강남콩에 대한 $SO_2$ 피해경감제로서 uniconazole의 효과에 관한 연구 (Efficacy of Uniconazole as a Phytoprotectant Against $SO_2$ Injury in Snap Bean)

  • This study was conducted to determine the efficacy of using uniconazole,[(E)-1-(4-chlorophenyl)-4,4-dimethyl-2-(1,2,4-triazole-1-yl)-1-penten-3-ol)] as a phytoprotectant against $SO_2$ injury in snap been (Phaseolus vulgaris L. 'Strike'). Thirteen days prior to $SO_2$ fumigation, plants were given a 100 ml soil drench of uniconazole solution at concentrations of 0.02, 0.10, 0.25 and 0.50 mg/pot. All four uniconazole concentrations were significantly effective in providing protection against $SO_2$ exposure(3 h at 1.5 ppm), but uniconazole treatment above 0.02 mg/pot severely reduced stem elongation, leaf enlargement, flowering date and pod number and weight. Uniconazole treatment had little or no effect on stomatal conductance but reduced transpiration rate on a whole plant basis by nearly 40%. This may reflect an alteration in canopy structure by reducing stem elongation and leaf enlargement. Although uniconazole did not increase the activities of superoxide dismutase(SOD) and peroxidase(POD) in non-$SO_2$-fumigated plants, it significantly increased those enzyme activities in $SO_2$-fumigated plants. Chlorophyll concentration on the basis of unit area was increased 50-60% by uniconazole. However, the difference was not detected on the basis of dry weight. $SO_2$ increased variable chlorophyll fluorescence (Fv) 48% after 1.5 h of exposure in non-uniconazole treated plants but decreased Fv in the plants after 3 h of exposure. By appliing uniconazole, it was possible to maintain high Fv values in the latter group of plants. These results suggest that the phytoprotective effects of uniconazole are related to its growth-retarding properties as an anti-gibberellin as well as the increase of activites of free radical scavengers such as SOD and POD.

GROWTH AND FODDER YIELD OF THE Gliricidia sepium PROVENANCES IN ALLEY CROPPING SYSTEM IN DRYLAND FARMING AREA IN BALI, INDONESIA

  • The design of this field experiment was a completely randomized block arrangement, consisted of 16 treatments (Gliricidia sepium provenances) and 6 blocks as replications with 12 plants per provenance. Of the 16 gliricidia provenances, six were from Mexico (M), four were from Guatemala (G), and one each was from Colombia (C), Indonesia (I), Nicaragua (N), panama (P), Costa Rica (R), and Venezuela (V). After 12 months establishment the gliricidia were lopped regularly 4 times a year, twice during the 4 months wet season and twice during the 8 months dry season at 150 cm height. There was variation (p < 0.05) in stem elongation from 22 to 80 cm, leaf retained from 118 to 209%, branch number from 13 to 24, fodder yield from 1,015 to 1,671 g DW/plant and wood yield from 792 to 1,662 g DW/plant among the provenances; and such variations were affected by the seasons. Belen (N14), Retalhuleu (G14) and Bukit Bali (I) provenances were ranked first, second and third, respectively, measured in terms of leaf retention, stem elongation, fodder and wood yields during the wet and dry seasons.

  • The field experiment was carried out to study the growth and fodder yield of the Gliricidia sepium provenances in fence system in dryland farming area in Bali, Indonesia for 24 months. The design of the experiment was a completely randomized block arrangement, consisted of 16 treatments (Gliricidia sepium provenances) and 12 blocks with 10 plants per provenance. Of the 16 gliricidia provenances, six were from Mexico (M), four were from Guatemala (G), and one each was from Colombia (C), Indonesia (I), Nicaragna (N), Panama (P), Costa Rica (R) and Venezuela (V). After 40 weeks establishment the gliricidia were lopped regularly 4 times a year, twice during the four month wet season and twice during the eight month dry season at 150 cm height. There were variations (p < 0.05) in stem elongation from 10 to 121 cm, leaf retained from 48 to 105%, leaf shedding from 53 to 86%, branch number from 4 to 7, fodder yield from 281 to 648 g DW/plant, and wood yield from 53 to 179 g DW/plant; and such variations were affected by the seasons. Retalhuleu provenance of Guatemala (G14), Belen provenance of Nicaragua (N14) and Bukit Bali provenance of Indonesia (I) were ranked first, second and third, respectively, measured in terms of stem elongation, leaf retention, fodder and wood yields during the wet and dry seasons.

  • A field experiment was carried out on a dryland farming area of southern Bali for 92 weeks, to study the growth and fodder yield of 16 provenances of Gliricidia sepium in guardrow system. The experimental design was completely randomized blocks of 16 treatments (Gliricidia sepium provenances) replicated 3 times, with 6 plants per provenance. Six provenances were from Mexico (M), four from Guatemala (G), and one each from Colombia (C), indonesia (I), Nicaragua (N), Panama (P), Costa Rica (R) and Venezuela (V). After 40 weeks establishment the gliricidia were lopped 4 times a year at 150 cm height, at 2 months intervals during the 4 month wet season and 4 month intervals during the 8 month dry season. Stem elongation varied from 21 to 81 cm, leaf retention from 39 to 240%, branch number from 12 to 35, fodder yield from 1,090 to 3,153 g DW/plant. and wood yield from 743 to 2,750 g DW/plant. Pontezuelo provenance of Colombia (C24), Belen provenance of Nicaragua (N14) and Retalhuleu provenance of Guatemala (G14) were ranked first, second and third, respectively, for stem elongation, leaf retention, fodder and wood yields, during the wet and dry seasons.

  • 2009년 전남 해남 지역의 논에서 벼 키다리병과 유사한 병징을 나타내는 피를 채집하여 병원균을 분리하였다. 이병 식물체는 줄기가 고사하여 하얗게 말라 죽고, 줄기 표면에 흰가루 모양의 포자가 형성되었다. 분리균은 균학적 특성과 분자생물학적 특성에 따라 Fusarium fujikuroi로 동정되었다. Translation elongation factor 1-alpha 유전자의 염기서열 분석 결과, GenBank에 등록된 F. fujikuroi와 상동성을 나타내었다. 벼와 피를 대상으로 침지접종에 의해 병원성을 확인하였으며, 이는 피가 벼 키다리병의 전염원으로서의 역할을 할 수 있음을 의미한다. 따라서 벼 키다리병의 방제와 건전종자의 채종을 위해서는 논에 발생한 이병식물체뿐 아니라 전염원으로 작용할 수 있는 피를 제거해야 할 것으로 생각된다.

반추위액의 미생물에 의한 In vitro 상에서의 리놀렌산과 리놀산의 Biohydrogenation (In vitro Biohydrogenation of Linolenic and Linoleic Acids by Microorganisms of Rumen Fluid)

  • 리놀렌산(C18:3)과 리놀산(C18:2)의 biohydrogenation 과 반추위 내에서의 이들 지방산의 bypass를 검토하기 위하여 3 구로 나누어 티모시 조사료를 반추위액과 함께 in vitro상에서 39℃, 0, 3, 6, 9, 24, 36 시간 동안 혐기 배양 시험을 하였다. 첫번째 시험의 목적은 C18:2과 C18:3의 in vitro biohydrogenation에 대한 조사료의 성장 단계(stem elongation, early heading, late heading, early flowering)와 질소 시비(0 and 120kg N ha-1)의 영향을 시험하는 것이었다. 수소첨가가능분획(hydrogenable fraction), C18:2과 C18:3의 효과적인 소실과 bypass는 stem elongation 시 수확된 티모시에서 높았고, 성숙함에 따라 일정하게 감소하였다. 질소시비구는 C18:3의 수소첨가가능분획, C18:2과 C18:3의 효과적인 소실과 bypass가 증가하였다. 그러나 C18:2과 C18:3의 소실율은 성숙과 질소시비(P≻0.1)에 의해 영향을 받지 않았다. 2번째 시험에서는 in vitro C18:2과 C18:3 biohydrogenation에 대한 티모시 보존 방법의 영향을 보았다. 사일리지는 C18:2과 C18:3를 가장 효과적으로 소실시켰으며, 건초는 가장 효과가 낮았다. C18:2과 C18:3의 biohydrogenation 된 양은 건초에서 보다 헤일리지와 사일리지에서 더 높았다. 티모시 헤일리지와 비교하였을 때 C18:3의 bypass는 신선 목초, 시든풀, 건초에서 더 높았다. C18:2의 bypass는 건초와 헤일리지에 비해 신선목초와 사일리지가 더 높았다. 3번째 시험에서는 C18:2과 C18:3의 소거와 bypass에 대한 티모시 헤일리지와 사일리지에 대한 개미산 첨가와 Lactobacillus plantarum 접종의 효과를 검토하였다. 개미산의 첨가는 헤일리지와 사일리지에 있어서 C18:3의 biohydrogenation 비율을 증가시켰으나 사일리지에 있어서 C18:2의 수소첨가 가능분획을 감소시켰다. 이러한 3가지 배양구의 결과는 티모시에 있어 C18:2과 C18:3의 수소첨가 가능분획과 bypass가 성숙도에 따라 감소하였고 질소시비에 따라 증가하였음을 보여 준다. 헤일리지와 사일리지에서 건초에서 보다 C18:2과 C18:3의 더 많은 양이 biohydrogenation 되었으며 C18:3의 반추위 소실은 헤일리지에서 보다 신선목초, 시든풀, 건초에서 더 높았다.

Effects of Medium Compositions and Plant Growth Regulators on in vitro Organogenesis in Cultured Explants of Platycodon grandiflorum Species

  • Platycodon grandiflorum (Bell flower) is an important plant that has traditionally been used as herbal medicine for the treatment of cough, phlegm, sore throats, lung abscesses, chest pains, dysuria, and dysentery. The present study was initiated to investigate the feasibility of inducing shoot and root organogenesis in cultured explants of P. grandiflorum in a range of culture media and through use of various plant growth regulators (PGRs). The plantlets (Stem containing one node) were isolated and cultured on different concentrations of Murashige and Skoog (MS) medium supplemented with PGRs. We found that proliferation and elongation of shoots and roots could be achieved on 1/4 MS for P. grandiflorum with wild and green petals and on 1/8 MS for P. grandiflorum with double petals. The highest levels of development and elongation of adventitious shoots and roots were observed when petal explants were cultured on 1/4 MS (pH 3.8) supplemented with 5% sucrose. Increasing the agar concentration reduced shoot growth and rooting potential; nevertheless, the highest number of shoots and roots was observed on 0.6% agar. In the case of growth regulators, 1/4 MS supplemented with $1mg\;L^{-1}$ 6-benzylaminopurine (BA) was found to be best for shooting, although higher concentrations of BA tended to reduce shoot and root elongation. The highest number of shoots was achieved on $0.5mg{\cdot}L^{-1}$ thidiazuron (TDZ) from double petal explants grown on 1/8 MS. However, root and shoot elongation were found to decrease when TDZ concentrations were increased. Low concentrations of kinetin, naphthalene acetic acid, indole acetic acid, and 3-indole butyric acid induced shoot and root proliferation and elongation. Taken together, our study showed that low concentrations of PGRs induced the greatest root formation and elongation, showing that the optimal concentration of PGRs for shoot proliferation was species-dependent.

  • 파속 식물체 내에 있는 활성물질을 추출 분리하여 생물검정하고, 이들 작물의 경엽을 직접 pot에 시용하여 벼를 재배하였을 때 생육 및 수량 구성 요소에 미치는 영향은 다음과 같다. 1. 벼 유묘의 발근 및 생육을 촉진시키는 물질은 마늘의 염기성 분획과 대파, 양파, 부추 및 마늘의 약산성 분획에서 공통적으로 억제 물질보다 촉진물질의 활성이 높았으며 억제물질은 그 외 분획에서 활성이 높았다. 제 2엽초의 신장촉진 혹은 억제 물질은 공시한 파속 식물에는 내재하지 않았다. 2. 이앙 후 벼 유묘의 고사률은 이앙 당일에 비해 이앙 10일 전에 경엽을 시용한 처리가 높았고 특히 대파와 양파에 비해 부추와 마늘 시용구에서 높았다. 이앙 10일 전 동엽시용은 유효분얼수를 현저히 촉진시켰으며 특히 대파의 50g에서 400g까지, 양파의 50g에서 200g까지는 이삭입수의 증가도 나타나 표준시비구에 비해 수량 증가효과를 가져왔다.

Peony Stem Rots by Neopestalotiopsis clavispora and Sclerotinia sclerotiorum, and Antifungal Propineb and Bacillus siamensis H30-3 against the Two Fungal Species

  • In July 2022, stem rot symptom was found in a peony plant grown in a pot under a greenhouse at Jinju, Gyeongnam Province, South Korea. Two fungal species were isolated from the infected peony stems and cultured on 1/2-strength potato dextrose agar for identification. The morphological characteristics of the fungal isolates were examined, and nucleotide sequences of the internal transcribed spacer region, β-tubulin and translation elongation factor 1-α were analysed. The pathogenicity of the two isolates was confirmed in detached peony leaves, according to Koch's postulates. To our knowledge, this is the report of Neopestalotiopsis clavispora and Sclerotinia sclerotiorum as the causal agents of peony stem rots. Antifungal activity of chemical fungicide propineb and rhizobacterium Bacillus siamensis H30-3 was shown against the two plant pathogenic fungi N. clavispora and S. sclerotiorum.Unidentified diffusible and volatile compounds from B. siamensis H30-3 could suppress in vitro mycelial growths of N. clavispora JJ 8-2-1 and S. sclerotiorum JJ 8-2-2.

Uniconazole처리가 토마토의 저온 및 고온 피해 경감에 미치는 효과 (Alleviation of Low and High Temperature Injury in Tomato Plants by Uniconazole)

  • 작물의 저온 및 고온 피해를 경감시킬 수 있는 방안을 강구하기 위하여 토마토 품종, Fireball과 Patio를 공시재료로 하여 0, 0.001, 0.01, $0.1mg{\cdot}pot^{-l}$의 uniconazole을 처리하였다. 주간/야간의 길이를 12시간으로 하고, 온도를 $25/25^{\circ}C$, $2.5/25^{\circ}C$, $25/2.5^{\circ}C$, $40/40^{\circ}C$로 조절하여 4일간 처리한 결과는 다음과 같다. 1. 가시피해율과 간장의 억제율은 $2.5/25^{\circ}C$ 처리에서 가장 높았고 엽장의 감소는 $40/40^{\circ}C$에서 가장 높았다. 주야간 고온($40/40^{\circ}C$)을 처리한 경우 개화를 지연시켰고 화뢰의 발달을 저해하고 낙뢰를 초래하였다. 온도처리에 따른 피해율은 Fireball에서 Patio에 비하여 높은 것으로 나타났다. 2. 처리농도에 관계없이 uniconazole은 식물체의 생장을 크게 억제하고 chlorophyll의 함량을 증가시켰으며 개화를 지연시키고 저온 및 고온에 의한 가시피해율을 크게 감소시켰다. 3. 일반적으로 uniconazole처리는 고온 및 저온에 의한 엽록소함량의 감소와 개화지연 및 화뢰의 낙뢰를 막아주는 효과는 크게 인정되지 않았다. 고농도 처리는 오히려 Fireball 품종에서 개화지연과 화뢰의 낙뢰를 조장하는 결과를 보였다. 4. Uniconazole의 저온 피해 경감은 free radical scavenger의 역할을 하는 물질의 생성 및 활성증대가 중요한 역할을 한 것으로 판단되나 고온피해에 대한 경감효과의 해석에는 더 진전된 연구가 요구된다.
