This experiment was carried out to investigate the possibility of practical use of intermittent soaking system(ISS) in hydroponicaly growing tomato. There were four treatments which were NFT, Aeroponics, and ISS with 10 and 15 minute soaking intervals. 1. Plant height is highest in NFT, but main root length is longest in ISS. While stem diameter and leaf number were not significantly difference. 2. Fresh and dry weight were heavier in NFT than those of other systems at early growth stage, but the heavier in ISS system, the growth more develop. 3. In 43 days after treatment, the number of flowerlet was largest in aeroponics, and then ISS and NFT in order. Fruit weight per plant was also heaviest in aeroponics. 4. Average yield per plant were 1929.1g in aeroponics, 1475.2g in ISS with 10 minute soaking interval, 1276.2g in NFT and 1084.8g in ISS with 15 minute soaking interval. 5. Average fruit number per plant were 11.0 in aeroponics, 9.7 in NFT, 8.3 in ISS with 15 minute soaking interval, and 7.7 in ISS with 10 minute soaking interval. Average fruit weight were 192.9g in aeroponics, 172.4g in ISS with 10 minute soaking interval, 134.0g in NFT and 126.0g in ISS with 15 minute soaking interval. 6. Average fruit length was longer in aeroponics and ISS with 10 minute soaking interval than in the NFT and ISS with 15 minute soaking interval. Average fruit width was similarly wider in NFT, aeroponics and ISS with 10 minute soaking interval than in ISS with 15 minute soaking interval.
This study was conducted to clarify the effects of the different concentration of the nutrient solution on the early growth and the nutritional contents of hydroponically grown Aster koraiensis, Hemerocallis fulva and Plantago asiatica at 70 days after transplanting in perlite culture. Balanced nutrient solution formulated by Japanese Horticultural Experiment Station was used as the standard concentration of the nutrient solution. Overall plant growth of Aster koraiensis and Hemerocallis fulva such as plant height, stem diameter, number of loaves, fresh and dry shoot and root weight were significantly increased in 1.5 times concentration of nutrient solution. Shoot and root fresh and dry weight of hydroponically grown Plantago asiatica were significantly increased in the higher concentration of the nutrient solution, however, number of loaves and root length were significantly increased in the standard and the lower concentration of the nutrient solution. The highest contents of calcium, magnesium and sodium in plants were shown in Aster koraienxis which were grown in the 1.5 times concentration of nutrient solution, and Hemerocallis fulva and Plantago asiatica in 0.25 times of that. The contents of potassium in Aster koraiensis and Hemerocallis fulva were significantly increased when the plants were grown in the 0.5 times concentration of the nutrient solution, and that in Plantago asiatica in the 1.5 times concentration of the nutrient solution. The contents of phosphoric acid in plants as affected by the different species of Namul and the different concentration of the nutrient solution were not significant.
Morphological characteristic during formation of tobacco crown gall tumor and genetic tumor, and their differential response to growth regulator were investigated in in vitro culture. Crown gall tumor was induced from tumor tissue transformed by infecting Agrobacterium tumefaciens C58. Genetic tumor was induced from tumor tissue which was induced spontaneously from reciprocal interspecific hybrids between Nicotiana glauca (2n=24) and Nicotiana langsdorffii (2n=18). Morphological characteristic of crown gall tumor, genetic tumor, and teratoma shoot was very similar, and they were actively proliferated on hormone-free medium. Typical tumor callus and teratoma shoot formed from crown gall tumor on the hormone-free medium. On the contrary, tumor callus derived from genetic tumor formed as a crown gall tumor callus on the medium supplemented with 0.5 mg/L of 2,4-D, and lots of teratoma shoots without any root formed on the hormone-free medium. Root development from the teratoma shoots was hardly obtained on the medium with IAA, GA and active carbon. However, teratoma shoots with roots, as normal shoots, were initiated occasionally on the hormone-free medium. These shoots also formed new genetic tumor on the stem, which leaves formed lots of teratoma shoot on the hormone-free medium in in vitro culture.
Single-node stem pieces ca. 1 cm in length containing a axillary bud were obtained from in vitro plants of potato (Solanum tuberosum L.). The influences by a position of the node and the existence of a leaf at the node were observed in the single-node culture on the 8% sucrose MS medium. The effect of CCC was also investigated for the microtuberization. The apical part node was excellent in the tuberization not to mention shoot length, fresh weight, diameter, the number of node on the in vitro culture of a single-node than the lower part. The differences in the diameter of a tuber formed in the part of the axillary bud on all treatments including the cultivation of the apical part node were not recognized. However, the fresh weight of the tuber showed high value in the tuber formed at the axillary bud of shoot apex part. At 20 days after cultivation, tuberization was promoted in the new stolen that developed from the bud of node with a leaf under SD condition of 8 hours at $20^{\circ}C$. The tuberization from axillary bud of the single-node without leaf was inhibited at high temperature of $28^{\circ}C$ regardless of daylength. Whereas, tuberization at $20^{\circ}C$ and $28^{\circ}C$ was similar without the difference under SD condition but the tuber formation ratio were low. CCC 500 mg/L promoted tuberization and the effect was also showed even under LD condition at $28^{\circ}C$. The inhibiton of tuberization under LD and high temperature condition could be solved by treatment with CCC.
Development of modern agricultural machinery and accompanying agricultural development cause soil compaction and reduce growth by stressing roots. Kalanchoe pinnata was used to investigate the impact of stress on rooting and changes in plant growth and reproduction. K. pinnata forms somatic embryos capable of asexual reproduction at the edge of leaves. Impact of root pressurization of K. pinnata on somatic embryogenesis and organ differentiation according to external stress factors was investigated by using a high concentration of agar and this phenomenon was studied histologically. Agar concentration in culture media ranged from 0.5%-1.5% to induce a compression effect on roots. The stem and leaf of K. pinnata were subjected to a microtechnique process to study changes in tissue. In vivo, K. pinnata produced 2nd and 3rd plantlets at edges of leaves from lack of water and excessive lighting conditions. In in vitro culture studies, the lower the concentration of agar, the higher the population and the higher the biomass, but plantlet did not occur in leaf bends. Conversely, as concentration of agar increased, increase in the number of individuals was low. Plantlet development occurred only in agar 1.5% medium. The difference in agar concentration was a stressor in the root of K. pinnata, and thus the pattern of asexual reproduction changed from the division method in root to a plantlet generation in leaf. This suggests root pressurization may act as stress and change in the plant reproduction pattern.
This study was carried out to elucidate the effects of mulching materials on the growth of potato and weed control at the experimental farm of Chungbuk National University from August 9 in 2008 to June 28 in 2009. Tested potato, cv. 'Superior', was grown under the different mulching materials such as pine tree leaf, oak tree leaf, rice straw, rice hull and sawdust, and control(non-mulching). We tested two times with autumn and spring culture season. The obtained results from this study were summarized as follows; There was significantly difference in plant height, stem length, leaf length, leaf weight and tuber weight in pine tree leaf mulching treatment in autumn season among the treatments. The dominant weeds were Echinochioa crus-galli var. fadmerntacca and Cyperus amvuricus in autumn culture season, while were Echinochioa crus-galli var. fadmerntacca and Digitaria sanguinalis in spring culture season. The appearance of weeds in all of mulching treatment was significant reduced compared to control. The soil moisture content was the highest in rice hull mulching treatment, and that of mulching treatments was significantly higher than control. The soil temperature of mulching treatments was lower than that of control by from $1.0^{\circ}C$to $2.8^{\circ}C$ in autumn season and from $0.7^{\circ}C$ to $2.3^{\circ}C$ in spring season. The soil temperature was low in the order of pine tree leaf, rice straw, oak tree leaf, rice hull, sawdust, and control.
A large patch disease caused by Rhizoctonia solani AG2-2 (IV) is a serious problem in Korean lawngrass (Zoysia japonica) sites including golf courses and sports fields in Korea. Antagonistic microorganisms against R. solani AG2-2 (IV) were isolated from various forest and crop soil sources in Southern Korea. Among the 61 isolates, I-009, FRIN-001-1, and YPIN-022 strains showing dramatic inhibition of the mycelial growth of R. solani AG2-2 (IV) in the pairing culture were selected as the most potential antagonistic microorganisms for this study. Based on the 16s RNA sequence comparison, I-009 and FRIN-001-1 isolates were identified as Bacillus spp., while YPIN-022 isolate belongs to the genus Pseudomonas. The greater inhibition (clear) zone between two edges of the selected and pathogenic microbes ranged from 11 to 15 mm in three selections, but the others averaged to 7 mm out of 30 mm distance. In another antifungal test using culture filtrate, those three isolates represented a range of 51.7 to 63.5% suppression potential. The selected isolates also inhibited significantly the stem-segment colonization by R. solani AG2-2 (IV) in vivo test by 28.1%, 43.0%, and 23.7% when inoculated with I-009, FRIN-001-1, and YPIN-022, respectively. The highest antagonistic activity for the large patch disease was demonstrated by the isolate FRIN-001-1, which will be useful for developing a bio-pesticide against Rhizoctonia.
This study was conducted to select useful medium material as a replacement for the rockwool in long term bag-culture of sweet pepper. The sole use of perlite and carbonized rice hull(CRH) as well as their mixture with various combinations were compared to the rockwool. The results are summarized as the followings : 1. Plant height and number of leaves did not significantly differ among media. However, in the mixture of CRH(1) : Perlite(1), stem diameter was thicker; plant weight and root weight were heavier, and T/R ratio was lower. 2. Although fruit length and number of fruits did not significantly differ among media, the mixture of CRH(1) : Perlite(1) provided longer fruit length, more fruits, heavier fruit weight, and greater yield. 3. Monthly yield was continuously increased from the first harvest in November to the harvest in May next year. The amount of increase in the fruit yield of the mixture of CRH(1) : Perlite(1) from the first harvest to the final harvest was significantly greater than the amount of increase of other media. 4. The amount of total nitrogen and phosphate was higher in mixture of CRH (1) : Perlite(1), while the amount of other elements did not significantly differ among mixtures.

In this study, the induction regeneration of callus from immature embryo in trifoliata orange (Poncirus trifoliata RAFIN.) were accomplished. The embryogenic calli were induced from the immature embryo derived from seed when the calli were irradiated for 16hr at about 2,000 Lux in $\frac{1}{2}$ MS medium supplemented with 3% sucrose, and 44.4$\mu$M BA. Regeneration to whole plants was the most successful in MS medium containing 5.0$\mu$M BA. The yellowish callus was developed at 2 to 3 weeks of culture and the callus was changed from yellow to green at 5 to 6 weeks culture. In vitro regeneration was directly induced from embryogenic callus in MS medium containing 3% sucrose and 5.0$\mu$M BA. Multishoot was formed at 16 weeks culture. Moreover, when the root-formed plantlet was transplanted to soil, they grew to a whole plant. The compact cultured-cells were observed by light microscope after 4 weeks of cultivation and the embryogenic clumps were formed about the 5 weeks. At the same time, the neighboring cells were liquefied. In addition, differentiation of leaf and stem from the callus was observed after 12 weeks. The developed oil sacs and the profacicular cambium of the immature leaf were observed after 18 weeks. Therefore, we can see the considerable changes of cell arrangements according to the developmental stages of calli from trifoliata orange.
