Use of Single-leaf Cutting in the Study of the Expression of Starch Synthesis and Modification Genes in Sweetpotato

  • The evaluation of source potential and sink strength is the generally large and laborious sample size required to adequately assess anyone of the parameters in field-grown sweetpotato. For this purpose we used the rooted single-leaf cuttings with petioles, because the source and sink organs are restricted in this system. The rooted single-leaf cutting of sweetpotato provides a unique source-sink model system, and is established within about 50 days after planting. In this study, the sink potential of sweetpotato tubers was examined based on the expression of genes for starch synthesis (AGPase) and modification (SBEII and GBSSI) in single rooted leaf plant. The gene expression patterns of GBSSI, SBEII and AGPase at various developmental stages and in different types of root tissues presented. These results suggest that the rooted single-rooted method can be used an ideal model system to study physiological and biochemical mechanisms in sweetpotato.

The Requirement of Ruminal Degradable Protein for Non-Structural Carbohydrate-Fermenting Microbes and Its Reaction with Dilution Rate in Continuous Culture

  • A continuous culture study was conducted to determine the impact of ruminal degradable soy protein (S-RDP) level and dilution rate (D) on growth of ruminal non-structural carbohydrate-fermenting microbes. Corn starch, urea and isolated soy protein (ISP) were used to formulate three diets with S-RDP levels of 0, 35 and 70% of total dietary CP. Two Ds were 0.03 and $0.06h^{-1}$ of the fermenter volume in a single-effluent continuous culture system. As S-RDP levels increased, digestibilities of dietary dry matter (DM), organic matter (OM) and crude protein (CP) linearly (p=0.001) decreased, whereas digestion of dietary starch linearly (p=0.001) increased. Increasing D from 0.03 to $0.06h^{-1}$ resulted in decreased digestibilities of dietary DM and OM, but had no effect on digestibilities of dietary starch (p=0.77) and CP (p=0.103). Fermenter pH, the concentration of volatile fatty acids (VFA) and daily VFA production were unaffected (p=0.159-0.517) by S-RDP levels. Molar percentages of acetate, propionate and butyrate were greatly affected by S-RDP levels (p=0.016-0.091), but unaffected by D (p=0.331-0.442). With increasing S-RDP levels and D, daily bacterial counts, daily microbial N production (DMNP) and microbial efficiency (MOEFF; grams of microbial N produced per kilogram of OM truly digested) were enhanced (p=0.001). The increased microbial efficiency with increasing S-RDP levels is probably the result of peptides or amino acids that served as a stimulus for optimal protein synthesis. The quantity of ruminal degradable protein from soy proteins required for optimum protein synthesis of non-structural carbohydrate-fermenting microbes appears to be equivalent to 9.5% of dietary fermented OM.

Comparative Gene Expression Analysis of Seed Development in Waxy and Dent Corn (Zea mays L.)

  • We used Illumina/HiSeq sequencing for analysis of gene expression profiling among four maize seed types (dent, CM3 and CM6; waxy, CM5 and CM19) at 10 DAP (days after pollination). A total of 88,993,000 (CM3), 103,817,340 (CM6), 103,139,640 (CM5), and 66,978,958 (CM19) sequence reads were generated with read lengths of about 0.9, 1.0, 1.0, and 0.7 billion bp, respectively. We obtained 69.1 (CM3), 71.0 (CM6), 71.2 (CM5), and 71.8% (CM19) high quality reads from the raw data and compared them with reference RNA sequences in a public DB (NCBI). It was revealed that mapped reads were 58%, 63%, 62%, and 62% of the EST reference in CM3, CM6, CM5 and CM19, respectively; and more than 51,000 genes were expressed based on RPKM criteria (over 0.25 value) in each CM3, CM6, CM5, and CM19 inbred line. In differentially expressed gene (DEG) analysis, we found that 3,527 genes were differentially expressed by at least two-fold with 1,709 upregulated in the two waxy inbred lines and 1,818 upregulated in the two dent inbred lines. We also detected genes for the sucrose and starch biosynthesis pathways based on BINs, and different expression patterns between waxy and dent inbred lines were shown for the gene set for starch synthesis, such as sh2, bt2, du1, wx1, and ae1. Although some genes were more expressed in dent lines, most genes for starch synthesis were much expressed in waxy lines. Especially, there was greater expression of the sus2 gene in both waxy lines compared with the dent lines.

Growth and Quality Characteristics in Response to Elevated Temperature during the Growing Season of Korean Bread Wheat

  • Wheat (Triticum aestivum L.) is the major staple foods and is in increasing demand in the world. The elevated temperature due to changes in climate and environmental conditions is a major factor affecting wheat development and grain quality. The optimal temperature range for winter wheat is between 15~25℃, it is necessary to study the physiological characteristic of wheat according to the elevated temperature. This study presents the effect of elevated temperature on the yield and quality of two Korean bread wheat (Baekkang and Jokyoung) in a temperature gradient tunnel (TGT). Two bread wheat cultivars were grown in TGT at four different temperature conditions, i.e. TO control (near ambient temperature), T1 control+1℃, T2 control+2℃, T3 control+3℃. The period from sowing to heading stage has accelerated, while the growth properties including culm length, spike length and number of spike, have not changed by elevated temperature. On the contrary, the number of grains per spike and grain yield was reduced under T3 condition compared with that of control condition. In addition, the. The grain filling rate and grain maturity also accelerated by elevated temperature (T3). The elevating temperature has led to increasing protein and gluten contents, whereas causing reduction of total starch contents. These results are consistent with reduced expression of starch synthesis genes and increased gliadin synthesis or gluten metabolism genes during late grain filling period. Taken together, our results suggest that the elevated temperature (T3) leads to reduction in grain yield regulating number of grains/spike, whereas increasing the gluten content by regulating the expression of starch and gliadin-related genes or gluten metabolism process genes expression. Our results should be provide a useful physiological information for the heat stress response of wheat.

Global Transcriptome-Wide Association Studies (TWAS) Reveal a Gene Regulation Network of Eating and Cooking Quality Traits in Rice

  • Eating and cooking quality (ECQ) is one of the most complex quantitative traits in rice. The understanding of genetic regulation of transcript expression levels attributing to phenotypic variation in ECQ traits is limited. We integrated whole-genome resequencing, transcriptome, and phenotypic variation data from 84 Japonica accessions to build a transcriptome-wide association study (TWAS) based regulatory network. All ECQ traits showed a large phenotypic variation and significant phenotypic correlations among the traits. TWAS analysis identified a total of 285 transcripts significantly associated with six ECQ traits. Genome-wide mapping of ECQ-associated transcripts revealed 66,905 quantitative expression traits (eQTLs), including 21,747 local eQTLs, and 45,158 trans-eQTLs, regulating the expression of 43 genes. The starch synthesis-related genes (SSRGs), starch synthase IV-1 (SSIV-1), starch branching enzyme 1 (SBE1), granule-bound starch synthase 2 (GBSS2), and ADP-glucose pyrophosphorylase small subunit 2a (OsAGPS2a) were found to have eQTLs regulating the expression of ECQ associated transcripts. Further, in co-expression analysis, 130 genes produced at least one network with 22 master regulators. In addition, we developed CRISPR/Cas9-edited glbl mutant lines that confirmed the role of alpha-globulin (glbl) in starch synthesis to validate the co-expression analysis. This study provided novel insights into the genetic regulation of ECQ traits, and transcripts associated with these traits were discovered that could be used in further rice breeding.

  • 본(本) 연구(硏究)는 경(硬), 연질(軟質) 소맥(小麥)의 성숙과정(成熟過程)에 따른 탄수화물(炭水化物) 특성(特性)의 변화(變化)에 대하여 검토하였다. 종실(種實)의 수분함량(水分含量)이 감소(減少)함에 따라 천립중(千粒重)과 용적중(容積重)은 증가(增加)되었다. 배유(胚乳)의 전분함량(澱粉含量)은 출적후(出積後) $35{\sim}40$일까지 급격히 증가(增加)하였으며, 특히 조숙종(早熟種)인 조광(早光)은 B-type 전분(澱粉)의 합성(合成)이 억제되었다. Amylose와 amylopectin 함양(含量)은 성숙기간(成熟期間)에 따라 증가(增加)되었으며, pentosan의 함량(含量)도 계속증가되었다. Amylograph break-down은 성숙기간중(成熟期間中) 계속 감소(減少)되었으며. 배유(胚乳)의 전분함량(澱紛含量)과 고도(高度)의 부동법(負桐關)이 있었다.

Effects of the Preheating Treatments of Raw Ginseng in the Model System on the Synthesis of the Maillard Type-Browning Reaction Products of Red Ginseng

  • During our investigations on the relationship between the browning reaction of ginseng root and two compounds (arginyl-fructosyl-glucose and arginyl-fructose) in the model system of steaming and heat-drying processes for the preparation of red ginseng, the preheating treatment of main roots of raw ginseng at 60∼70$^{\circ}C$ prior to the steaming and heat-drying processes was found to bring about the gelatinization of starch granules. The enzymatic hydrolysis of gelatinized starch to maltose, a marked formation of maltose, and the increase of both free arginine and total amino acids, resulting the acceleration of the Maillard type-browning reaction of ginseng root during the steaming and heat-drying processes, and the rise of brown color intensity of red ginseng. These results show that the preheating treatment may be effective for the decrease of inside white of red ginseng.

Proteomic Analysis of Drought Stress-Responsive Proteins in Rice Endosperm Affecting Grain Quality

  • Drought stress is one of the major abiotic stresses in agriculture worldwide. We report here a proteomic approach to investigate the impact of post-fertilization drought on grain quality in rice seed endosperm (Oryza sativa cv. IR-64). Plants were stressed for 4 days at 3 days before heading. Total proteins of endosperm were extracted and separated by two-dimensional gel electrophoresis. Not many protein spots showed differential accumulation in drought-stressed samples. More than 400 protein spots were reproducibly detected, including three that were up-regulated and five down-regulated. Mass spectrometry analysis and database searching helped us to identify six spots representing different proteins. Functionally, the identified proteins were related to protein synthesis and carbohydrate metabolism, such as Granule-Bound Starch Synthase (GBSS, Wx protein), which is thought to play a very important role in starch biosynthesis and quality, a very crucial factor in determining rice grain quality.

  • 토양으로부터 역가높은 CGTase를 분비하는 호알칼리성 미생물을 분리하였으며, 동정 결과 Bacillus circulans로 판정되었다. 배양액 중의 CGTase를 ammonium sulfate 침전, DEAE-Sephadex 그리고 Sephadex G-100 column chromatography로 분리, 정제하여 단일 단백질 band를 얻었다. 정제된 CGTase의 분자량은 약 93,000, 최적 pH와 온도는 6.0, $50^{\circ}C$였으며, pH와 온도안정성은 5.5-11, $65^{\circ}C$까지였다. Soluble starch를 기질로 할 때의 $V_{max}$$K_{m}$ 값은 각각 0.16$\mu$mole $\beta$-CD/min, 14.3mg soluble starch/mi이였고 24시간 반응액의 $\alpha$-:$\beta$-:${\gamma}$-CD 의 생성비율은 1:8.1:1.9로서 $\beta$-CD를 우선적으로 합성하였다. 기질로 glucose와 maltose를 사용하였을 때 CD합성작용이 없었으며, sweet potato 그리고 cornstarch를 사용하였을 때 가장 높은 CD합성작용을 보였다. 어느 수준 이상의 과다한 CGTase 첨가경우에는 $\alpha$-CD생성이 급격히 증가하였다. 또한 정제된 CGTase는 stevioside에로의 당전이성을 갖고 있었다.

Purification and Characterization of Two Extracellular Glucoamylase Isozymes from Lipomyces kononenkoae CBS 5608 Mutant

  • Two forms of glucoamylase (GI and GII) from starch-grown Lipomyces kononenkoae CBS 5608 mutant were purified to apparent homogeneity by means of ultrafiltration, Sephacryl S-200 gel filtration and DEAE Sephadex A-50 chromatography. The apparent molecular weight was calculated as ca. 150 kDa for GI and ca. 128 kDa for GII, respectively. Both enzymes were glycoproteins with isoelectric points of 5.6 (GI) and 5.4 (GII). They had a pH optimun of 4.5 and were stable from pH 5 to 8. The temperature optimum for both enzymes was $60^{\circ}C$, but they were rapidly inactivated above $70^{\circ}C$. The $K_m$ values toward starch were estimated to be 6.57 mg per ml for GI and 4.52 mg per ml for GII, and the $V_{max}$ values were 16.28 ${\mu}M$ per mg for GI and 32.25 ${\mu}M$ per mg for GII, respectively. The $K_m$ and $V_{max}$ values of GII for ${\alpha}-$ or ${\beta}-cyclodextrin$ were estimated to be 0.15 mg per ml and 2.0 mg per ml, respectively ($K_m$) and 1.02 ${\mu}M$ per mg or 1.02 ${\mu}M$ per mg, respectively ($V_{max}$). Neither enzyme exhibited pullulanase activity but they released only glucose from starch or cyclodextrin. Amino acid analysis indicated that both glucoamylases were enriched in proline and acid amino acids. Glucoamylase GII strongly cross-reacted with a monoclonal antibody raised against GI enzymes, and the two enzymes shared very similar amino acid composition. Western blot analysis indicated that L. kononenkoae CBS 5608 mutant produced two forms of glucoamylase on starch, and that synthesis of them was subject to glucose repression.
