  • An adult female ringed seal died suddenly and was subsequently examined for diagnostic purposes. The animal's lungs demonstrated mild non-collapsibility and multifocal white to yellow patches. Histopathological examination revealed multifocal pulmonary histiocytosis. Alveoli were filled with numerous foamy macrophages cytoplasm and scattered multinucleated giant cells containing cholesterol clefts. The foamy cytoplasm of the macrophages stained with oil red O stain. Further, lipid droplets within the cytoplasm were detected by electron microscopy. To the author's knowledge, this is the first case report describing the histochemical staining and electron microscopic findings associated with endogenous lipid pneumonia in ringed seal.

  • Although culture is the gold standard method to identify mycobacteria, its use in tuberculous lymphadenitis (TBL) is limited due to formalin fixation of the submitted specimens. We evaluated the performance of quantitative real-time PCR (q-PCR) for Mycobacterium Tuberculosis (MTB) in granulomatous lymphadenitis using formalin-fixed paraffin-embedded (FFPE) tissues. From 2000 to 2010, a total number of 117 cases of lymph node samples with granulomatous inflammation which were surgically removed and fixed in formalin were studied. Hematoxylin & Eosin (H&E) and Ziehl-Neelsen-stained (ZN) slides were reviewed. qPCR using Real TB-Taq$^{(R)}$ was performed for all cases to identify Mycobacterium tuberculosis. Thirteen non-tuberculous lymphadenopathy cases were used as negative control. Cervical lymph nodes were more frequently affected (60%, 70/117) than other sites. ZN stain for acid fast bacilli was positive in 19 (16.24%) cases. qPCR for tuberculosis was positive in 92 (78.63%) cases. Caseous necrosis was found in 103 (88.03%) cases. While the ZN stain and qPCR were both negative in all control cases, the qPCR showed a significantly higher positive rate (78.63% vs. 16.24%) compared to ZN stain in histologically diagnosed TBL. Quantitative real-time PCR proves to be more sensitive than ZN stain for diagnosis of tuberculous lymphadenitis.

  • The effect of oven firing on the color stability and surface texture of extrinsic stains used characterization and color modification of metal cermic restoration were studied by comparing different temperatures and techniques. They were autoglaze technique, applied glaze technique, two step low-fusing glaze technique and one step low-fusing glaze technique. Autoglaze technique and applied glaze technique were air-fired to manufacturer's recommended temperature. Two step low-fusing glaze technique is separated staining and overglazing. Thin slurry of stain powder were air-fired to $1500^{\circ}F$, Overglaze powder powder was then mixed with seam medium and air-fired to $170^{\circ}F$. One step low-fusing glaze technique is combined staining and overglazing. Thin slurry of stain and glaze powder were air-fired to $170^{\circ}F$. The obtained results were as fellows. 1. Slightly significant color differences by colorimeter were found between different stain application and fusion techniques(P<0.05). 2. Two step low-fusing glaze method showed the most rough surface, especially ceramco orage stain(P<0.05). 3. Surface roughness increased gradually in order of autoglaze, applied glaze, one step low-fusing glaze but they were not significantly different(P<0.05). 4. When two step low-fusing glaze was applied, both color measurement and surface texture were sinificantly different from other groups(P<0.05).

  • The purpose of the current study was to investigate histologic changes in the alveolar bone of the lower molar region subsequent to the loss of their opposite molars, and to characterize chemical alterations by utilization of histochemical procedures. Twenty five rats(Sprague Dawley), approximately 150-200gm body weight, were used in this experiment. In the treated animals, upper molars were removed. The animals were decapitated by groups at the following intervals after teeth removals: 10th, 20th, 50th, 70th and 100th day. The normal, untreated rats were used as controls. The molar region of lower jaw, including the intact alvelar bone and teeth was dissected and specimens were decalcified in 3% formic acid. After the tissues were fully decalcified, the specimens were embedded in celloidin and sectioned in mesiodistal plane. These sections were stained in the following staining methods. Mallory azan stain and hematoxylin-eosin stain were utilized for structural evaluation. Polysaccharides were demonstrated by means of the PAS reaction. Acidmucopolysaccharides were studied by means of the colloidal iron stain. Alloxan-Schiff reaction was used for protein. The results were as follows: 1) In the control animals, bone resorption was noted in the distal alveolar bone proper and bone apposition was shown in the mesial alveolar bone proper. But in the treated animals, bone apposition was observed on the mesial and distal walls of the alveolus and osteoclastic activity was not noted in any walls. 2) Bone apposition was most prominent from the 10th to 20th day after treatment. 3) Appositional growth of cementum along the surface of root was prominent from the 50th to 70th day after treatment. 4) In the area where osteoblastic activity was apparent, osteoblasts were stained strongly in the PAS and alloxan-Schiff reaction. A plastic resorption line showed strong alloxan-Schiff reaction. 5) In the colloidal iron stain, the alveolar wall adjacent to the cementum apposition area was stained more strongly than the other areas.

  • The purpose of this study was to evaluate the effect of orthodontic force on periodontal cellular activity by immunoperoxidase stain of epidermal growth factor, one of the tissue hormone. And supplementarily, to investigate of the changes of periodontal structures, periodontium was stained by H-E, Masson's Trichrome, P. A. S. stain after orthodontic force application. The experimental animals were four young adult dogs of average 8 month old. The fixed orthodontic appliance was cemented on mandibular right 4th premolar and 1st molar of each animal as experimental site. Mandibular left 4th premolar area of the same animal was used as control. The appliance consist of two silver crown soldered with 0.030' tube, $0.018\times0.022'$ S.S. sectional arch wire, and 0.009' open coil spring for manifestating of orthodontic force for bodily tooth movement of mandibular 4th premolar toward mesial direction. Experimental group was sacrificed at 1, 2, 3, 5 weeks from beginning of the experiment, and was investigated immunohistochemically and bistochemically by several staining methods. Findings were as follows: 1. The degree of EGF staining in control group was highest in epithelium of periodontium, and osteoclasts, osteoblasts and fibroblasts around the capillary were stained at higher level in periodontium. Generally, control group shows positive distribution of EGF all around the periodontal area. 2. The degree of EGF staining in control and 5 week group were similar, and did not show the significant different level between tension and pressure side. 3. All of 1, 2, 3 week group showed the same staining degree and distribution of EGF, and the tension side was more positive reaction of EGF stain than the pressure side. 4. The features of collagen fiber and periodontal fiber arrangement observed by H-E, Masson's Trichrome and P. A. S. stain revealed that oblique periodontal fibers were strectched in tension side, compressed in pressure side of all experimental group. Some fiber group in pressure side of 5 week group recovered the regular arrangement along the capillaries.

  • Objective : To assess the effects of progesterone and acetyl-L-carnitine used after treated with Isolate gradient before semen cryopreservation-thawing on sperm parameters and membrane integrity. Material and Methods : From April 2001 to July 2001, ten normal male partner of couples who were visited in vitro fertilization (IVF) clinics. the semens were treated with $Isolate^{(R)}$ gradient before cryopreservation, spermatozoa was incubated with progesterone (1, 5 and $10{\mu}M$), acetyl-L-carnitine (2.5, 5 and $10{\mu}M$), or both (progesterone, $1{\mu}M$; and acetyl-L-carnitine, $5{\mu}M$) for 30 min. Results: There were no differences in sperm parameters and vital stain among isolate only treated group, progesterone (1, 5 and $10{\mu}M$), acetyl-L-carnitine (2.5, 5 and $10{\mu}M$) and both (progesterone, $1{\mu}M$; and acetyl-L-carnitine, $5{\mu}M$). But, in high concentration of acetyl-L-carnitine ($10{\mu}M$) treated group, sperm parameters and vital stain were decreased. The statistical method was used ANOVA (Kruskal-Wallis test) and p value was <0.01. Conclusions : Neither progesterone nor acetyl-L-carnitine show to be protective effect on the cryodamage assessed by sperm parameters and vital stain (eosin-Y stain) in normal sperm. High concentration of acetyl-L-carnitine ($10{\mu}M$), however, was harmful effect on cryoprevention.

  • We examined the effects of Whadamcheongwha-tang(WDCWT) extract on the acetic-acid induced antigastric ulcer in rats. These experiments investigated the numerical changes of gastrin and histamine secreting cells of the gastric mucosa by immunohistochemical method, and the changes of mucin of gastric mucosa by PAS-AB stain methods after the oral administration of WDCWT extract(1.0ml/day) and omeprazole(0.2mg/day) for 1, 3 and 6 weeks. The result are as follows; 1. When WDCWT extract was administrated for 1, 3, 6 weeks, in result, gastrin secreting cells in gastric mucosa were increased compared to the control group. 2. When WDCWT extract was administrated for 1, 3, 6 weeks, in result, the density of immunoreactive gastrin cells was increased compare to the control group. 3. When WDCWT extract was administrated for 1, 3, 6 weeks, in results, the changs of mucosal thickness stained by PAS/PAS-AB was increased compared to the control group. 4. When WDCWT extract was administrated for 1, 3, 6 weeks, in results, the density of PAS stain was decreased compare to control group, but density of AB stain was increased compare to control group. The results suggest that WDCWT extract inhibits a gastric acid secretion in rat gastric mucosa, and is useful in the treatment of the hyperacidity and gastric ulcer.
