품질 보증을 위한 멸균법이 수종 생약의 지표성분 함량에 미치는 영향 (Effects of Sterilizing Methods on the Content of Index .Constituents of Herbal Medicines)

  • The purpose of this study was to develop the best methods to sterilize herbal medicines which is frequently used and known to have high susceptibility to microbial contamination. We used dry heat, gamma irradiation and alcohol gas treatment for sterilization, and evaluated these methods in terms of the followings; i) the efficacy of sterilization, and ii) the chemical alteration of index constituents of herbal medicines. Treatment with dry heat effectively eliminated the contaminated microorganisms, and did not significantly alter the content of berberine chloride, paeoniflorin and amygdalin in Phellodendron Bark, Peony Root and Apricot Kernel, respectively: However it seriously changed the color and morphology which are essential criteria to estimate a measure of quality of herbal medicines. Treatment with gamma irradiation showed a strong sterilizing effect, and no alteration of the content of index constituents, color and morphology: Alcohol gas treatment resulted in similar effects as those in gamma irradiation. Collectively; these results suggest that treatment with gamma irradiation or alcohol gas may be useful methods for sterilizing herbal medicines without a decrease in their microbial quality.

Treatment of Microencapsulated ${\beta}$- Galactosidase with Ozone : Effect on Enzyme and Microorganism

  • The present study was designed to examine the effect of ozone treatment in microencapsulated ${\beta}$-galactosidase on inactivation of the enzyme and sterilization of microorganism. The efficiency was the highest as 78.4% when the ratio of polyglycerol monostearate (PGMS) was 15:1. Activities of lactase remaining outside the capsule were affected by ozone treatment. With the increase of ozone concentration and duration of ozone treatment, the activity reduced significantly. In sensory aspect, with 2% microcapsule addition, no significant difference in sweetness was found compared with a market milk during 12 d storage. Above result indicated that the additional washing process of lactase was not necessary to inactivate the residual enzyme. In a subsequent study, the vegetative cells of microorganisms were completely killed with 10 ppm for 10 min treatment by ozone. The present study provides evidence that ozone treatment can be used as an inactivation and a sterilization process. In addition, these results suggest that acceptable milk products containing lactase microcapsules made by PGMS can be prepared with ozone treatment.

  • Comparison of ethylene oxide(E.O) fumigation and gamma irradiation on the sterilization, microbiological, physicochemcial and sensory quality of dried fishery products (shucked shellfish and mussel powder) were investigated. The viable cell count of mesophilic total bacterial was reduced by over 2 to 3 log cycles with irradiation of 3 to 5 kGy, and those samples were completely sterilized by irradiation dose of 7 to 10 kGy. In view of reduction of microorganisms, D10 value of each samples were ranged from 1. 36-1. 46 kGy. fungi, mesophilic spores, acid tolerant bacteria and coliforms were sterilized at 5 to 7 kGy irradiation but E.O. fumigation proved insulfficient to eliminate the mesophilic total bacteria and fungi. In the physicochemical and sensory quality of samples, such as the amino acid, TBA value, TMA-N, color difference and overall acceptability irradiated sample with optimum dose was similar to those of the nonirradiated sample, while E.O. fumigated sample was remarkably deteriorated in the physicochemical and sensory quality of samples.

Nonthermal Sterilization of Animal-based Foods by Intense Pulsed Light Treatment

  • The consumption of meat has been increasing, leading to a dynamic meat and meat processing industry. To maintain the quality and safety of meat products, various technologies have been explored, including intense pulsed light (IPL) technology. Several factors affect the inactivation of microorganisms by IPL treatment, including light intensity (fluence), treatment duration, pulse frequency, and the distance between the lamp and the samples. Meat products have been studied for IPL treatment, resulting in microbial reductions of approximately 0.4-2.4 Log. There are also impacts on color, sensory attributes, and physico-chemical quality, depending on treatment conditions. Processed meat products like sausages and ham have shown microbial reductions of around 0.1-4 Log with IPL treatment. IPL treatment has minimal impact on color and lipid oxidation in these products. Egg products and dairy items can also benefit from IPL treatment, achieving microbial reductions of around 1-7.8 Log. The effect on product quality varies depending on the treatment conditions. IPL technology has shown promise in enhancing the safety and quality of various food products, including meat, processed meat, egg products, and dairy items. However, the research results on animal-based food are not diverse and fragmentary, this study discusses the future research direction and industrial application through a review of these researches.

  • The purpose of this study was to develop the best methods to sterilize Moutan Root Bark which is frequently used as a herbal medicine, and known to have high susceptibility to m icrobial contamination. We used dry heat, gamma irradiation and alcohol gas treatment for sterilization, and evaluated these methods in terms of the followings; I) the efficacy of sterilization, and ii) the chemical alteration of a major component of the harbal medicines. Treatment with dry heat effectively got rid of contaminated microorganisms, and did not significantly alter the content of paeonol. However it seriously changed the color and morphology which are an essential criterion to estimate a measure of quality of medicinal herbs. Treatment with gamma irradiation showed a strong sterilizing effect, and no alteration of the content of paeonol, color and morphology. Alcohol gas treatment resulted in similar effects as those in gamma irradiation. Collectively, these results suggest that appropriate sterilizing methods to guarantee the microbial quality of this herbal medicine may be those using gamma irradiation or alcohol gas.

음이온 처리된 몇 가지 새싹채소의 생장과 세균 수 (Growth of and Bacterial Counts on Several Edible Sprouts Exposed to Spray Ionization)

  • 새싹은 영양분이 풍부한 식품이다. 하지만 생산 과정에서 종피에 있는 미생물에 의한 오염 가능성이 존재한다. 이 실험의 목적은 음이온 처리가 새싹의 생장과 살균에 미치는 효과를 구명하는 것이다. 음이온 처리는 4종류의 새싹의 생장과 살균효과에 긍정적인 효과를 보였다. 대조구에 비해 음이온 처리한 적양배추와 케일 새싹의 배축길이는 약 1.26배 증가하였으며, 상추, 적양배추, 케일 유근의 길이는 1.4~1.6배 증가하였다. 모든 새싹의 생체중은 음이온 처리했을 때 대조구에 비해 16.0~38.5% 유의적으로 증가하였다. 유근의 활력 또한 음이온 처리가 대조구에 비해 유의적으로 높은 수치를 보였다. 음이온 처리된 상추, 적양배추, 다채 새싹의 일반 세균 수는 대조구에 비해 각각 41%, 66%, 19% 감소하였으며, 배수되는 물의 세균 수 또한 감소되었다. 결국 음이온처리는 새싹의 생장을 향상시켰으며, 동시에 살균하는데도 효과적이었다.

Analysis of Microbial Contamination and Antibacterial Effect Associated with Toothbrushes

  • The purpose of this study was to propose a method for the effective management of toothbrush contamination. Toothbrush microbial contamination was analyzed according to the duration of toothbrush use, frequency of toothbrush use per day, and toothbrush storage location. We also analyzed the microbial reduction effect of vinegar, antimicrobial mouth rinse, bamboo salt, and baking soda, which are sterilization materials that can be easily used every day. We collected 45 toothbrushes from university dormitories from May to June 2018. To determine the degree of microbiological contamination with general bacteria, coliform bacteria, and Staphylococcus aureus, bristle samples were cultured at $36^{\circ}C$ for 24 hours using 3M$^{TM}$ Petrifilm plates and then measured based on Petrifilm evaluation criteria. Toothbrush microorganisms were analyzed according to the duration of use, frequency of use per day, storage location, and effect of each sterilization material. General bacteria, coliforms, and S. aureus contamination increased with frequency and duration of use (p<0.05). In particular, S. aureus showed a statistically significant increase to 36.15 CFU/ml after 1 month, 504.23 CFU/ml after 2 months, and 2,386.67 CFU/ml after 3 months (p<0.05). We found that 1% vinegar was the most effective substance for reducing general bacteria, coliforms, and S. aureus. In addition, 1% antimicrobial mouth rinse solution applied for 5 minutes was the most effective in reducing S. aureus. It is crucial to recognize the importance of toothbrush care and store toothbrushes in a dry place and replace them periodically. We recommend use of vinegar and antimicrobial mouth rinse solution to disinfect toothbrushes. These should be applied as a 1% solution for at least 1 minute. Proper care of toothbrushes is important in maintaining oral health as well as overall health. Instructions on toothbrush care should be given when teaching children or adults how to brush teeth.

선박평형수 처리장치의 조합에 따른 살균효과에 관한 연구 (A Study on the Sterilization Effect of Ballast Water according to the Combination of Types of Treatment Apparatus)

  • 2차 오염이 없는 물리적 선박 평형수 처리 장치의 조합별 비교를 통하여 비교적 우수한 조합을 밝혀내는 것이 본 연구의 목적이다. 2008년 IMO에서는 선박 평형수 문제를 제제하기 위해 배출 선박 평형수 내 미생물의 농도를 일정 농도로 규제하게 되었다. IMO 규제 농도를 맞추기 위한 멸균 방법으로는 약품을 이용한 화학적 처리, 전기분해를 통한 차아염소산 처리, UV나 오존 및 플라즈마를 이용하는 방법, membrane을 이용하는 방법 등이 있다. 하지만 어느 방법이나 부분적으로 2차오염이나 선체부식 및 살균효율 등의 단점을 가지고 있는 실정이다. 아울러 이런 단점들을 피해가면서 조류와 미생물을 효율적으로 멸균 시킬 수 있는 방법은 현재까지 미비한 실정이다. 본 연구에서 적용된 처리장치는 전단응력을 이용하여 2차 오염을 발생시키지 않으면서 조류와 미생물을 살균시킬 수 있는 물리적 살균처리장치이다. 본 연구에서는 이 장치의 다양한 type별 조합으로 실험하여 살균 가능한 최저 회전속도에 따른 최고 유량 등을 규명하여 상업적 처리 장치의 설계를 위한 기초자료로 삼고자하였다.

일회용 주방용품의 미생물 오염도평가 및 방사선 살균 (Microbiological Contamination Level and Radiation Sterilization in Disposable Kitchen Utensil)

  • 일회용 주방용품의 미생물학적 안전성 확보를 위한 연구의 일환으로 17개 제품군, 총 51종의 일회용 주방용품에 대한 미생물 오염도 평가와 함께 오염된 미생물을 효과적으로 살균할 수 있는 방안으로 감마선 살균 방법을 적용하였다. 일반호기성세균, 대장균군, 곰팡이 Salmonella의 오염도 평가 결과, 대장균군과 Salmonella는 거의 검출되지 않았으나 일반호기성 세균은 전체 시료의 약$50\%$에서 $10^1{\sim}10^2\;CFU/100\;cm^2$ 수준으로 검출되어 일회용 주방용품의 위생화가 필요함을 확인하였다. 일회용 주방용품에 오염된 미생물의 감마선 살균 결과 3 kGy의 조사선량에서 미생물이 검출되지 않았다.

자외선 살균이 청징 탁주의 저장 중 품질에 미치는 효과 (Effect of UV Sterilization on Quality of Centrifuged Takju during Storage)

  • 총 110 가닥의 테플론 튜브와 9개의 자외선(UV)램프(총 1395 W)로 구성된 허니컴 방식의 UV살균기를 제작하고 원심분리를 통해 생탁주의 고형분을 제거시킨 청징탁주(CT)를 순환시켜 살균하였다. 미생물살균과 관능적 품질 등을 고려한 전반적인 품질평가 결과, 최적의 살균조건은 2 L/min의 유속에서 1.5분간 살균하는 것이 최적의 조건으로 확인되었다. 이러한 살균조건에서 약 일반세균수와 진균수를 5~6 log cycle 감소시킬 수 있었다. 자외선으로 살균된 청징탁주는 $30^{\circ}C$에서 8일 동안 저장하며 pH, 아미노태질소 함량, 산도, 환원당 함량, 일반세균수 및 진균수 등의 품질 평가를 수행하였다. 자외선으로 살균된 청징탁주의 진균수는 저장 4일 후에 이르러서야 $10^8\;CFU/mL$에 도달하였고 일반세균수는 6일 후에 이르러서야 $10^8\;CFU/mL$에 도달하여 생육이 억제되었음을 확인할 수 있었다. 또한 환원당 함량을 제외하고는 자외선으로 살균된 청징탁주의 pH, 아미노태질소 함량, 산도는 저장 중에 변화를 나타내지 아니하였다. 또한 저장 기간 동안 청징탁주와 자외선으로 살균된 청징탁주 간에는 L, a 및 b 값 간에 차이를 보이지 않아 색도의 변화에는 자외선 살균이 영향을 미치지 않은 것으로 확인되었다.
