Conservation treatment on the Kim-gu's relics-Baekbumilgi and bood stained clothes stained with blood (백범 김구 선생 유품의 과학적 보존처리)

  • We preformed many treatments to preserve Kim-gu's relics (Baekbum-ilgi and the clothes stained with blood). The first we performed re-paper mount of binding area of baekbumilgi, and fumigated all relics to sterilize microorganism. Then all relics were sealed with biaxially oriented polyvinyalcohol film(BO-PVA film) and $N_2$ gas filled up in the BO-PVA film bags. We investigated the paper condition of Baekbum-ilgi, its brightnees was 25-35%, water content was 9.4%, and acidity was pH 4.3. These results indicated that the paper was very weak. On the other hand, We analyzed his blood type and genetic type with the solidified blood of the clothes. In result, his blood type was AB and were classified 6 genetic types such that HUMTH01 was 7-9 type, HumTPOX was 9-11 type, HumCSF1PO was 12-13 type, HumF13A01 was 4-4type, HumFESFPS was 12-12, and HumvWA was 18-18 type.

Hygiene of fish feed I. Effect of gamma radiation on the pathogenic bacteria of fish (어류양식용 사료의 위생화 I. 어류유래 병원성 세균에 대한 방사선 조사의 효과)

  • The gamma-radiation sensitivity of three kinds of pathogenic bacteria to fish was investigated. $D_{10}$ values (irradiation dose required to inactivate 90% of the population) of Edwardsiella tarda, Vibrio anguillarum and Streptococcus faecalis were 0.08, 0.10 and 0.44 kGy, respectively. The inactivation factors were 4.50 - 24.30 and 6.75 - 36.45 at radiation doses of 2 and 3 kGy. Raw moist pellets were inoculated with Edwardsiella tarda, Vibrio anguillarum or Streptococcus faecalis. Inoculated feed samples were packaged in air and irradiated at 5 kGy. This dose was effective in controlling the inoculated and general bacteria in fish feed. We consider gamma-radiation to be an effective method to sterilize pathogenic bacteria in fish feed.

A Study on Inactivation of Pathogenic Bacteria for Nutrient Solution Recycling Using Advanced Oxidation Processes

  • This study was conducted to test the applicability of hydroxy radical reactor system, which applied advanced oxidation processes, to sterilize pathogenic bacteria for nutrient solution recycling in closed hydroponics. Removal efficiency was tested on 25 L of nutrient solution maxed with 10 mL culture solution of bacteria, E. coli, and R. solanacearum in a pilot tank. The testing conditions included various levels of hydroxy radicals resulting from air flow rates of 40, 80, and $120L\;min^{-1}$, and 12 hours processing time. The removal of bacteria, E. coli, and R. solanacearum by hydroxy radical in nutrient solution was significantly increased with an increase in the flow rate of the air from $40L\;min^{-1}$ to $120L\;min^{-1}$. The optimum removal efficiency was achieved at an air flow rate of $120L\;min^{-1}$ for 2 hours treatment. There were no significant differences in removal efficiency among bacteria, E. coli, and R. solanacearum for tested level and time of hydroxy radical. These results verified the efficiency of hydroxy radical in removing the pathogenic bacteria and the applicability of hydroxy radical reactor system in the field.

Effects of Sterilization for Quality Control and Content of baicalin in Scutellariae Radix (멸균법에 따른 황금의 미생물학적 품질보증과 바이칼린 함량에 미치는 영향)

  • The purposes of this study was to develop the best one among the methods using dry heat, gamma irradiation and alcohol gas, to sterilize Scutellariae Radix selected based on high frequency of circulation between merchants and high susceptibility to microbial contamination, through comparing these methods in terms of the followings; ⅰ) the efficacy of sterilization in comparison with cost, and ⅱ)the alteration of chemical components of these herbal medicines. The results of this study will be taken advantage of establishment of the maximum limit of microorganism contaminated in herbal medicines. From the results of this study we conclude the followings: ⅰ) The sterilizing method using dry heat may be inappropriate, because it seriously changed their color and morphology which is an essential criterion to estimate a measure of quality between merchants and consumers, although it effectively got rid of contaminated microorganism and did not alter major components, baicalin; ⅱ) The sterilizing method using gamma irradiation may be appropriate, because it showed a strong sterilizing effect, and no alteration of chemical components, color and morphology; ⅲ)The sterilizing method using alcohol gas may also be appropriate, because it had a similar effects as in the case of gamma irradiation. Collectively, we suggest that appropriate sterilizing methods to guarantee the microbial quality of this herbal medicine may be those using gamma irradiation and alcohol gas.

Effects of Sterilizing Methods on the Content of Index .Constituents of Herbal Medicines (품질 보증을 위한 멸균법이 수종 생약의 지표성분 함량에 미치는 영향)

  • The purpose of this study was to develop the best methods to sterilize herbal medicines which is frequently used and known to have high susceptibility to microbial contamination. We used dry heat, gamma irradiation and alcohol gas treatment for sterilization, and evaluated these methods in terms of the followings; i) the efficacy of sterilization, and ii) the chemical alteration of index constituents of herbal medicines. Treatment with dry heat effectively eliminated the contaminated microorganisms, and did not significantly alter the content of berberine chloride, paeoniflorin and amygdalin in Phellodendron Bark, Peony Root and Apricot Kernel, respectively: However it seriously changed the color and morphology which are essential criteria to estimate a measure of quality of herbal medicines. Treatment with gamma irradiation showed a strong sterilizing effect, and no alteration of the content of index constituents, color and morphology: Alcohol gas treatment resulted in similar effects as those in gamma irradiation. Collectively; these results suggest that treatment with gamma irradiation or alcohol gas may be useful methods for sterilizing herbal medicines without a decrease in their microbial quality.

Characterization of Phage-Resistant Strains Derived from Pseudomonas tolaasii 6264, which Causes Brown Blotch Disease

  • Pseudomonas tolaasii 6264 is a representative strain that causes bacterial blotch disease on the cultivated oyster mushroom, Pleurotus ostreatus. Bacteriophages are able to sterilize the pathogenic P. tolaasii strains, and therefore, they can be applied in creating disease-free mushroom cultivation farms, through a method known as "phage therapy". For successful phage therapy, the characterization of phage-resistant strains is necessary, since they are frequently induced from the original pathogenic bacteria in the presence of phages. When 10 different phages were incubated with P. tolaasii 6264, their corresponding phage-resistant strains were obtained. In this study, changes in pathogenic, genetic, and biochemical characteristics as well as the acquired phage resistance of these strains were investigated. In the phylogenetic analyses, all phage-resistant strains were identical to the original parent strain based on the sequence comparison of 16S rRNA genes. When various phage-resistant strains were examined by three different methods, pitting test, white line test, and hemolytic activity, they were divided into three groups: strains showing all positive results in three tests, two positive in the first two tests, and all negative. Nevertheless, all phage-resistant strains showed that their pathogenic activities were reduced or completely lost.

  • This is made of domestic and foreign coal activated carbon of five species, physicochemical adsorption efficient about sterilize products and micro harmful substances and is a result of the economic evaluation. The most well-developed micropores bed volume 123,409 of AC-1 activate carbon appeared to be the best next AC-2, AC-3, AC-4, AC-5 followed by activated carbon was investigated. PFOA and PFOS in the BV 96,000 when evaluating foreign types of adsorption activated carbon adsorption capacity was greater when more than PFOA, PFOS showed that the adsorption well. The economic evaluation of activated carbon usage in chloroform (CUR) was most excellent as a AC-1 4.3 g/day, the next AC-2, AC-3, AC-4, AC-5 there are two types of foreign economic order appears to have appeared, but the current domestic market when applying the price AC-1, AC-3, AC-2, AC-4, AC-5 order was investigated.

Status of Health Hygiene of Some Beauty Shop in Taegu City (대구시내 미용실의 보건위생 실태)

  • The objective of this study was to improve sanitary conditions at the beauty shops by informing the owners and employees to be aware of the public health. The data were obtained by the Physical investigation (August 1 to 30, 2001) of the sanitary Problems at 112 beauty shops in Daegu Metrpolitan area and by the surveys from the employees and customers. By analysis these data with SAS/PC+(ver. 6.12) and $x^2-test$, we have obtained the following results. 1. The numbers of the beauty shop workers (67.86% of the shops) were two including the owner, and only 18.75% of the workers had the regular public health training session. Even at the training sessions, no public health specialist was available as an instructor. 2. Sanitary conditions of the floor (92.76%), washer (88.93%) and rest room (74.11%) at the beauty shops were satisfactory. However, only 3.57% of the beauty shop used the vacuum cleaner that would be far more effective than the bloom to remove hairs from the floor. 3. About 91.07% (102 shops) did not or poorly sterilize equipments, and 91.76% (103 shops) did not store the sterilized or non-sterilized equipments separately. The shops that had the ultraviolet sterilizer that is required by the public health regulation were 53.57% (60 shops), and only 2.67% (3shops) actually used it. 4. About 82.14% (72 shops) used Oxyclean and detergent for washing the fabrics such as towels and gowns. About 11.61% (13 shops) used only detergent and 3.57% (4 shops) used the boiling sterilization method. No shop used the customer gown once per customer, and only 2.68% (3 shops) washed the used gown daily.

Disinfection effect of corona discharged plasma water on fish pathogens (코로나 방전 플라즈마 처리수에 의한 어류 병원체 소독 효과)

  • Fish culture is constantly threatened by various infectious diseases which are largely transmitted by water. Plasma technology is being used to sterilize polluted water in many industries. In this study, two bacterial pathogens Aeromonas salmonicida and Streptococcus iniae, and a virus (viral hemorrhagic septicemia virus, VHSV) were subjected to plasma water that was produced by a corona discharge system. Growth of A. salmonicida was greatly inhibited from 105.61 CFU/ml in positive control to 103.51 CFU/ml in treated group by only 60 sec contact with plasma water. Similarly, S. iniae was inhibited from 105.85 CFU/ml to 103.40 CFU/ml. VHSV titer also decreased from 104.1 TCID50/ml to 101.45 TCID50/ml by the same treatment. Activation of water by the plasma was confirmed by the existence of ozone in the plasma water. These results suggest that plasma water could efficiently disinfect fish pathogens, possibly by the action of reactive oxygen species contained in the plasma water.

Antibacterial Effects on Bacillus Stearothermophilus by Adding Natural Grapefruit Seed Extracts in Soymilk (두유에서 자몽씨 추출물의 Bacillus 균에 대한 항균효과)

  • We evaluated grapefruit seed extract for antibacterial activity at varying time intervals and concentration levels against heat tolerant and spore-forming Bacillus stearothermophilus, mesophilic Bacillus subtilis, and food poisoning Staphylococcus aureus. Grapefruit seed extract showed growth inhibitory activity against B. stearothemophilus and B. subtilis, and S. aureus at the level of 0.01 tp 0.03% in nutrient broth. However, when applied to soymilk in a market, grapefruit seed extract at the level above 0.2% effectively inhibited the growth of B. stearothermophilus, However, it failed to completely sterilize the test organisms. On the other hand, B. subtilis and S. aureus were completely sterilized at the level of 0.2% within 48 hrs and 72 h, respectively. The higher concentration of grapefruit seed extracts showed more effective antibacterial activities against the test organisms, but caused deteriorated organoleptic quality and emulsion stability. We could conclude, by applying grapefruit seed extract (0.015%) with thermal treatment (10 min at $121^{\circ}C$) that the microbial stability of commercial soymilk was enhanced greatly.
