Acceptability on the Sweetness of Stevioside as a Natural Sweetner (천연감미료(天然甘味料) Stevioside의 감미(甘味)에 관한 수응력(受應力) 시험(試驗))

  • The acceptability of purified stevioside as a sweetner was carried out. The content of stelvioside was $7.30{\sim}17.8%$ of the dry leaf of stevia harvested in Korea and the purified stevioside showed $100{\sim}150$ folds sweeter than sucrose. Improvement of acceptability of the purified stevioside by mixing with other sweetners was as the following order: sucrose>glucose>fructose= invert sugar >saccharin-Na. When 2 kinds of sweetner were mixed stevioside the improvement was the same as one sweetner was mixed with stevioside. The mixture of the stevioside and sodium chloride did not give any off-taste.

  • Stevioside 는 남미 파라과이 원산의 국화와 식물인 Ste-via rebaudiana Bertoni 유래의 천연대체감미료서 식품, 의약품, 주류 산업에 널리 활용되고 있다. 이와 같은 stevio-side 의 구조와 특성, 제조방법, 그리고 사용현황 등을 기술하였으며, 당전이 stevioside의 종류 제조방법을 정리하였다. 본 연구실에서 개발한 생전분을 당공여체로 히용하는 분쇄마찰매체 효소반응계와 팽윤 extrusion 전분을 당공여체로 이용하는 불균일상 효소반응계에서의 stevioside 당전이반응의 특성과 산업적 활용 가능성을 검토하였다. 상기 두 종류의 불균일상 효소반응계는 높은 수율, 반응속도, 낮은 maltooligo 당의 축적, 용이한 당전이 stevioside 의 분리정제 등 기존의 액화 전분을 당공여체로 이용하는 반응계에 비하여 많은 장점이 예상되는 산업적 활용 가능성이 높고 고효율 반응계임을 알 수 있었다. 끝으로 stevioside 감미료의 장래 전망에 관하여 예측하였다.

  • The purpose of this study was to search for a mixture of stevioside and sucrose, the sweetness of which in soft drink such as cocoa and ginger tea were almost equal the sweetness of commomly used 7% sucrose in the drinks. Results obtained by the present experiment were as fellows: 1) Mixture of 50% stevioside 40mg (0.04%) (A) and 2.0% sucrose, 95% stevioside 40mg (0.04%) (B) and 1.0% sucrose, and 50% and 95% stevioside 60mg (0.06%) (A,B) respectively, the sweetness of which in ginger tea corresponded to that of reference in the drink. 2) Mixture of 50% stevioside 40mg (0.04%) (A) and 2.0% sucrose, 95% stevioside 20mg (0.02%) (B) and 2.0% sucrose, and 50% stevioside 40-60mg (0.04-0.06%) (A) and 95% stevioside 40-60mg (0.04-0.05%) (B), the sweetness of which in cocoa is equal to that of standard sucrose in the drink. 3) The mixture contains low energe, and also the after-taste of stevioside was not detected in the above drinks.

  • Some sensory properties of synthetic sweeteners are limiting factors for use in low calorie foods of soft drinks. By combining synthetic sweeteners(Neohesperidin dihydrochacone(NHDC), stevioside) with sucrose, sorbitol or xylitol, these limitations can be overcome. Using trained taste panelists and magnitude estimation methods, synergistic effects were investigated in binary sweetener mixtures, The results showed that synergism, as much as 28% to 69%,was noted at all concentration in xylitol-stevioside, NHDC-stevioside mixtures. Synergistic effects were found only at high concentration levels in mixtures of sucrose-NHDC, sucrose-stevioside, and sorbitol-stevioside, ranging from 11% to 22%.By taking advantage of synergistic effects, ginseng tea and orange flavored beverages, sweetened with either xylitol-stevioside of NHDC-stevioside, were prepared and the sensory quality was compared with that of sucrose containing beverages. It was found that sensory characteristics were judged to be very similar for all formulations. The result suggests the possibility of using of using these sweetener mixtures as sugar substitutes in ginseng tea and orange drink.

Reaction Mechanism of Transglycosylation of Stevioside in the Attrition Coupled Reaction System Using Raw Starch as a Glycosyl Donor (생전분을 당공여체로 한 분쇄마찰매체 함유 효소반응계에서의 Stevioside의 당전이 반응 기작)

  • Transglycosylation of stevioside in the attrition coupled heterogeneous reaction system using raw starch as a glycosyl donor has significant advantages over conventional reaction systems using liquefied starch as a donor. The transglycosylation of stevioside under the presence of organic solvent showed that transglycosylation reaction occurs via two steps ; initially from raw starch to cyclodextrin(CD), and then followed by transglycosylation of produced CD. Comparison of the transglycosylation efficiency of c$\alpha $-, $\beta $, $\gamma $-CDs indicated that $\alpha $-, $\beta $-CD are mainly utilized as a glycosyl donor for following reaction. The reaction mechanism of transglycosylation between stevioside and CD proceeded according to random sequential bireactant mechanism. The equilibrium constant of transglycosylation reaction of cyclodextrin glucanotransferase wase also evaluated. The structure of transglycosylated stevioside was confirmed by TLC, and it was found that glycosyl group(G$_{1}, $ ~ G$_{4}$-glycosidic bond.

  • Kinetic equations for transglycosylation of stevioside in the attrition coupled reaction system using raw starch as a glycosyl donor were derived considering that the reaction was carried out through two steps; production of cyclodextrin(CD) from raw starch in the attrition coupled reaction system and then transglycosylation of glycosyl residues to stevioside from produced CD. Kinetic constants of derived equation were evaluated. The simulation result showed that the derived kinetic equations could predict the experimental data reasonably well and that can be utilized for optimization and scale-up of transglycosylation reactor and process developments.

  • Stevia rebaudiana Bertoni M. can be cultivated in Korea as a new sweetness resource. It seems possible that stevioside the sweet substance extracted from the leaves of Stevia rebaudiana Bertoni M. can be used instead of saccharin. In order to obtain the fundamental data due to natural sweetening materials used of as additives to food stuffs such as coffee and tea etc. It was investigated to reduce the calories from coffee and tea without reducing the conventional sweetness, drinks sweetened with mixtures of stevioside and sucrose were prepared. The results obtained are as follows: 1) Mixture of 50% stevioside 40 mg (0.04%) (A) and 1.5% sucrose, 95% stevioside 20 mg (0.02%) (B) and 2.0% sucrose, and 50% stevioside 60 mg (0.06%) (A) and 95% stevioside 40 mg (0. 04%) (B), the sweetness of which in coffee corresponded to that of 7% sucrose in the drink, was prepared. 2) Mixture of 50% stevioside 60 mg (0.06%) (A) and 1.5, 2.0, 2.5% sucrose, 95% stevioside 40 mg (0.04%) (B) and 1.5, 2.0% sucrose, and 50% 80 mg (0.08%), 95% 60 mg (0.06%) (A,B), respectively the sweetness of which in tea corresponded to that of 7% sucrose in the above drink, was prepared. 3) Sucrose saving of 21.42-28.57% compared to equally sweet sucrose-sweetened coffee and tea could be achieved without deterioration of other taste qualities.

  • The standard operation procedure of mouse lymphoma L5178Y $tk^{+/-}-3.7.2C$ gene mutation assay (MOLY) has been regarded as a sensitive in vitro mammalian cell gene mutation assay that is capable of detecting clastogens as well as mutagens. Using MOLY, one of natural sweetner, stevioside (5mg/ml) and its aglycon, steviol ($340{\;}\mu\textrm{g}/ml$) were evaluated the mutagenicity. Stevioside and steviol did not induce mutagenicity in MOLY. On the other hand, stevioside (250mg/kg, B.W.) and steviol (200mg/kg, B.W.) were also evaluated their ability to induce micronuclei in regenerating hepatocytes and bone marrow cells of ddY mice. From these results, stevioside and steviol did not induce any mutagenic effect both MOLY and in vivo micronucleus test.

Optimization of Transglucosylation Reaction of Stevioside (스테비오시드 당전이 반응의 최적화)

  • Stevioside was transglucosylated to improve its sweetness. Eighteen conditions were tested using the tables of orthogonal arrays of $L_{18}\;(2^{1}{\times}3^{7})$. Statistical analysis showed that the transglucosylation rate was significantly affected by temperature, the ratio of co-substrate (maltodextrin) to stevioside, pH, DE of maltodextrin and concentration of stevioside, in their order. Optimum conditions selected for temperature, ratio of co-substrate to stevioside, pH and DE of maltodextrin were $80^{\circ}C$, 1.0, 6.0 and 15, respectively. Glycosyl-stevioside with 68% yield of transglucosylation was produced at the optimum condition and found to have better quality in sweetness than stevioside and rebaudioside A.

  • 김치의 단맛을 내기 위하여 일반 제조에 이용괴고 있는 sucrose 대신 stevioside나 sorbitoal과 같이 당의 종류를 달리하여 제조한 김치의 이화학적 성분. 미생물의 변화 및 관능적 특서에 미치는 영향을 조사하였다 pH와 적정산도는 stevioside와 sorbitol이 첨가된 처리구가 sucrose 가 첨가된 처리구보다 산의 생성 억제효과가 있는 것으로 나타났다 총균수의 경우 전반적으로 당의 종류와 저장온도와는 관계없이 미생물의 생육 억제효과가 없는 것으로 나타났으나 젖산균의 경우 1$0^{\circ}C$에서는 균의 생육 억제효과는 없는 반면 5$^{\circ}C$에 저장한 김치의 경우 stevioside와 sorbitol이 첨가된 김치는 젖산균의 생육에 억제효과가 있는 것으로 나타났다 관능검사를 실시한 결과 stevioside 첨가된 김치가 sucrose 첨가구에 비하여 기호도에서는 전반적으로 우수한 것으로 나타났다.
