Characterization of dihydroflavonol 4-reductase cDNA in tea [Camellia sinensis (L.) O. Kuntze]

  • Tea leaves are major source of catechins—antioxidant flavonoids. Dihydroflavonol 4-reductase (DFR, EC 1.1.1.219) is one of the important enzymes that catalyzes the reduction of dihydroflavonols to leucoanthocyanins, a key ''late'' step in the biosynthesis of catechins. This manuscript reports characterization of DFR from tea (CsDFR) that comprised 1,413 bp full-length cDNA with ORF of 1,044 bp (115-1,158) and encoding a protein of 347 amino acids. Sequence comparison of CsDFR with earlier reported DFR sequences in a database indicated conservation of 69-87% among amino acid residues. In silico analysis revealed CsDFR to be a membrane-localized protein with a domain (between 16 and 218 amino acids) resembling the NAD-dependent epimerase/dehydratase family. The theoretical molecular weight and isoelectric point of the deduced amino sequence of CsDFR were 38.67 kDa and 6.22, respectively. Upon expression of CsDFR in E. coli, recombinant protein was found to be functional and showed specific activity of 42.85 nmol $min^{-1}$ mg $protein^{-1}$. Expression of CsDFR was maximum in younger rather than older leaves. Expression was down-regulated in response to drought stress and abscisic acid, unaffected by gibberellic acid treatment, but up-regulated in response to wounding, with concomitant modulation of catechins content. This is the first report of functionality of recombinant CsDFR and its expression in tea.

  • In order to move the body of a walking robot translationally, and step over the obstacles, the walking robot must have at least 3 degrees of freedom for each leg. Therefore each leg of the general walking robots can be composed of 3-link system with 3 revolute joints. In this paper, the colsed form of inverse kinimatic solutions is shown for this general 3R linkage. Moreover, in order to have efficient walking volume in rough terrain, the workspace of each log is obtained considering the twist angles and the offsets in D-H parameters. When we design a walking robot, the information of the walking volume is needed for planning desired trajectories of the feet effectively. Appropriate knowledge of the walking volume can also be used to maximize linear or angular velocity of minimize power of stress. However, since it is impossible to obrain the information of walking volume in 3-D space directly from the kinematic equations, the walking volume can be searched through the edge detection algorithm using the triangle tracer with closed from inverse kinematic solutions. In this study, we present the closed form inverse kinematic solutions for 3R linkage model, and the walking volume of 6 legged walking robot which is modeled after the darking bettle, Eleodes obscura sulcipennis, through the method of edge detection for an arbitrary 2 dimensional shape using triangle tracer.

  • Micro-defects induced by design and production failure or working environments are known as the cause of SCC(Stress Corrosion Cracking) in aged structures. Therefore, the evaluation of structural integrity based on micro-cracks is required not only a manufacturing step but also in-service term. So we introduce a new nondestructive inspection method using the magneto-optical film to detect micro-cracks. The method has some advantage such as high testing speed, real time data acquistion and the possibility of remote sensing by using of a magneto-optical film that takes advantage of the change of magnetic domains and domain walls. This paper introduces the concept of the new nondestructive inspection method using the magneto-optical film, also proves the possibility of this method as a remote testing system under oscillating load considering application on real fields by applying the method to four types of specimens.

  • Since Modern bridges have a tendency to make the spans continuous and longer, the effect of concrete shrinkage and creep is very important and must be evaluated appropriately for the durability and safety of steel-concrete composite bridges. However, highway design specification in current use prescribes $180^{1\;2}$ as the final shrinkage strain. which is for less value than one resulted from many experimental researches and cause some problems in the construction of composite bridges due to the understimation of shrinkage strain. Thus, in this paper nonlinear analysis with time-steps applying the CEB-FIP(90) provision have been conducted for plate girder bridge, box girder bridge and Preflex beam bridge and the linear equivalent shrinkage strain for the design of composite bridges. which produces the stress equal to the values from the nonlinear analysis, has been calculated by comparing the results with the values following highway design specification. The results yield appropriately double values than $180^{1\;2}$ which highway design specification prescribes.

  • Plate has cutout inner bottom and girder and Door etc. in hull construction absence is used much, and this is strength in case must be situated, but establish in region that high stress interacts sometimes fatally in region that there is no big problem usually by purpose of weight reduction, a person and freight movement, piping etc.. Because cutout‘s existence is positioning in this place, and, elastic bucking strength by load causes large effect in ultimate strength. Therefore, perforated plate elastic bucking strength and ultimate strength is one of important design criteria to decide structural elements size at early structure design step of a ship. Therefore, we need reasonable & reliable design formula for elastic bucking strength of the perforated plate. The author computed numerically ultimate strength change about several aspect ratios, cutout dimension, and plate thickness by using ANSYS Finite element analysis code based on finite element method in this paper.

  • It is significant technique to increase competitiveness that solar cells have a high energy conversion efficiency and cost effectiveness. When making high efficiency crystalline Si solar cells, evaporated Ti/Pd/Ag contact system is widely used in order to reduce the electrical resistance of the contact fingers. However, the evaporation process is no applicable to mass production because high vacuum is needed. Furthermore, those metals are too expensive to be applied for terrestrial applications. Ni/Cu/Ag contact system of silicon solar cells offers a relatively inexpensive method of making electrical contact. Ni silicide formation is one of the indispensable techniques for Ni/Cu/Ag contact sytem. Ni was electroless plated on the front grid pattern, After Ni electroless plating, the cells were annealed by RTP(Rapid Thermal Process). Ni silicide(NiSi) has certain advantages over Ti silicide($TiSi_2$), lower temperature anneal, one step anneal, low resistivity, low silicon consumption, low film stress, absence of reaction between the annealing ambient. Ni/Cu/Ag metallization scheme is an important process in the direction of cost reduction for solar cells of high efficiency. In this article we shall report an investigation of rapid thermal silicidation of nickel on silngle crystalline silicon wafers in the annealing range of $350-390^{\circ}C$. The samples annealed at temperatures from 350 to $390^{\circ}C$ have been analyzed by SEM(Scanning Electron Microscopy).

Microbial Risk Assessment in Reclaimed Wastewater Irrigation on a Paddy Field (하수의 농업적 재이용에 따른 논 담수 내 미생물 위해성 평가)

  • Water stress has become a major concern in agriculture. Korea suffers from limited agricultural water supply, and wastewater reuse has been recommended as an alternative solution. A study was performed to examine the effects of microorganism concentration in the ponded-water of a paddy rice field with reclaimed-water irrigation for evaluating the microbial risk to farmers and neighborhood children. Most epidemiological studies were performed based on an upland field, and they may not directly applicable to paddy fields. Beta-Poisson model was used to estimate the microbial risk of pathogen ingestion. Their risk value increased significantly high level after irrigation and precipitation. It implies that agricultural activities such as plowing, and fertilizing, and precipitation need be practiced a few days after irrigation considering health risks. The results about field application of the microbial risk assessment using E. coli showed difference according to monitoring time and treatment plot. Result of the microbial risk assessment showed that risk values of ground-water and reclaimed secondary wastewater irrigation were lower than directly use of wastewater treatment plants' effluent. This paper should be viewed as a first step in the application of quantitative microbial risk assessment of E. coli to wastewater reuse in a paddy rice farming.

Promoter Methylation Status of Two Novel Human Genes, UBE2Q1 and UBE2Q2, in Colorectal Cancer: a New Finding in Iranian Patients

  • Background: The ubiquitin-proteasome system (UPS) degrades a variety of proteins which attach to specific signals. The ubiquitination pathway facilitates degradation of damaged proteins and regulates growth and stress responses. This pathway is altered in various cancers, including acute lymphoblastic leukemia, head and neck squamous cell carcinoma and breast cancer. Recently it has been reported that expression of newly characterized human genes, UBE2Q1 and UBE2Q2, putative members of ubiquitin-conjugating enzyme family (E2), has been also changed in colorectal cancer. Epigenetics is one of the fastest-growing areas of science and nowadays has become a central issue in biological studies of diseases. According to the lack of information about the role of epigenetic changes on gene expression profiling of UBE2Q1 and UBE2Q2, and the presence of CpG islands in the promoter of these two human genes, we decided to evaluate the promoter methylation status of these genes as a first step. Materials and Methods: The promoter methylation status of UBE2Q1 and UBE2Q2 was studied by methylation-specific PCR (MSP) in tumor samples of 60 colorectal cancer patients compared to adjacent normal tissues and 20 non-malignant controls. The frequency of the methylation for each gene was analyzed by chi-square method. Results: MSP results revealed that UBE2Q2 gene promoter were more unmethylated, while a higher level of methylated allele was observed for UBE2Q1 in tumor tissues compared to the adjacent normal tissues and the non malignant controls. Conclusions: UBE2Q1 and UBE2Q2 genes show different methylation profiles in CRC cases.

A Basic Study on Crushability of Sands and Characteristics of Particle Strength (모래의 파쇄성과 단입자강도 특성에 관한 기초적 연구)

  • Particle crushing is an important and essential factor in interpreting the strength and deformation properties of granular materials in the case of geotechnical problems related to soil crushability. As a recent field problem, the exploitation of offshore oil reserves in tropical and sub-tropical coastal shelf areas has shown that the behaviour of soils containing carbonates is markedly different from predominantly silica sands. In this study, as a first step in making a mechanical framework of granular materials incorporating the soil crushability, single particle fragmentation tests were carried out on four different types of sands in order to clarify the characteristics of the single particle fragmentation strength as related to soil crushability. The single particle strength was considered with the influence of the particle shapes, the amount of mineral components and the particle sizes. The soil particle strength corresponding $D_{50}$ of soil distribution curve has shown the lower value, the more the carbonate component and the more angular the particle shape.
