Characterization of Two Forms of Glucoamylase from Traditional Korean Nuruk Fungi, Aspergillus coreanus NR 15-1

  • Some characteristics of two forms of glucoamylase (glucan 1 A-$\alpha$-glucosidase, EC 3. 2. I. 3) purified from Aspergillus coreanus NR 15-1 were investigated. The enzymes were produced on a solid, uncooked wheat bran medium of A. coreanus NR 15-1 isolated from traditional Korean Nuruk. Two forms of glucoamylase, GA-I and GA-II, were purified to homogenity after 5.8-fold and 9.6-fold purification, respectively, judged by disc- and SDS-polyacrylamide gel electrophoresis. The molecular mass of GA-I and GA-II were estimated to be 62 kDa and 90 kDa by Sephadex G-1OO gel filtration, and 64 kDa and 91 kDa by SDS-polyacrylarnide gel electrophoresis, respectively. The optimum temperatures of GA-I and GA-II were 60$^circ$C and 65$^circ$C, respectively, and the optimum pH was 4.0. The activation energy (Ea value) of GA-I and GA-II was 11.66 kcal/mol and 12.09 kcal/mol, respectively, and the apparent Michaelis constants (K_{m}) of GA-I and GA-II for soluble starch were found to be 3.57 mg/ml and 6.25 mg/ml, respectively. Both enzymes were activated by 1 mM Mn^{2+} and Cu^{2+}, but were completely inhibited by 1 mM N­bromosuccinimide. The GA-II was weakly inhibited by 1 mM p-CMB, dithiothreitol, EDTA, and pyridoxal 5-phosphate, but GA-I was not inhibited by those compounds. Both enzymes had significant ability to digest raw wheat starch and raw rice starch, and hydrolysis rates of raw wheat starch by GA-I and GA-II were 7.8- and 7.3-fold higher than with soluble starch, respectively.

  • Evaluation of drug-excipient compatibility is one of the basic steps in the preformulation of pharmaceutical dosage forms. Some reactive excipients have been known so far which may cause stability problems for drug molecules in pharmaceutical dosage forms. The aim of this study was to evaluate drugexcipient compatibility of gliclazide with some common pharmaceutical excipients, known for their ability to incorporate in drug-excipient interactions. Binary mixtures were prepared using lactose, magnesium stearate, polyvinylpyrrolidone, sodium starch glycolate, polyethylene glycol 2000 and dicalcium phosphate. Based on the results; gliclazide was incompatible with all tested excipients; but not with dicalcium phosphate. DSC (Differential Scanning Calorimetry) results were in accordance with HPLC (High Pressure liquid chromatography) data and were more predictive than FTIR (Fourier Transform Infrared Spectroscopy). Drug and reactive excipients incompatibility was fully discussed and documented. It is advisable to avoid incompatible excipients or carefully monitor the drug stability when incorporating such excipients in final formulation designs.

Isozyme Variability in Three Species of Freshwater Planorbid Snails in Korea : Gyraulus convexiusculus , Hippeutis cantori and Segmentina hemisphaerula (한국산 또아리물달팽이과 ( family Planorbidae ) 3종에서의 동위 효소 변이)

  • A horizontal starch gal electrophoresis for enzyme proteins extracted from 3 species of Korean planorbid snails; Gyraulus convexiusculus, Hippeutis cantori and Segmentina hemisphaerula was carried out in order to elucidate their genetic relationships.The results from 12 enzymes employed in three different kinds of buffer systems are summarized as follows:1) Two loci from each enzyme of aldehyde oxidase, esterase, glucose phosphate isomerase. isocitrate dehydrogenase, leucine aminopeptidase, malate dehyogenase, peptidase and xanthine oxidase were detected, and only one locus was observed from each of the following four enzymes: 6-phosphogluconate dehydrogenase, glyceraldehyde-phosphate dehydrogenase, glutamate oxaloacetate transaminase and glycerol-3-phosphate dehydrogenase.2) Most of loci in 3 species of planorbid snails employed showed homozygous and monomorphic banding patterns and some of them were specifis as genetic markers among different species. However, a few of loci (EST-1. EST-2 and GPI-2)showed polymorphic banding patterns. 3)Hippeutis cantori and Segmentina hemisphaerula were more closely clustered in a dendrogram with the genetic iddentity value of 0.431, and these two species were lineated with Gyraulus convexiusculus as another cluster at the value of 0.294.In summarizing the above results, three species of Korean planorbid snails employed in this study mostly showed monomorphic enzyme protein banding patterns and genetic differences specific among 3 species.

  • Limitation of photosynthesis in detached Chinese cabbage (Brassica campestris L.) leaves was induced by feeding of mannose (25 mM) for 12 h in the light, and changes in the basic thylakoid functions under this condition were investigated. The acid soluble phosphate contend and CO2 uptake rate was decreased by 66% and 67%, respectively. However, the starch content was increased by 24% compared to those of controls. From the fast induction curves of chlorophyll fluorescence, dark level fluorescence (Fo) slightly increased while intermediate plateau fluorescence level (FI) to peak level fluorescence (Fp) transient was significantly decreased with a slight decrease in the Fo-to-FI transient. This data means that reduction of secondary electron acceptor of PSII (QB) might be more severely inhibited than that of primary electron acceptor of PSII (QA) by decrease in phosphate level. The strong decline of (Fv)m//Fm ratio suggests that efficiency of excitation energy capture by PSII was decreased markedly. The quenching of Fo (qO), an indicator of state transition, was also occurred over the slow induction kinetics of chlorophyll fluorescence. From quenching analysis, fluorescence was dominantly quenched by nonphotochemical quenchings (qE+qT). These results showed that the capture and transfer efficiency of excitation energy to PSII reaction center in thylakoid was decreased with the decline of leaf phosphate level, and that the state transition was occurred during the induction of photosynthesis under these conditions.

  • The effect of a polyphosphate having $P_2O_5$ content of 67% on the firming rate of nonwaxy (Akibare and Milyang 30) and waxy cooked rice stored at room temperature was investigated. The phosphate retarded the firming rate of Akibare and Milyang 30 by 14.0 and 27:00, respectively. The phosphate reduced the starch components available for crystallization of cooked nonwaxy rice. The phosphate exerted no effect on the firming rate of cooked waxy rice.

Studies on the Production of Foods and Feeds Yeast from the Hydrolyzate of Corn Starch Cake (옥수수 전분박(澱粉粕)을 이용(利用)한 식사료(食飼料) 효모생산(酵母生産)에 관한 연구(硏究))

  • To meet the need of protein feed and fine more efficient ways of returning waste to resources, we have carried out the study of the production of yeast for foods and feeds from the corn starch cake. The present study includes the method for acid-hydrolysis, the selection of yeast capable of utilizing hydrolyzate of the corn starch cake, and culture condition of Candida tropicalis under the liquid culture and the semisolid culture. Obtained results were as follows. 1. Hydrochloric acid was more excellent on the hydrolysis of the corn starch cake than sulfuric acid, and the yield of sugar was maximum, 57.2%, when the corn starch cake was hydrolyzed with 1.0% of hydrochloric acid at 2.0kg/cm for 30 minutes. 2. As the acid solution content was increased, more sugar was liberatedfrom the mixture, until the acid solution-substrate ratio reached 10:1. Beyond this point, no further increase was observed. To prepare the cultural medium of semisolid fermentation, a acid solution to substrate ratio of 3:1 appeared to be optimum. 3. Out of 6 yeast strains, Candida tropicalis had excellent growth on the hydrolyzate of the corn starch cake, and optimum temperature and initial pH were $30^{\circ}C$ and 6.0 respectively. 4. Optimum liquid medium of Candida tropicalis is ures 0.3%, potassium phosphate monobasic 0.15g and magnesium sulfate 0.04g in 100ml of the hydrolyzate of the corn starch cake, while optimum semisolid medium is ammonium chloride 0.4g, potassium phosphate monobasic 0.1%, magnesium sulfate 0.04%. 5. Candida tropicalis could assimilate the sugar in the hydrolyzate up to more than 88.75%, and a yield of dry yeast reached 19.13% to the corn starch cake under the liquid culture. 6. Compared to the that of the untreated corn starch cake, the cellulose content of the semisolid fermented cake decreased by 3.76% to 14.7%, whereas dry yeast contents increased by 13.89%.

Tribological Characteristics of Phosphorated Starch Based Electrorheological Fluid (인산화 전분 ER 유체의 트라이볼로지 특성)

  • This experimental study presents tribological characteristics under boundary lubrication contacts associated with electrorheolocal (ER) fluid. ER fluid is prepared by using phosphorated starch particles and silicone oil. Experimental apparatus of tribological tester is designed and constructed to evaluate tribological characteristics of pin specimens. Wear tests under boundary lubrication of ER fluid are experimentally performed under consideration of several operational factors such as normal load, sliding distance, sliding speed and specimen materials: steel, copper and aluminum. After wear test, microscopic surface changes of the worn pin specimens are analyzed in order to investigate measured wear characteristics by using the scanning electron microscope (SEM) as well as surface profilometer. In addition, the chemical wear characteristics are investigated by using energy dispersive x-ray spectroscopy (EDS). Moreover, friction coefficient measurements under different materials of pin specimens are conducted for the tribological investigations. In order to verify the effect of starch phosphate particles in ER fluid, the wear test results with ER fluid are compared with test results with only silicone oil. The results clearly present that the phosphorated starch based ER fluid shows the stabilized wear as well as friction characteristics after run-in operations, but the wear rate under ER fluid is increased.

  • Cultivation condition of yeast on the utilization of fermentable substrate from the cellulosic wastes such as rice hull, rice straw and corn starch cake was investigated. The results obtained were summarized as follows;1. Corn starch cake was respectively added to rice hull and rice straw in order to increase sugar concentration in the hydrolyzate, and then hydrolyzed. As the result, concentration of sugar in hydrolyzed solution of rice hull was 9.12%, in that of rice straw was 7.98%. 2. It was found that calcium carbonate as a neutralizer was the most effective to prepare the culture broth of yeast. 3. An optimal growth of Hansenula subpelliculosa GFY-2 was observed in the medium prepared by adding 0.3% of ammonium sulfate, 0.4% of potassium phosphate dibasic, 0.02% of magnesium sulfate, sodium chloride and calcium chloride to hydrolyaed sugar solution, respectively. 4. Hansenula subpellicuiosa GFY-2 cultured in the substrate solution which of rice hull and rice straw added to corn starch cake was assimilated more than 90% of sugar in the hydrolyzate within 48 hours. The yeast cells yielded in rice hull was 46.5%, and that of rice straw 45.4% to utilized sugars.

Medium Optimization for the Protease Production by Bacillus licheniformis Isolated from Cheongkookjang (청국장에서 분리된 Bacillus licheniformis의 Protease 생산을 위한 배지 최적화)

  • Bacillus licheniformis fermenting soybean product with highest score in consumer acceptance had been isolated from homemade Cheongkookjang. In order to develop the medium composition, effects of ingredients including nitrogen sources, carbon sources, metal ions and phosphate were examined for protease production of the isolate. Potato starch increased the protease productivity, while glucose repressed it. Yeast extract was the most effective nitrogen source for enzyme production. The calcium was found to increase protease activity slightly while cell growth and enzyme production was completely inhibited by divalent ions such as $Zn^{2+}$, $Cu^{2+}$ and $Co^{2+}$. The maximum protease productivity was reached approximately 800 unit/mL in the optimized medium consisting of potato starch (1.5%), yeast extract (1.5%), $CaCl_2$(0.7%), $K_2HPO_4$(0.03%) and $KH_2PO_4$(0.03%). The protease activity of culture filtrate was gradually decreased after incubation for 28 h.

Optimal Culture Condition and Extracellular Enzyme Activity of Metarhizium anisopliae (녹강균(Metarhizium anisopliae)의 최적 배양조건 및 효소활성)

  • In order to use Metarhizizmn~ anisopliae as a biological pesticide, effect of envrionmental factors on nlycelial growth, spore formation, and extracellular enzyme activity in culture broth of M. anisopliae DGUM 35001 was determined. Optimal temperature was 26^{\circ}C.$ and optimal pH ranged from 5 to 9. Among the complex media tested, MCM and SDPY media were the most favorable for mycelial growth. When Czapek-Dox agar was used as a mnimal medium, glucose and sucrose among the saccharides were very excellent source of carbohydrate. Among the biopolyners tested. chitin was the most favorable source for mycelial growth and produced high aerial inycelia. Urea and ammonium phosphate as an inorganic nitrogen source and bacto-peptone and soytone as an organic nitrogen source enhanced the mycelial growth When serine as a source of amino acid was supplemented, excellent mycelial growth was shown. Large amount of spores could be obtained from the aerial mycelia of starch medium. When the culture broth was filtrated and then the concentrate with ammonium sulfate was used as a crnde enzyme solution, high enzyme activities of amylase and protease were shown. However, lipase and chitinase activities were comparatively low.
