumins may be responsible for better stimulatory effect.

  • 양송이를 재배할 때 복토의 최적 pH와 칼슘원의 효과를 구명코저 본 시험을 실시하였다. 양송이 복토의 최적 pH는 7.5이었다. 중성내지 약알카리성토양에서 칼슘의 촉진적 효과가 뚜렷하였다. 즉, 복토에 첨가되는 수산화칼슘 및 탄산칼슘의 효과는 pH교정뿐만 아니라 칼슘의 생장촉진효과도 중요하다고 본다.

  • We used the saponin Rgi extracted from Panax ginseng to study its effects on lymphocytes of 10 young and 19 elderly persons. The proliferate response of Iymphocytes cocultured for 72h with PHA and saponin was measured by using MTT method and the'H-TdR incorporation procedure. PHA and Rgl had stimulative effects on the phenotype of Iymphocytes (p<0.001). Rgl also increased the fluidity of lymphocyte membrane of the aged (p<0.001). The CD2s and CDfsRA positive cells of Iymphocytes in the elderly were lower than those of the young people,8.6clo $\pm$ 2.7olo vs 10.43% : 3.5%, 20.95% $\pm$ 15.5clQ vs 50.86% :4.3olo, respectively. More CDfsRO positive cell lymphocy populations were seen in the aged. The CEfsRO positive cells of the young people were 39.63% $\pm$ 3.2%. We discussed the cause of declined immune function of Lymphocytes of aged person and the mechanism of the effect of P. ginseng on Lymphocytes. 저출력레이저조사가 섬유아세포와 면역세포의 증식 및 유전자발현에 미치는 영향 (Effects of Low Level Laser on the Proliferation and Gene Expression of Fibroblasts and Immune Cells)

  • The growth and synthetic activities of fibroblasts are regulated by cytokines and growth factors derived from activated inflammatory cells. Stimulatory effect of low level laser (LLL) radiation on wound healing seems to be in part due to direct stimulatory action on cell proliferation and synthetic activities of fibroblasts. Also indirect stimulatory effect on the fibroblast function through inflammatory or immune cells is another possible mechanism of biostimulatory action of LLL. This study was performed to determine the growth rate of human gingival fibroblasts obtained biopsy and culture, fibroblast cell line, and immune cell line by using $[^3H]-$ thymidine incorporation test. And gene expression pattern was also analyzed by using the DNA probe such as Hsp70, IL-1$\beta$, MIP-1$\alpha$ and actin cDNA. Proliferation rate of gingival fibroblast was increased by LLL irradiation, but no more effect was added by LPS or IL-1$\beta$ pretreatment Enhanced Hsp70 gene expression was found from gingival fibroblasts and fibroblast cell line COS by LLL irradiation., which was not more increased by LPS or IL-1$\beta$ pretreatment. LLL-irradiated promyelcytic cell line HL-60 and macrophage cell line RAW264.7 showed significant stimulatory effect of proliferation rate when compared with respective control. However there were no changes in growth rate of other immune cell tested in this study, such as B cell line WR19n.l and 230, helper T cell line Jurkat and Hut78, cytolytic T cell line CTLL-r8. By LLL-irradiation Hsp70 gene expression was increased in RAW246.7 and HL-60, not in CTLL-R8. And IL-1$\beta$ and MIP-1$\alpha$ gene expression were induced only from LLL-irradiated RAW264.7. These results led us to presume that LLL radiation may affect to the immune cells, especially to macrophage, through which it might promote wound healing process.

인삼(Panax ginseng C.A. Meyer) Saponin 성분이 흰쥐의 장기에서 Polyamine 대사에 미치는 영향

  • In order to study effects of Korean ginseng (Panax ginseng C.A. Meyer) saponin fraction on polyamine metabolism in rat organs, Korean ginseng saponin fraction was administrated to rats for 1, 2, 3, 4, 6 and 12 months and brain, liver, prostate, spleen and testis were removed from these rats. Two enzyme activities were measured from those organs; ornithine decarboxylase (ODC), which is a regulatory enzyme of putrescence biosynthesis and S-adenosylmethionine decarboxylase (SAMDC), which is also a regulatory enzyme of spermidine and spermine biosynthesis. The amounts of polyamine were also determined. As for prostate and testis organs, Korean ginseng saponin fraction was innocuous for ODC and SAMDC activities from rats fed for 1 and 2 months. However, after 3 months, the stimulatory effect on the activities of two enzyme gradually increased in test groups and reached its maximum in 12 months. The contents of spermidine and spermlne of test groups in prostate and testis were also much higher than those of control groups. Another stimulatory effect on the activities of two enzymes was observed in liver and reached its maximum in 6 months. In the other organs such as brain and spleen, the enzymes were turned out to be not affected by feeding Korean ginseng saponin fraction. From the cumulative results, the stimulatory effect of Korean ginseng saponin fraction on polyamine metabolism was observed in prostate, testis and liver.
