The main purpose of the ball valve ball is to be moved by the rotation of the stem when fully open or completely closed. In this study the heat of the initial model, which used a structure interaction analysis technique, tried to examine the structural safety of the high temperature for the ball valve. In the initial model the stress of the exiting sheet was more than the yield strength. We selected two design shapes with variables of length and thickness for the optimization of the sheet. The Kriging interpolation method was applied to a meta-model-based optimization technique. As a result, it was possible to find a thickness and length for the sheet within the yield strength. This was done by measuring the value of the capacity coefficient of the valve and evaluating the performance of the ball valve.
For the domestic digital contents, it is particularly important that the planning and mediating functions should cover the whole industries so that in order for dividends and assistance to reach all the way back to the primary contents-creating level, where economic risks are especially high. A proactive cooperation system or framework between the government, the businesses and research institutions, which would allow for free flow of interaction among these players, can play that role. We can broadly define cooperation frameworks into two; one between the central and the local governments and one among private-sector groups, such as companies, associations and groups. These player sin the framework have differing, sometimes, conflicting views regarding the digital contents industry whether the digital contents should aim for skilled applicability or the abstract or whether the emphasis should rest on public interest or profitability. Immature competition and lack of trust among these players also give rise to such inefficiencies as overlap in investments, inexpertness and inefficient use of resources. We have proposed and realized the National Integrated Cooperation System to bridge these gaps among the major players in the industry. The National Integrated Cooperation System rests on the following major functions. The first major function of the NICS is to decipher any ambiguity that may be embedded in external inputs by stratified role and bias. The second function is to create cooperative groups that will deal with the ambiguities based on its consequent situation. The third is a feedback function that will draw out a new cooperative way by re-feeding the capacities and the conflicts that stem from the existing organizations and strategies into cooperation and adjustment process. Our NICS has compared and evaluated with England and Australia digital content industry models under AHP(Analytic Hierarchy Process) method. NICS has turned out to be well designed and have strong points based on OECD innovation and cooperation criteria.
Loop-mediated isothermal amplification (LAMP) developed by Notomi et al. (2000) has made it possible to amplify DNA with high specificity, efficiency and rapidity under isothermal conditions. The ultimate products of LAMP are stem-loop structures with several inverted repeats of the target sequence and cauliflower-like patterns with multiple loops shaped by annealing between every other inverted repeats of the amplified target in the similar strand. Because the amplification process in LAMP is achieved by using four to six distinct primers, it is expected to amplify the target region with high selectivity. However, evaluation of reaction accuracy or quantitative inspection make it necessary to append other procedures to scrutinize the amplified products. Hitherto, various techniques such as turbidity assessment in the reaction vessel, post-reaction agarose gel electrophoresis, use of intercalating fluorescent dyes, real-time turbidimetry, addition of cationic polymers to the reaction mixture, polyacrylamide gel-based microchambers, lateral flow dipsticks, fluorescence resonance energy transfer (FRET), enzyme-linked immunosorbent assays and nanoparticle-based colorimetric tests have been utilized for this purpose. In this paper, we reviewed the best-known techniques for evaluation of LAMP amplicons and their applications in molecular biology beside their advantages and deficiencies. Regarding the properties of each technique, the development of innovative prompt, cost-effective and precise molecular detection methods for application in the broad field of cancer research may be feasible.

Bracken fern [Pteridium aquilinem (L.) kuhn (Dennstaedtiaceae)] is one of the most common species on the planet. It has been consumed by humans and animals for centuries. Use by some human groups is because they believe bracken fern is good for health as plant medicine. However, it is also one of the few known plants that can cause tumors in farm animals. Many interested groups have focused their attention on bracken fern because of these interesting features. In order to evaluate the biological effects of exposure to this plant in cellular level, human cancer cell lines were treated with the fern dichloromethane extracts and the genotoxic and cytotoxic effects were studied. Anti-proliferative/cytotoxic effects were evaluated by cell count, MTT assay and flow cytometry methods with three different cancer cell lines, TCC, NTERA2, and MCF-7, and two normal cells, HDF1 and HFF3. Pro-apoptotic effects of the extracts were determined by DAPI staining and comet assay, on TCC cancer cells compared to the normal control cell lines. Cellular morphology was examined by light microscopy. Our present study showed that the extract caused DNA damage and apoptosis at high concentrations ($200{\mu}g/mL$) and also it may induce cell cycle arrest (G2/M phase) at mild concentrations (50 and $30{\mu}g/mL$) depending on the cell type and tumor origin. These results indicate that bracken fern extract is a potent source of anticancer compounds that could be utilized pharmaceutically.
The role of moisture absorptive capacity of pre-silage material and its relationship with silage effluent in high moisture by-product feedstuffs (HMBF) is assessed. The term water retention capacity which is sometimes used in explaining the rate of effluent control in ensilage may be inadequate, since it accounts exclusively for the capacity of an absorbent incorporated into a pre-silage material prior to ensiling, without consideration to how much the pre-silage material can release. A new terminology, 'potential water retention capacity' (PWRC), which attempts to address this shortcoming, is proposed. Data were pooled from a series of experiments conducted separately over a period of five years using laboratory silos with four categories of agro by-products (n = 27) with differing moisture contents (highest 96.9%, lowest 78.1% in fresh matter, respectively), and their silages (n = 81). These were from a vegetable source (Daikon, Raphanus sativus), a root tuber source (potato pulp), a fruit source (apple pomace) and a cereal source (brewer's grain), respectively. The pre-silage materials were adjusted with dry in-silo absorbents consisting wheat straw, wheat or rice bran, beet pulp and bean stalks. The pooled mean for the moisture contents of all pre-silage materials was 78.3% (${\pm}10.3$). Silage effluent decreased (p<0.01), with increase in PWRC of pre-silage material. The theoretical moisture content and PWRC of pre-silage material necessary to stem effluent flow completely in HMBF silage was 69.1% and 82.9 g/100 g in fresh matter, respectively. The high correlation (r = 0.76) between PWRC of ensiled material and silage effluent indicated that the latter is an important factor in silage-effluent relationship.
Luteolin is a naturally occurring flavonoid present in many plants with diverse applications in pharmacology. Despite several studies elucidating its significant anti-cancer activity against various cancer cells, the mechanism of action in skin cancer is not well addressed. Hence, we investigated the effects of luteolin in HaCaT (human immortalized keratinocytes) and A375 (human melanoma) cells. The radical scavenging abilities of luteolin were determined spectrophotometrically, prior to a cytotoxic study (XTT assay). Inhibitory effects were assessed by colony formation assay. Further, the capability of luteolin to induce cell cycle arrest and apoptosis were demonstrated by flow cytometry and cellular DNA fragmentation ELISA, respectively. The results revealed that luteolin possesses considerable cytotoxicity against both HaCaT and A375 cells with $IC_{50}$ values of 37.1 ${\mu}M$ and 115.1 ${\mu}M$, respectively. Luteolin also inhibited colony formation and induced apoptosis in a dose and time-dependent manner by disturbing cellular integrity as evident from morphological evaluation by Wright-Giemsa staining. Accumulation of cells in G2/M (0.83-8.14%) phase for HaCaT cells and G0/G1 (60.4-72.6%) phase for A375 cells after 24 h treatment indicated cell cycle arresting potential of this flavonoid. These data suggest that luteolin inhibits cell proliferation and promotes cell cycle arrest and apoptosis in skin cancer cells with possible involvement of programmed cell death, providing a substantial basis for it to be developed into a potent chemopreventive template for skin cancer.

Background: CD40-activated B (CD40-B) cells might be an attractive source of autologous antigen-presenting cells (APCs) for immunotherapy due to the convenience to obtain from peripheral blood and expand in vitro. Moreover, CD40-B cells were found to be comparable with DCs in their capacity to raise antigen-specific CD8+ T cells. Here, we have established K562 cells expressing CD40L to expand CD40-activated B cells used for APCs. Methods: After activation of B cell by K562/CD40L, CD40-B cells were examined by counting B cell numbers. Surface expression of CD54, CD80, CD86 and HLA class II was measured by flow cytometry. The CD40-B cells were tested for its function as APC by mixed lymphocyte reactions (MLR) and by induction of T cell responses specific for pp65 peptide in vitro. Results: The expansion of B cells by K562/CD40L increased about 6-folds compared with anti-CD40 or K562. Furthermore, the expression of CD54, CD80, CD86 and HLA class II was up-regulated by K562/CD40L. B cells by K562/CD40L showed comparable antigen presentation activity with mature DCs as shown in MLR, INF-${\gamma}$ ELISPOT assay. Conclusion: These results suggest that K562/CD40L could be used to generate activated B cells as potent APCs which could be useful for cellular vaccination and adoptive immunotherapy.
Sclerotinia sp. (isolate BWC98-105) causes stem blight and root rot in Leghum sp., and is presently being evaluated as a potential mycoherbicide for the control of Trifoliorium repens. Bioassays have shown that Sclerotinia sp. produces phytotoxic substance which is biologically active against T. repens. Two biologically active compounds, designated as compoundsI and II, were produced in vitro from the culture filtrate of BWC98-105 isolate Sclerotium sp. Compounds I and II were purified by means of liquid-liquid extraction and $C_{18}$ open column chromatography (300 ${\times}$ 30 mm, i.d). To determine the purity, the purified compounds were analyzed by RP-HPLC. The analytical RP-HPLC column was a TOSOH ODS-120T (150 ${\times}$ 4.6 mm i.d, Japan), of which the flow rate was set at 0.7 mL/min using the linear gradient solvent system initiated with 15 % methanol to 85 % methanol for 50 min with monitoring at 254 nm. Under these RP-HPLC conditions, compounds I and II eluted at 3.49 and 4.13 min, respectively. Compound II was found to be most potent and host specific. However, compound I had a unique antibiotic activity against phytopathogenic bacteria like bacterial leaf blight (Xanthomonas oryzae) on rice, where it played a less important role in producing toxicity on T. repens. No toxin activity was detected in the water fraction after partitioning with several organic solvents. However, toxin activity was detected in the ethyl acetate and butanol fractions. In the leaf bioassay using compound II, the disease first appeared within 4-5 h as water soaked rot, which subsequently developed into well-defined blight affecting the whole plant.

This study was carried out to determine the effects of wind speed on physiological responses in 'Fuji' apple (Malus pumila Miller). Two levels of wind blowing (3 and $5m{\cdot}s^{-1}$) were produced by large electric fans. Photosynthetic rate was reduced by one-way wind blowing treatment at $5m{\cdot}s^{-1}$, compared to the mild wind control, and this reduction was more obvious with stronger wind and increasing duration of wind application. The reduction in photosynthesis by the wind treatments was correlated with that in the proportion of opened stomates and stomatal conductance. The one-way wind treatment at $5m{\cdot}s^{-1}$ caused a leaf browning and leaf fall, and this negative effect became more serious with increasing time of exposure to the wind treatments. The sap flux through stem increased in all wind treatments compared to the natural mild wind.

In the study, a design process for ensuring optimal clearance in a fuel injection pump(FIP) is suggested. Structure analysis and hydrodynamic lubrication analysis are performed to determine the optimal clearance. The FIP is simulated using Hypermesh, Abaqus 6.12 to evaluate the reduction of clearance when the maximum supply pressure is applied. The reduction in clearance is caused by the difference in the deformations between the barrel and plunger. When the deformation of the plunger is larger than that of the barrel, a reduction in clearance at the head part occurs. On the other hand, the maximum clearance reduction equals the maximum deformation in the stem part, because the deformation of barrel does not occur in this region. The clearance of FIP should be designed to be larger than maximum reduction of clearance in order to avoid contact between the plunger and barrel. In addition, the two-dimensional Reynolds equation is used to evaluate lubrication characteristics with variations of viscosity, clearance and nozzle for a laminar, incompressible, unsteady state flow. The equation is discretized using the finite difference method. The lubrication characteristics of FIP are investigated by comparing film parameter, which is the ratio of the minimum film thickness and surface roughness. The optimal clearance of FIP is to be designed by considering the maximum reduction in clearance, lubrication characteristics, machining limits and tolerance of clearance.
