Interactions of Virulent and Avirulent Fusarium species on Clonal Asparagus Plantlets and Mechanisms Involved in Protection of Asparagus with Avirulent Fusarium Species Against Stem and Crown Rots (아스파라거스에서 병원성 및 비병원성 Fusarium균의 상호작용과 비병원성 Fusarium을 이용한 아스파라거스 줄기 및 뿌리썩음병 방제 기작 연구)

  • 병원성 Fusarium에 의한 아스파라거스 감염은 비병원성 Fusarium을 5일과 7일 전에 접종하였을 때 방제효과가 있었다. 비병원성 F. oxysporum은 F. moniliforme에 대하여 방제효과가 있었고, F. solani는 F. oxysproum에 대하여 방제효과가 있음이 밝혀졌다. 실험에 사용된 Fusarium 균들은 모두 주근과 측근의 말단 부위, 상처부위, 그리고 표피의 세포벽 사이를 통하여 감염하였다. 경우에 따라 감염하는 동안 appressorium과 유사한 구조를 형성하기도 하였고, 직접 감염하는 경우도 있었다. 병원성 그리고 비병원성 Fusarium 균 모두 공통적으로 생장점 부위를 통하여 감염하였다. 병원성이 강한 Fusarium 균의 경우 비병원성 균들보다 감염의 속도가 빨랐고 더욱 생장이 왕성하였다. F. solani는 생장속도나 기주 조직 침입속도가 매우 느렸다. 기주 감염의 결과 처음에는 cortical rot을 유발시켰고 나중에는 tracheary elements를 감염하고 결국은 조직의 괴사를 유발하는 것이 관찰되었다. 비병원성 F. oxysporum은 표피조직에 두터운 균사층을 형성하였고, 이는 병원성 Fusarium 균에 대한 방제효과를 나타내는 원인을 제공한 것으로 여겨진다. F. solani는 측근의 생성을 촉진시켜 표면적을 증대시킨 것으로 여겨진다. 결론적으로 AVFO와 F. solani를 이용하여 아스파라거스에 발생하는 병원성 Fusarium균의 침입을 저지할 수 있는 생물적 방제가 가능함이 밝혀졌다.

Pathogenicity of Didymella bryoniae on the Seedlings of Cucurbits (오이류 유묘에 대한 덩굴마름병균의 병원성)

  • The objective of the study is to determine differences between cucurbits in the pathogenicity of Didymella bryoniae isolated from the naturally infected seeds of cucumber and pumpkin. Primary seedling infection of cucumber(Cucumis sativus L.), oriental melon(Cucumis melo var. makuwa Makino), pumpkin(Cucurbita pepo L.) and watermelon (Citrullus vulgaris Shrad.) occurred on the radicle, hypocotyl and cotyledons and symptoms on each crop were very similar. Infection of the radicle generally caused pre-emergence rot, while infection on the hypocotyl and cotyledons provided further inoculum for infection of the first true leaves and the stem. In cross inoculation tests, all isolates of D. bryoniae could infect cucumber, oriental melon, pumpkin and watermelon at different growth stages and there were not much differences in pathogenicity or susceptibility between isolates of the pathogen and crops tested. The susceptibility of cucumber and pumpkin was markedly influenced by prevailing humid conditions.

In vitro Inhibition Effect of Plant Extracts, Urine, Fertilizers and Fungicides on Stem Rot Pathogen of Sclerotium rolfsii

  • Twenty plant extracts were tested against mycelial growth, sclerotium formatiom and dry weight of mycelium with sclerotia of Sclerotium rolfsii Sacc. The highest(90 mm) mycelial growth was measured in Adhatoda vasica, Tegetes erecta, Allium cepa, and Curcuma longa. The lowest(25 mm) was in Azadirachta indica. No mycelial growth was found in any concentration of cow, buffalo, and goat urine. The highest(90 mm) and the lowest(15 mm) mycelial growth were measured in Biomil and Urea, respectively. No mycelial growth was observed in Zinc. The highest(60 mm) and the lowest(2 mm) mycelial growth were recorded in Macuprex(Dodine; 65% WP) and Boron(100% Boric acid and 17% Boron) respectively. Mycelial growth was totally inhibited in Rovral(Iprodione; 50% WP).

Stub Dieback of Carnation Caused by Fusarium graminearum

  • A disease survey on the carnation (Dianthus caryophyllus L.) wilt was conducted during the high temperature period (June through August) and the low temperature period (February through May) in 58 greenhouses of its major cultivation areas, including Pusan, Kimhae, and Changwon in Korea from 1998 to 1999. The disease incidence was averaged 5.4% and 11.9% in the low and high temperature periods, respectively. Severe damage was found in summer with high incidences of around 50% in some greenhouses. Close examination of the symptoms and isolation of the causal agent revealed that there was a new disease different from Fusarium wilt caused by Fusarium oxysporum f. sp. dianthi, which was determined as the stub dieback caused by F. was cetermined as the stub dieback caused by F. graminearum (teleomorph : Gibberella zeae). The stub dieback symptoms involved brown rot of stem that started usually from the portion of cutting without discoloration of inner vascular tissues. Seven out of 38 isolates from the wilted plants were identified as F. graminearum, while the others as F. oxysporum f. sp. dianthi. Mycological characteristics of the stub dieback pathogen including colony color, absence of microconidia, and the shape of macroconidia, were consistent with F. graminearum previously described. This is the first report of the carnation stub dieback in Korea.

Genome-wide Association Analyses for Resistance to Phytophthora sojae and Pseudomonas amygdali pv. tabaci in Soybean

  • Phytophthora root and stem rot (PRSR) and wildfire disease (WFD) of soybean are frequently observed in the field of South Korea. The most environmentally friendly way to control PRSR and WFD is to use soybean varieties with resistance to Phytophthora sojae (P. sojae) and Pseudomonas amygdali pv. tabaci. Plant germplasm is an important gene pool for soybean breeding and improvement. In this study, hundreds of soybean accessions were evaluated for the two pathogens, and genome-wide association analyses were conducted using 104,955 SNPs to identify resistance loci for the two pathogens. Of 193 accessions, 46 genotypes showed resistance reaction, while 143 did susceptibility for PRSP. Twenty SNPs were significantly associated with resistance to P. sojae on chromosomes (Chr.) 3 and 4. Significant SNPs on Chr.3 were located within the known Rps gene region. A region on Chr. 4 is considered as a new candidate resistance loci. For evalation of resistance to WFD, 18, 31,74,36 and 34 genotypes were counted by a scale of 1-5, respectively. Five SNP markers on Chrs 9,11,12,17 and 18 were significantly associated with resistance to P. amygdali pv. tabaci. The identified SNPs and genomic regions will provide a useful information for further researches and breeding for resistance to P. sojae and P. amygdali pv. tabaci.

A Shinpung Typed Large Grain and High-Yielding Peanut Variety 'Daan' (단경 소분지 대립 다수성 땅콩 신품종 '다안')

  • A new peanut variety 'Daan'(Arachis hypogaea ssp. hypogaea L.) was developed at the Department of Southern Area Crop Science, National Institute of Crop Science (NICS) in Milyang 2014. This was developed from the cross between 'Iksan 31' with Virginia typed short stem and 'Dakwang' with Shinpung-typed larger grain. 'Daan' which is a Shinpung plant type had 13 branches per plant and its length of main stem was 44cm. Each pod has two grains with brown testa and long ellipse-shaped large kernel. Its yield components showed 34 pods per plant, 127 g of 100-seed-weight and 75% of pod shelling ratio in the regional yield trials (RYT). Seed quality showed 47.8% of crude oil and 28.3% of protein content. This variety showed resistant to early leaf spot and had more resistant to late leaf spot, stem rot and lodging, compared to reference variety. In the regional yield trials for 3 years 'Daan' was more productive than reference variety by 16% with 5.00 MT/ha for grain production.

Overwintering pattern of larvae of Chilo suppressalis Walker in the bioenergy crop Miscanthus sacchariflorus cv. Geodae 1 (바이오에너지작물 거대억새 가해 해충 이화명나방 유충 월동양상)

  • The rice stem borer (Chilo suppressalis Walker) was one of the most destructive pest of rice for the 1960s and 1970s in Korea. Recently, it is newly recognized as a potential risk factor to the biomass yield of bioenergy crops. The current research was firstly conducted to investigate overwintering larvae population density and pattern of rice stem borer attacking Miscanthus sacchariflorus cv. Geodae 1 which is referred to as an ideal lignocellulosic bioenergy crop in Korea. Population density of larvae per $1m^2$ in stems and rhizomes at the Miscanthus experimental plots and rates of damage (wormhole, abscission) of M. sacchariflorus cv. Goedae 1 were investigated from October 2012 to March 2013. The population of larvae per $1m^2$ in stems of Miscanthus were 23, 4, 1, and 1 in October, November, December 2012, and January 2013, respectively. Over the same period, the population of larvae in basal stem rots and rhizomes were increased, whereas decreased in stems. Interestingly, the positions of larvae for overwintering in Miscanthus were confirmed to 5~10 cm below the soil surface such as basal stem rot and rhizome, whereas the most common overwintering position known in rice is a part of stem on the ground such as rice straw and rice stubble. It would suggest that the larvae gradually moved to bottom of stems and rhizomes in soil in line with decline in temperature. Moreover, the damage rates of stems per $1m^2$ were up to more than 50% in some places. In conclusion, this might be the first report that rice stem borer could affect the productivity of biomass of Miscanthus in case of mass cultivation. Moreover, it should be necessary to make a decision in insect control management for this bioenergy feedstock and other related crops.

Effect of On-site Postharvest Hot Water Treatment on Storage Quality of Commercial Greenhouse Satsuma Mandarin (현장 열수처리에 따른 온실재배 온주감귤 상품의 저장 중 품질특성 변화)

  • Greenhouse satsuma mandarins (Citrus unshiu Marc., cv. Gungchun) of an early harvesting cultivar were treated by hot water showering at 65$^{\circ}C$ for 10 s at a commercial scale in a packing house and then stored at 5$^{\circ}C$ for 3 weeks and subsequently at 18$^{\circ}C$ for 1 week (simulated shelf-life) to examine the potential use of hot water treatment (HWT) as an environmentally benign method to maintain mandarin quality characteristics during postharvest storage and sale. The respiration rate just after heat treatment or during storage was at a similar level in both the treated and untreated fruit. HWT also had no detrimental effects on quality attributes including pH, titratable acidity, soluble solids content, weight loss, firmness, and peel color. The development of stem-end rot, mold decay, and black rot was lower in the heat-treated fruit compared to those in the untreated control. A sensory evaluation showed that HWT markedly improved fruit appearance, making the fruit cleaner and glossier. The results suggested that HWT can be applied to satsuma mandarin as an effective pretreatment to maintain postharvest quality during storage and marketing.

Effect of Potassium on Rice Plant Growth on an Akiochi Soil (추락답토양(秋落沓土壤)에 생육(生育)한 수도(水稻)에 대(對)한 가리(加里)의 효과)

  • In application of manganese and silica to Akiochi soil, growth of rice plant with potassium was remarkably increased, but the growth without potassium was stunted and the number of the tiller was relatively reduced in comparison with that receiving potassium. The minus potassium plant developed tiny brown spots on the leaves and color of the leaves was dark green. They had smaller leaves and shorter stem while heading advanced. Roots with potassium were long and thick, and new ones developed, while roots without K were very poor, the branch and hair roots very thin, and severe root rot was observed. With minus potassium treatment, the root rot of Jin Heung(commercial variety) was more severe than Nonglim 6(Akiochi resistant). Growth of plant without potassium was very poor, and potassium content of the plant was very low, while nitrogen, phosphorus, silica, iron and manganese contents were higher than potassium supplied plant. Application of $Mn+SiO_2$ to Akiochi soil lowered iron content of plant, and iron content of the plant were also reduced by application of potassium. From the results obtained it can be concluded that root damage of minus potassium plant grown in Akiochi soil supplied $Mn+SiO_2$ was caused by potassium deficiency. Potassium, and $Mn+SiO_2$ applications lowered iron content in the plant grown on Akiochi soil.

Genome Wide Association Study for Phytophthora sojae Resistance with the Two Races Collected from Main Soybean Production Area in Korea with 210 Soybean Natural Population

  • Recently days, soybean production in paddy field is increasing, from 4,422 ha in 2016 to 10,658 ha in 2021 in Korea. It is easy for Phytophthora stem and root rot (PSR) occurring in paddy field condition, when it is poorly drained soils with a high clay content, and temporary flooding and ponding. Therefore PSR resistant soybean cultivar is required. The objective of this study is to identify QTL region and candidate genes relating to PSR resistance of the race in main soybean cultivation area in Korea. 210 soybean materials including cultivars and germplasm were used for inoculation and genome-wide association study (GWAS). Inoculation was conducted using stem-scar method with 2 replications in 2-year for the race 3053 from Kimje and 3617 from Andong. 210 materials were genotyped with Soya SNP 180K chip, and structure analysis and association mapping were conducted with QTLMAX V2. The results of inoculation showed that survival ratio ranged from 0% to 96.7% and mean 9.7% for 3053 and ranged from 0% to 100% and mean 7.6% for 3617. Structure analysis showed linkage disequillibrium (LD) was decayed below r2=0.5 at 335kb of SNP distance. Significant SNPs (LOD>7.0) were identified in Chr 1, 2, 3, 4, 5, 11, 14, 15 for 3053 and Chr 1, 2, 3, 7, 10, 14 for 3617. Especially, LD blocks (AX-90455181;15,056,628bp~AX-90475572;15,298,872bp) in Chr 2 for 3053 and 3067 were duplicated. 29 genes were identified on these genetic regions including Glyma.02gl47000 relating to ribosome recycling factor and defense response to fungus in Soybase.
