Studies on the mechanism of the cardiovascular effect of intraventricular 5-hydroxytryptamine in rabbit

  • An attempt was made to investigate the effect of intracerebroventricular 5-hydroxytrypatamine (5-HT) on the cardiovascular system in urethane-anesthetized rabbit and to elucidate the mechanism of its action. 5-HT given into a lateral ventricle caused clearly a dose-dependent decrease inboth arterial blood pressure and in heart rate. The bradycardia and hypotension induced by 5-HT were significantly attenuated by the prior injection of ketanserin, cyproheptadine or clonidine. Pretreatment of atropine with bilateral vagotomy did not affect both bradycardia and hypotension. Propranolol weakened markedly the breadcardia of 5-HT but did not influence the depressor response of 5-HT. These experimental results suggest that intraventricular 5-HT cause the hypotension and bradyardia in rabbits through the stimulation of serotonergic receptors in brain, which is seemed to be associated to inhibition of sympathetic tone.

  • Observing after enforcing the medication and the acupuncture theraphy on the CVA patient sick with the cerebral embolus by organic pathology of the heart and whose desease was from heat symptoms caused by exessive pathogenic factors of which caused from affection due to exogeneous factors, we obtained the results as follows; 1. Serious condition could be relieved by removing acute symptoms according to the principle of the 'In emergency causes treat the acute symptoms first, when these being relieved treat it's fundament cause'. 2. The lesion could be disable for which the traditional medication theraphy through the differentiation of symptoms-complexes had removed the symptoms caused by organic pathology of heart. 3. The acupuncture stimulation through the differentiation of symptoms seemed to influence electric conduction system. Through this case, If we objectify the diagnosis and the medical treatment, the part of the curing diseases can progress more effectively.

Effects of Piroxicam on Pharmacodynamics and Pharmacokinetics of Nifedipine in Spontaneously Hypertensive Rats (피록시캄이 니페디핀의 약력학 및 약동학에 미치는 영향)

  • Because nonsteroidal anti-inflammatory drugs are reported to cause fluid retention and hypertension by inhibition of prostaglandin synthesis, the effects of piroxicam on pharmacodynamics and pharmacokinetics of nifedipine were studied in male spontaneously hypertensive rats. They received nifedipine (0.5 mg/kg) alone or combined with piroxicam (5 mg/kg) intravenously. Plasma levels norepinephrine, an index of sympathetic stimulation, were measured prior to each treatment and 5 min after drug administration. Changes in blood pressure were examined serially and blood samples for analysis of nifedipine were also taken for 6 hr following drug administration. Plasma nifedipine concentration were assayed by HPLC and pharmacokinetic parameters were calculated. Blood pressure was reduced (p<0.01), but plasma norepinephrine level was increased (p<0.05) by nifedipine administration. Anti-hypertensive effect of nifedipine was potentiated (p<0.05) by piroxicam coadministration, but effect of nifedipine on plasma norepinephrine level was not affected. In case of rats received nifedipine and piroxicam, plasma nifedipine concentrations were higher (p<0.05) than those from rats received nifedipine alone at 2,3,4,5 and 6 hours following drug administration. The area under the plasma concentration vs. time curve was increased (p<0.05), while the elimination rate constant was decreased (p<0.01) by piroxicam coadministration. No significant differences were observed in the plasma clearance, apparent volume of distribution and elimination half-life. Thus, piroxicam not only potentiated antihypertensive effect of nifedipine, but also altered nifedipine pharmacokinetics in the rats. It is concluded that the potentiation of nifedipine antihypertensive effect might correlate with the increment of its plasma concentration by piroxicam coadministration.

  • The purpose of this study was to determine the changes of skin temperature and subjective fatigue of the thoracic vertebrae by the chiropractic adjustments. Stimulation of the sympathetic nervous system will cause the changes in the skin blood flow which can be detected by measuring the skin surface temperature. This study was to see whether chiropractic adjustments could affect the activity of the sympathetic nervous system as reflected by changes in skin temperature of the thoracic. Skin temperature and subjective fatigue on 16 subjects was measured before, within 10 sec, 10min, 20min and 30 min after a thoracic adjustment with Digital Infrared Thermal Imaging(DITI) and Visual Analogue Scale(VAS). The adjustments consisted of a straight posterior-to-anterior high-speed, low-amplitude thrust to the transverse process of T3-T10 using a reinforced hypothenar contact. The average temperature and fatigue changed when the spine was considered as an entire unit. These results illustrate that the blood flow through the soft tissue can be affected by specific adjustments to the spine. This study might be served as an useful baseline data for the changes of the circulation and fatigue after the chiropractic adjustments.

Effect of dental fear on dental use behavior and treatment need degree (치과공포가 치과이용행태 및 치료요구도에 미치는 영향)

  • This study was conducted in order to know whether DFS questionnaire which can measure dental fear is suitable for dental fear measuring method and to know effect of dental fear on dental use behavior and treatment need degree. This survey was conducted from May 1, 2007 to May 31, 2007. A total of 281 surveys eligible among all the collected surveys were used in the analysis. Collected data were analyzed by using SPSS 12.0. The obtained results were as follows: 1. Based on the reliability analysis, we concluded that the questionnaire of dental fear was reliable, and the Cronbach' s alpha coefficients of the all subscales were more than 0.6. 2. Among the dental fear of 3 factors, treatment fear cause stimulation factor was higher than the others. Most of all, following two situations(when preparation was made and when people hear dental instrument operating sound) are the highest in the treatment fear. And women are higher than men in every item. 3. In the Dental treatment need degree caused by dental fear degree, high fear group was $4.76{\pm}1.954(M{\pm}SD)$ which was higher than that of low fear group.(low fear group was $4.07{\pm}2.184(M{\pm}SD)$. In conclusion, dental fear and dental treatment need degree are related each other.

Long-Term Follow Up of Refractory Myotonia Associated with Hyperadrenocorticism in a Maltese Dog

  • A 12-year-old, female Maltese was referred with uncontrolled hyperadrenocorticism (HAC). Despite well management of HAC through treatment with trilostane and serial monitoring with ACTH stimulation tests for over three years, stiffness of the neck and limbs progressively worsened over time. Neurological test was performed, which showed no abnormalities of cranial nerves. Proprioception was delayed but the cause appeared to be due to stiffness of limb muscles. Muscle tone had increased over time and stiffness had worsened to the extent where it made walking difficult. MRI scans showed no orthopedic or spinal diseases, and pituitary microadenoma was confirmed with pituitary gland measurement of 6 × 6.4 × 4.5 mm (H × W × L). Electromyography presented random discharges with fluctuating amplitude and frequency, which were consistent with myotonic discharges. There were no improvements of myotonic signs despite treatment for HAC with trilostane. Supplementation of L-carnitine and coenzyme Q-10 to mitigate muscle stiffness, following diazepam and methocarbamol to help with muscle rigidity, failed to show any positive effect and the dog died a sudden death, 1,182 days after the initial visit.

The Influence of Illuminance Variation by a Daylight Dimming Control System on Visual Comfort Under Clear Sky Conditions (청천공 조건에서 조광제어 시스템 적용시 조도변화에 대한 시각적 만족도 분석)

  • This study examines the influence of light variation on visual responses in a small office space where a daylight dimming control system is applied. Field measurements and survey were performed in a full-scale mocked-up small office space under clear sky day conditions. Maximum fluctuation range for desktop was 133.5 lx and it just happened once for entire monitoring period. For the majority of time, the fluctuation range did not exceed 50 lx, which did not cause visual discomfort to subjects. The daylight dimming control system successfully kept required illuminance levels for an office environment when desktop illuminance by daylight ranged up to 300 lx. The most serious contributor to the sensation of glare was direct daylight through window. The dimming of electric light was not a significant contributor to visual discomfort for the subjects under clear sky day conditions. Visual comfort was significantly influenced by the sensation of glare, feeling for visual stimulation and distraction, and required illuminance level.

  • Background and purpose: Seven Emotions consist of Joy(喜), Anger(怒), Anxiety(憂), Thought(思), Sorrow(悲), Fear(恐), Fright(驚). If Seven Emotion is excessive, its extreme mental stimulation causes physical illness. There was no study of the Seven Emotion Disease in detail for now. Therefore the purpose of this study is to pigeonhole the Seven Emotion Disease. Methods: We extract the sentences about the Seven Emotion and related words in Donguibogam. We classify the sententences into Joy(喜), Anger(怒), Anxiety(憂), Thought(思), Sorrow(悲), Fear(恐), Fright(驚), Frustration, Mental Exhaustion, Character. We analysis pattern of Symptoms Derived from Seven Emotions. Results and Conclusions Seven Emotion give rise to various type of symptom. In special Anger cause more illness than other Seven Emotion.

Toll-Like Receptor-Mediated Free Radical Generation in Clonorchis sinensis Excretory-Secretory Product-Treated Cholangiocarcinoma Cells

  • Clonorchiasis, caused by direct contact with Clonorchis sinensis worms and their excretory-secretory products (ESPs), is associated with chronic inflammation, malignant changes in bile ducts, and even cholangiocarcinogenesis. Our previous report revealed that intracellular free radicals enzymatically generated by C. sinensis ESPs cause NF-${\kappa}B$-mediated inflammation in human cholangiocarcinoma cells (HuCCT1). Therefore, the present study was conducted to examine the role of upstream Toll-like receptors (TLRs) on the initial host innate immune responses to infection. We found that treatment of HuCCT1 cells with native ESPs induced changes in TLR mRNA levels in a time-dependent manner, concomitant with the generation of free radicals. ESP-mediated free radical generation was markedly attenuated by preincubation of the cells with TLR1-4-neutralizing antibodies, indicating that at least TLR1 through 4 participate in stimulation of the host innate immune responses. These findings indicate that free radicals triggered by ESPs are critically involved in TLR signal transduction. Continuous signaling by this pathway may function in initiating C. sinensis infection-associated inflammation cascades, a detrimental event leading to progression to more severe hepatobiliary diseases.

Clostridium difficile Toxin A Upregulates Bak Expression through PGE2 Pathway in Human Colonocytes

  • Clostridium difficile toxin A is known to cause colonic epithelial cell apoptosis, which is considered the main causative event that triggers inflammatory responses in the colon, reflecting the concept that the essential role of epithelial cells in the colon is to form a physical barrier in the gut. We previously showed that toxin A-induced colonocyte apoptosis and subsequent inflammation were dependent on prostaglandin E2 ($PGE_2$) produced in response to toxin A stimulation. However, the molecular mechanism by which $PGE_2$ mediates cell apoptosis in toxin A-exposed colonocytes has remained unclear. Here, we sought to identify the signaling pathway involved in toxin A-induced, $PGE_2$-mediated colonocyte apoptosis. In non-transformed NCM460 human colonocytes, toxin A exposure strongly upregulated expression of Bak, which is known to form mitochondrial outer membrane pores, resulting in apoptosis. RT-PCR analyses revealed that this increase in Bak expression was attributable to toxin A-induced transcriptional upregulation. We also found that toxin A upregulation of Bak expression was dependent on $PGE_2$ production, and further showed that this effect was recapitulated by an Prostaglandin E2(PGE2) receptor-1 receptor agonist, but not by agonists of other EP receptors. Collectively, these results suggest that toxin A-induced cell apoptosis involves $PGE_2$-upregulation of Bak through the EP1 receptor.
